Abstract
본 발명은 화학식 I의 아미드, 그의 호변이성질체 및 이성질체 형태, 그의 거울상이성질체 및 부분입체이성질체 형태 및 그의 생리학적으로 허용가능한 염에 관한 것이다. 또한, 본 발명은 칼파인 억제제로서 그의 제조 및 그의 용도에 관한 것이다.

Description
본 발명은 효소, 특히 칼파인 (=칼슘-의존성 시스테인 프로테아제)과 같은 시스테인 프로테아제 및 그의 동위효소 및 카텝신, 예를 들어 B 및 L의 억제제인 신규한 아미드에 관한 것이다.
칼파인은 이른바 시스테인 프로테아제 군으로부터의 세포내 단백질 분해 효소이며 많은 세포에서 발견된다. 칼파인은 증가된 칼슘 농도에 의해 활성화되며, 칼슘 이온의 μ-몰 농도에 의해 활성화되는 칼파인 I 또는 μ-칼파인 및 칼슘 이온의 m-몰 농도에 의해 활성화되는 칼파인 II 또는 m-칼파인으로 구별된다 (P.Johnson, Int. J. Biochem. 1990, 22(8), 811-22). 오늘날 여전히 다른 칼파인 동위효소가 가정되고 있다 (K.Suzuki 등, Biol. Chem. Hoppe-Seyler, 1995, 376 (9), 523-9).

증가된 칼파인 수준이 여러 가지 병태생리 과정, 예를 들어: 심장의 허혈 (예, 심근경색), 신장 또는 중추신경계의 허혈 (예, "졸증"), 염증, 근육 이영양증, 눈의 백내장, 중추 신경계에 대한 손상 (예, 외상), 알쯔하이머 질병 등에서 측정되었다 (참조, 상기 K.K Wang의 문헌). 증가되고 지속적인 세포내 칼슘 수준과 상기 질병들과의 관계가 추측된다. 그 결과, 칼슘-의존성 과정이 과다활성화되고 더 이상 생리적으로 조절될 수 없다. 따라서, 칼파인의 과다활성화는 또한 병태생리 과정을 개시시킬 수 있다.
따라서, 칼파인 효소의 억제제가 상기 질병들의 치료를 위해 유용할 수 있는 것으로 가정된다. 여러 연구가 이를 뒷받침한다. 즉, 문헌 (Seung-Chyul Hong 등, Stroke 1994, 25(3), 663-9) 및 문헌 (R.T.Bartus 등, Neurological Res. 1995, 17, 249-58)은, 예컨대 뇌졸증 후에 발생하는 급성 신경변성 질환 또는 허혈에서 칼파인 억제제의 신경보호 작용을 나타내고 있다. 유사하게, 실험적인 뇌 외상후에, 칼파인 억제제는 발생되는 기억력 결핍 및 신경지배 장애로부터의 회복을 개선시켰다 (K.E.Saatman 등, Proc. Natl. Acad. Sci. USA, 1996, 93, 3428-3433). C.L. 엘데스테인(Edelstein) 등 (Proc. Natl. Acad. Sci. USA, 1995, 92, 7662-6)은 저산소증에 의해 손상된 신장에 대한 칼파인 억제제의 보호 작용을 밝혔다. 요시다, 껜 이스찌(Yoshida, Ken Ischi) 등 (Jap. Circ. J. 1995, 59(1) 40-8)은 허혈 또는 재관류에 의해 발생되는 심장 손상후에 칼파인 억제제의 바람직한 효과를 나타낼 수 있었다. 칼파인 억제제는 β-AP4 단백질의 방출을 억제하기 때문에, 알쯔하이머 질병을 위한 치료제로서의 잠재적인 용도가 제안되었다 (J.Higaki 등, Neuron, 1995, 14, 651-59). 인터루킨-1α의 방출은 또한 칼파인 억제제에 의해 억제되었다 (N.Watanabe 등, Cytokine 1994, 6(6), 597-601). 또한, 칼파인 억제제가 종양 세포에 대해 세포독성 효과를 나타내는 것으로 밝혀졌다 (E.Shiba 등, 20th Meeting Int. 칼파인 억제제의 또다른 가능한 용도는 문헌 (K.K.Wang, Trends in Pharmacol. Sci., 1994, 15, 412-8)에 기재되어 있다.
칼파인 억제제는 이미 문헌에 기재되어 있다. 그러나, 이들은 주로 비가역적이거나 또는 펩티드 억제제이다. 일반적으로, 비가역적 억제제는 알킬화 물질이고, 체내에서 비-선택적으로 반응하거나 불안정하다는 단점을 갖고 있다. 즉, 이러한 억제제들은 종종 바람직하지 못한 부작용, 예컨대 독성을 나타내며, 따라서 그들의 용도에서 제한되거나 이용될 수 없다. 비가역적 억제제중에는, 예를 들어 에폭시드 E 64 (E.B.McGowan 등, Biochem. Biophys. Res. Commun. 1989, 158, 432-5), α-할로케톤 (H.Angliker 등, J. Med. Chem. 1992, 35, 216-20) 또는 디술파이드 (R.Matsueda 등, Chem. Lett. 1990, 191-194)가 포함될 수 있다.
시스테인 프로테아제의 공지된 다수의 가역적 억제제, 예컨대 칼파인은 펩티드 알데히드, 특히 디펩티드 및 트리펩티드 알데히드, 예를 들어 Z-Val-Phe-H (MDL 28170) (S.Mehdi, Trends in Biol. Sci. 1991, 16, 150-3)이다. 생리학적 조건하에서, 펩티드 알데히드는 그의 높은 반응성 때문에 종종 불안정하며, 빠르게 대사될 수 있으며, 독성 작용을 일으킬 수 있는 비특이적 반응을 야기시킬 수 있는 단점이 있다 (J.A. Feherntz and B. Castro, Synthesis 1983, 676-78).
