본 발명은 세포 항원을 코딩하는 뉴클레오티드 서열을 갖는 미생물에 관한 것으로, 상기 미생물의 게놈 내에(서)는 하기의 성분들이 삽입되고 발현될 수 있다: I) 종양 세포의 항원의 적어도 하나의 에피토프(epitope)를 코딩하는 뉴클레오티드 서열 및/또는 종양이 발생되는 조직 세포에 대해 특정적인 항원의 적어도 하나의 에피토프를 위한 뉴클레오티드 서열; 선택적으로, II) 면역 체계의 세포를 자극하는 단백질을 코딩하는 뉴클레오티드 서열; IIIA) 성분 I) 및 선택적으로는 박테리아의 외부면에 있는 성분 II)의 발현 생성물의 발현을 가능케 하는 및/또는 성분 I) 및 선택적으로는 성분 II)의 발현 생성물의 분비를 가능케 하는 전달 시스템을 위한 뉴클레오티드 서열; 및/또는 IIIB) 포유 동물 세포의 시토졸 내에 미생물을 용해하기 위해 그리고 용해된 미생물 내에 포함된 플라스미드를 세포내 방출하기 위해 사용되는 단백질을 위한 뉴클레오티드 서열; 그리고 IV) 미생물 내에서 활성화될 수 있고, 조직-세포-특정적이지만 세포-특정적이지 않은 활성화 서열로 구성된 그룹 중에서 선택되는, 성분 I) 내지 IIIB)의 하나 또는 다수의 성분을 발현시키기 위한 활성화 서열. 각각의 성분 I) 내지 IV)는 개별적으로 또는 다중으로, 각각 동일하게 또는 상이하게 배열될 수 있다. 본 발명은 또한 약제를 제조하기 위해 사용되는 상기와 같은 미생물의 용도에 관한 것이다.
Description
악성 종양 질병이 대부분 치명적으로 진행하는 주원인은, 악성 암세포를 인식하여 파괴하는 신체 자생적 면역 체계의 무능력이다. 공업 국가에서 암 질병은 치명적인 결과를 낳는 가장 빈번하게 발생하는 질병에 속한다. 따라서, 독일에서만 매년 210,000명 이상의 사람이 신종 악성 종양의 발생으로 사망하며(출처: WHO, 1997년의 숫자), 이와 같은 숫자는 인구 100,000명 당 255명 이상이 매년 사망하는 비율이다.
본 발명의 토대는, 신종 악성 종양을 발생시키는 분자 메커니즘에 대한 인식을 보다 새롭게 하는 것이다. 이미 암 형성에 대한 초기 연구에서, 세포 성장 및/또는 세포 분화의 통제 측면에서 특징적인 변화가 이루어졌다(폰텐, Cancer Surv 32: 5-35., 1998). 상기 변화에는, 최근에 확인되었고 또한 모두 종양 항원인 신호 형질 도입(signal transduction) 및 세포 사이클 통제용 단백질이 중요하게 관여했다.
종양 항원은 대충 3가지 계급으로 분류된다(파르돌, Nat Med 4: 525-531., 1998): i) 예를 들어 EGF-R, HER-2와 같이 변성된 및/또는 과발현된 형태로 종양 세포 내에 존재하는 종양-특정적인 신항원(neo-antigen), ii) MAGE 단백질 군의 대표 단백질 또는 CEA와 같은 종양-특정적인 태아 항원(embryonal antigen), iii) 티로시나아제, Mart-1/멜란-A 및 gp100과 같은 종양 조직-특정적인 분화 항원(differetiation antigen).
종양 백신 물질의 효과를 위해서는 CD8+ T-세포의 효과적인 유도가 결정적인 의미를 갖는데, 그 이유는 종양 세포가 대부분 MHC 계급 II 분자를 전혀 투여하지 않고, 세포 내에 존재하는 종양 항원이 대부분 MHC 계급 I을 제한하기 때문이다. 종양 환자의 경우에는, 당연히 존재하는 CD8+의 모집단, T-세포(CTL)가 종양 세포를 인식하여 제거하기에는 명확하게 충분치 않다(자페, Ann. N.Y. Acad. Sci. 886: 67-72, 1999). 더 나아가 종양 특정적인 T-세포는 종종 상이한 메커니즘(면역성 결여(anergy), 내성(tolerance), 중화(neutalization))에 의해서 종양 조직을 효과적으로 공격할 수 없다(스미스 등, Nat Immunol 2: 293-299., 2001). 그렇기 때문에, 효과적인 종양 백신 물질은 상기 면역성 결여 또는 내성을 끊어야만 하고, 충분한 개수의 활성화된 특정 CTL 그리고 또한 특정 항체를 유도해야만 한다. 특정 항체의 역할은, 이미 통상적으로 이용 가능한 헤르셉틴과 같이 그룹 (a)의 종양 항원에 대항하는 모노클로날 항체(mAb), 즉 HER-2에 대항하는 mAb를 효과적으로 사용할 때 나타난다(콜로머 등, Cancer Invest 19: 49-56., 2001).
