Abstract
본 발명은 5-HT1A수용체에 대한 높은 친화도를 화학식 I의 페녹시에틸아민 유도체, 그의 제조 방법, 이를 함유하는 제약 조성물 및 특히 위산 분비 억제제 또는 항구토제로서의 그의 용도에 관한 것이다.

화학식 I에서, Ar은 1종 이상의 치환체로 치환된 페닐이고, R은 직쇄 또는 분지쇄 알킬, 알케닐 또는 알키닐기, 포화 또는 불포화 모노시클릭 또는 폴리시클릭 시클로알킬, 시클로알킬알킬 또는 알킬시클로알킬기; 피리딜 또는 이소퀴놀릴기; 1종 이상의 치환체에 의해 치환될 수 있는 페닐 중에서 선택되는 탄소 원자수 1 내지 10의 탄화수소기이다.
Description
신경전달자 세로토닌 (5-히드록시트립타민, 5-HT)은 많은 의학 문제의 병인학 및 치료에서 매우 중요하다 (R.A. Glennon, J. Med. Chem., 30, 1 [1987]). 많은 5-HT 수용체가 이미 확인되었고 이 수용체는 세로토닌 기능의 선택적 처리를 위한 관심의 대상이 되었다. 서브타입 5-HT1A는 상기 수용체에 대해 작용하는 화합물이 불안증, 우울증, 수면 장애 또는 심근 문제의 치료에 사용될 수 있다는 측면에서 특히 관심의 대상이 되고 있다 (Brain 5-HT1Areceptors: Behavioural and Neurochemical Pharmacology; Editors: C.T. Dourish, S. Ahlenius, P.H. Huston; Ellis Horwood, Ltd., Chichester [1987]). 또한, 5-HT1A중화제는 항구토제로서 (U. Wells, M. Ravenscroft, P. Bhandari, P.L.R. Andrews, Med. Sci. Symp., Ser. 5, 179 [1993]; F. Okada, Y. Torii, H. Saito, N. Matsuki, Ipn. J. Pharmacol., 64, 109 [1994]) 또는 항분비제로서 (J. S. Gidda, J. M. Schaus, EP 455,510 A2; D. C. Evans, J. S. Gidda, Gastroenterology, 104, A76 [1993]) 사용될 수 있다.
본 발명은 5-HT1A수용체에 대한 높은 친화도를 갖는 신규 페녹시에틸아민의 유도체, 그의 제조 방법, 이를 함유하는 제약 조성물 및 위산 분비 억제제 또는 항구토제로서의 용도에 관한 것이다.
또한, 본 발명의 목적은 하기 화학식 I의 화합물 및 그의 염이다.
상기 식에서,
Ar은 1종 이상의 치환체로 치환된 페닐이고,
R은 직쇄 또는 분지쇄 알킬, 알케닐 또는 알키닐기, 포화 또는 불포화 모노시클릭 또는 폴리시클릭 시클로알킬, 시클로알킬알킬 또는 알킬시클로알킬기; 피리딜 또는 이소퀴놀릴기; 1종 이상의 치환체에 의해 치환될 수 있는 페닐 중에서 선택되는 탄소 원자수 1 내지 10의 탄화수소기이다.
보다 구체적으로, 본 발명의 목적은 Ar로 나타낸 페닐기를 포함할 수 있는 치환체(들)이 할로겐, 저급 알킬, 저급 알콕시, 시아노, 니트로, 히드록시, C(O)NR1R2, C(O)NHOR3, NHC(O)R4, NHC(O)NHR5, CH2NHC(O)NHR6, CH2OR7, CH2NHC(O)R8, CO2R9, NHCO2R10, C(O)R11및 SR12기 (여기서, R1, R2, R3, R4, R5, R6, R7및 R8은 독립적으로 수소 원자 또는 저급 알킬이고, R9, R10, R11및 R12는 독립적으로 저급알킬임) 중에서 선택되고, R로 나타낼 수 있는 페닐기를 포함할 수 있는 치환체(들)이 저급 알킬, 저급 알콕시, 할로겐, 히드록시, 니트로, 아미노 또는 아실아미노기 중에서 선택되는 것을 특징으로 하는, 상기 정의된 화학식 I의 화합물이다.
상기 정의에서, 할로겐은 불소, 염소, 브롬 또는 요오드, 바람직하게는 불소 또는 요오드 원자를 나타낸다. 저급 알킬은 바람직하게는 직쇄 또는 분지쇄의 탄소 원자수 1 내지 6의 알킬기, 특히 탄소 원자수 1 내지 4의 알킬기, 예를 들면 메틸, 에틸, 프로필, 이소프로필, 부틸, 이소부틸, 2급 부틸 및 3급 부틸이다.
알케닐기로는 비닐, 알릴 및 부테닐기를 예로 들 수 있다. 알키닐기로는 에티닐 또는 프로파르길기를 들 수 있다.
아실아미노기로는 아세틸아미노 및 프로피오닐아미노기를 들 수 있다.
저급 알콕시기는 상기 나타낸 알킬기에 대응할 수 있다. 메톡시, 에톡시 또는 이소프로필옥시가 바람직하다.
포화 모노시클릭 시클로알킬기는 탄소 원자수 3 내지 7의 시클로알킬기, 예를 들면 시클로프로필, 시클로부틸, 시클로펜틸, 시클로헥실 또는 시클로헵틸기 중에서 선택할 수 있다.
일 또는 다포화 시클로알킬기는 시클로부텐, 시클로펜텐, 시클로헥센, 시클로펜탄디엔 또는 시클로헥사디엔기 중에서 선택할 수 있다.
폴리시클릭 시클로알킬기의 예는 비시클로-[2,2,1]-헵틸 또는 아다만틸이다.
화학식 I의 화합물은 산, 특히 약리학상 허용되는 산과 부가염을 형성할 수 있다.
염의 예는 하기 실시예에서 기술된다.
본 발명의 특별한 목적은 Ar이 히드록시, 메톡시, -C(O)NHMe, -NHC(O)Me, -NHCO2R'10(여기서, R'10은 메틸 또는 에틸기임), -NHCONH2, -C(O)NHOH 중에서 선택되는 치환체에 의해 치환된 페닐기이고, R은 시클로펜틸, 시클로헥실, 시클로헵틸, 시클로펜틸메틸, 아다만틸, t-부틸, 네오펜틸, 페닐 또는 플로오로페닐기임을 특징으로 하는, 상기 화학식 I의 화합물이다.
Ar을 나타낼 수 있는 페닐기의 치환체는 위치 2 또는 3에 존재하는 것이 바람직하다.
보다 구체적으로는, 본 발명의 목적은 하기 실시예에서 설명되는 화합물, 특히 하기 화합물 및 이들 화합물의 유기산 또는 무기산과의 염이다.

