Abstract
본 명세서에는 세포 투과성 펩티드 및 유효성분의 컨쥬게이트(conjugate), 및 그 용도가 개시된다. 특히, 세포 투과성 펩티드로서 서열번호 1 내지 서열번호 156 중 어느 한 서열을 갖는 펩티드, 서열번호 1 내지 서열번호 156 중 어느 한 서열의 단편인 펩티드 또는 상기 펩티드 서열과 80% 이상의 서열 상동성을 갖는 펩티드를 포함하는 컨쥬게이트 및 그것을 포함하는 조성물이 개시된다.
Description
본 명세서에는 텔로머라제로부터 유래된 세포 투과성 펩티드, 그 세포 투과성 펩티드와 유효성분과의 컨쥬게이트 및 상기 컨쥬게이트를 포함한 조성물에 관한 것이다.
저분자 물질, 핵산, 단백질 또는 나노입자 등은 분자 수준의 치료 물질로서 큰 잠재력을 가지고 있으나, 조직 침투 및 세포막의 침투 저항성 때문에 사용이 제한 될 수 밖에 없다. 상기 물질들을 세포 내로 이동 시켜 줄수 있는 시스템의 개발은 분자적 방법의 치료에서 화두가 되어 왔다. 저분자 물질, 핵산, 또는 나노입자의 경우, 다양한 시약, 전기천공법(electroporation) 또는 열충격(heatshock) 등의 방법으로 세포 내 수송을 가능케 하였으나, 단백질의 경우, 활성을 그대로 유지하면서 세포 내로 이동하는 것은 매우 어려운 문제로 해결의 실마리를 얻지 못하고 있었다. 그런데, 1980년대에 HIV의 세포막을 투과하는 연구과정에서 특정 11개의 아미노산 서열로 이루어진 HIV-TAT 단백질이 세포 내로의 전달과정에서 중요한 역할을 수행하는 것으로 밝혀져 1990년대부터 본격적으로 단백질의 세포내 전달 연구가 진행되었다.
텔로미어(telomere)는 염색체의 말단에 반복적으로 존재하는 유전 물질로서, 해당 염색체의 손상이나 다른 염색체와의 결합을 방지한다고 알려져 있다. 세포가 분열할 때마다 텔로미어의 길이는 조금씩 짧아지는데, 일정한 횟수 이상의 세포 분열이 있게 되면 텔로미어는 매우 짧아지고, 그 세포는 분열을 멈추고 죽게 된다. 반면 텔로미어를 길게 하면 세포의 수명이 연장된다고 알려져 있으며, 그 예로 암세포에서는 텔로머라제(telomerase)라는 효소가 분비되어 텔로미어가 짧아지는 것을 막기 때문에, 암세포가 죽지 않고 계속 증식할 수 있다고 알려져 있다.

단백질, 핵산, 펩티드 또는 바이러스 등은 치료 물질로서 큰 잠재력을 가지고 있으나, 분자 수준의 크기를 가지므로 조직 및 세포막을 침투하지 못해 그 사용이 매우 제한적이었다. 또한 크기가 작은 물질이라 하더라도, 그 성질이나 구조상 세포막의 지질 이중층을 투과하지 못 하는 경우가 많다. 이에, 전기 천공법(electroporation) 또는 열 충격(heatshock) 등으로, 단백질, 핵산, 펩티드 또는 바이러스 등을 세포 내로 이동시키려는 시도가 있었으나, 세포막에 손상을 주지 않음과 동시에 상기 물질들의 활성을 그대로 유지하면서 상기 물질들을 세포 내로 이동시키기는 곤란하였다. 그러던 중 인간 면역 결핍 바이러스(Human Immuno-deficiency Virus: HIV)로부터 유래된 TAT(Trans-Activating Transcriptional activator) 단백질이 거대 활성 물질들을 세포 내로 이동시킬 수 있는 세포 투과성 펩티드(Cell Penetrating Peptide)로 작용할 수 있음이 알려지면서, 이에 대한 연구가 활발히 진행되었다. 구체적으로 세포 내 독성을 야기한다고 알려진 TAT 단백질과는 달리 생체 내 독성을 야기하지 않으면서도, 더욱 효과적으로 단백질, 핵산, 펩티드 또는 바이러스 등과 같은 거대 분자를 표적 세포 내로 이동시킬 수 있는 물질에 대한 연구가 진행되었다. 이에 본 발명자들은 지속적인 연구를 통해, 텔로머라제로부터 유래된 펩티드가 세포 독성이 거의 없으면서도 세포 투과성 펩티드로서 우수한 효과를 가짐을 발견하였다.