일본 특허 제08183771호 (캐나다 특허, 1996, 제605307호) 및 유럽 특허 제520336호는 칼파인 억제제로서 4-피페리디노일아미드 및 1-카르보닐피페리디노-4-일아미드로부터 유래된 알데히드를 개시한다. 국제 특허 공개 제97/21690호에서, N-술포닐프롤린아미드로부터 유래된 알데히드가 제조되었다. 국제 특허 공개 제96/06211호는 Y가 R1-X와 같은 추가의 임의의 라디칼을 갖지 않는 크산틴 유도체인 것을 제외한 화학식 I과 유사한 알데히드 유도체를 개시한다. 그러나, 본원에서 청구되고, 헤테로방향족 치환기를 갖는 화학식 I의 아미드로부터 유래되는 알데히드는 이전에 개시된 적이 없다.
펩티드 케톤 유도체는 또한 시스테인 프로테아제, 특히 칼파인의 억제제이다. 따라서, 예를 들어, 세린 프로테아제의 경우에 있어서 케토기가 CF3와 같은 전자-끌게 기에 의해 활성화되는 케톤 유도체는 억제제로서 공지되어 있다. 세린 프로테아제의 경우에 있어서, CF3또는 유사한 기로 활성화된 케톤을 갖는 유도체는 거의 활성이지 않거나 불활성이다 (M. R. Angelastro et al., J. Med. 케토벤즈아미드가 이미 문헌에 공지되어 있다. 즉, 케토에스테르 PhCO-Abu-COOCH2CH3가 국제 특허 공개 제91/09801호, 동 제94/00095호 및 동 제92/11850호에 기재되어 있다. 유사한 페닐 유도체 Ph-CONH-CH(CH2Ph)-CO-COCOOCH3를 문헌 (M.R.Angelastro 등, J.Med. Chem. 1990, 33, 11-13)에서 찾아볼 수 있으나, 이는 단지 약한 칼파인 억제제이다. 이러한 유도체는 또한 문헌 (J.P.Burkhardt, Tetrahedron Lett., 1988, 3433-36)에 기재되어 있다. 그러나, 헤테로고리 치환된 아미드의 중요성은 지금까지 결코 연구된 바 없다.

화학식 I의 화합물은 라세미체로서 또는 거울상이성질적으로 순수한 화합물로서 또는 부분입체이성질체로서 사용될 수 있다. 거울상이성질적으로 순수한 화합물을 원한다면, 예를 들어 적절한 광학적 활성 염기 또는 산을 사용하여 화학식 I의 화합물 또는 그의 중간체에 통상적인 라세미체 분할을 수행함으로써 수득될 수 있다. 다른 한편, 거울상이성질체 화합물은 통상적으로 입수가능한 화합물, 예를 들어 페닐알라닌, 트립토판 및 티로신과 같은 광학적 활성 아미노산을 사용하여 유사하게 제조될 수 있다.
본 발명은 또한 화학식 I의 화합물과 메조메리 또는 호변이성질체인 화합물, 예를 들어 화학식 I의 알데히드 또는 케토기가 에놀 호변이성질체로서 존재하는 화합물에 관한 것이다.
본 발명은 또한 화학식 I의 화합물과 적합한 산 또는 염기와의 반응으로 얻을 수 있는 화학식 I의 화합물의 생리학적으로 허용가능한 염에 관한 것이다. 적합한 유기 및 무기산은 예를 들면 문헌 (Fortschritte der Arzneimittelforschung, 1996, Birkhauser Verlag, vol. 10, pp. 224-285)에 기재되어 있다. 이는 예를 들어, 염산, 시트르산, 타르타르산, 락트산, 인산, 메탄술폰산, 아세트산, 포름산, 말레산, 푸마르산, 말산, 숙신산, 말론산, 황산 등 또는 수산화나트륨, 수산화리튬, 수산화칼륨, α, α, α-트리스(히드록시메틸)메틸아민, 트리에틸아민 등이 있다.
본 발명에 따른 아미드 I은 합성 반응식 1에 개략된 다양한 방법으로 제조될 수 있다.
헤테로고리 카르복실산 II는 적합한 아미노알콜 III와 결합하여 상응하는 아미드 IV를 생성한다. 문헌 (C.R.Larock, Comprehensive Organic Transformations, VCH Publisher, 1989, page 972f 또는 Houben-Weyl, Methoden der organischen Chemie [Methods of Organic Chemistry], 제4판, E5, V장)에 기재된 통상의 펩티드 결합 방법이 사용된다. 반응은 바람직하게는 "활성화된" 산 유도체 II (여기서, 산기 COOH가 COL로 전환됨)를 이용하여 수행된다. L은 예를 들어 Cl, 이미다졸 및 N-히드록시벤조트리아졸과 같은 이탈기이다. 이어서 이 활성화된 산은 아민을 이용하여 아미드 IV로 전환된다. 이 반응은 염화메틸렌, 테트라히드로푸란 및 디메틸포름아미드와 같은 무수 불활성 용매 중에 -20 내지 +25 ℃의 온도에서 수행된다.
이러한 알콜 유도체 IV는 본 발명에 따른 알데히드 유도체 I으로 산화될 수 있다. 예를 들어, 스웬 (Swern) 및 스웬 (Swern)-유사 산화 (T.T. Tidwell, Synthesis 1990, 857-70), 하이포염화나트륨/TEMPO (S.L. Harbenson et al,. 상기 참조) 또는 Dess-Martin (J. Org. 별법으로, 카르복실산 II는 아미노히드록삼산 유도체 VI와 반응하여 벤즈아미드 VII를 생성한다. 이러한 경우에 있어서, IV의 제조에서와 동일한 반응 방법이 사용된다. 히드록삼산 유도체 VI은 히드록시아민과 반응함으로써 보호된 아미노산 V로부터 얻을 수 있다. 이 방법에 있어서, 상기 기술된 아미드 제조 방법이 사용된다. 예를 들어, Boc와 같은 보호기 X의 제거는 트리플루오로아세트산을 사용하여 종래의 방법으로 수행된다. 이와 같이 얻은 아미도히드록삼산 VII는 환원에 의해서 본 발명에 따른 알데히드 I으로 전환될 수 있다. 이 방법에 있어서, 예를 들어 테트라히드로푸란 또는 에테르와 같은 불활성 용매 중에서 -60 내지 0 ℃의 온도에서 환원제로서 수소화리튬알루미늄이 사용된다.
