Description
뉴클레오시드 운반 억제

화학식(I)의 신규한 화합물은 3-(아미노카보닐)-N-(2,6-디클로로-페닐)-4-[4-(4-플루오로페닐)-4-(3-피리디닐)-부틸]-1-피페라진아세트아미드, 및 N-(2-아세틸-6-클로로페닐)-4-[5,5-비스(4-플루오로페닐)-4-펜테닐]-3-[(메틸-아미노)카보닐]-1-피페라진아세트아미드이다.
화학식(I)의 화합물은 EP-0,068,544, EP-A-0,285,219, US-4,968,684, US-5,026,853 및 EP-A-0,455,789에 기술된대로 제조될 수 있다. 일반적으로, 이들은 화학식(II)의 중간체를 화학식(III)의 중간체로 N-알킬화시키거나, 화학식(IV)의 중간체를 화학식(V)의 중간체로 N-알킬화시켜 제조할 수 있다.

화학식(I)의 화합물의 통증-경감(relieving) 성질은 본 화합물의 급성 및 만성 통증-경감 효과를 예시하는 실시예 B.1에 기술된 "기니아-피그에 대한 포르말린 시험"에서 예시된다.

특히, 본 화합물은 신경병성 통증과 같은 만성 비-암 통증을 치료하는데에 유용하다. 신경병성 통증은 말초 및 중추 신경계의 손상으로 일어난다. 이는 종종 체성 감각(somatosensory) 결함과 관련되어 있고, 통증의 분포는 대부분 체성 감각의 기능장애의 지역과 관련되어 있다. 통증의 시작은 원인이 되는 사건후에 수개월 또는 수년까지 지연될 수 있다. 증상 및 신경학상의 결함에 있어서 상당한 가변성을 갖는 신경병성 통증의 수개 원인이 있다. 예로는 외상성 손상 압박(traumatic injury compression), 허혈, 독소, 영양 결핍, 감염 및 간 및 신장의 합병증에 기인한 말초 신경 손상이다.

화학식(I)의 화합물이 진통제와 배합하여 사용될 때, 진통제의 투여량은 단독으로 사용될 때의 투여량과 다를 수 있다. 따라서, 화학식(I)의 화합물이 진통제와 함께 사용될 때, 후자의 투여량은 진통제가 단독으로 사용될 때 이용된 투여량과 동일하거나, 보다 흔히는 그보다 낮을 수 있다. 적절한 투여량은 본 분야에 숙련된자에 의해서 쉽게 인지될 것이다.
화학식(I)의 화합물의 상기 용도로 비추어 볼때, 본 발명은 또한 통증을 겪는 온혈 동물을 치료하는 방법을 제공하고, 상기 방법은 통증을 완하시키는데에 유효한 화학식(I)의 화합물의 치료량을 경구 또는 전신 투여하는 것을 포함한다.