서열번호 1 내지 서열번호 156에 기재된 펩티드는 아래 표 1과 같다. 서열번호 157은 인간의 텔로머라제 전장 단백질의 서열이다. 아래 표 1의 "이름"은 펩티드를 구별하기 위해 명명한 것이다. 본 발명의 다른 일측면에서, 서열번호 1 내지 서열번호 156에 기재된 펩티드 중 하나 이상의 펩티드는 텔로머라제에 포함된 펩티드 중 해당 위치의 펩티드를 선별해 합성한 "합성 펩티드"를 포함한다. 본 명세서에서 "pep"이라 함은 서열번호 1 내지 서열번호 156 중 어느 한 서열을 갖는 펩티드, 또는 그 서열과 80% 이상의 서열 상동성을 갖는 펩티드, 또는 상기 서열의 단편을 총칭하는 용어이다.

본 발명의 일측면은 서열번호 1 내지 서열번호 156 중 어느 하나를 포함하는 펩티드 또는 상기 펩티드 서열과 80% 이상의 서열 상동성을 갖는 펩티드, 또는 그 단편을 코딩하는 폴리뉴클레오티드를 제공한다. 상기 폴리뉴클레오티드를 이용하여 펩티드를 대량 생산할 수 있다. 예컨대, 펩티드를 코딩하는 폴리뉴클레오티드를 포함하는 벡터를 숙주 세포에 넣어 배양함으로써, 펩티드를 대량 생산할 수 있다.
본 명세서에 개시된 펩티드는 80% 이상, 85% 이상, 90% 이상, 95% 이상, 96% 이상, 97% 이상, 98% 이상, 99% 이상의 서열 상동성을 갖는 펩티드를 포함할 수 있다. 또한, 본 명세서에 개시된 펩티드는, 서열번호 1 내지 서열번호 156 중 어느 하나를을 포함하는 펩티드 또는 그 단편들과 1개 이상의 아미노산, 2개 이상의 아미노산, 3개 이상의 아미노산, 4개 이상의 아미노산, 5개 이상의 아미노산, 6개 이상의 아미노산 또는 7개 이상의 아미노산이 변화된 펩티드를 포함할 수 있다.
본 발명의 일측면에서, 아미노산 변화는 펩티드의 물리화학적 특성이 변경되도록 하는 성질에 속한다. 예를 들어, 펩티드의 열안정성을 향상시키고, 기질 특이성을 변경시키고, 최적의 pH를 변화시키는 등의 아미노산 변화가 수행될 수 있다.

펩티드의 다른 유형의 아미노산 변이체는 항체의 글리코실화 패턴이 변화된 것이다. 변화란 의미는 펩티드에서 발견된 하나 이상의 탄수화물 잔기의 결실 및(또는) 펩티드 내에 존재하지 않는 하나 이상의 글리코실화 부위의 부가를 나타낸다.