또한, 본 발명의 목적은
A) 하기 화학식 II의 화합물을 하기 화학식 III의 화합물과 반응시켜 하기 화학식 IV의 화합물을 생성시킨 후, 화학식 IV의 화합물을 프탈이미드기 제거 반응시켜 하기 화학식 V의 화합물을 생성시키고, 화학식 V의 화합물을 RCO2H 산에서 유도된 아실화제로 처리하여 하기 화학식 VI의 화합물을 제조하거나, 또는
B) 하기 화학식 VII의 화합물을 화학식의 N-벤질에탄올아민과 반응시켜 하기 화학식 VIII의 화합물을 제조한 후, 화학식 VIII의 화합물을 하기 화학식 IX의 화합물로 전환시키고, 화학식 IX의 화합물을 화학식 ArOH의 페놀 유도체와 반응시켜 상기 정의된 화학식 VI의 화합물로 전환시키고,
이와 같이 제조된 화학식 VI의 화합물로부터 벤질기를 분절시킴으로써 화학식 I의 화합물로 전환시키고, 필요한 경우 화학식 I의 화합물을 대응하는 산의 작용에 의해 산 부가염으로 전환시키는 것으로 이루어지는,
화학식 I의 화합물의 제조 방법이다.
상기 식에서,
Ar 및 R은 상기한 의미를 갖고,
X 및 Y는 할로겐 또는 슈도-할로겐이다.
상기 반응식에서, Ar 및 R은 상기 정의한 바와 같고, X는 클로로, 브로모, 요오도, 메탄술포닐옥시, 벤젠술포닐옥시 또는 p-톨루엔술포닐옥시, 달리 표현하면 할로겐 또는 슈도-할로겐기와 같은 출발기이다.
화학식 II의 화합물과 화학식 III의 화합물의 반응에 의한 화학식 IV의 화합물의 제조는 산 수용체, 예를 들면 탄산칼륨 또는 탄산나트륨의 존재 하에 극성 용매, 예를 들면 아세토니트릴 또는 디메틸포름아미드 중에서 가열함으로써 용이하게 실시할 수 있다. 화학식 IV의 프탈이미드는 통상의 방법에 따라, 예를 들면 바람직하게는 에탄올과 같은 용매 중에서 히드라진 히드레이트와 함께 가열하여 화학식 V의 1급 아민으로 전환시킬 수 있다. 이와같이 제조된 화학식 V의 1급 아민은 적합한 산 염화물 또는 산 수소화물 또는 펩티드 커플링 반응에 일반적으로 사용되는 것과 같은 다른 활성화된 산 유도체일 수 있는 산 RCO2H의 유도체와 반응시켜 화학식 VI의 아미드로 전환시킬 수 있다. 반응은 불활성 용매, 예를 들면 에테르, 테트라히드로푸란 또는 디클로로메탄 중에서 및 일반적으로 산 수용체, 예를 들면 3급 아민, 예를 들면 트리에틸아민 또는 디-이소프로필에틸아민의 존재 하에 실시된다.
상기 반응식에서, Ar 및 R은 상기 정의한 바와 같고, Y는 X와 동일하거나 상이하고 클로로, 브로모, 요오도, 메탄술포닐옥시, 벤젠술포닐옥시 또는 p-톨루엔술포닐옥시, 달리 표현하면 할로겐 또는 슈도-할로겐기와 같은 출발기이다.
화학식 VIII의 화합물은 화학식 VII의 화합물을 알콜과 같은 극성 용매 중에서 산 수용체, 예를 들면 3급 아민 또는 무기 염기, 예를 들면 탄산칼륨 또는 탄산나트륨의 존재 하에 N-벤질에탄올아민과 함께 가열함으로써 제조할 수 있다. 별법으로, 화학식 VIII의 화합물은 화학식 VII의 화합물을 용매 부재 하에 바람직하게는 질소 분위기 중에서 과량의 N-벤질에탄올아민과 함께 60 내지 90℃로 가열하여 용이하게 제조할 수 있다. 이와같이 제조된 화학식 VIII의 화합물은 예를 들면 트리에틸아민 또는 디-이소프로필에틸아민과 같은 유기 염기의 존재 하에 디클로로메탄과 같은 불활성 용매의 존재 하에 메탄술포닐의 염화물과 반응시켜 화학식 IX (Y=Cl)의 염화물로 전환시킬 수 있다. 화학식 VI의 화합물은 화학식 IX의 화합물을 수산화나트륨, 수산화칼륨 또는 수소화나트륨과 같은 염기를 사용하여 적합한 ArOH 페놀로부터 생성된 페녹사이드 음이온과 반응시켜 제조할 수 있다. 이 반응은 비양성자성 용매 중에서, 바람직하게는 이극성 비양성자성 용매 중에서, 예를 들면 디메틸포름아미드 중에서 실시된다.
화학식 I의 화합물은 화학식 VI의 화합물을 공지의 탈벤질화 방법, 예를 들면 촉매에 의한 수소화 또는 비닐 클로로포르메이트 또는 α-클로로에틸 클로로포르메이트와 같은 클로로포르메이트와 반응시킨 후 가수분해 또는 가메탄분해 (methanolysis)에 의해 탈보호시켜 제조한다. 또한, 문헌 (Protective Groups in Organic Synthesis, T.W. Green, P.G.M. Wuts; 2nd edition, J. Wiley and Sons, Inc, pp. 364-66 [1991])에 기재된 바와 같은 다른 탈벤질화 방법은 화학식 VI의 화합물의 아릴 코어(들)와의 치환에 적합한 정도로 사용될 수 있다.
또한, 화학식 I의 화합물의 염화는 하기 실시예에서 나타낸 통상의 방법에 따라 실시된다.