무엇보다도 언급된 Th1 면역 반응에 의해 대항될 수 있는 특정 세균성 전염병에 대항하는 백신 물질 캐리어로서 독소가 약화된 세포내 박테리아가 적합하다는 내용은 이미 공지되어 있다(헤쓰 및 카우프만, FEMS Immunology & Medical Microbiology 23: 165-173, 1999). 상기 반응은 CTL 그리고 IFN-g를 분비하는 현존 특정적 CD4+ T-세포(또한 T-보조 세포, Th)를 특징으로 한다(압바스 등, Nature 383: 787-793, 1996). 다른 그룹들은, 재조합된 박테리아가 이종성(heterologous) 종양을 막을 수 있다는 사실을 보여주었다(메디나 등, Eur J Immunol 29: 693-699., 1999; 판 등, Cancer Res 59: 5264-5269., 1999; 우드락 등, J Immunother 22: 251-259., 1999; 파글리아 등, Blood 92: 3172-3176., 1998; 파글리아 등, Eur J Immunol 27: 1570-1575., 1997; 판 등, Nat Med 1: 471-477., 1995; 판 등, Cancer Res 55: 4776-4779., 1995). 상기 경우에는 물론 동물들은 대체-항원에 대해 면역성을 나타냈고, 그 다음에는 상기 항원을 발현시키는 종양 세포가 적응되었다.
그러나 상기 종양 체계는 임상적인(clinic) 종양과 비교될 수 없는데, 그 이유는 상기 모델에서는 종양 항원에 대한 내성이 형성되었기 때문이다.
다수의 상이한 종양 백신들은 이미 임상적으로 실험되었다. 그러나 지금까지는 어떤 백신도 그리고 어떤 백신화 방법도 종양 질병 치료 분야에서 돌파구를 얻어내지 못했다. 이와 같은 배경에서는 또한 새로운 종양 치료 방법에 대한 필요성도 매우 크다.
박테리아 내에 삽입된 핵산 서열의 발현 생성물을 상기 박테리아의 세포막 상에서 발현시키거나 혹은 상기 박테리아로부터 분비시키는 것은 공지되어 있다. 이와 같은 기술의 토대는 그람 음성 박테리아의 타입 I 분비 체계의 원형인 에셰리키아 콜리 헤몰리신 체계 HlyAs이다. 상기 HlyAs에 의해서는 분비 작용 벡터가 발생되었고, 상기 분비 작용 벡터는 살모넬라 엔테리카, 예르시니아 엔테로콜리티카 및 비브리오 콜레아 내에서 단백질 항원의 효과적인 제거를 가능케 한다.
상기와 같은 분비 작용 벡터는 HlyA-신호 펩티드를 위한, 헤몰리신-분비 장치를 위한, hlyB 및 hlyD 그리고 hly-특정적인 프로모터(promoter)를 위한 뉴클레오티드 서열에 결합된 임의의 단백질 항원의 cDNA를 포함한다. 상기 분비 작용 벡터에 의해 프로모터는 상기 박테리아의 표면 상에서 발현될 수 있다. 상기와 같이 유전적으로 변형된 박테리아는 삽입된 핵산으로부터 발현된 단백질을 세포 내부에 포함하는 박테리아보다 훨씬 더 강력한 면역 방어 체계를 백신으로서 유도한다(도너 등 EP 1015023 A, 겐트쉐프 등, Gene, 179: 133-140, 1996; Vaccine 19: 2621-2618, 2001, 헤쓰 등 PNAS 93: 1458-1463, 1996). 그러나 상기 체계의 단점은, hly-특정적인 프로모터를 사용함으로써 박테리아의 표면 상에서 발현된 단백질의 양이 매우 적다는 것이다.
살모넬라 및 리스테리아와 같은 캐리어 박테리아를 이용하여 포유 동물 세포 내부로 플라스미드-DNA를 전달하기 위한 기술이 개발되었다. 상기 플라스미드 내부에 포함된 유전자는, 상기 유전자가 유카리온 프로모터의 통제하에 형성된 경우에도 포유 동물 세포 내에서 발현될 수 있었다. 리스테리아 모노시토겐 세균 내에는, 임의의 유카리온 프로모터의 통제하에서 임의의 항원을 위한 뉴클레오티드 서열을 포함하는 플라스미드가 삽입되었다. 특정 리시스-유전자를 위한 뉴클레오티드 서열을 삽입함으로써, 리스테리아 모노시토겐 세균은 항원을 나타내는 세포의 시토졸 내에서 용해되어 자신의 플라스미드를 방출하게 되며, 이와 같은 작용에 의해서는 후속적으로 플라스미드 코딩된 단백질이 발현, 처리 및 표시되고, 상기 단백질의 면역성이 현저하게 상승된다(디트리히 등, Nat. Biotechnol 16: 181-185, 1998; Vaccine 19: 2506-2512, 2001).
세포 내에 침범하는 박테리아의 독성 약화된 변종들이 발생되었다. 예를 들어 리스테리아 모노시토겐, 살모넬라 엔테리카 sv. 티피무리움 및 티피, 그리고 마이코 박테리아 bovis에 의해서 상기와 같은 변종들은 이미 타이푸스 및 튜버클로스에 대한 상화성이 우수한 생명체 접종 물질로서 사용되었다. 약화된 변종을 포함한 상기 박테리아는 일반적으로 면역을 자극하고, 우수한 세포 면역 반응을 발생시킬 수 있다. 예를 들어L. monocytogenes은 TH1 세포의 활성화를 특이한 정도만큼 능가하여 세포 파괴적인 림포 세포의 증식을 자극한다. 상기 박테리아는 분비된 항원을 시토졸 항원을 나타내는 세포 (APC ; 대백혈구 및 수상 세포) 내부로 직접 전달하며, 상기 세포는 공통으로 자극하는 분자를 발현시키고 T-세포의 효과적인 자극을 개시한다. 리스테리아는 부분적으로 파고조말 분획으로 분해되기 때문에 상기 캐리어 박테리아에 의해 생성된 항원은 한편으로는 MHC 계급 II 분자를 통해 나타날 수 있고, 그럼으로써 T-보조 세포를 유도할 수 있다. 다른 한편으로 리스테리아는 파고좀으로부터 방출된 후에 APCs의 시토졸 내에서 복제된다; 따라서, 상기 박테리아로부터 생성되고 분비된 항원은 바람직하게 MHC 계급 I-경로를 통해 나타나고, 그럼으로써 상기 항원에 대한 CTL 반응이 유도된다. 또한 대백혈구, 천연 식세포(NK) 및 뉴트로필과 리스테리아의 상호 반응에 의해 상기와 같은 시토킨의 발현이 유도되었고 (TNF-alpha, IFN-gamma, I1-2, IL-12; Unanue, Curr.Opin.Immunol, 9: 35-43, 1997; Mata and Paterson, J Immunol 163: 1449-14456, 1999), 상기 발현을 위해 안티-종양적인 효과가 검출되었다. 그럼으로써, 종양 항원의 발현을 위해 형질 도입된 L. 모노시토겐의 투여에 의해, 실험 종양의 성장이 항원 특정적으로 저지될 수 있었다(판 등 Nat Med 1: 471-477, 1995, Cancer Res 59: 5264-5269, 1999; 푀스트 등 Natl Cancer Inst 87: 581-586, 1995; 베아티 및 파터슨, J Immunol 165: 5502-5508, 2000).