A. 최종 화합물의 제조
실시예 A.1
6.7 g의 1,1'-(4-요오도부틸리덴)비스[4-플루오로벤젠], 5.2 g의 3-[(메틸아미노)카보닐]-N-(2,4,6-트리메틸페닐)-1-피페라진아세트아미드, 2.3 g의 탄산 나트륨 및 38 ml의 DMF의 혼합물을 70℃에서 18시간 동안 교반하였다. 반응 혼합물을 냉각시키고 빙수에 부었다. 생성물을 DCM으로 추출하였다. 추출물을 물로 세척하고, 건조시키며, 여과하고, 증발시켰다. 잔류물을 용리제로서 트리클로로메탄 및 메탄올(부피로 95:5)의 혼합물을 사용하여 실리카겔상에서 칼럼 크로마토그래피에 의해 정제하였다. 순수 분획물을 모으고 용리제를 증발시켰다. 잔류물을 건조시켜, 4.23 g의 4-[4,4-비스(4-플루오로페닐)부틸]-3-[(메틸아미노)-카보닐]-N-(2,4,6-트리메틸페닐)-1-피페라진아세트아미드(화합물 1, mp.82.3℃)를 수득하였다.
실시예 A.2
5.8 g의 4-[4,4-비스(4-플루오로페닐)부틸]-N-메틸-2-피페라진-카복스아미드, 3.6 g의 2-클로로-N-(2,4,6-트리메틸페닐)아세트아미드, 2.12 g의 탄산 나트륨 및 150 ml MIK의 혼합물을 18시간 동안 교반하고 환류시켰다. 반응 혼합물을 냉각시키고 물로 세척했다. 유기 상을 건조시키고, 여과하며, 증발시켰다. 잔류물을 용리제로서 트리클로로메탄 및 메탄올(부피로 95:5)의 혼합물을 사용하여 실리카 겔상에서 칼럼-크로마토그래피에 의해 정제하였다. 순수 분획을 모으고, 용리제를 증발시켰다. 잔류물을 건조시켜, 4.34 g의 4-[4,4-비스(4-플루오로페닐)부틸]-2-[(메틸아미노)카보닐]-N-(2,4,6-트리메틸페닐)-1-피페라진아세트아미드(화합물 2, mp. 90.2℃)를 수득하였다.
실시예 A.3
1-[4-(4-플루오로페닐)-4-(3-피리디닐)부틸]-2-피페라진카복스아미드(6.75 g), 2-클로로-N-(2,6-디클로로페닐)아세트아미드(5.24 g), 탄산 나트륨(3.16 g) 및 DMF(143 ml)의 혼합물을 70℃에서 4 시간동안 교반하였다. 반응 혼합물을 증발시키고, 물을 잔류물에 첨가시켰다. 실시예 A.4
DMF(75 ml)중의 (±)-1-[5,5-비스(4-플루오로페닐)-4-펜테닐]-N-메틸-2-피페라진 카복스아미드(4 g), N-(2-아세틸-6-클로로페닐)-2-클로로아세트아미드(3.2 g) 및 트리에틸 아민(75 ml)의 혼합물을 70℃에서 3시간 동안 교반하였다. 용매를 증발시켰다. 잔류물을 실리카겔상에서 칼럼 크로마토그래피(용리제:CH2Cl2/CH3OH 97/3)에 의해 정제하였다. 원하는 분획을 모으고, 용매를 증발시켰다. 잔류물을 2-프로판올에 용해시키고, 에탄디오산 염(1:2)으로 전환시켰다. 염을 여과하고 건조시켜(진공; 70℃), 3.72 g의 (±)-N-(2-아세틸-6-클로로페닐)-4-[5,5-비스(4-플루오로페닐)-4-펜테닐]-3-[(메틸아미노)카보닐]-1-피페라진아세트아미드 에탄디오에이트(1:2)(화합물 18, mp 140℃)를 수득하였다.

B. 약리학적 실시예
실시예 B.1
화학식(I)의 화합물의 통증-완화 가능성을 기니아 피그상에서 "포르말린 시험"을 사용하여 분석하였다.
암컷 기니아 피그(280-320 g)를 밤새 굶기고, 체중을 측정하여 관찰 우리에 넣었다. 화학식(I) 화합물을 복강내 주사하고 1시간후, 5% 포르말린 용액(0.05 ml)을 기니아 피그의 오른쪽 뒷발에 주사하였다. 5분동안 주춤하고(flinching) 핥는 반응을 계수하였다. 다음 25분에 걸쳐 안반사, 및 근긴장(muscle tone)에 대한 효과를 기록하였다. 기니아 피그의 왼쪽 뒷발에 또한 포르말린 5% 용액 0.05 ml를 주사하고 동일하게 관찰하여 기록하였다. 그후에 동물을 희생시켰다.
통증-유도된 반응 수가 50% 이상 감소되는 화합물 1 내지 18의 측정된 최저 유효 투여량은 체중 1 kg당 0.63 내지 40 mg 범위이었다.