본 발명의 화합물은 유리한 약리학적 특성을 갖는다. 본 발명자들은 본 발명의 화합물이 5-HT1A수용체에 대한 높은 친화성을 갖는다는 것을 발견하였다.
따라서, 본 발명의 화합물은 다양한 방법으로 치료용으로 사용할 수 있다.
본 발명의 화합물은 위산 분비 및 예를 들면 시스-플라티넘에 의한 구토를 억제할 수 있다. 따라서, 본 발명의 화합물은 항구토제로서 또는 위산 분비의 저하가 필요하거나 바람직한 질병, 예를 들면 위궤양 또는 십이지장 궤양 또는 위염, 위식도 역류, 위성 소화불량, 졸링거-엘리슨 (Zollinger-Ellison) 신드롬 또는 구토증의 치료에 사용될 수 있다.
또한, 본 발명의 화합물은 위 공복 및 장 운동성에 대해 영향을 준다. 예를 들면, 이 화합물은 변비, 수술후 이완 또는 경증 위아토니에 대해 작용하는데 사용할 수 있다.
또한, 본 발명의 화합물은 특정 신경계 질환, 예를 들면 불안증, 우울증, 불면증과 같은 수면 장애, 특정 약물 의존증, 알쯔하이머 질병, 식욕 감퇴와 같은 섭생 질환 또는 현기증에 대하여 사용할 수 있다. 또한, 본 발명의 화합물은 심혈관계 질병, 특히 고혈압의 치료에 사용할 수 있다.
본 발명의 화합물의 약리학적 특성은 하기 실시예에서 입증된다.
상기 약리학적 특성은 화학식 I의 화합물을 제약학적 용도에 적합하게 만든다. 또한, 본 발명은 약물로서의 상기 정의된 화학식 I의 화합물 및 상기 화학식 I의 화합물의 유기산 또는 무기산과의 제약상 허용되는 산 부가염 및 활성 성분으로서 상기 정의된 약물을 1종 이상 포함하는 제약 조성물을 목적으로 한다.
또한, 본 발명은 본 발명의 화합물 또는 이의 제약상 허용되는 산 부가염을 제약상 허용되는 부형제와 함께 포함하는 제약 조성물에 관한 것이다. 제약 조성물은 고형, 예를 들면 분말제, 과립제, 정제, 겔제 또는 좌제의 형태일 수 있다. 적합한 고상 부형제는 예를 들면 인산칼슘, 스테아르산 마그네슘, 탈크, 당, 락토스, 덱스트린, 전분, 젤라틴, 셀룰로스, 메틸 셀룰로스, 소듐 카르복시메틸 셀룰로스, 폴리비닐 피롤리딘 및 왁스일 수 있다.
또한, 본 발명의 화합물을 포함하는 제약 조성물은 액제, 에멀젼제, 현탁액제 또는 시럽제와 같은 액체 형태일 수 있다. 적합한 액체 부형제는 예를 들면 물, 글리세롤 또는 글리콜과 같은 유기 용매 및 오일 또는 제약상 허용되는 지질에 첨가된 이들의 물 중의 다양한 비율의 혼합물일 수 있다. 멸균 액체 화합물은 근내, 복강내 또는 피하 주사에 사용할 수 있고, 멸균 화합물은 또한 정맥내 투여될 수도 있다.
또한, 본 발명의 목적은 항구토제, 위산 분비 저하용 약물, 위 공복 촉진용 약물, 장내 수송 촉진용 약물, 불안증, 우울증 또는 수면 장애 치료용 약물 및 심혈관 질병 치료용 약물의 제조를 위한 상기 정의된 화학식 I의 화합물의 용도이다.
또한, 본 발명의 목적은 신규 산업적 생산물, 특히 화학식 I의 화합물 제조를 위한 신규 산업적 생산물인 상기 화학식 IV, V, VI, VIII 및 IX의 화합물을 제공하는 것이다.
본 발명의 출발 물질, 특히 화학식 II, III 및 VII의 화합물은 공지 화합물이거나 공지 화합물로부터 제조할 수 있다. 예를 들면, N-벤질에탄올아민은 ACROS사에서 시판하는 화합물이다.
화학식 II의 화합물은 대응하는 페녹시 에틸 아민으로부터 전통적인 방법에 의해, 예를 들면 벤즈아미드의 매개 및 리튬 알루미늄 히드라이드에 의한 환원에 의해 또는 이와 균등한 방법에 의해 제조할 수 있다. 별법으로, 통상의 방법에 따라 환원 아민화를 사용할 수 있다.
페녹시에틸에틸아민은 통상의 방법에 따라, 예를 들면 염기성 매질 중에서 클로르아세토니트릴과 페놀의 반응, 이어서 문헌 [Chim. Ther. 8(3) 259-270 (1973)]의 방법에 따라 리튬 알루미늄 히드라이드에 의한 니트릴의 환원에 의해 제조할 수 있다.
화학식 III의 화합물은 시판되고 있거나 당업계의 숙련가에게 공지된 방법에 의해 제조할 수 있다. X가 브롬 원자인 화학식 III의 화합물은 ACROS사에서 시판하고 있다.
X가 또다른 할로겐 원자, 예를 들면 염소 또는 요오드 또는 슈도-할로겐, 예를 들면 메실, 토실 또는 페닐술포닐기인 화학식 III의 화합물은 메이브리지 (Maybridge)사에서 시판하는 대응하는 알콜로부터 통상의 방법에 의해 제조할 수 있다.
화학식 VII의 화합물은 알콜로부터 메실레이트를 형성하는 공지의 방법, 예를 들면 문헌 [J. Org. Chem. 35(9), 3195-6 (1970)]의 방법에 따라 ω-히드록시부틸 아미드로부터 제조할 수 있다.
하기 실시예는 상기 방법을 예시하고자 하는 것으로서 어떠한 경우에도 본 발명의 범위를 제한하는 것으로 생각하지 않아야 한다.

