Abstract
본 발명은 실크 펩타이드를 포함하는 치매, 뇌졸증 및 인지 기능 개선용 조성물에 관한 것이다. 본 발명의 일 실시예에 따른 인지 개선용 조성물은 천연물 유래의 추출물을 활용하여 인지 기능의 개선에 효과가 있는 조성물을 제공할 수 있으며, 본 발명의 실크 펩타이드 및 천연 추출물을 포함하는 인지 기능 개선용 조성물은 약제학적 조성물로 제공되어, 종래 실크 펩타이드만을 이용하는 경우에 비해, 인지 기능의 개선 효과가 높아진 조성물을 제공할 수 있다. 또한, 본 발명의 기능성 식품 조성물에 의하는 경우, 인지 기능의 개선 효과를 내면서도 기호도 높은 기능성 식품 조성물로 제공될 수 있다.
Description
본 발명은 실크 펩타이드를 포함하는 치매, 뇌졸증 및 인지 기능 개선용 조성물에 관한 것이다. 보다 구체적으로, 실크 단백질의 분해에 의해 생성된 실크 펩타이드를 유효 성분으로 포함하여, 치매 및 뇌졸증의 예방 또는 개선 효과 및 인지 기능 개선 효과가 우수한 조성물에 관한 것이다.
뇌졸중은 뇌혈관이 터지거나 막혀서 결과적으로 국소 뇌조직의 기능 이상을 가져오는 뇌혈관 질환으로 흔히 중풍이라고도 하며, 우리나라에서는 사망원인 중 선두를 달리고 있다. 더욱이 산업화와 의학의 발전에 의한 평균수명의 연장으로 인하여 그 발병률이 점차 증가하는 추세이다. 뇌졸중은 뇌의 어느 부위에서나 발생할 수 있고 따라서 신체의 거의 모든 기능에 장애를 초래할 수 있다. 의학적으로 뇌졸중은 크게 뇌의 혈관이 막혀서 특정 부위에 혈액 순환이 안되어 나타나는 '허혈성 뇌졸중'과 뇌출혈에 의한 '출혈성 뇌졸중'으로 분류할 수 있으며, 그 중 성인병의 원인으로 알려진 고혈압, 동맥경화증과 밀접한 연관성이 있는 허혈성 뇌졸중의 발생 비율이 더 높은 편이다.
허혈성 뇌졸중은 목 부분에 있는 경동맥, 추골뇌저동맥부터 뇌 안의 가느다란 동맥에 이르기까지 어디든지 혈관이 막히면 발병하게 되며, 이로 인해 그 혈관이 지배하던 뇌 조직이 죽는 현상, 즉 '뇌경색(infarction)'이 발생하게 된다. 일단 발생된 뇌경색 부위는 그 기능을 되살릴 수 없으며, 따라서 뇌경색에 의한 중추신경계 장애는 회복되지 않고 영구적으로 남게된다. 그러므로 뇌졸중 치료에 있어서 제일 중요한 것은 뇌졸중 발병 위험인자로 알려진 고혈압, 당뇨병, 고지혈증 등의 예방법과 더불어 뇌 허혈 자체에 대한 예방이다. 또한 뇌졸중 발병에 의해 뇌경색이 왔을 때 뇌부종을 감소시키고, 허혈 상태인 곳은 적절하게 순환되게 하여 이차적인 뇌손상을 줄이는데 그 초점을 두고 있다.
현재 신경 세포의 보호를 위해 사용하고 있는 물질로는 흥분성 아미노산 길항제인 갱글리오사이드(ganglioside), 니모디핀(Nimodipine), GABA 항진제인 클로메치아졸(clomethiazole) 등이 있고 황산 마그네슘과 글리신 길항제는 2상 임상 연구중이며, 피라세탐(piracetam)이 현재 대규모 임상 연구가 진행중이다. 그러나 현재 시도되고 있는 신경 세포 보호제들은 허혈 진행 과정의 각기 다른 단계에 작용하는 제제들로, 이러한 여러 가지 과정에 동시에 작용하는 복합요법의 개발이 필요하지만 부작용과 약물 상호간섭의 문제를 해결해야 하는 과제를 안고 있다.
또한, 허혈성 뇌졸중의 증상은 특별한 예후 없이 급작스레 찾아오므로 뇌 허혈 발병 당시에 투여되는 약물을 통하여 치료를 기대하기보다는 지속적인 허혈 예방과 허혈 후의 신경세포의 아폽토시스의 억제를 위한 기능성 식품의 섭취가 더욱 효과적이라 판단되고 있다.
신경 퇴행성 질환의 일종인 파킨슨병(Parkinson's disease, PD)은 운동 및 인지 장애를 일으키는 퇴행성 뇌 신경 질환으로 1817년 제임스 파킨슨에 의해 처음 보고되었다. 이 질환의 발병률은 미국에서 인구 10만 명 당 100-150명 정도로 약 75만-100만 명의 환자가 보고되고 있으며, 매년 6만여 명이 새로이 진단되는 추세로, 국내에서도 인구의 노령화에 따라 그 발병률 및 유병률이 계속 증가할 것으로 예상된다. 조직병리학적으로는 흑질(substantia nigra)에 위치하는 도파민 신경세포의 소실이 특징적으로 나타나며, 그 신경 섬유의 투사 부위인 미상핵 (caudate nucleus)과 조가비핵(putamen)의 도파민이 감소하여 특징적인 진전 (tremor), 운동지연(bradykinesia), 경직(rigidity), 자세의 장애(disturbance of posture) 등 운동 및 인지 기능 장애가 나타난다.