종양 항원을 코딩하는 뉴클레오티드 서열이 삽입된 독성 약화된Salmonella enterica균주는 종양 항원을 발현시키는 박테리아성 캐리어로서 구강 접종 후에 상이한 실험 종양에 대해 특정적인 보호 작용을 할 수 있었다(메디나 등, Eur J Immunol 30: 768-777, 2000, 촐러 및 크리스트 J Immunol 166: 3440-34450, 2001; 시앙 등, PNAS 97: 5492-5497, 2000).
재조합된Salmonella균주는 또한 바이러스 감염(HPV)에 대한 예방 백신으로서도 작용하였고(베니아콥 등, Infect Immun 67: 3674-3679, 1999), 종양 바이러스(HPV)에 의해 사멸되지 않게 된 쥐 종양의 치료에 효과적이었다(레바즈 등, Virology 279: 354-360, 2001).
본 발명은 이질적인 뉴클레오티드 서열을 갖는 미생물, 약제, 특히 백신(vaccine)을 제조하기 위한 상기 미생물의 용도, 이질적인 뉴클레오티드 서열을 갖는 플라스미드 그리고 상기와 같은 미생물을 제조하기 위한 방법에 관한 것이다.
본 발명의 기술적 과제
본 발명의 기본이 되는 기술적 과제는, 특히 종양 예방 및 종양 치료 면에서 종양에 대한 면역 내성을 극복하는 개선된 백신인 약제를 하는 제공하는 것이다.
본 발명의 기본 개념
상기 기술적인 문제점을 해결하기 위해 본 발명은 세포 항원을 코딩하는 뉴클레오티드 서열을 갖는 미생물을 교시하며, 상기 미생물의 게놈 내에(서)는 하기의 성분들이 삽입되고 발현될 수 있다: I) 종양 세포의 하나의 항원 또는 다수의 항원의 적어도 하나의 에피토프(epitope)를 코딩하는 뉴클레오티드 서열 및/또는 종양이 발생되는 조직 세포에 대해 특정적인 하나의 항원 또는 다수의 항원의 적어도 하나의 에피토프를 위한 뉴클레오티드 서열; II) 선택적으로, 면역 체계의 세포를 자극하는 단백질을 코딩하는 뉴클레오티드 서열; IIIA) 성분 I) 및 선택적으로는 박테리아의 외부면에 있는 성분 II)의 발현 생성물의 발현을 가능케 하는 및/또는 성분 I) 및 선택적으로는 성분 II)의 발현 생성물의 분비를 가능케 하는 전달 시스템을 위한 뉴클레오티드 서열; 및/또는 IIIB) 포유 동물 세포의 시토졸 내에 미생물을 용해하기 위해 그리고 용해된 미생물 내에 포함된 플라스미드를 세포내 방출하기 위해 사용되는 단백질을 위한 뉴클레오티드 서열; 그리고 IV) "미생물 내에서 활성화될 수 있고, 조직-세포-특정적이지만 세포-특정적이지 않은 활성화 서열"로 구성된 그룹 중에서 선택되는, 성분 I) 내지 IIIB)의 하나 또는 다수의 성분을 발현시키기 위한 활성화 서열. 이 경우 각각의 성분 I) 내지 IV)는 개별적으로 또는 다중으로, 각각 동일하게 또는 상이하게 배열될 수 있으며, 본 발명은 또한약제를 제조하기 위해 사용되는 상기와 같은 미생물의 용도를 교시한다.
따라서 본 발명의 대상은, 재차 미생물의 외부 막에서 발현되거나 또는 분비되는 세포 항원을 코딩하는 뉴클레오티드 서열의 캐리어로서 기능하는 미생물 및 종양에 대한 면역 내성을 극복하기 위해 사용되는 상기 미생물의 용도, 및 정상 세포 및/또는 종양 세포의 세포 항원을 코딩하는 뉴클레오티드 서열의 캐리어로서 기능하는 미생물을 포함하는 새로운 종양 백신이다. 결국 본 발명에 의해서는, 종양 지향적인 면역 반응이 개시된다.
