Claims (12)

  1. 프로인슐린 전구물질 유사체를 트립신 또는 트립신-형 엔도펩티다제 존재하의 인슐린 등전점 미만의 pH에서 천연 아미노산의 에스테르 또는 천연 아미노산 에스테르의 유도체와 반응시킨 다음, 에스테르기와 경우에 따라서 존재할 수도 있는 다른 보호기물을 제거시킴을 특징으로 하는 프로인슐린 전구물질 유사체로부터 인슐린을 제조하는 방법.
  2. 제 1 항에 있어서, 아미노산 유도체가 ala-OtBu 또는 thr(tBu)OtBu인 방법.
  3. 제 1 항에 있어서, pH 3.8 내지 5.5에서 수행하는 방법.
  4. 제 1 항에 있어서, pH3.8 내지 5.0에서 수행하는 방법.
  5. 제 1 항에 있어서, 효소/기질 비율이 1 : 1 내지 1 : 100 중량비인 방법.
  6. 제 1 항에 있어서, 효소/기질 비율이 1 : 4 내지 1 : 10 중량비인 방법.
  7. 제 1 항에 있어서, 50 내지 500배몰 과잉의 아미노산 에스테르 또는 그 유도체의 존재하에 수행하는 방법.
  8. 제 1 항에 있어서, 100 내지 250배몰 과잉의 아이노산 에스테르 또는 그 유도체의 존재하에 수행하는방법.
  9. 제 1 항에 있어서, 반응온도가 +2내지 +37℃인 방법.
  10. 제 1 항에 있어서, 반응온도가 +4내지 +10℃인 방법.
  11. 제 1 항에 있어서, pH 3.8 내지 5.5, 효소/기질의 비율 1 : 1 내지 1 : 100 중량비, 반응온도 +2 내지 +37℃ 및 50 내지 500배몰 과잉의 아미노산 에스테르 또는 그의 유도체를 사용하여 반응을 수행하는 방법.
  12. 제 1 항에 있어서, pH3.8 내지 5.0, 효소/기질의 비율 1 : 4 내지 1 : 10 중량비: 반응온도 +4 내지 +10℃ 및 100 내지 250배몰 과잉의 아미노산 에스테르 또는 그의 유도체의 존재하에 반응을 수행하는 방법.