마지막 방법과 유사하게, 에스테르 IX (Y=COOR', COSR')과 같은 카르복실산 또는 산 유도체가 또한 제조될 수 있으며, 환원에 의해서 본 발명에 따른 알데히드 I으로 전환될 수 있다. 이러한 방법은 문헌 (R.C.Larock, Comprehensive Organic Transformations, VCH Publisher, 1989, page 619-26)에 기재되어 있다.
케토아미드 또는 케토에스테르기를 보유하는 본 발명에 따른 헤테로고리 치환된 아미드 I의 제조는 합성 반응식 2 및 3에 개략된 다양한 방법으로 수행될 수 있다.
적절하다면, 카르복실산 에스테르 IIa는 실온 또는 25 ∼ 100 ℃와 같은 승온에서 수성 매질 중에서 또는 물과 알콜 또는 테트라히드로푸란과 같은 유기 용매와의 혼합물 중에서 수산화리튬, 수산화나트륨 또는 수산화칼륨과 같은 염기 또는 산을 사용하여 산 II로 전환된다.
이러한 산 II는 α-아미노산 유도체에 연결되며, 예를들어 문헌 (Houben-Weyl, Methoden der organischen Chemie [Methods of Organic Chemistry], 제 4 판, E5, V장, 및 C.R.Larock, Comprehensive Organic Transformations, VCH Publisher, 1989, Ch.9)에 기재된 통상의 조건이 사용된다.
예를 들어, 카르복실산 II는 "활성화된" 산 유도체 IIb=Y-COL (식중, L은 Cl, 이미다졸 또는 N-히드록시벤조트리아졸과 같은 이탈기임)로 전환되고, 이어서 아미노산 유도체 H2N-CH(R3)-COOR과의 반응에 의해 유도체 XI로 전환된다. 이 반응은 염화메틸렌, 테트라히드로푸란 및 디메틸포름아미드와 같은 무수 불활성 용매 중에서 -20 내지 +25 ℃의 온도에서 수행된다.
대체로 에스테르인 유도체 XI는 상기 기재된 가수분해와 유사하게 케토카르복실산 XII으로 전환된다. 케토에스테르 I'는 다킨-웨스트 (Dakin-West)의 방법과 유사한 반응으로 제조되며, 반응은 자오자오 리(ZhaoZhao Li) 등의 방법 (J.Med.Chem., 1993, 36, 3472-80)에 따라 수행된다. 이 방법에서, XII과 같은 카르복실산을 예를 들어 테트라히드로푸란과 같은 용매 중에서 승온 (50∼100 ℃)에서 옥살산 모노에스테르 클로라이드와 반응시키고, 이렇게 수득된 생성물을 25∼80 ℃의 온도에서 에탄올 중에서 나트륨 에톡시드와 같은 염기와 반응시켜 본 발명에 따른 케토에스테르 I'이 수득된다. 케토에스테르 I'를 상기 기재된 바와 같이 예를 들면 본 발명에 따른 케토카르복실산으로 가수분해될 수 있다.
케토아미드 I'을 수득하기 위한 반응은 또한 자오자오 리(ZhaoZhao Li) 등 (상기 참조)의 방법과 유사하게 수행된다. I'에서의 케토기는 예를 들면 염화메틸렌과 같은 불활성 용매 중에서 실온에서 트리플루오르화 붕소 에테르화물과 같은 루이스 산 촉매하에 1,2-에탄디티올의 첨가에 의해 보호되며, 디티안이 수득된다. 이러한 유도체를 0∼ 80 ℃의 온도에서 알콜과 같은 극성 용매 중에서 아민 R3-H과 반응시키면, 케토아미드 I (R4=Z 또는 NR7R8)이 수득된다.
별법을 반응식 3에 나타낸다. 케토카르복실산 II을 통상의 펩티드 결합 방법 (상기 참조, Houben-Weyl)하에서 아미노히드록시카르복실산 유도체 XIII (XIII의 제조에 대해, 문헌 S.L.Harbenson 등, J.Med.Chem. 1994, 37, 2918-29 참조)와 반응시키면 아미드 XIV가 수득된다. 이러한 알콜 유도체 XIV는 본 발명에 따른 케토카르복실산 유도체 I로 산화될 수 있다. 이를 위하여 여러 가지 통상의 산화 반응 (참조, C.R.Larock, Comprehensive Organic Transformations, VCH Publisher, 1989, 604면), 예를 들면 실온 또는 -50 ∼ 25 ℃의 온도에서 염화메틸렌 또는 테트라히드로푸란과 같은 용매 중에서 적절하다면 디메틸 술폭시드를 첨가하여 디메틸 술폭시드/피리딘-삼산화황 착물 (T.T. Tidwell, Synthesis 1990, 857-70) 또는 하이포염화나트륨/TEMPO (상기 참조, S.L.Harbenson 등)의 스웬 및 스웬-유사 산화반응이 사용될 수 있다.
XIV가 α-히드록시 에스테르 (X=O-알킬)이라면, 이것은 카르복실산 XV로 가수분해될 수 있으며, 반응은 상기 방법과 유사하게 수행되지만, 바람직하게는 실온에서 물/테트라히드로푸란 혼합물 중에서 수산화리튬을 사용하여 수행된다. 다른 에스테르 또는 아미드 XVI의 제조는 이미 기재된 결합 조건하에서 알콜 또는 아민과의 반응에 의해 수행된다. 알콜 유도체 XVI은 다시 산화되어 본 발명에 따른 케토카르복실산 유도체 I이 수득될 수 있다.