상세하게는, 본 발명에 따른 미생물은 하기의 성분들을 포함한다: I) 종양 세포의 적어도 하나의 세포 단백질의 적어도 하나의 항원의 적어도 하나의 에피토프를 코딩하는 적어도 하나의 뉴클레오티드 서열 및/또는 선택적으로는 종양이 발생되는 조직 세포에 대해 특정적인 적어도 하나의 항원의 적어도 하나의 에피토프를 위한 뉴클레오티드 서열, II) 선택적으로는 면역 체계의 세포를 자극하는 적어도 하나의 단백질을 위한 적어도 하나의 뉴클레오티드 서열, IIIA) 성분 I)에 의해 코딩된 세포 항원을 세포막을 유지하면서 발현시키거나 또는 분비하기 위한 및 성분 II)에 의해 코딩된 면역 자극 단백질을 분비하기 위한 전달 시스템을 위한 적어도 하나의 뉴클레오티드 서열, IIIB) 선택적으로는 미생물을 시토졸 내에 용해시킴으로써 미생물 내에 포함된 플라스미드가 시토졸 내부로 방출되도록 하는 리신을 위한 뉴클레오티드 서열, IV) 성분 I) 및 II)를 발현시키기 위해 미생물 내에서 활성화 가능한 또는 세포 비특정적으로, 종양 세포 특정적으로, 조직 세포 특정적으로 혹은 기능 특정적으로 활성화 가능한 활성화 서열을 위한 적어도 하나의 뉴클레오티드 서열.
바람직한 실시 형태
본 발명에 따른 미생물의 성분들은 하기에서 상세하게 기술된다.
성분 I)
성분 I)은 하나의 종양 세포의 적어도 하나의 세포 단백질의 또는 적어도 하나의 종양 유전자로 변성된 세포 단백질의 적어도 하나의 항원의 적어도 하나의 에피토프를 위한 적어도 하나의 뉴클레오티드 서열이다. 상기 세포 단백질의 종양 유전자 변종은 자신의 원래의 세포 기능의 상실 또는 획득에 영향을 미칠 수 있다. 또한 상기 세포 단백질은 "수용체 분자 또는 상기 분자의 부분, 특히 상기 수용체의 세포외, 세포막간 전이적인 또는 세포내 부분; 접착 분자 또는 상기 분자의 부분 및 특히 상기 접착 분자의 세포외, 세포막간 전이적인 또는 세포내 부분; 신호 형질 도입용 단백질; 세포 사이클 통제용 단백질; 분화 단백질; 태아 단백질; 및 바이러스 유도된 단백질"로 이루어진 그룹으로부터 선택될 수 있다. 상기와 같은 세포 항원은 세포 내에서 세포 성장 및 세포 분열의 조절 기능을 담당하고, 예를 들어 MHC-계급 I 분자에 의해서 정상 세포의 세포막 상에 나타난다. 종양 세포 내에서 상기 세포 항원은 종종 과도하게 발현되거나 특정적으로 변성된다. 상기와 같은 변종들은 종양 유전자 억제 유전자의 기능 한정 또는 종양 유전자에 대한 원형 종양 유전자의 활성화를 결과적으로 야기할 수 있으며, 단독으로 혹은 과도 발현과 공동으로 실제로 종양 성장에 참여할 수 있다. 상기와 같은 세포 항원은 종양 세포의 세포막 상에 나타나고 그에 따라 항원을 세포막 상에 나타내지만, 환자의 종양 질병에 영향을 미치는 면역 반응을 야기하지는 않는다. 라프(미국-5,156,841)에 의해서는 이미 종양 형성 단백질, 즉 종양 유전자의 발현 생성물을 종양 백신용 면역원으로서 사용하는 내용이 기술되었다. 상기 문헌이 명확하게 참조된다.
본 발명에 따른 세포 항원 및 상기 항원의 종양 유전자 변종에 대한 예는 i) 예를 들어 Her-2/new, 안드로겐-수용체, 에스트로겐-수용체, 미드카인-수용체, EGF-수용체, ERBB2, ERBB4, TRAIL-수용체, FAS, TNFalpha-수용체와 같은 수용체, ii) 예를 들어 c-Raf (Raf-1), A-Raf, B-Raf, Ras, Bcl-2, Bcl-X, Bcl-W, Bfl-1, Brag-1, Mcl-1, Al, Bax, BAD, Bak, Blc-Xs, Bid, Bik, Hrk, Bcr/abl, Myb, C-Met, IAP1, IAO2, XIAP, ML-IAP LIVIN, Survivin, APAF-1과 같은 신호 형질 도입 단백질 및 상기 단백질의 종양 유전자 변종; iii) 예를 들어 Cyclin - D(1-3), - E, - A, - B, - H, Cdk-1, -2, -4, -6, -7, Cdc25c, P16, p15, p21, p27, p18, pRb, p107, p130, E2F (1-5), GAAD45, MDM2, PCNA, ARF, PTEN, APC, BRCA, P53 및 동족체와 같은 세포 사이클 통제용 단백질 및 상기 단백질의 종양 유전자 변종, iv) 예를 들어 C-Myc, NFkB, c-Jun, ATF-2, Spl과 같은 트란스스크립션 인자 및 상기 인자의 종양 유전자 변종, v) 예를 들어 카르시노 태아 항원, 알파-페토 단백질, Mage, PSCA와 같은 태아 단백질, vi) 예를 들어 Mart, Gp100, 티로시나아제, GRP, TCF-4와 같은 분화 항원, vii) 예를 들어 하기의 바이러스와 같은 바이러스성 항원: HPV, HCV, HPV, EBV, CMV, HSV.
