Abstract
Description
도 2는 다른 종의 HSTF (heat shock transcription factor)와 본 발명에 의한 CaHSTF1 cDNA로부터 유추된 아미노산 서열을 비교한 도면이다. Capsicum annuum (CaHSTF1), Arabidopsis thaliana (AT-HSFB1, AT-HSFB2A, AT-HSFB2B, AT-HSFB3, HSFB4)의 서열을 나타낸다.
도 3은 아미노산 서열의 유사성에 기초하여 본 발명에 의한 CaHSTF1과 다른 HSTF와의 예측되는 진화적 관계를 나타내는 계통수이다. 수평선의 길이는 연관된 서열 사이의 비유사성을 나타내고, 선 아래의 숫자는 근접한 노드로부터의 상대적 거리를 나타낸다.
도 4a 및 도 4b는 본 발명에 의한 CaHSTF1 프로모터 부위의 식물 조절 DNA 요소를 나타내는 도면과, 본 발명에 의한 CaHSTF1의 프로모터의 염기서열(서열번호 3)을 나타내는 도면이다.
도 5a 및 5b는 TMV-P0 접종에 따른 CaWRKYd 침묵 식물에서의 본 발명에 의한 CaHSTF1 유전자 발현 양상을 보인 도면이다.
도 6a ~ 6d는 24℃에서 TMV-P0의 접종에 의한 본 발명에 의한 CaHSTF1, CaPR-1 및 CaHSP18의 발현을 RT-PCR 및 qRT-PCR로 분석한 결과를 나타낸 도면이다. 총 RNA는 접종 후 0, 24, 48, 72 시간에 잎으로부터 추출되었다 (HAI). 대조군으로 이용된 잎은 버퍼와 카보런덤만으로 모조-접종 (mock-inoculation) 되었다. 수치는 CaActin 유전자의 발현으로 표준화된 mRNA 수준을 나타낸다.
도 7a 및 7b는 본 발명에 의한 CaHSTF1 유전자가 칠레 고추 (Capsicum annuum) 식물에서 침묵한 것을 나타낸 사진과 CaHSTF1 유전자가 침묵한 식물에서 HR lesion이 대조군에 비해 감소한 것을 나타낸 그래프이다. 2주 된 칠레 고추 유묘에 아그로인필트레이션 (agroinfiltration)에 의해 pTRV1 및 pTRV2-CaHSTF1 벡터가 접종되었다. 4주 후, 상부 잎들에 TMV-P0가 접종되었고, 사진은 24℃에서 TMV-P0가 접종된 후 3일 후 pTRV2 및 pTRV2-CaHSTF1의 잎을 나타낸다.
도 8a ~ 8f는 24℃에서 TMV-P0 접종에 따른 CaHSTF1 침묵 식물에서의 고추 병 관련 (Capsicum annuum pathogenesis - related, CaPR) 유전자의 발현 양상을 나타낸다. qRT-PCR을 이용하여 pTRV2 및 pTRV2-CaHSTF1 침묵 식물 잎에서의 CaPR 유전자 발현을 분석한 결과를 나타낸다. 총 RNA는 TMV-P0 접종 후 24, 48, 및 72 시간에 잎으로부터 추출되었다. 수치는 CaActin 유전자의 발현으로 표준화된 mRNA 수준을 나타낸다.
도 9a ~ 도 9g는 고온 처리에 의한 Capsicum annuum CaHSTF1 , 고추 병 관련 (Capsicum annuum pathogenesis - related, CaPR) 유전자 및 고온 관련(Capsicum annuum heat shock protein18 , CaHSP18) 유전자의 발현양상을 qRT-PCR로 분석한 결과를 나타낸다.

도 13a 및 13b는 방어반응에 관련된 신호전달 분자에 의해 Capsicum annuum CaHSTF1이 유도되어 발현되는 것을 qRT-PCR로 분석한 결과를 나타낸다.
도 13a는 살리실산 (SA)으로 처리된 칠레 고추 식물 (Capsicum annuum cv. Bukang)에서 CaHSTF1의 전사가 확인된 결과를 나타낸다. 고추 잎에 10mM SA가 분무되거나 모조(mock)로 10% 에탄올이 처리되었다.
도 13b는 에테폰 (ethephon)으로 처리된 칠레 고추 식물 (Capsicum annuum cv. Bukang)에서 CaHSTF1의 전사가 확인된 결과를 나타낸다. 고추 잎에 10mM 에테폰이 분무되거나 모조(mock)로 10% 에탄올이 처리되었다.
도 14a 및 14b는 GFP :: CaHSTF1 융합 유전자의 개략적 구조 및 GFP :: CaHSTF1 융합 단백질의 세포 내 위치를 나타낸다.
도 14a는 GFP 대조군의 세포 내 위치를 나타낸다.
도 14b는 GFP::CaHSTF1 융합 단백질의 세포 내 위치를 나타낸다. 상기 GFP::CaHSTF1 융합 구조체는 PEG-매개 형질전환에 의해 고추 원형질체로 도입되었다. 도입된 유전자의 발현은 형광 또는 광학 현미경관찰로 24시간 후에 확인되었다.

Claims (9)
- 서열번호 2로 표시되는 아미노산 서열을 갖는 단백질을 코딩하는 분리된 유전자(CaHSTF1 ).
- 제1항에 있어서, 상기 유전자는 서열번호 1로 표시되는 염기서열을 갖는 것을 특징으로 하는 유전자.
- 서열번호 2로 표시되는 아미노산 서열을 갖는 분리된 단백질.
- 제1항 기재의 유전자를 포함하는 재조합 벡터.
- 제1항 기재의 유전자 또는 제4항 기재의 재조합 벡터에 의해 형질전환된 식물세포.
- 제1항 기재의 유전자 또는 제4항 기재의 재조합 벡터에 의해 형질전환된 식물체.
- 서열번호 3으로 표시되는 염기서열을 갖는, 제1항 기재의 유전자 CaHSTF1의 프로모터.
- 제7항의 프로모터를 포함하는 재조합 벡터.
- 서열번호 1로 표시되는 염기서열을 갖는, 식물의 방어관련 식물특이 전사인자 유전자 CaHSTF1를 TRV 벡터에 재조합하여 구축된 VIGS (virus induced gene silencing)용 벡터 pTRV2-CaHSTF1.