카르복실산 에스테르 II의 제조는 이미 임의의 경우에 있어서 기재되거나 종래의 화학 방법에 따라서 수행된다.
X가 결합인 화합물은 종래의 방향족 결합 예를 들어, 붕산 유도체 및 할로겐화물과의 팔라듐 촉매와 스즈키 (Suzuki) 결합 또는 할로겐화 방향족의 구리-촉매 결합으로 제조된다. 알킬-가교된 라디칼 (X=-(CH2)m-)은 케톤 동족체의 환원 또는 유기리튬, 예를 들어 오르토-페닐옥사졸리딘 또는 기타 유기금속 화합물의 알킬화에 의해 제조될 수 있다 (I.M. Dordor et al., J. Chem. Soc. Perkin Trans. I, 1984, 1247-52).
에테르-가교된 유도체는 상응하는 알콜 또는 페놀을 할로겐화물과 알킬화하여 제조한다.
술폭사이드 및 술폰은 상응하는 산화로 얻을 수 있다.
알켄- 및 알킨-가교된 화합물을 방향족 할로겐화물 및 적절한 알켄 및 알킨으로부터 예를 들어, 헤크 (Heck) 반응으로 제조하였다 (I. Sakamoto et al., Chem. Pharm. Bull., 1986, 34, 2754-59).
칼콘 (chalcone)은 아세토페논과 알데히드의 축합으로부터 형성되며, 임의로 수소화에 의해 유사한 알킬 유도체로 전환될 수 있다.
아미드 및 술폰아미드는 상기 기재된 방법과 유사하게 아민 및 산 유도체로부터 제조된다.
본 발명의 헤테로고리 치환된 아미드 I은 시스테인 프로테아제의 억제제, 특히 칼파인 I 및 II 및 카텝신 B 및 L과 같은 시스테인 프로테아제이다.
헤테로고리 치환된 아미드 I의 억제 활성은 문헌에 통상적인 효소 시험을 사용하여 측정하였으며, 효소 활성의 50%가 억제된 억제제 농도 (=IC50)를 활성의 스케일로 측정하였다. 아미드 I을 칼파인 I, 칼파인 II 및 카텝신 B의 억제 활성에 대하여 이러한 방법으로 측정하였다.
카텝신 B 시험
카텝신 B 억제를 문헌 (S.Hasnain 등, J.Biol.Chem. 1993, 268, 235-40)의 방법과 유사하게 측정하였다.
억제제 및 DMSO (최종 농도: 100 μM 내지 0.01 μM)로 부터 제조된 2 ㎕의 억제제 용액을 88 ㎕의 카텝신 B (500 μM 완충액중에 5 유니트로 희석된 인간 간으로부터의 카텝신 B (Calbiochem))에 용해시켰다. 이 혼합물을 실온 (25 ℃)에서 60 분동안 예비배양한 다음, 10 ㎕의 10 mM Z-Arg-Arg-pNA (10 % DMSO를 가진 완충액중)의 첨가에 의해 반응을 출발시켰다. 반응을 30 분동안 미소적정 플레이트 판독기에서 405 nM에서 감시하였다. 이어서 최대 구배로부터 IC50을 결정하였다.
칼파인 I 및 II 시험
칼파인 억제제의 억제 특성의 시험은 50 mM 트리스 HCl, pH 7.5; 0.1 M NaCl; 1 mM 디티오트레이톨; 0.11 mM CaCl2을 사용하여 완충액 중에서 수행되며, 형광발생 칼파인 기질 Suc-Leu-Tyr-AMC (DMSO중에 용해된 25 mM, 바켐 (Bachem)/스위스연방)이 사용된다. 인간 μ-칼파인을 적혈구로부터 단리하고, 여러 번의 크로마토그래피 (DEAE-세파로스, 페닐-세파로스, 슈퍼덱스 200 및 블루 세파로스) 단계 후에, SDS-PAGE, 웨스턴 블롯 분석 및 N-말단 시퀀싱에 따라 평가된 > 95 %의 순도를 가진 효소를 수득한다. 분열 생성물 7-아미노-4-메틸코우마린 (AMC)의 형광을 λex= 380 nm및 λex= 460 nm에서 스펙스-플로오로로그 (Spex-Fluorolog) 형광계에서 감시한다. 60 분의 측정 범위에서, 기질의 분열은 직선형이며, 실험을 12 ℃의 온도에서 수행하는 경우 칼파인의 자기촉매 활성이 낮다. 억제제 및 칼파인 기질을 DMSO 용액으로서 실험 배치에 첨가하고, 이때 DMSO는 최종 농도가 2 %를 넘지 않아야 한다.
실험 배치에서, 10 ㎕의 기질 (250 μM 최종) 및 이어서 10 ㎕의 μ-칼파인 (2 ㎍/ml 최종, 즉 18 nM)을 완충액을 함유한 1 ml 큐벳에 첨가한다. 기질의 칼파인-매개 분열을 15 내지 20 분동안 측정한다. 이어서, 10 ㎕의 억제제 (DMSO 중에서 50 ∼ 100 μM)을 첨가하고, 분열의 억제를 추가 40 분동안 측정한다.
가역적 억제에 대해 종래의 방정식에 따라 Ki값을 결정한다:

칼파인은 세포내 시스테인 프로테아제이다. 칼파인에 의한 세포내 단백질의 분해를 막기 위해서는 칼파인 억제제가 세포막을 통과해야 한다. 일부 공지된 칼파인 억제제, 예컨대 E 64 및 류펩틴만이 어렵게 세포막을 통과하며, 따라서 이들은 양호한 칼파인 억제제이긴 하지만 세포내에서 단지 불량한 작용을 나타낸다. 양호한 막 접근성을 가진 화합물을 찾아내는 것이 목적이다. 칼파인 억제제의 막 접근성의 증명으로서, 본 발명자들은 인간 혈소판을 사용한다.