대안적으로 또는 추가적으로, 성분 I)은 각각의 종양이 발생되는 정상 조직세포에 대해 특정적인 적어도 하나의 항원을 위한 적어도 하나의 뉴클레오티드 서열일 수 있다. 상기와 같은 특정적인 항원의 예로서는 i) 예를 들어 안드로겐 수용체, 에스트로겐 수용체, 락토페린 수용체와 같은 수용체, ii) 예를 들어 염기성 Myelin, 알파-락트알부민, GFAP, PSA, Fibrillary acid 단백질, 티로시나아제, EGR-1, MUC1과 같은 분화 항원이 있다.
성분 II)
성분 II)는 면역 체계의 세포를 자극하는 적어도 하나의 단백질을 위한 적어도 하나의 뉴클레오티드 서열이다. 단백질의 선택에 의해, 성분 I)의 발현 생성물에 대한 면역 반응은 강화될 수 있고/있거나 (세포 면역 반응을 위한) Th1 세포의 활성화를 위해서 또는 (체액과 관련된 면역 반응을 위한) Th2 세포의 활성화를 위해서 조절될 수 있다. 면역을 자극하는 단백질은 예를 들어 i) M-CSF, GM-CSF, G-CSF와 같은 시토킨, ii) IFN-알파, -β, 감마와 같은 인터페론, iii) IL-1, -2, -3, -4, -5, -6, -7, -9, -10, -11, -12, -13, -14, -15, -16, 인간 발암 물질 억제 인자(LIF)와 같은 인터류킨, iv) Rantes, Monocyte chemotactic and activating factor(MCAF), 대백혈구 inflammatory 단백질-1 (MIP-1-알파, -β), 뉴트로필 활성 단백질-2 (NAP-2), IL-8이다.

성분 IIIA)는 미생물 외부면 상에서 성분 I) 및, 선택적으로는, II)의 발현 생성물의 발현을 가능케 하는 적어도 하나의 전달 시스템을 코딩하는 적어도 하나의 뉴클레오티드 서열이다. 이 경우 각각의 성분들은 선택적으로 분비될 수 있거나 또는 미생물의 세포막 상에서, 즉 세포막을 유지하면서 발현될 수 있다. 이와 같은 전달 시스템은 예를 들어 i) E. coli의 헤몰리신 전달 신호(hly-특정적인 프로모터의 통제하에 HlyA, HlyB 및 HlyD를 포함하는 뉴클레오티드 서열); 하기의 전달 신호가 사용될 수 있다: 분비를 위해 - HlyB 및 HlyD 단백질의 존재하에 C-말단 HlyA-전달 신호; 세포막을 유지하는 발현을 위해 - HlyB-단백질의 존재하에 C-말단 HlyA-전달 신호, ii) E. coli의 헤몰리신 전달 신호(hly-특정적이지 않은 세균성 프로모터의 통제하에 HlyA, HlyB 및 HlyD를 포함하는 뉴클레오티드 서열), iii) Caulobacter crescentus의 S-층 단백질(Rsa A)를 위한 전달 신호; 하기의 전달 신호가 사용될 수 있다: 분비 및 세포막을 유지하는 발현을 위해 - C-말단 RsaA-전달 신호, iv) 에셰리키아 콜리의 TolC-단백질을 위한 전달 신호; 하기의 전달 신호가 사용될 수 있다: 세포막을 유지하는 발현을 위해 - TolC의 N-말단 전달 신호(E. coli의 적분 세포막 단백질은 예를 들어 콜리신 E1의 흡수(모로나 등, J Bacteriol 153: 693-699., 1983) 및 콜리신 V의 분비(파트 등, J Bacteriol 173: 7549-7556., 1991)와 같은 기능 외에 U3-파아지용 수용체로서도 이용되는(오스틴 등, J Bacteriol 172: 5312-5325., 1990) E. coli의 외부 세포막의 기공을 형성하는 다기능성 단백질이다); 상기 단백질은 E. coli에서 뿐만 아니라 다수의 그람 음성 박테리아에서도 발견된다(비이너, Structure Fold Des 8: R171-175., 2000); 외부 세포막에서의 위치 설정 및 확대된 사건들은 TolC를 예를 들어 면역 반응을 발생시키기 위해 이형적 항원을 나타낼 수 있는 이상적인 후보자로 만들어준다).
성분 IIIB)
성분 IIIB)는 포유 동물 세포의 시토졸 내에서 발현되어 숙주 세포의 시토졸 내에 있는 플라스미드를 방출하기 위해 미생물을 세포 용해시키는 적어도 하나의 세포 용해성 단백질을 코딩하는 뉴클레오티드 서열이다. 상기와 같은 세포 용해성 단백질(엔돌리신)은 예를 들어 PLY551(뢰쓰너 등, Mol Microbiol 16: 1231-41, 1995)과 같은 리스테리아-특정적인 리시스-단백질 및/또는 리스테리아 프로모터의 통제하에 있는 리스테리아-특정적인 홀린이다.
본 발명의 바람직한 실시예는 상이한 성분들 IIIB)의 조합, 예컨대 홀린을 갖는 리시스-단백질의 조합이다.
성분 IIIA 및/또는 IIIB는 구성적으로 활성일 수 있다.
성분 IV)
성분 IV)는 성분 I) 및, 선택적으로는, II)를 발현시키기 위한 적어도 하나의 활성화 서열을 위한 적어도 하나의 뉴클레오티드 서열이다.