펩티드의 글리코실화는 전형적으로 N-연결되거나 O-연결된 것이다. N-연결된이란 탄수화물 잔기가 아스파라긴 잔기의 측쇄에 부착된 것을 말한다. 트리펩티드 서열 아스파라긴-X-세린 및 아스파라긴-X-트레오닌 (여기서, X는 프롤린을 제외한 임의의 아미노산임)은 탄수화물 잔기를 아스파라긴 측쇄에 효소적 부착시키기 위한 인식 서열이다. 따라서, 이들 트리펩티드 서열 중의 하나가 폴리펩티드에 존재함으로써, 잠재적인 글리코실화 부위가 생성된다. 본 명세서에서,"세포 투과성 펩티드(Cell Penetrating Peptide, CPP)"는 인비트로(in vitro) 및/또는 인비보(in vivo)에서 이동 대상(cargo)을 세포 내로 이동시킬 수 있는 펩티드를 의미한다. 본 명세서에서, "이동 대상(cargo)"은 세포 투과성 펩티드와 결합하여 세포 내로 이동할 수 있는 물질을 모두 포함하며, 예를 들어 세포 투과 효율을 높이길 원하는 모든 물질, 구체적으로 약물, 화장품 또는 건강 식품의 유효 물질, 더 구체적으로 일반적인 경로를 통해서는 세포 내로 이동이 용이하지 않은 물질, 보다 더 구체적으로 단백질, 핵산, 펩티드, 미네랄, 포도당을 예로 들 수 있는 당, 나노 입자, 생물학적 제제, 바이러스, 조영물질 또는 기타 화학 물질을 포함하나, 이에 제한되는 것은 아니다. 본 명세서에서, "약물"은 질병, 상처 또는 특정 증상을 완화, 예방, 치료 또는 진단하기 위한 물질을 포함하는 광범위한 개념이다.

본 발명의 또 다른 일측면에서, 이동 대상이 DNA 또는 RNA인 경우, 펩티드에는 SH 기(thiol 기)를 도입하고 DNA 또는 RNA에는 말레이미드 기(maleimide group)를 도입한 후, 펩티드의 SH 기와 DNA 또는 RNA의 말레이미드 기를 결합시킴으로써, 펩티드에 이동 대상을 결합시킬 수 있다.
본 발명의 또 다른 일측면에서, 이동 대상이 단백질 또는 펩티드인 경우, 이동 대상을 발현하는 DNA에 운반 펩티드를 발현하는 DNA를 결합시킨 후 이를 발현시킴으로써, 이동 대상과 펩티드의 융합 단백질 형태로 운반 펩티드와 이동 대상을 결합시킬 수 있다. 융합 단백질에 의한 결합의 구체적인 예는 다음과 같다: 융합 단백질을 생산하기 위한 프라이머(primer) 제작시, 이동 대상을 발현하는 뉴클레오티드 앞에 운반 펩티드를 코딩하는 뉴클레오티드를 붙인 다음, 수득한 뉴클레오티드를 제한 효소로 pET 벡터를 예로 들 수 있는 벡터에 삽입하고, BL-21(DE3)을 예로 들 수 있는 세포에 도입(transformation)하여 발현시킨다. 이때, IPTG(isopropyl-1-thio-β-D-galactopyranoside)와 같은 발현 유도제를 처리하여 융합 단백질이 효과적으로 발현되도록 할 수 있다. 이후, His tag 정제(purification)와 같은 방법을 통해 발현시킨 융합 단백질을 정제한 후, PBS를 이용하여 투석하고, 키트에 넣어 예컨대, 2,000 내지 4,000 rpm에서 5 내지 20 여분간 원심 분리하여 농축시킬 수 있다.
본 발명의 일측면에서, 운반 펩티드는 염색 물질 또는 형광 물질, 구체적으로 플루오레세인 이소티오시아네이트(FITC, fluorescein isothiocyanate) 또는 녹색 형광 단백질(Green Fluorescent Protein, GFP)과 결합할 수 있다. 본 발명의 일측면에서 FITC는 운반 펩티드의 N-말단 또는 C-말단 Lys의 아미노기(NH3 +)와 결합할 수 있다. 말단에 Lys가 존재하지 않는 펩티드인 경우 Lys를 포함하는 링커에 의해 펩티드와 FITC를 결합시킬 수 있다.

