Abstract
NEW MATERIAL:The insulin derivative of formulaI(R1 is H or hydroxy-protecting group; R2 is carboxy-protecting group; R3 is guanidino-protecting group).
EXAMPLE: Pig-originated[B22-Arg(1,2-dihydroxycyclohexylene), B30-Thr-t-butyl ester]-insulin.
USE: Intermdiate for the synthesis of remedy for diabetes. It can be converted to human insulin by removing the protecting groups. Useful for the synthesis of human insulin from pig insulin.
PROCESS: The compound of formulaIis prepared by reacting the compound of formula II with the L-threonine derivative of formula III in the presence of trypsin or an enzyme having similar activity to trypsin. The compound of formula II can be obtained e.g. by introducing a protecting group to the guanidino group of Arg at 22-position of the B-chain of pig insulin, and severing the 30- position amino acid with an enzyme.