혈소판에서 티로신 키나아제 pp60src의 칼파인-매개 분해
혈소판의 활성화후에, 티로신 키나아제 pp60src를 칼파인에 의해 분열시킨다. 이는 문헌 (Oda 등, J. Biol. Chem., 1993, Vol. 268, 12603-12608)에 의해 상세히 조사되었다. 이 문헌의 내용에는, pp60src의 분열이 칼파인의 억제제인 칼펩틴에 의해 방해될 수 있다고 나타나 있다. 본 발명의 물질의 세포 유효성을 이 문헌에 따라 시험하였다. 시트레이트로 처리된 신선한 인간 혈액을 200 g에서 15 분동안 원심분리하였다. 혈소판-다량 함유 혈장을 모으고, 혈소판 완충액 (혈소판 완충액: 68 mM NaCl, 2.7 mM KCl, 0.5 mM MgCl2×6 H2O, 0.24 mM NaH2PO4×H2O, 12 mM NaHCO3, 5.6 mM 글루코스, 1mM EDTA, pH 7.4)로 1:1 희석하였다. 원심분리 및 혈소판 완충액으로 세척 단계 후에, 혈소판을 107세포/ml로 조절하였다. 인간 혈소판의 단리를 RT에서 수행하였다.
분석 혼합물에서, 단리된 혈소판 (2 ×106)을 상이한 농도의 억제제 (DMSO에 용해됨)와 함께 37 ℃에서 5 분동안 예비배양하였다. 이어서, 혈소판을 1 μM 이온운반체 A 23187 및 5 mM CaCl2로 활성화시켰다. 5 분동안 배양한 후에, 혈소판을 13000 rpm에서 짧게 원심분리하고, 펠릿을 SDS 시료 완충액 (SDS 시료 완충액: 20 mM 트리스-HCl, 5 mM EDTA, 5 mM EGTA, 1 mM DTT, 0.5 mM PMSF, 5 ㎍/ml 류펩틴, 10 ㎍/ml 펩스타틴, 10 % 글리세롤 및 1 % SDS)중에 취하였다. 단백질을 12 % 농도 겔에서 분리하고, pp60src 및 그의 52 kDa 및 47 kDa 분열 생성물을 웨스턴 블롯팅에 의해 동정하였다. 사용된 다클론성 토끼 항체 항-Cys-src (pp60c-src)는 바이오몰 훼인케미칼리엔 (Biomol Feinchemikalien) (함부르크)로부터 구입되었다. 이러한 1차 항체를 염소로부터의 HRP-결합 2 차 항체 (뵈링거 만하임 (Boehringer Mannheim), FRG)를 사용하여 검출하였다. 웨스턴 블롯팅을 공지된 방법에 따라 수행하였다.
pp60src의 분열의 정량을 밀도계측에 의해 수행하였으며, 사용된 대조는 비활성화된 혈소판 (대조 1: 비 분열) 및 이온운반체 및 칼슘으로 처리된 혈소판 (대조 2: 100 % 분열에 상응)이다. ED50값은 색 반응의 강도가 50 % 감소된 억제제의 농도에 상응한다.
피질 뉴론에서의 글루타메이트-유발 세포 사멸
문헌 (Choi D.W., Maulucci-Gedde M.A. 및 Kriegstein A.R., "Glutamate neurotoxicity in cortical cell culture", J.Neurosci. 1989, 7, 357-368)에서와 같이 시험을 수행하였다.

국소적 외부 적용을 위하여, 예를 들면 분말, 연고 또는 스프레이에서, 활성 화합물은 통상의 농도로 함유될 수 있다. 일반적으로, 활성 화합물은 0.001 내지 1 중량%의 양, 바람직하게는 0.001 내지 0.1 중량%의 양으로 함유된다.
내부 투여의 경우에, 제제는 개별적인 투여량으로 투여된다. 체중 kg 당 개별적인 투여량에서 0.1 내지 100 mg이 제공된다. 제제는 질환의 특성 및 심각성에 의존하여 1 일 1 회 이상의 투여량으로 투여될 수 있다.

활성 화합물에 추가로 제제에 함유되는 물질 및 제약 제제의 제조에서 사용되는 물질은 각각의 활성 화합물과 상용성이고 독성학적으로 허용가능한 것이다. 제약 제제는 통상의 방식으로, 예를 들어 활성 화합물을 다른 통상의 부형제 및 희석제와 혼합함으로써 제조된다.

(S)-4(N-(1-나프틸메틸)카르바모일)-N-(3-페닐프로판-1-알-2-일)피리딘-2-카르복스아미드
a) 에틸 4-(N-(1-나프틸메틸)카르바모일)피리딘-2-카르복실레이트
4.9 g (25 mmol)의 2-에톡시카르보닐피리딘-3-카르복실산 (N. Finch et al., J. Med. a) 6-아미노니코틴산 히드로클로라이드

a) 에틸 2-메틸-1-(2-나프틸)메틸이미다졸-4-카르복실레이트
4.6 g (29.8 mmol)의 에틸 2-메틸이미다졸-4-카르복실레이트 및 6.6 g (29.8 mmol)의 2-브로모메틸나프탈렌을 방법 8a와 유사하게 반응시켜 5.7 g (65%)의 생성물을 얻었다.
b) 2-메틸-1-(2-나프틸)메틸이미다졸-4-카르복실산
5.5 g (18.7 mmol)의 중간체 화합물 9a를 방법 8b와 유사하게 가수분해하여 3.2 g (65%)의 생성물을 얻었다.
c) (S)-N-(1-카르바모일-1-히드록시-3-페닐프로판-2-일)-2-메틸-1-(2-나프틸메틸)이미다졸-5-카르복스아미드
1 g (3.8 mmol)의 중간체 화합물 9b 및 1.2 g (3.8 mmol)의 (2S),(3R,S)-3-아미노-2-히드록시-3-페닐부티르아미드 트리플루오로아세테이트를 방법 1c와 유사하게 반응시켜 0.65 g (39%)의 생성물을 얻었다.
d) (S)-N-(1-카르바모일-1-옥소-3-페닐프로판-2-일)-2-메틸-1-(2-나프틸메틸)이미다졸-5-카르복스아미드
0.6 g (1.4 mmol)의 중간체 생성물 9c를 방법 1d와 유사하게 산화하여 0.42 g (71%)의 생성물을 얻었다.

