Description
Claims (33)
- a) 서열 2의 아미노산 서열을 갖는 단백질을 코딩하는 DNA 서열,b) 엄격한 조건하에 (a)의 서열과 혼성화하는 DNA 서열, 또는c) (a) 또는 (b)의 서열과 동의성이 있는 (degenerative) DNA 서열.
- 제1항에 있어서, 서열 1의 서열인 DNA 서열 또는 이의 단편.
- 제2항에 있어서, 서열 1의 위치 212 내지 214의 ATG로 출발하여 위치 5468 내지 5470의 TGA에서 종결하는 서열을 포함하는 DNA 서열 또는 이의 임의의 단편.
- 제3항에 있어서, 서열 1의 위치 212 내지 214의 ATG로 출발하여 위치 3731 내지 3734의 ATCT에서 종결하는 서열을 포함하는 DNA 서열 또는 이의 임의의 단편.
- 제3항에 있어서, 서열 1의 위치 308 내지 310의 GTC로 출발하여 위치 5468 내지 5470의 TGA에서 종결하는 서열을 포함하는 DNA 서열 또는 이의 임의의 단편.
- 제3항에 있어서, 서열 1의 위치 308 내지 310의 GTC로 출발하여 위치 3731 내지 3734의 ATCT에서 종결하는 서열을 포함하는 DNA 서열 또는 이의 임의의 단편.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 서열 2에 따른 아미노산 서열의 60% 미만, 바람직하게는 45% 미만, 더욱 바람직하게는 25% 미만이 변화되는 뉴클레오티드 치환, 부가 또는 결실을 포함하는 DNA 서열 또는 이의 단편.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 보존적 아미노산 치환되는 뉴클레오티드 치환을 포함하는 DNA 서열.
- 제1항 내지 제8항 중 어느 한 항의 DNA 서열에 의해 코딩되는 효소.
- 서열 2에 따른 아미노산 서열 또는 이의 단편을 포함하는 효소.
- 서열 2에 정의된 바와 같은 서열을 갖는 β-갈락토시다제.
- 제10항에 있어서, 서열 2에 정의된 바와 같은 Met(1) 내지 Gly(1752)의 서열 또는 이의 단편을 갖는 효소.
- 제12항에 따른 성숙한 β-갈락토시다제.
- 제10항에 있어서, 서열 2에 정의된 바와 같은 Met(1) 내지 Ile(1174)의 서열 또는 이의 단편을 갖는 효소.
- 제14항에 따른 트랜스갈락토실화 효소.
- 제14항에 있어서, 서열 2에 정의된 바와 같은 Ala(33) 내지 Ile(1174)의 서열 또는 이의 단편을 갖는 효소.
- 제16항에 따른 성숙한 트랜스갈락토실화 효소.
- 제14항 내지 제17항 중 어느 한 항에 있어서,a) 37℃에서 100 g/L 락토스 용액 중에서 β-갈락토시다제 활성에 대한 트랜스갈락토실화 활성의 비가 1 : 1 이상임,b) 37℃에서 100 g/L 락토스 용액을 사용한 배치 (batch) 반응에서 25% 이상의 갈락토-올리고당류 생산을 촉매함,c) 37℃에서 100 g/L 락토스 용액을 사용한 배치 반응에서 갈락토-올리고당류 생산을 촉매하며 갈락토-올리고당류의 농도가 최고인 반응 시간에 유리 형태로 존재하는 락토스로부터 15% 미만의 갈락토스 생산을 촉매함의 특징 중 하나 이상을 보유하는 트랜스갈락토실화 효소.
- 제1항 내지 제8항 중 어느 한 항의 DNA 서열을 포함하는 재조합 벡터.
- 제19항에 있어서, 발현 벡터인 벡터.
- 제1항 내지 제8항 중 어느 한 항의 DNA 서열을 포함하는 숙주 세포.
- 제19항 또는 제20항의 벡터를 포함하는 숙주 세포.
- 제21항 또는 제22항에 있어서, 박테리아 세포, 효모 세포 또는 진균 세포인 세포.
- 제23항에 있어서, 비피도박테륨 (Bifidobacterium), 락토콕쿠스 (Lactococcus), 락토바실러스 (Lactobacillus), 스트렙토콕쿠스 (Streptococcus), 류코노스톡 (Leuconostoc), 에쉐리히아 (Escherichia), 바실러스 (Bacillus), 스트렙토마이세스 (Streptomyces), 사카로마이세스 (Saccharomyces), 클루이베로마이세스 (Kluyveromyces), 칸디다 (Candida), 토룰라 (Torula), 토룰롭시스 (Torulopsis) 및 아스페르길루스 (Aspergillus)로 구성된 군에서 선택된 세포.
- 제24항에 있어서, 비피도박테륨 브레브 (Bifidobacterium breve), 비피도박테륨 롱검 (Bifidobacterium longum), 비피도박테륨 인판티스 (Bifidobacterium infantis), 비피도박테륨 비피둠 (Bifidobacterium bifidum) 및 락토콕쿠스 락티스 (Lactococcus lactis)로 구성된 군에서 선택된 세포.
- 요구르트, 치즈, 발효시킨 우유 제품, 식이보조제 및 건강보조 (probiotic) 식품으로 구성된 군에서 선택된 제품의 생산을 위한, 제21항 내지 제25항 중 어느 한 항의 세포의 용도.
- 제21항 내지 제25항 중 어느 한 항의 세포를 포함하는 유제품.
- 갈락토-올리고당류 생산을 위한, 제14항 내지 제18항 중 어느 한 항의 트랜스갈락토실화 효소 또는 제21항 내지 제25항 중 어느 한 항의 세포의 용도.