본 발명의 바람직한 구체예들은 본 발명을 수행하기 위해 발명자에게 알려진 가장 최적의 모드를 포함한다. 바람직한 구체예들의 변이들이 앞선 기재를 읽으면 당업자에게 명백하게 될 수 있다. 본 발명자들은 당업자들이 그러한 변이를 적절히 이용하길 기대하고, 발명자들은 본 명세서에 기재된 것과 다른 방식으로 본 발명이 실시되기를 기대한다. 따라서, 본 발명은, 특허법에 의해 허용되는 것과 같이, 첨부된 특허청구범위에서 언급된 발명의 요지의 균등물 및 모든 변형들을 포함한다. 더욱이, 모든 가능한 변이들 내에서 상기 언급된 구성요소들의 어떤 조합이라도 여기서 반대로 명시하거나 문맥상 명백히 모순되지 않는 한 본 발명에 포함된다. 본 발명은 예시적인 구체예들을 참조하여 구체적으로 나타내어지고 기술되었지만, 당업자들은 하기 청구범위에 의해 정의되는 발명의 정신 및 범위를 벗어나지 않고서도 형태 및 디테일에서 다양한 변화가 행해질 수 있음을 잘 이해할 것이다.
발명의 실시를 위한 형태
실시예 1: 펩티드의 합성
서열번호 1 내지 서열번호 156의 펩티드를 종래에 알려진 고상 펩티드 합성법에 따라 제조하였다. 구체적으로, 펩티드들은 ASP48S(Peptron, Inc., 대한민국 대전)를 이용하여 Fmoc 고상 합성법(solid phase peptide synthesis, SPPS)을 통해 C-말단부터 아미노산 하나씩 커플링함으로써 합성하였다. 다음과 같이, 펩티드들의 C-말단의 첫번째 아미노산이 수지에 부착된 것을 사용하였다. 예컨대 다음과 같다:
NH2-Lys(Boc)-2-chloro-Trityl Resin
NH2-Ala-2-chloro-Trityl Resin
NH2-Arg(Pbf)-2-chloro-Trityl Resin
펩타이드 합성에 사용한 모든 아미노산 원료는 N-term이 Fmoc으로 보호(protection)되고, 잔기는 모두 산에서 제거되는 Trt, Boc, t-Bu (t-butylester), Pbf (2,2,4,6,7-pentamethyl dihydro-benzofuran-5-sulfonyl) 등으로 보호된 것을 사용하였다. 예컨대 다음과 같다:
Fmoc-Ala-OH, Fmoc-Arg(Pbf)-OH, Fmoc-Glu(OtBu)-OH, Fmoc-Pro-OH, Fmoc-Leu-OH, Fmoc-Ile-OH, Fmoc-Phe-OH, Fmoc-Ser(tBu)-OH, Fmoc-Thr(tBu)-OH, Fmoc-Lys(Boc)-OH, Fmoc-Gln(Trt)-OH, Fmoc-Trp(Boc)-OH, Fmoc-Met-OH, Fmoc-Asn(Trt)-OH, Fmoc-Tyr(tBu)-OH, Fmoc-Ahx-OH, Trt-Mercaptoacetic acid.
커플링 시약(Coupling reagent)으로는 HBTU[2-(1H-Benzotriazole-1-yl)-1,1,3,3-tetamethylaminium hexafluorophosphate] / HOBt [N-Hydroxxybenzotriazole] /NMM [4-Methylmorpholine] 를 사용하였다. Fmoc 제거는 20%의 DMF 중 피페리딘(piperidine in DMF)을 이용하였다. 합성된 펩타이드를 Resin에서 분리 및 잔기의 보호기 제거에는 절단 칵테일(Cleavage Cocktail) [TFA (trifluoroacetic acid) /TIS (triisopropylsilane) / EDT (ethanedithiol) / H2O=92.5/2.5/2.5/2.5] 를 사용하였다.
아미노산 보호기가 결합된 출발 아미노산이 고상 지지체에 결합되어 있는 상태를 이용하여 여기에 해당 아미노산들을 각각 반응시키고 용매로 세척한 후 탈보호하는 과정을 반복함으로써 각 펩티드를 합성하였다. 합성된 펩티드를 수지로부터 끊어낸 후 HPLC로 정제하고, 합성 여부를 MS로 확인하고 동결 건조하였다.