(S)-N-(1-카르바모일-1-옥소-3-페닐프로판-2-일)-1-(2-나프틸메틸)이미다졸-2-카르복스아미드
a) 부틸 1-(2-나프틸)메틸이미다졸-2-카르복실레이트
5.0 g (29.7 mmol)의 부틸 이미다졸-2-카르복실레이트 및 6.6 g (29.7 mmol)의 2-브로모메틸나프탈렌을 방법 8a와 유사하게 반응시켜 6.4 g (71%)의 생성물을 얻었다.
b) 1-(2-나프틸)메틸이미다졸-2-카르복실산
6.2 g (20.1 mmol)의 중간체 화합물 10a를 방법 8b와 유사하게 가수분해하여 4.2 g (83%)의 생성물을 얻었다.
c) (S)-N-(1-카르바모일-1-히드록시-3-페닐프로판-2-일)-1-(2-나프틸메틸)이미다졸-2-카르복스아미드
1.1 g (4.4 mmol)의 중간체 화합물 10b 및 1.3 g (4.4 mmol)의 (2S),(3R,S)-3-아미노-2-히드록시-3-페닐부티르아미드 트리플루오로아세테이트를 방법 1c와 유사하게 반응시켜 1.3 g (70%)의 생성물을 얻었다.
d) (S)-N-(1-카르바모일-1-옥소-3-페닐프로판-2-일)-1-(2-나프틸-메틸)이미다졸-2-카르복스아미드
1.0 g (2.3 mmol)의 중간체 화합물 10c를 방법 1d와 유사하게 산화하여 0.73 g (74%)의 생성물을 얻었다.

(S)-1-벤질-N-(1-카르바모일-1-옥소-3-페닐프로판-2-일)-이미다졸-2-카르복스아미드
a) 부틸 1-벤질이미다졸-2-카르복실레이트
5.4 g (32.1 mmol)의 부틸 이미다졸-2-카르복실레이트를 4.1 g (32.1 mmol)의 벤질 클로라이드와 방법 8a와 유사하게 반응시켜 7.3 g (78%)의 생성물을 얻었다.
b) 1-벤질이미다졸-2-카르복실산
7 g (27.1 mmol)의 중간체 화합물 11a를 수산화나트륨 용액을 사용하여 방법 8b와 유사하게 가수분해하여 3.7 g (68%)의 생성물을 얻었다.
c) (S)-1-벤질-(1-카르바모일-1-히드록시-3-페닐프로판-2-일)-이미다졸-2-카르복스아미드
1.0 g (5.1 mmol)의 중간체 화합물 11b 및 1.6 g (5.1 mmol)의 (2S),(3R,S)-3-아미노-2-히드록시-3-페닐부티르아미드 트리플루오로아세테이트를 방법 1c와 유사하게 반응시켜 1.1 g (58%)의 생성물을 얻었다.
d) (S)-1-벤질-N-(1-카르바모일-1-옥소-3-페닐프로판-2-일)-이미다졸-2-카르복스아미드
1.0 g (2.3 mmol)의 중간체 화합물 11c를 방법 1d와 유사하게 산화시켜 0.79 g (80%)의 생성물을 얻었다.

(S)-N-(1-카르바모일-1-옥소-3-페닐프로판-2-일)-1-(2-나프틸메틸)이미다졸-5-카르복스아미드
a) 에틸 1-(2-나프틸메틸)이미다졸-5-카르복실레이트
2.4 g (17.1 mmol)의 부틸 이미다졸-5-카르복실레이트를 4.1 g (32.1 mmol)의 벤질 클로라이드와 방법 8a와 유사하게 반응시켜 7.3 g (78%)의 생성물을 얻었다.
b) 1-(2-나프틸메틸)이미다졸-5-카르복실산
3 g (10.7 mmol)의 중간체 화합물 12a를 수산화나트륨 용액을 사용하여 8b의 방법과 유사하게 가수분해하여 1.9 g (73%)의 생성물을 얻었다.
c) (S)-N-(1-카르바모일-1-히드록시-3-페닐프로판-2-일)-1-(2-나프틸메틸)이미다졸-5-카르복스아미드
1.0 g (4.0 mmol)의 중간체 화합물 12b 및 1.2 g (4.0 mmol)의 (2S),(3R,S)-3-아미노-2-히드록시-3-페닐부티르아미드 트리플루오로아세테이트를 방법 1c와 유사하게 반응시켜 0.85 g (51%)의 생성물을 얻었다.
d) (S)-N-(1-카르바모일-1-옥소-3-페닐프로판-2-일)-1-(2-나프틸메틸)이미다졸-5-카르복스아미드
0.8 g (1.9 mmol)의 중간체 화합물 12c를 방법 1d와 유사하게 산화하여 0.41 g (52%)의 생성물을 얻었다.