아세토니트릴 200 ml 중의 N-(4-브로모부틸)프탈이미드 9.87 g (0.035 몰)의 용액을 탄산칼륨 9.1 g (0.066 몰)을 포함하는 아세토니트릴 170 ml 중의 N-벤질[2-(2-메톡시페녹시)에틸]아민 8.5 g (0.033 몰)의 고온 용액 (60℃)에 첨가하였다. 혼합물을 교반하고 6시간 동안 환류로 가열한 후 정치시켜 실온으로 냉각시키고 여과한 후에 용매를 감압하에 증발시켰다. 잔사를 디클로메탄 500 ml 중에 용해시키고 각각 물 50 ml로 3회 세척하여 황산마그네슘 상에서 건조시켰다. 용매를 여과 및 증발시켜 오일을 수득하고 이를 용출제로서 에틸 아세테이트/헵탄 (1/4, 3/7 순으로)을 사용하여 실리카겔 상에서 플래시 크로마토그래피에 의해 정제하여 화학식 IV의 화합물 7.2 g (48%)을 오일의 형태로 수득하였다.

N-벤질-N-[2-(2-메톡시페녹시)에틸]-1,4-부탄디아민
(V, Ar = 2-메톡시페닐)
에탄올 20 ml 중의 히드라진 수화물 0.83 ml (0.017 몰)의 용액을 에탄올 70 ml 중의 N-[4-{벤질[2-(2-메톡시페녹시)에틸]아미노}부틸]프탈이미드 7.2 g (0.016 몰)의 용액에 첨가하고, 용액을 교반하여 3시간 동안 환류로 가열하였다. 실온으로 냉각시키고 용매를 감압하에 증발시킨 후 잔류 오일을 1N 염산 40 ml로 처리하였다. 형성된 침전물을 여과 제거하고 물로 세척한 후 여액 및 세척액을 탄산칼륨 16 g으로 염기성으로 만들었다. 분리된 오일을 디클로로메탄 (3 x 80 ml)을 사용하여 추출하고 추출물을 황산마그네슘 상에서 건조시켰다. 용매를 감압 하에 여과하고 증발시켜 이론상의 화합물 4.4 g (86%)을 오일의 형태로 수득하였다. 에테르 중의 염화수소 기체의 포화 용액을 사용하여 상기 유리 염기에 대해 에테르화 용액 처리하고 에탄올로부터 재결정화한 후에 N-벤질-N-[2-(2-메톡시페녹시)에틸]-1,4-부탄디아민을 디히드로클로레이트화염 (mp 200-202℃)으로서 수득할 수 있었다.