서열번호 164의 Pep 1 (EARPALLTSRLRFIPK) 을 예로 들어 구체적인 과정을 설명하면 다음과 같다.
1) 커플링
NH2-Lys(Boc)-2-chloro-Trityl Resin 에 보호된 아미노산(8당량)와 커플링 시약 HBTU(8당량)/HOBt(8당량)/NMM(16당량) 을 DMF에 녹여서 첨가한 후, 상온에서 2시간 동안 반응하고 DMF, MeOH, DMF순으로 세척하였다.
2) Fmoc 탈보호
20%의 DMF 중의 피페리딘(piperidine in DMF) 을 가하고 상온에서 5분 간 2회 반응하고 DMF, MeOH, DMF순으로 세척하였다.
3) 1과 2의 반응을 반복적으로 하여 펩타이드 기본 골격 NH2-E(OtBu)-A-R(Pbf)-P-A-L-L-T(tBu)-S(tBu)-R(Pbf)L-R(Pbf)-F-I-P-K(Boc)-2-chloro-Trityl Resin)을 만들었다.
4) 절단(Cleavagge) : 합성이 완료된 펩타이드 Resin에 절단 칵테일(Cleavage Cocktail) 을 가하여 펩타이드를 Resin에서 분리하였다.
5) 얻어진 mixture에 Cooling diethyl ether를 가한 후, 원심 분리하여 얻어진 펩타이드를 침전시킨다.
6) Prep-HPLC로 정제 후, LC/MS로 분자량을 확인하고 동결하여 powder로 제조하였다.
실시예 2: CPP-FITC 컨쥬게이트의 제조
(1) FITC-CPP 컨쥬게이트의 제조
서열번호 1 내지 서열번호 156의 펩티드들을 FITC와 접합시킨 컨쥬게이트를 다음과 같이 제조하였다. 예를 들어 서열번호 157의 pep1과 FITC의 컨쥬게이트, 즉 FITC-링커-pep1을 다음과 같이 제조하였다.
상기 실시예 1과 같이 하여 얻어진 펩타이드 기본 골격 NH2-링커-E(OtBu)-A-R(Pbf)-P-A-L-L-T(tBu)-S(tBu)-R(Pbf)L-R(Pbf)-F-I-P-K(Boc)-2-chloro-Trityl Resin)에 FITC를 반응시켰다. 구체적으로, 플루오레세인-5-이소씨오시아네이트(fluorescein-5-isothiocyanate, FITC) (8당량)과 N,N-디이소프로필에틸아민(N,N-Diisopropylethylamine, DIPEA)(16당량) 을 DMF에 녹여서 첨가한 후, 상온에서 2시간 동안 반응하고 DMF, MeOH, DMF순으로 세척하였다. 그 결과, FITC-링커-E(OtBu)-A-R(Pbf)-P-A-L-L-T(tBu)-S(tBu)-R(Pbf)L-R(Pbf)-F-I-P-K(Boc)-2-chloro-Trityl Resin를 얻었다. 여기서 링커는 는 6-아미노헥사노산 (6-aminohexanoic acid, Ahx)이다. 상기 합성이 완료된 펩타이드 Resin에 TFA/TIS/H2O=95/2.5/2.5 을 가하여 컨쥬게이트를 Resin에서 분리하였다. 얻어진 혼합물에 쿨링 디에틸 에테르(Cooling diethyl ether)를 가한 후, 원심 분리하여 얻어진 컨쥬게이트를 침전시켰다. 그 후 Prep-HPLC로 정제 후, 분석적(analytical) HPLC로 순도를 확인하고, LC/MS로 분자량을 확인하였다. 분자량 확인을 통해 얻어진 물질이 FITC-pep1임이 입증되었다. 그 후 동결건조를 하였다. 서열번호 1 내지 서열번호 156의 펩티드들을 FITC와 접합시킨 컨쥬게이트 또한 서열번호 157의 pep 1과 같이 제조하였다.