발현이 세포막을 유지하는 방식으로 미생물의 외부면 상에서 이루어지면, 상기 활성화 서열이 바람직하게 선택될 수 있음으로써, 상기 활성화 서열이 미생물 내에서 활성될 수 있다. 이와 같은 활성화 서열은 예를 들어: i) 예컨대 E. coli의 베타-lactamase Gen 또는 tetA Gen의 "Ribosomal binding site"(RBS)를 갖는 프로모터 구역과 같이 구성적으로 활성인 프로모터 구역(버스비 및 에브라이트, Cell 79: 743-746., 1994), ii) 유도 가능한 프로모터, 바람직하게는 세포 내부로의 흡수 후에 작용하는 프로모터이다. 상기 프로모터에는 L. monocytogen의 actA 프로모터(디트리히 등, Nat. Biotechnol. 16: 181-185, 1998) 또는 S. typhimurium의pagC 프로모터(부만, Infect Immun 69: 7493-7500., 2001)가 속한다.
미생물이 세포의 시토졸 내부로 세포 용해된 후에 플라스미드가 상기 미생물로부터 방출되면, 활성화 서열은 세포 특정적이지 않고, 조직 세포 특정적이며, 세포 사이클 특정적이거나 또는 기능 특정적이다. 바람직하게는, 대백혈구, 잔기형 세포 및 림프구 내에서 특이하게 활성되는 활성화 서열이 선택된다.
본 발명의 의미에서의 미생물은 바이러스, 박테리아 또는 단세포 기생 생물이며, 상기 생물들은 미생물에 이질적인 뉴클레오티드 서열을 전달하기 위해서 통상적으로 이용된다.
본 발명의 한 특이한 실시예에서 미생물은 그람-양성 또는 그람-음성 박테리아이며, 바람직하게는 예를 들어 에셰리키아 콜리, 살모넬라, 예르시니아 엔테로콜리티카, 비브리오 콜레라, 리스테리아 모노시토겐, 시겔라와 같은 박테리아이다.
바람직하게는 독성이 약화된 박테리아가 사용된다.
본 발명에 상응하는 상기 성분들은 당업자에게 공지된 방법으로 미생물 내에 삽입된다. 미생물이 박테리아인 경우에는, 상기 성분들이 플라스미드 내부로 삽입되고, 플라스미드는 박테리아 내부로 전달된다. 이에 적합한 기술 및 플라스미드는 당업자에게 충분히 공지되어 있다.
본 발명의 대상은 본 발명에 따른 미생물 또는 상기 미생물의 세포막 외피를 포함하는 약제 조성물이다. 상기 세포막 외피의 제조는 예를 들어 특허 출원서 EP-A-0 540 525호에 기술된 방법에 따라 이루어진다. 상기와 같은 약제 조성물은 예를 들어 약학 분야에서 잘 알려진 용액 속에 있는 본 발명에 따른 미생물의 현탁액으로서, 주입(injection)에 적합하다.
본 발명의 추가의 대상은 본 발명에 따른 미생물을 포함하는 약제 조성물의 투여이다. 이와 같은 투여는 국부적으로 또는 시스템적으로, 예컨대 세포성 표피 내부로, 피하 조직 내부로, 근육 조직 내부로, 체공 내부로, 장기 내부로, 종양 내부로 또는 혈액 순환계 내부로 이루어진다.
본 발명의 특이한 대상은 세포 증식적 질병의 예방 및/또는 치료를 위한 본 발명에 따른 약제 조성물의 경구적 투여 또는 직장내 투여이다. 이와 같은 투여 방법은 1회 또는 다수회 이루어질 수 있다. 각각의 투여 때마다 10 내지 10^9의 범위에서 본 발명에 따른 미생물이 투여된다. 상기 개수의 본 발명에 따른 미생물의 투여가 충분한 면역 반응을 야기하지 않으면, 주입될 개수를 높일 수 있다.
본 발명에 따른 미생물의 투여 후에는, 성분 I)을 나타내는 세포, 예컨대 종양 세포에 대한 내성 또는 종양이 생성되는 조직 세포에 대한 내성이 극복되어, 종양 및/또는 조직 세포 지향적이고 세포에 유독한 면역 반응이 개시된다.
성분 I)의 선택에 따라, 상기 세포에 유독한 면역 반응은 전적으로 종양 지향적이거나 또는 종양 세포가 생성되는 조직 세포를 포함한 종양 세포 지향적이기도 하다.
본 발명의 대상은 세포 증식적 질병의 예방 또는 치료를 위해 본 발명에 따른 약제 조성물을 투여하는 것이다. 상기와 같은 세포 증식적 질병에는 종양 질병, 백혈병, 바이러스에 의해 야기되는 질병, 만성적인 염증, 이식된 장기의 반발 및 자동 면역 질병이 속할 수 있다.
성분 I)이 종양 세포로부터 및 종양이 생성되는 조직 세포로부터 발현되는 적어도 하나의 세포 항원인 본 발명의 한 특이한 실시예에서는, 본 발명에 따른 약제 조성물이 갑상선, 유방, 위, 신장, 난소, 모반, 전립선, 목 또는 방광의 종양을 예방 또는 치료하기 위해서 투여된다.
본 발명은 실시예를 참조하여 하기에서 자세히 설명된다.
실시예 1: C-Raf를 발현시키는 살모넬라를 이용하여 면역성을 제공함으로써 BxB 쥐에서 면역 반응을 유도함
Raf는 신호 케스케이드의 다른 단백질과의 협력에 의해 세포 성장 및 세포 생존을 통제하는 통상적으로 세포에 유독한 세린/테레오닌 키나제(PSK)이다(케르크호프 및 라프, Oncogene 17: 1457-1462., 1998; 트로프마이르 및 라프, Recent Results Cancer Res 143: 245-249., 1997). 상응하는 수용체에 대한 성장 인자의 결합은 통상적으로 Ras의 활성화, PSK- 및 티로신 키나제 MEK를 통한 다수의 인산화 단계를 거친 Raf 및 PSK ERK의 후속하는 활성화를 통해 세포핵 내에서 유전자 복제가 활성된다(케른호프 및 라프, Oncogene 17: 1457-1462., 1998). 상기 사슬의 제 1 고리, 즉 작은 G-단백질 Ras는 모든 사람 종양의 약 30% 내에 변형된 상태로 존재한다(차코스 및 스판디도스, Crit Rev Oncol Hematol 26: 65-75., 1997). Raf는 Ras의 효과 인자(effector)이고, 다수의 사람 종양 내에 과도하게 발현된 상태로 존재한다(나우만 등, Recent Results Cancer Res 143: 237-244., 1997).