N-[4-{벤질[2-(2-메톡시페녹시)에틸]아미노}부틸]벤즈아미드
(VI, Ar = 2-메톡시페닐, R = 페닐)
테트라히드로푸란 25 ml 중의 벤조일 클로라이드 0.9 ml (0.0076 몰)의 용액을 교반하고 냉각시키면서 (5-10℃) 테트라히드로푸란 25 ml 중의 트리에틸아민 1.05 ml (0.0076 몰)의 용액에 첨가하였다. 10℃에서 30분, 이어서 실온에서 1.5시간 정치시킨 후에 용매를 감압 하에 증발시켰다. 잔사를 디클로로메탄 중에 용해시키고, 용액을 물로 세척하고 황산 마그네슘 상에서 건조시켰다. 감압 하에 용매를 여과하고 증발시켜 오일을 수득하고, 용출제로서 에틸 아세테이트/헥산 (1/1, 이어서 3/2)을 사용하여 실리카겔 상에서 플래시 크로마토그래피에 의해 정제하여 이론상 화합물 2 g (61%)을 오일의 형태로 수득하였다.

탄소 상의 팔라듐(10%)으로 이루어지는 촉매 0.8 g을 메탄올 30 ml 중의 N-[4-{벤질[2-(2-메톡시페녹시)에틸]아미노}부틸]벤즈아미드 2 g (0.0046 몰)의 용액에 첨가하고 혼합물을 실온에서 1.5 psi에서 수소화시켰다. 수소 흡수가 종결된 후에 (약 3시간) 촉매를 여과 제거하고 용매를 감압에서 증발시켜 화합물 I 1.2 g (76%)을 오일의 형태로 수득하였다. 에테르 중의 기상 염산의 포화 용액을 사용하여 상기 유리 염기에 대해 에테르화 용액을 처리하여 화합물 번호 1의 디히드로클로레이트 1.0 g (59%)을 백색 결정 형태로 수득하였다 (mp 118-120℃).
실시예 2

1-아다만틸카르보닐의 클로라이드 2.14 g (0.011 몰) 및 테트라히드로푸란 40 ml 중의 트리에틸아민 1.54 ml (0.011 몰)을 실시예 1의 방법에 따라 제조된 N-벤질-N-[2-(2-메톡시페녹시)에틸]-1,4-부탄디아민 3.28 g (0.01 몰)의 냉각된 용액 (5℃)에 소량씩 첨가하였다. 30분 후에 반응 혼합물을 실온으로 재가열하고 1시간 후에 용매를 감압하에 증발시켰다. 잔사를 디클로메탄 중에 용해시키고 물로 세척하여 황산마그네슘 상에서 건조시켰다. 용매를 여과 및 증발시켜 오일을 수득하고 용출제로서 에틸 아세테이트/헥산 (1/2, 3/4 순으로)을 사용하여 실리카겔 상에서 플래시 크로마토그래피에 의해 정제하여 예상 화합물 3 g (61%)을 오일의 형태로 수득하였다.

탄소 상의 팔라듐 (10%)으로 구성되는 촉매 1.5 g 을 메탄올 30 ml 중의 N-[4-{벤질[2-(2-메톡시페녹시)에틸]아미노}부틸]-1-아다만타미드 2.85 g (0.058 몰)의 용액에 첨가하고 혼합물을 실온에서 1.5 psi에서 수소화시켰다. 수소 흡수가 종결된 후에 (약 3시간) 촉매를 여과 제거하고 용매를 감압에서 증발시켜 화합물 I 2 g (87%)을 오일의 형태로 수득하였다. 상기 유리 염기 0.6 g의 용액을 아세톤 중의 푸마르산 0.17 g의 용액으로 처리하여 화합물 번호 2의 푸마레이트 0.5 g (59%)을 백색 결정 형태로 수득하였다 (mp 122-124℃).
실시예 3
N-[4-{2-(2-니트로페녹시)에틸}아미노부틸]벤즈아미드

제1 단계
N-[4-{벤질(2-히드록시에틸)아미노}부틸]벤즈아미드
(VIII, R = 페닐)
N-벤질아미노 에탄올 60.5 g (56.8 ml, 0.4 몰) 및 N-[4-(메탄술포닐옥시)부틸]벤즈아미드 51.5 g (0.19 몰)의 혼합물을 질소 분위기 중에서 75-80℃에서 2시간 동안 가열하였다. 반응 혼합물을 냉각시키고 디클로메탄 400 ml 중에 용해시키고 용액을 각각 물 100 ml로 3회 세척한 후 황산마그네슘 상에서 건조시켰다. 용액을 여과하고 용매를 감압에서 제거하여 오일을 수득한 후 디-이소프로필 에테르를 사용하여 결정화시켜 화합식 VIII의 화합물 51.5 g (83%)을 백색 결정의 형태로 수득하였다 (mp 61-62℃).
제2 단계
N-[4-{벤질(2-클로로에틸)아미노}부틸]벤즈아미드
(IX, R = 페닐, Y = Cl)
메탄술포닐 클로라이드 14.8 g (10 ml, 0.13 몰)을 교반하면서 디클로로메탄 300 ml 중의 N-[4-{벤질(2-히드록시에틸)아미노}부틸]벤즈아미드 35 g (0.11 몰)의 냉각된 용액 (5℃)에 적가하였다. 용액을 실온으로 만든 후 1.5시간 후에 트리에틸아민 13 g (18 ml, 0.13 몰)을 첨가하고 1.5 시간 동안 계속 교반하였다. 반응 혼합물을 빙수 150 ml, 염으로 포화된 물 100 ml (2회)로 세척하고 황산 마그네 상에서 건조시켰다. 용매를 감압 하에 여과하고 증발시켜 예상 화합물 36.6 g (99%)을 담갈색 오일의 형태로 수득하고 4℃에서 유지시켜 결정화시켰다.