실시예 13
(S)-2-(2-나프틸)에텐-1-일)-N-(3-페닐프로판-1-알-2-일)-피리딘-3-카르복스아미드
a) 에틸 2-(2-나프틸에텐-1-일)피리딘-3-카르복실레이트 히드로클로라이드
10 g (43.5 mmol)의 에틸 2-브로모피리딘-3-카르복실레이트, 8.7 g (56.5 mmol)의 2-비닐나프탈렌, 15 mL (0.11 mol)의 트리에틸아민, 0.36 g의 팔라듐 (II) 아세테이트 및 0.96 g의 트리-o-톨릴포스핀을 150 mL의 디메틸포름아미드에 용해하였다. 이후, 추가의 1 mL의 물을 첨가하고, 전체 혼합물을 3 시간 동안 환류하였다. 이후, 전체 혼합물을 에테르로 추출하였다. 유기상을 물로 추가로 세척하고, 건조하여 진공하에 농축하였다. 잔류물을 아세톤으로 용해하고, 디옥산으로 용해시킨 염화수소로 처리하였다. 이후, 생성물을 에테르를 첨가하여 침전시켰다. 8.7 g (67%)의 생성물을 얻었다.
b) 2-(2-나프틸에텐-1-일)피리딘-3-카르복실산
8.5 g (28 mmol)의 중간체 생성물 13a를 70 mL의 테트라히드로푸란에 용해하고, 140 mL의 2 M 수산화나트륨 용액으로 처리하였다. 전체 혼합물을 8 시간 동안 환류하였다. 이후, 배치를 빙수에 붓고, 아세트산으로 중화하였다. 서서히 결정화된 생성물을 감압하에 여거하고, 건조하였다. 5.6 g (73%)의 생성물을 얻었다.
c) (S)-2-(2-나프틸)에텐-1-일-N-(3-페닐프로판-1-올-2-일)피리딘-3-카르복스아미드
2 g (7.3 mmol)의 중간체 13b 및 1.1 g (7.3 mmol)의 (2S),(3R,S)-3-아미노-2-히드록시-3-페닐부티르아미드 트리플루오로아세테이트를 방법 1c와 유사하게 반응시켜 2.1 g (71%)의 생성물을 얻었다.
d) (S)-2-(2-나프틸)에텐-1-일)-N-(3-페닐프로판-1-알-2-일)피리딘-3-카르복스아미드
1.9 g (4.7 mmol)의 중간체 화합물 13c를 방법 1d와 유사하게 산화시켜 0.56 g (30%)의 생성물을 얻었다.
MS: m/e =406 (M+)
실시예 14
(S)-N-(3-페닐프로판-1-알-2-일)-2-(4-피리딜)에텐-1-일)-피리딘-3-카르복스아미드
a) 에틸 2-(4-피리딘)에텐-1-일피리딘-3-카르복실레이트
11.5 g (49.9 mmol)의 에틸 2-브로모피리딘-3-카르복실레이트 및 6.8 g (64.9 mmol)의 4-비닐피리딘을 방법 13a와 유사하게 반응시켜 7.0 g (49%)의 생성물을 얻었다.
b) 2-(4-피리딜)에텐-1-일피리딘-3-카르복실산
7.0 g (27.5 mmol)의 중간체 14a를 50 mL의 테트라히드로푸란에 용해하고, 100 mL의 2 M 수산화나트륨 용액으로 처리하였다. 전체 혼합물을 2 시간 동안 환류하였다. 유기 용매를 진공하에 제거하고, 얻은 수성상을 아세트산으로 산성화하였다. 수성상을 농축하고, 잔류물을 크로마토그래피 (용출액: 에틸아세테이트/메탄올/아세트산 = 50/50/1)하여 정제하였다. 5.5 g (89%)의 생성물을 얻었다.
c) (S)-N-(3-페닐프로판-1-올-2-일)-2-(4-피리딜)에텐-1-일피리딘-3-카르복스아미드
1.5 g (6.6 mmol)의 중간체 14b 및 1.0 g (6.6 mmol)의 (S)-2-아미노-3-페닐프로판올을 방법 1c와 유사하게 반응시켜 1.7 g (72%)의 생성물을 얻었다.
d) (S)-N-(3-페닐프로판-1-알-2-일)-2-(4-피리딜)에텐-1-일피리딘-3-카르복스아미드
1.5 g (4.2 mmol)의 중간체 화합물 14c를 방법 1d와 유사하게 산화시켜 0.71 g (48%)의 생성물을 얻었다.
MS : m/e = 357 (M+)
하기 실시예를 상기 실시예 및 방법과 유사하게 제조하였다.

Claims (19)
- 하기 화학식 I의 아미드 및 그의 호변이성질체 및 이성질체 형태, 거울상이성질체 및 부분입체이성질체 형태, 및 생리학적으로 허용가능한 염.<화학식 I>상기 식에서,R1은 3 개 이하의 라디칼 R5로 추가로 치환될 수 있는, 페닐, 나프틸, 퀴놀릴, 피리딜, 피리미딜, 피라질, 피리다질, 이미다졸릴, 티아졸, 퀴나질, 이소퀴놀릴, 퀴나질, 퀴녹살릴, 티에닐, 벤조티에닐, 벤조푸라닐, 푸라닐 및 인돌릴일 수 있으며,R2는 염소, 브롬, 불소, C1-C6-알킬, C1-C6-알케닐, C1-C6-알키닐, C1-C6-알킬페닐, C1-C6-알케닐페닐, C1-C6-알키닐페닐, 페닐, NHCO-C1-C4-알킬, NHSO2-C1-C4-알킬, -NHCO-페닐, -NHCO-나프틸, NO2, -O-C1-C4-알킬 및 NH2(여기서, 방향족 고리는 추가로 1 내지 2 개의 라디칼 R5를 가질 수 있으며, 2 개의 R2는 함께 -CH=CH-CH=CH- 사슬일 수 있고, 융합된 벤조 고리를 형성할 수 있으며, 여기에 1 개의 R5로 치환될 수 있음)이며,R3는 분지쇄 또는 비분지쇄 C1-C6-알킬이며, 추가로 S-CH3라디칼 또는 페닐, 시클로헥실, 시클로헵틸, 시클로펜틸, 인돌릴, 피리딜 또는 나프틸 고리 (여기에 2 개 이하의 라디칼 R5(여기서, R5는 수소, 분지쇄 또는 