쥐 모델 테스트를 위해 전이 유전자 쥐가 사용되며, 상기 쥐는 완전한 분자또는 구성적으로 활성인 키나제 활동 범위(BxB)를 과도하게 발현시킨다(케르크호프 등, Cell Growth Differ 11: 185-190., 2000). 그럼으로써 상기 쥐들은 약 반 년 후에는 자발적으로 폐 종양을 발생시킨다.
백신의 발생을 위해 인간 c-Raf cDNA가 HlyA를 갖는 PCR "인-프레임(in-frame)" 방식으로 플라스미드 pMOhly 1로 분리 증식되었다(도 1). 그 다음에 상기 플라스미드 pMO-Raf는 방향성 신진대사시에 결함을 갖는 독성이 약화된 살모넬라에 이식되었다(호이제트 및 슈토커, Nature 291: 238-239, 1981). c-Raf를 지향하는 항체를 이용한 면역 블로팅(blotting) 방법에서는, pMOhy-Raf 이식된 SL7207-박테리아를 갖는 박테리아 세포 용해물 속에서 c-Raf HlyAs 융합 단백질이 검출될 수 있었다.
BxB 전이 유전자 쥐는 7-10주의 연령에서 구강으로 살모넬라를 주입하여 면역성을 나타냈으며 (DOSIS 5X10^9), 이 경우 접종은 5일의 간격으로 2회 반복되었다. 최종 면역 반응 45일 후에는 5x10^5개의 살모넬라를 정맥 주입하는 방법으로 추가 접종이 이루어졌다. 통제를 위해서, c-Raf를 코딩하는 DNA가 쥐의 근육 조직 내에 투여되었다.
최종 면역 반응 5-7일 후에는 세륨 샘플을 추출하여 WESTERN-블롯 방법에 의해 항체 반응이 분석되었다. 이를 위해 1:200으로 희석된 세륨이 세포막에 대해서 분리된 단백질 및 블로팅된 c-Raf의 단백질로 이식되거나 또는 이식되지 않은 박테리아와 혼성되었다. 결합된 세륨 항체의 검출은 쥐-IgG에 대해 특정적인 항체의 도움으로 이루어졌다. 통제 쥐와 달리 쥐들에서는 면역된 상태로 pMohly-Raf 이식된 SL7207 c-Raf 특정적인 이소타입 IgG의 항체가 유도되었다. 그럼으로써, 기술된 살모넬라에 의한 면역화는 자체-내성을 극복할 수 있고, IgG로의 항체 이소타입 교체를 위해 필수적인 CD4+ T-세포를 유도한다고 나타났다.
CD8+ T-세포 반응을 분석하기 위해, C57BL-6 쥐가 동일한 프로토롤에 따라 면역되었다. 최종 면역 반응 7일 후에는 비장 세포가 격리되었고, 상기 세포는 Raf-과발현 EL-4 세포로 자극되었다. 자극 시작 1시간 후에는 폐 전달이 브레펠딘 A에 의해 차단되었고, 추가 4시간 후에는 세포가 CD 8 및 IFN-g 특정적인 항체로 염색되었으며, 스루풋(throughput) 세포 미터법으로 분석되었다(미트루커 등, Infect Immun 70: 199-203., 2002). pMO-Raf 면역된 쥐에서만 Raf-특정적인 항체 반응이 검출될 수 있었다.
종양성 활성을 검출하기 위해 10, 12 및 14개월 된 늙고 면역된 및 면역되지 않은 BxB 쥐들이 죽었고, 폐 덩어리의 무게가 측정되었다. 폐 덩어리는 종양의 크기를 위한 직접적인 치수이다. 14개월 후에 SL-pMO-Raf로 면역된 그룹에서는, c-Raf(SL-pCMV-raf)를 코딩하는 있는 그대로의 DNA로 면역된 그룹을 포함한 통제 그룹에서보다 훨씬 더 많은 쥐들이 폐 덩어리의 축소를 경험했다. 통상적으로, 치료되지 않은 동물들에서의 종양 성장은 돌이킬 수 없다(케르크호프 등, Cell Growth Differ 11: 185-190., 2000). 상기 데이터는, 상기 실험에서 SL-pMO-Raf의 접종이 동물들을 종양 형성으로부터 보호할 수 있었고, 본 발명이 종양 백신으로서 적합하다는 사실을 보여준다.
상기 실험들은 또한, 본 발명에 기술된 캐리어 체계에 의해서 원칙적으로는자체-내성이 극복될 수 있고, c-Raf에 내성을 갖는 동물들에서 c-Raf 특정적인 항체 반응 및 T-세포 반응이 유도될 수 있다는 사실을 보여준다.
동일한 실험 체계에 의해 백신으로서 살모넬라가 제조될 수 있으며, 상기 살모넬라는 (예를 들어 B-Raf 및 A-Raf와 같은) C-Raf의 동형체, 변성된 C-Raf, B-Raf 또는 A-Raf, 정상의 또는 변성된 C-Raf, B-Raf 또는 A-Raf의 에피토프, 또는 정상의 및/또는 변성된 C-Raf, B-Raf 또는 A-Raf의 에피토프의 조합을 발현시킨다. Raf의 활성 손실과 동시에 일어나는 변성에 대한 예는 Ras-결합 활동 범위의 변성, 키나제-활동 범위의 변성 및/또는 인산화 장소의 변성이다.