2-니트로페놀 0.56 g (0.004 몰)을 질소 분위기 중에서 무수 디메틸포름아미드 10 ml 중의 수소화나트륨 (오일 중의 60% 분산액 160 mg, 0.004 몰)의 교반된 현탁액에 소량씩 첨가하였다. 수소 방출이 종결되었을 때, 교반하면서 무수 디메틸포름아미드 5 ml 중의 N-[4-{벤질(2-클로로에틸)아미노}부틸]벤즈아미드 1.6 g (0.0046 몰)의 용액을 적가하였다. 반응 혼합물을 1.5시간 동안 65-70℃로 가열한 후 다시 실온으로 냉각시키고 분쇄된 얼음 상에 부었다. 혼합무을 에테르 50 ml로 추출하고 유기상을 분리하여 물 20 ml로 3회 세척한 후 황산 마그네슘 상에서 건조시켜 오일을 수득하고, 용출제로서 에틸 아세테이트/헵탄 (3/1)의 혼합물을 사용하여 실리카겔 상에서 플래시 크로마토그래피에 의해 정제하였다. 예상 화합물 0.94 g (52%)을 오일의 형태로 수득하였다.

무수 디클로로에탄 1.8 ml 중의 α-클로로에틸클로로포르메이트 0.28 g (0.002 몰)의 용액을 질소 분위기 중에서 빙조 중에서 냉각시키면서 무수 디클로로에탄 9 ml 중의 N-[4-{벤질[2-(2-니트로페녹시)에틸]아미노}부틸]벤즈아미드 0.82 g (0.18 몰)의 용액에 적가하였다. 2시간 후에 용매를 감압에서 증발시키고 잔사를 메탄올 10 ml 중에 용해시키고 환류로 1시간 동안 가열하였다. 용매를 감압 농축시키고 에틸아세테이트 10 ml을 첨가하였다. 형성된 침전물을 여과에 의해 모으고, 에틸아세테이트 및 디에틸에테르로 세척한 후 건조시켜 화합물 I 0.55 g (76%)을 히드로클로레이트의 형태로 수득하였다 (mp 159.5-160℃).
실시예 4
N-[4-{2-(2-메틸아미노카르보닐페녹시)에틸]아미노}부틸]벤즈아미드

제1 단계
N-[4-{벤질[2-(2-메틸아미노카르보닐페녹시)에틸]아미노}부틸]벤즈아미드
(VI, Ar = 2-MeNHC(O)C6H4, R = 페닐)
실시예 3의 방법에 따라 제조된 N-[4-{벤질(2-클로로에틸)아미노}부틸]벤즈아미드 8.1 g (0.023 몰)의 용액을 디메틸포름아미드 50 ml 중의 2-메틸아미노카르보닐페놀 3.0 g (0.02 몰), 수산화나트륨 0.8 g (0.02 몰) 및 물 4 ml로부터 반응계 내에서 제조된 소듐 2-메틸아미노카르보닐-페녹사이드의 교반된 용액에 첨가하였다. 혼합물을 4시간 동안 85℃로 가열한 후 실온으로 냉각시키고 얼음 및 물의 혼합물에 부었다. 혼합물을 에틸아세테이트 250 ml로 추출하고 유기상을 2% 수산화나트륨 수용액 100 ml, 염수 50 ml (3회)로 세척한 후 황산마그네슘 상에서 건조시켰다. 용매를 감압에서 여과 및 증발시켜 황색 오일을 수득하고 용출제로서 에틸 아세테이트를 사용하여 실리카겔 상에서 플래시 크로마토그래피에 의해 정제하여 화합물 VI 5.2 g (57%)을 오일의 형태로 수득하였다.