비분지쇄 C1-C4-알킬, -O-C1-C4-알킬, OH, Cl, F, Br, I, CF3, NO2, NH2, CN, COOH, COO-C1-C4-알킬, -NHCO-C1-C4-알킬, -NHCO-페닐, -NHSO2-C1-C4-알킬, -NHSO2-페닐, -SO2-C1-C4-알킬, -(CH2)n-NR12R13및 SO2-페닐임)로 치환될 수 있음)이,X는 결합, -(CH2)m-, -(CH2)m-O-(CH2)o-, -(CH2)o-S-(CH2)m-, -(CH2)o-SO-(CH2)m-, -(CH2)o-SO2-(CH2)m-, -CH=CH-, -C≡C-, -CO-CH=CH-, -(CH2)o-CO-(CH2)m-, -(CH2)m-NHCO-(CH2)o-, -(CH2)m-CONH-(CH2)o-, -(CH2)m-NHSO2-(CH2)o-, -NH-CO-CH=CH-, -(CH2)m-SO2NH-(CH2)m-, -CH=CH-CONH- 및이며, CH=CH 이중 결합의 경우에는 각각 E 또는 Z 형 중의 하나일 수 있으며,R1-X는 함께 또한및이며,Y는 피리딘, 피리미딘, 피라진, 이미다졸 및 티아졸과 같은 불포화 헤테로사이클 고리이며,R4는 수소, COOR6및 CO-Z (여기서, Z는 NR7R8임) 및,,이며,R6는 수소, 선형 또는 분지쇄 C1-C6-알킬 (여기서, 그자체가 1 또는 2 개의 라디칼 R9으로 임의로 치환될 수 있는 페닐 고리로 치환될 수 있음)이며,R7은 수소, 분지쇄 또는 비분지쇄 C1-C6-알킬이며,R8은 수소, 분지쇄 또는 비분지쇄 C1-C6-알킬 (여기서, 추가로 라디칼 R9를 가질 수 있는 페닐 고리 및,,,,,로 치환될 수 있음)이며,R9은 수소, 분지쇄 또는 비분지쇄 C1-C4-알킬, -O-C1-C4-알킬, OH, Cl, F, Br, I, CF3, NO2, NH2, CN, COOH, COO-C1-C4-알킬, -NHCO-C1-C4-알킬, -NHCO-페닐, -NHSO2-C1-C4-알킬, -NHSO2-페닐, -SO2-C1-C4-알킬 및 -SO2-페닐이며,R10은 수소, 분지쇄 또는 비분지쇄 C1-C6-알킬 (여기서, 그자체가 추가로 1 또는 2 개의 라디칼 R9로 치환될 수 있는 페닐 고리로 치환될 수 있음)이며,R11은 수소, 분지쇄 또는 비분지쇄 C1-C6-알킬 (여기서, 그자체가 임의로 1 또는 2 개의 라디칼 R9로 치환될 수 있는 페닐 고리로 치환될 수 있음)이며,n은 숫자 0, 1 또는 2이며,m 및 o는 서로 독립적으로 숫자 0, 1, 2, 3 또는 4이다.
- 제1항에 있어서,R3가 벤질, CH2CH2CH2CH3및 CH2CH2CH2CH2CH3이며,Y가 피리딘이고R4가 CO-NR7NR8이며,R7이 수소이고R8이 CH2CH2, CH2CH2CH2및 CH2CH2CH2CH2이고,R9이 수소이며,n이 0 및 1이고,남은 모든 변수들이 제1항과 동일한 의미를 갖는 화학식 I의 아미드.
- 제1항에 있어서,R3가 벤질, CH2CH2CH2CH3및 CH2CH2CH2CH2CH3이며,Y가 피리딘이고R4가 수소이며,R9이 수소이고n이 0 및 1이고,남은 모든 변수들이 제1항과 동일한 의미를 갖는 화학식 I의 아미드.
- 제1항에 있어서,R3가 벤질, CH2CH2CH2CH3및 CH2CH2CH2CH2CH3이며,Y가 이미다졸 및 티아졸이고R4가 CO-NR7NR8이며,R7이 수소이고R8이 CH2CH2, CH2CH2CH2및 CH2CH2CH2CH2이고,R9이 수소이며,n이 0 및 1이고,남은 모든 변수들이 제1항과 동일한 의미를 갖는 화학식 I의 아미드.
- 제1항에 있어서,R3가 벤질, CH2-피리딘, CH2CH2CH2CH3및 CH2CH2CH2CH2CH3이며,Y가 이미다졸 및 티아졸이고,R4가 수소이며,R9이 수소이고,n이 0 및 1이고,남은 모든 변수들이 제1항과 동일한 의미를 갖는 화학식 I의 아미드.
- 질환의 치료를 위한 제1항 내지 제5항 중 어느 한 항에 따른 화학식 I의 아미드의 용도.
- 시스테인 프로테아제의 억제제로서의 제1항 내지 제5항 중 어느 한 항에 따른 화학식 I의 아미드의 용도.
- 제6항에 있어서, 칼파인 및 카텝신, 특히 칼파인 I 및 II 및 카텝신 B 및 L과 같은 시스테인 프로테아제의 억제제로서의 용도.
- 칼파인 활성이 증가하는 질병의 치료용 약제의 제조를 위한, 제1항 내지 제5항 중 어느 한 항에 따른 화학식 I의 아미드의 용도.
- 신경변성 질병 및 신경 손상의 치료용 약제의 제조를 위한, 제1항 내지 제5항 중 어느 한 항에 따른 화학식 I의 아미드의 용도.

- 종양 및 그의 전이의 치료용 약제의 제조를 위한, 제1항 내지 제5항 중 어느 한 항에 따른 화학식 I의 아미드의 용도.
- 인터루킨-1 수준이 증가된 질병의 치료용 약제의 제조를 위한, 제1항 내지 제8항 중 어느 한 항에 따른 화학식 I의 아미드의 용도.
- 염증 및 류마티스성 질환의 치료를 위한, 제1항 내지 제5항 중 어느 한 항에 따른 화합물의 용도.
- 개별 투여량당 제1항 내지 제5항 중 어느 한 항에 따른 1종 이상의 화학식 I의 아미드와 함께 통상의 제약학적 보조제를 포함하는, 경구, 비경구 및 복강내 사용을 위한 제약 제제.