실시예 2: PSA를 발현시키는 살모넬라를 이용하여 면역성을 제공함으로써 BALB/c 쥐에서 면역 반응을 유도함
조직 특정적 항원, 특히 상당 부분이 종양 세포로부터 합성되고 발현되는 항원의 존재는 상기 마아커의 진단학적인 이용 가능성 외에 치료 방식을 위해 가능한 공격점도 형성한다. 전립선 발암 물질에 대해서는 지금까지 3개의 중요한 항원이 확인되었다: PSA(Prostate specific Antigen), PSMA(Prostate specific Membrane-Atigen) 및 PSCA(Prostate Stem-Cell-Antigen). PSA가 이미 초반의 종양 형성시에 과도하게 발현된 상태로 존재하고(와트 등, Proc Natl Acad Sci U S A 83: 3166-3170., 1986; 왕 등, Prostate 2: 89-96., 1981) 그럼으로써 발암 물질 진단에 기여하는 한편(라브리 등, J Urol 147: 846-851; discussion 851-842., 1992), PSCA-발현은 대부분 국부적으로 진행되었고, 분화 해소되었으며, 전이된 종양 단계에서 비로소 증가되었다(구 등, Oncogene 19: 1288-1296., 2000; 라이터 등, Proc NatlAcad Sci U S A 95: 1735-1740., 1998). 장기 특유성은 전립선 발암 물질에 대한 면역 치료 개발시에 PSA 뿐만 아니라 PSCA도 잠재적인 목적 항원으로 만든다(라이터 등, Proc Natl Acad Sci U S A 95: 1735-1740., 1998; 홋지 등, Int J Cancer 63: 231-237., 1995; Armbruster, Clin Chem 39: 181-195., 1993).
상기 테스트에서는, PSA를 분리시키는 살모넬라가 벡터 pMOHLY 1을 토대로 하여 BALB/c 쥐 내에서 면역 반응을 유도할 수 있는지가 나타나야 한다. 이 목적을 위해 먼저 중합체-사슬 반응(PCR)에 의해서 2개의 NsiI-절단 장소가 PSA의 c-DNA-서열 내부로 삽입됨으로써, 증폭된 분획이 목적 벡터 내부로 "인-프레임(in-frame)" 방식으로 삽입될 수 있다. 증폭을 위해서는 645개 염기쌍(bp)의 분획이 선택되었다. 프라이머(primer)로서는 5'-GTGGATTGGTGATGCATCCCTCATC-3' 및 5'-CAGGGCACATGCATCACTGCCCCA-3'이 이용되었다. PCR-생성물은 먼저 "블런트-엔드 (blunt-end)" 방식에 의해 벡터 pUC18로 분리 증식되었고, 나중에 NsiI-절단 장소를 통해 목적 벡터 pMOhlyl과 결찰되었다. 정확한 삽입은 제한적 소화에 의해 통제되었고, 서열화에 의해 확인되었다(도 2).
상기 살모넬라 균주에 의해서는 BALB/c 쥐가 3주의 간격을 두고 3회 1x107의 양만큼 비강으로 면역 반응했다. 면역 응답은 WESTERN-블롯 분석 및 세포내 세포 분열 염색에 의해 검출된다.
본 발명은 종양을 치료하기 위해 사용되는 세포 항원을 코딩하는 뉴클레오티드 서열의 캐리어로서 기능하는 미생물에 이용될 수 있다.

- 통제되지 않은 세포 분열 또는 감염에 의해 야기되는 질병, 바람직하게는 종양 질병, 특히 전립선 발암 물질, 난소 발암 물질, 유방 발암 물질, 위 발암 물질, 신장 발암 물질, 갑상선 발암 물질, 피부 종양, 목 종양, 방광 종양, 침샘 종양 또는 림프샘 종양, 백혈병, 바이러스성 혹은 세균성 감염, 만성적인 염증, 이식된 장기의 반발 및/또는 자동 면역 질병을 특히 예방 및/또는 치료하기 위한 약제를 제조하기 위한, 제 1 항 내지 제 11 항 중 어느 한 항에 따른 미생물의 용도.
- 제 12 항에 있어서,종양이 생성되는 건강한 조직의 종양을 제거하기 위한 미생물의 용도.
- 제 12 항 또는 제 13 항에 있어서,상기 약제가 국부적, 비경구적, 경구적 또는 직장내 투여를 위해 사용되는 용도.
- 제 12 항 내지 제 15 항 중 어느 한 항에 따른 약제를 제조하기 위한 방법으로서,제 1 항 내지 제 11 항 중 어느 한 항에 따른 미생물에 경구적, i.m., i.v., i.p., 직장내 또는 국부적 투여를 위해, 하나 또는 다수의 생리학적으로 상화적인 캐리어 물질이 생리학적으로 효과적인 양만큼 사용되는, 약제의 제조 방법.
- 제 1 항에 따른 성분 I) 내지 IV)를 포함하는 플라스미드 또는 발현 벡터.
- 제 1 항 내지 제 11 항 중 어느 한 항에 따른 유기체를 제조하기 위한 방법으로서,제 16 항에 따른 플라스미드 또는 발현 벡터가 형성되고, 상기 플라스미드 또는 발현 벡터에 의해 미생물이 변환되는, 유기체의 제조 방법.