탄소 상의 팔라듐 (10%)으로 구성되는 촉매를 메탄올 200 ml 중의 N-[4-{벤질[2-(2-메틸아미노카르보닐페녹시)에틸]아미노}부틸]벤즈아미드 6.6 g (0.015 몰)의 용액에 첨가하고 대기압 및 실온에서 수소화시켰다. 수소 흡수가 종결된 후에 (약 3시간) 촉매를 여과 제거하고 여액을 감압에서 증발시켰다. 잔사를 아세톤 15 ml 중에 용해시키고 아세톤 100 ml 중의 푸마르산 용액 1.04 g (0.009 몰)로 처리하였다. 침전에 의해 분리한 오일상 고형물을 이소프로판올 50 ml을 첨가하여 결정화시켰다. 백색 결정 생성물을 여과에 의해 수거하여 아세톤으로 세척한 후 건조시켜서 화합물 번호 18을 헤미푸마레이트의 형태로 2.7 g (38%) 수득하였다 (mp 159-161℃).
상기 방법을 통하여 본 발명의 화합물을 유리 염기 또는 산 부가염의 형태로 수득할 수 있다. 본 발명 화합물이 산 부가염의 형태로 수득되는 경우에 유리 염기는 산 부가염 용액을 염기화시켜 수득할 수 있다. 그 반대로, 상기 방법의 생성물이 유리 염기인 경우에 산 부가염, 특히 제약상 허용되는 산 부가염은 유리 염기를 적합한 유기 용매 중에 용해시키고 이 용액을 유리 염기로부터 산 부가염을 제조하는 통상의 방법에 따라 산으로 처리하여 수득할 수 있다.
산 부가염의 예는 무기산, 예를 들면 황산, 염산, 브롬화수소산 또는 인산, 또는 유기산, 예를 들면 타르타르산, 푸마르산, 말레산, 시트르산, 카프릴산, 옥살산, 벤조산, 메탄술폰산, p-톨루엔술폰산, 벤젠술폰산, 숙신산 또는 아세트산으로부터 유도된 염이다.
비대칭 중심을 갖는 본 발명의 화합물의 경우에, 라세미체 혼합물 및 개개의 광학 활성 이소머도 본 발명의 범위에 포함되는 것으로 간주된다.
하기 표 1은 상기 방법에 따라 제조된 주요 화합물을 나타낸 것으로 본 발명의 범위를 제한함이 없이 본 발명을 예시하고자 하는 것이다. 화합물 번호 1, 2, 8 및 18은 상기 실시예 1, 2, 3 및 4의 생성물에 대응한다. 다른 생성물은 동일한 방법을 사용하여 제조할 수 있다.

본 발명의 화합물의 약리학적 연구
1) 본 발명 화합물의 5-HT1A수용체에 대한 친화도
5-HT1A세로토닌성 수용체에 대한 본 발명 화합물의 친화도는 문헌 [Perutka et al., J. Neurochem., 47, 529 (1986)]의 방법에 따라 쥐의 뇌 피질과 연결된 [3H]8-히드록시-2(디-n-프로필아미노)테트라린 ([3H]8-OH-DPAT)의 억제를 측정하여 결정하였다.
스프라그 돌리 (Sprague Dawley) 쥐 수컷의 뇌 피질을 pH 7.4의 트리스-HCl 중에 균질화시키고 4℃ 10분 동안 40,000 g에서 원심분리하였다. 펠렛을 동일 완충액 중에 넣어 현탁시키고 37℃에서 10분 동안 배양한 후 균질액을 다시 4℃에서 10분 동안 400,000 g에서 원심분리하였다.
농도 100 pM 내지 100 μM의 비표지 경쟁자를 사용하여 [3H]8-OH-DAPT의 경쟁 억제 시험을 3회 실시하였다.
쥐 뇌 피질 멤브레인 (습윤성, 10 mg/ml)을 4 mM CaCl2, 10 μM 파르길린 및 0.1% 아스코르브산을 포함하는 pH 7.4의 50 mM 트리스-HCl 중에서 [3H]8-OH-DAPT 1 nm과 함께 20℃에서 30분 동안 배양하였다.
결합된 [3H]8-OH-DAPT을 Brandel 세포 회수기를 사용하여 Whatman GF/B 필터글라스로 즉시 여과하였다. 필터를 0 내지 4℃의 동일한 완충액으로 3회 세척하고 그 방사성을 액체 신틸레이션 분광계로 측정하였다.
특이 결합은 전체 결합으로부터 1 μM 8-OH-DAPT의 존재 하에 제시된 결합을 빼어서 계산하였다. 결합 특성은 Ligand 프로그램 (Munston and Rodbard, Anal. Biochem. 107, 220 [1980])을 사용하여 컴퓨터에 의한 비선형 회귀의 상호 분석에 의해 분석하였다.

2) 위산 분비 억제 활성
본 발명 화합물의 위산 분비 억제 활성을 문헌 [Shay et al., H. Shay, D.C. Sun, M. Gruenstein, Gastroenterology, 26, 906 [1954])의 방법에 따라 시험하였다. 체중 약 200 g의 스프라그 돌리 쥐 수컷에게 물을 자유롭게 섭취하게 하고 실험 전에 24시간 동안 단식시켰다. 에테르 마취 하에 유문을 결찰시키고 시험 약물을 복강 경로 (i.p.) 또는 십이지장 경로 (i.d.)에 의해 동시에 투여하였다. 복강을 다시 봉합하고 4시간 후에 동물을 희생시켰다. 위 분비액을 회수하여 1,500 g에서 15분 동안 원심분리하고, 상등액의 용량을 측정하고 0.05 N 수산화나트륨을 자동 적정하여 활성을 측정하였다. 대조군을 각 실험에 포함시켰다.
산 분비 (염산 용량 농도 X) 50% 억제에 필요한 투여량 (ED50)을 문헌 [J.T. Litchfield, F.A. Wilcoxon, J. Pharmacol, 96, 99 [1949])에 기재된 방법을 사용하여 계산하였다. 본 발명의 대표적 화합물에 대해서 얻어진 결과를 표 3에 나타내고 임상에 사용되는 대조 화합물인 피렌제핀 (항콜린성), 라니티딘 (히스타민성 H2수용체의 길항제) 및 오메프라졸 (양성자 펌프 억제제)와 비교하였다.