(2) CPP-FITC 컨쥬게이트의 제조
상기 실시예 1과 같이 펩타이드 기본 골격(NH2-E(OtBu)-A-R(Pbf)-P-A-L-L-T(tBu)-S(tBu)-R(Pbf)L-R(Pbf)-F-I-P-K(Dde)-2-chloro-Trityl Resin)을 제조하였다. 제조된 펩타이드 기본 골격의 C-term에 FITC를 선택적으로 도입하기 위해서 N-term을 Boc으로 보호하였다. 그 후 Di-터트-부틸 디카보네이트(30당량)와 DIPEA(30당량) 을 DMF에 녹여서 첨가한 후, 상온에서 2시간 동안 반응하고 DMF, MeOH, DMF순으로 세척하였다. 그 결과, Boc-E(OtBu)-A-R(Pbf)-P-A-L-L-T(tBu)-S(tBu)-R(Pbf)L-R(Pbf)-F-I-P-K(Dde)-2-chloro-Trityl Resin을 얻었다. 그런 다음, C-term K의 잔기에 FITC를 붙이기 위하여 2% DMF 중의 히드라진(hydrazine in DMF)을 처리하여 C-term Lys의 잔기 보호기인 Dde를 제거하였다. 그 결과 Boc-E(OtBu)-A-R(Pbf)-P-A-L-L-T(tBu)-S(tBu)-R(Pbf)L-R(Pbf)-F-I-P-K(NH2)-2-chloro-Trityl Resin를 얻었다. 그런 다음, FITC(8당량)과 DIPEA(16당량) 을 DMF에 녹여서 첨가한 후, 상온에서 2시간 동안 반응하고 DMF, MeOH, DMF순으로 세척하였다. 그 결과, Boc-E(OtBu)-A-R(Pbf)-P-A-L-L-T(tBu)-S(tBu)-R(Pbf)L-R(Pbf)-F-I-P-K(FITC)-2-chloro-Trityl Resin를 얻었다. 상기 합성이 완료된 펩타이드 Resin에 TFA/TIS/H2O=95/2.5/2.5을 가하여 펩타이드를 Resin에서 분리하였다. 얻어진 혼합물에 쿨링 디에틸 에테르를 가한 후, 원심 분리하여 얻어진 펩타이드를 침전시켰다. 그런 다음 Prep-HPLC로 정제 후, 분석적(analytical) HPLC로 순도를 확인하고, LC/MS로 분자량을 확인하였다. 그 결과 얻어진 물질이 pep1-FITC임이 입증되었다. 그 후 동결건조하였다. 서열번호 1 내지 서열번호 156의 펩티드-FITC 컨쥬게이트들 또한 상기 pep1-FITC와 같은 방법으로 제조하였다.
실시예 3: pep-FITC 컨쥬게이트의 세포 투과능 실험
(1) HeLa 세포주에서의 세포 투과능 시험
세포주 배양
ATCC로부터 얻은 HeLa 세포주를 MEM(Minimum Essential Medium)에 10% 우태아혈청 (Invitrogen, USA)과 Earle's salts, non-essential amino acids, sodium pyruvate 및 100 ㎍/㎖ 페니실린과 100 units/㎖ 스트렙토마이신(streptomycin)를 첨가하여 37℃, 5% CO2 배양기에서 배양하였다.
유세포 분석기(Flow cytometry) 분석
상기 세포주에 본 발명의 서열번호 1 내지 156의 펩타이드들, pep (CPP)을 처리하여 control과 uptake 정도를 비교하기 위해 유세포 분석기(Flow cytometry) 분석을 실시하였다.