3) 본 발명 화합물의 항구토 활성 - 시스-플라티넘에 의해 유도된 구토의 억제
시스-플라티넘에 의해 유도된 구토의 억제는 문헌 [J.M. Barnes, N.M. Barnes, B. Costall, R.J. Naylor, F.D. Tattersall, NEUROPHARMACOLOGY, 27 [8] 783 [1988])에 기재된 프로토콜의 변형을 사용하여 흰족제비에서 평가하였다. 체중 0.5 kg 내지 1.5 kg의 알비노 흰족제비 또는 유러피안 긴털 족제비 암수 모두를 펜토바르비톤 30 내지 40 mg/kg을 복강내 주사하여 마취시키고 폴리에틸렌 주사관을 좌측 경정맥 내에 삽입하였다. 2일 후에 시험 화합물을 정맥내 투여하고 30분 후에 시스-플라티넘 30 mg/kg을 정맥내 투여하였다. 동물을 6시간 동안 관찰하여 구토의 발생 횟수를 기록하였다. 대조군을 각 실험에 포함시켰다.
본 발명의 대표적 화합물에 대해서 얻어진 결과를 표 4에 나타내고 임상에 사용되는 대조 화합물인 온단세트론 (5-HT3길항제) 및 메토클로프라미드 (D2길항제)와 비교하였다.

4) 장내 수송에 대한 본 발명 화합물의 효과
장내 수송에 대한 본 발명 화합물의 효과를 측정하기 위해 문헌 [Br. J. Pharmacol, 14, 26-34 (1959)]의 방법에 따라 시험하였다. 단식 전의 체중이 약 200 g인 스프라그 돌리 쥐 수컷에게 증류수 100 ml 중에 용해된 목탄 10 g 및 아라비아 고무 2.5 g으로 구성된 먹이를 경구 투여하였다. 15분 후에 동물을 희생시켜 목탄 수송 시간을 측정하였다.
시험 화합물을 목탄 투여 30분 전에 피하 투여하였다. 화합물 번호 18을 사용하여 얻은 결과를 표 5에 나타내었다.

5) 위 공복에 대한 본 발명 화합물의 효과
위 공복에 대한 본 발명 화합물의 효과를 문헌 [Arch. Int. Pharmacodyn., 246, 286-294 (1980)]의 방법에 따라 시험하였다. 페놀 레드 0.5 mg/ml을 포함하는 1.5% 메틸 셀룰로스 용액 1.5 ml을 체중 약 200 g의 미리 단식시킨 스프라그 돌리 쥐에 경구 투여하였다. 15분 후에 동물을 희생시키고 유문 및 식도를 묶었다. 위 내용물을 0.1 N 소다 용액 100 ml을 포함하는 테플론 비이커 중에 수거하여 혼합물을 균질화시켰다. 20% 트리플루오로아세트산 수용액 0.5 ml을 균질액 5 ml에 첨가하여 단백질을 침전시켰다. 원심분리 후에 상부에 상승한 용액의 일부를 분리하여 0.5 N 소다 수용액 4.5 ml을 첨가하고 560 nm에서 분광광도계를 사용하여 페놀 레드 함량을 측정하였다.
시험 화합물을 페놀 레드 투여 15분 전에 피하투여하였다.

Claims (15)
- 하기 화학식 I의 화합물 및 그의 염.화학식 I상기 식에서,Ar은 1종 이상의 치환체로 치환된 페닐이고,R은 직쇄 또는 분지쇄 알킬, 알케닐 또는 알키닐기, 포화 또는 불포화 모노시클릭 또는 폴리시클릭 시클로알킬, 시클로알킬알킬 또는 알킬시클로알킬기; 피리딜 또는 이소퀴놀릴기; 1종 이상의 치환체에 의해 치환될 수 있는 페닐 중에서 선택되는 탄소 원자수 1 내지 10의 탄화수소기이다.
- 제1항에 있어서, Ar로 나타낸 페닐기를 포함할 수 있는 치환체(들)이 할로겐, 저급 알킬, 저급 알콕시, 시아노, 니트로, 히드록시, C(O)NR1R2, C(O)NHOR3, NHC(O)R4, NHC(O)NHR5, CH2NHC(O)NHR6, CH2OR7, CH2NHC(O)R8, CO2R9, NHCO2R10, C(O)R11및 SR12기 (여기서, R1, R2, R3, R4, R5, R6, R7및 R8은 독립적으로 수소 원자 또는 저급 알킬이고, R9, R10, R11및 R12는 독립적으로 저급알킬임) 중에서 선택되고, R로 나타낼 수 있는 페닐기를 포함할 수 있는 치환체(들)이 저급 알킬, 저급 알콕시, 할로겐, 히드록시, 니트로, 아미노 또는 아실아미노기 중에서 선택되는 것을 특징으로 하는 화학식 I의 화합물.