세포주를 6-well plate에 분주하여 배지에 10% 우태아혈청 (Invitrogen, USA)과, 100 ㎍/㎖ 페니실린과 100 units/㎖ 스트렙토마이신(streptomycin)를 첨가하여 37℃, 5% CO2 배양기에서 12시간 배양하였다. PBS로 세척한 후, MEM(Minimum Essential Medium)에 1시간 동안 starvation을 시킨다. 각 운반 펩티드 20 uM을 처리하여 37℃에 1시간 배양하였다. PBS로 세척과정을 3번 반복하고 Trypsin-EDTA를 37℃에서 10분간 처리하여 세포 바깥쪽에 붙은 운반 펩티드를 제거한다. 냉장된 PBS로 세포를 수거한 후 3번 세척 과정을 원심분리를 통해서 반복하였다. 그 후 4% Paraformaldehyde가 포함된 0.5㎖ PBS에 suspension시켜 FACS Calibur(Becton Dickinson)로 형광을 분석하였다. 운반펩티드를 처리하지 않은 세포(Control)와 다양한 접합 펩티드의 cellular uptake 양상을 MFI(mean fluorescence intensity)로 비교 분석하였다.
그 결과는 도 1 내지 도 23에 나타난 바와 같다. 도 1 내지 도 23에 나타난 분석 결과는 하기의 표 3에 상세히 나타내었다.

(2) Huh7 세포주에서의 세포 투과능 시험
세포주 배양
인간의 간세포 암의 세포주인 Huh7 (human hepatocellular carcinoma cell lien의 부유 세포주를 사용하였다.(ATCC(American Type Cell Culture)로부터 구입) Huh7 세포주는 RPMI 1640 배지를 사용한다. 배지에는 10% 우태아혈청(Invitrogen, USA)과, 100 ug/ml penicillin, 100 units/ml streptomycin를 첨가하여 37℃, 5% CO2 배양기에서 배양하였다.
유세포 분석을 통한 세포침투능 스크리닝
펩타이드들의 세포침투능을 살펴보고자 서열번호 1 내지 서열번호 156의 펩타이드를 처리한 Huh7 세포주에서 유세포 분석을 하였다. 분석방법은 상기 (1)의 Hela 세포주 분석에 기재된 바와 같다. 분석 결과는 도 24 내지 도 43에 나타내었다.
(3) 사람 T 림프구 세포주에서의 세포 투과능 시험
세포주 배양
인간 T 림프구 세포주 Jurkat (Human T-cell leukemia cell line의 부유 세포주를 사용하였다. (ATCC(American Type Cell Culture)로부터 구입) Jurkat 세포주는 RPMI 1640 배지를 사용한다. 배지에는 10% 우태아혈청(Invitrogen, USA)과, 100 ug/ml penicillin, 100 units/ml streptomycin를 첨가하여 37℃, 5% CO2 배양기에서 배양하였다.
사람 유래 림프구(lymphocyte) 분리는 건강한 사람에게서 혈액을 채혈 (50 ml)한 후 Biocoll Separating Solution (Biochrom AG, Berlin, Germany)를 사용하여 peripheral blood mononuclear cells (PBMC) 층 및 림프구를 회수한다.
사람 T 림프구 세포주의 세포 침투능 분석
펩타이드들의 세포침투능을 살펴보고자 서열번호 1 내지 서열번호 15656의 펩타이드를 처리한 사람 T 림프구 세포주에서 유세포 분석을 하였다. 분석방법은 상기 (1)의 Hela 세포주 분석에 기재된 바와 같다. 분석 결과는 도 44 내지 도 58에 나타내었다.
(4) 세포생존율 및 독성 분석
한편, 상기 실시예 3(1)에서 배양된 Hela 세포주를 96-well plate)에 분주하여 배지에 10% 우태아혈청 (Invitrogen, USA)과, 100 ㎍/㎖ penicillin과 100 units/㎖ streptomycin를 첨가하여 37℃, 5% CO2 배양기에서 12시간 배양하였다. PBS 세척 후, MEM(Minimum Essential Medium)에 1시간의 starvation을 시켰다. 각 운반 펩티드 20 uM을 처리하여 37℃에 24시간 배양한 후 MTT assay 방법을 이용하여 세포생존율 및 독성을 분석하였다. 그 결과는 도 59 내지 도 72에 나타난 바와 같다.