파킨슨병에 사용되는 주된 치료 약물은 뇌에서 부족해진 도파민의 기능을 보충해 주는 약제 또는 그 외 신경 세포의 파괴를 예방 혹은 지연시키고자 하는 목적이나 기타 우울증 등의 부수적인 증상을 조절하기 위한 약물 치료 등이 있다. 대표적인 약물로 도파민 전구 물질인 레보도파(levodopa, L-dopa) 성분의 마도파(Madopar), 도파민 수용체 효현제 (agonist) 성분의 브로미딘(bromidine), 리슈라이드(lisuride), 그리고 항 아세틸콜린성 약물인 알탄(artane), 코젠틴 (cogentin) 등의 다양한 신경약리학적 보상성 치료법이 개발되어 있다. 이중 도파민의 전구체인 레보도파는 부족한 뇌 내 도파민의 농도를 보충시킴으로써 파킨슨병의 증상을 개선시키는데 가장 효과적으로 사용되고 있으나, 3 내지 5년 이상 장기간 투여하면 약물효과 시간이 점점 짧아지거나(wearing-off), 약물의 효과에 대한 운동조절 기능의 변동이 심해지는 현상(on-off 현상), 이상운동증 (diskinesia) 등의 부작용이 나타난다(Freed et. al., N. Engl. J. Med. 327:1549-55(1992)).
그밖에도 파킨슨병의 외과적 치료법으로 시상절단술(thalamotomy)과 담창구절단술(pallidotomy), 심부 뇌자극술(deep brain stimulation), 신경세포 이식술(Neuronal cell transplantation)등이 시행되고 있다. 그러나 치료효과의 지속 기간이 환자에 따라 큰 차이를 나타내며, 드물게 수술에 따른 발성부전(hypophonia), 구음장애(dysarthria; 말더듬증), 기억력 감쇠 등의 부작용을 동반한다(Ondo et. al., Neurology 50:266-270 (1998); Shannon et. al., Neurology 50:434-438(1998)).
알츠하이머병에 대한 최근 치료 기조는 대뇌피질(cerebral cortex)과 해마 (hippocampus)에서 기능 손상된 콜린성 시그널링 및 전달(cholinergic signaling and transmission)에 의해 알츠하이머병이 유래된다는 가능성에 중심을 두고 있다(Bartus et al., Science. 217(4558): 408-14(1982)); 및 Coyle et al., Science. 219(4589):1184-90(1983)). 이런 뇌의 영역은 기억 및 지능과 연결되어 있으므로, 뇌의 이들 부분의 기능적인 결함은 기억 및 판단에 대한 확실한 손상을 주고 지적 능력을 상실케 한다. 신경 신호전달 (neuronal signaling)에 손상이 이루어지는 정확한 과정이 논쟁 대상이 되고는 있지만, 감각성 플라크 (senile plaque) 및 신경원 섬유 농축체 (neurofibrillary tangle: NFT)이 신경 손상의 주 원인으로 여겨진다. 아밀로이드 베타 (Aβ)의 축적으로 인한 감각성 플라크는 이병의 가장 큰 특징으로, 알츠하이머병은 사후 부검으로 확진이 가능하다 (Khachaturian, Arch. Neurol. 42(11):1097-105(1985)).
알츠하이머 병의 경우, 콜린성 시그널링의 손상을 억제할 수 있도록 아세틸콜린의 양을 증가시키거나, 아세틸콜린이 장기간 존재할 수 있도록 하거나 또는 신경세포의 전달에 아세틸콜린이 더욱 효과적으로 작용하도록 하는 약물과 치료법이 제시되고 있어서, 알츠하이머병 환자들의 아세틸콜린 활성도를 높이는 많은 화합물들이 사용되고 있다. 현재 가장 효과적인 접근 방법은 시냅스에서 아세틸콜린을 빠르게 분해하여 신경 신호 전달을 막는 아세틸콜린에스테라아제의 활성을 억제하는 방법이다. 실제로 이런 저해제(예: tacrine, donepezil 및 rivastigmine)들은 현재 FDA에서 인정한 알츠하이머병 치료 약물로 시장에서 유통되고 있다. 많은 경우 상기 약물들은 병의 파괴적인 진행을 막는데 유효하나, 신경계의 병 이전 상태로의 회복에는 잘 적용되지 않고 있다.
비록 알츠하이머 병의 영향을 줄이기 위한 치료 방법에 대한 개발이 많이 진행되고 있으나, 현재로써는 일시적인 증상의 개선을 제공하는 것이다. 결론적으로 현재 알츠하이머병 치료는 병의 진행 과정을 되돌리는 것이 아니라 병의 증상을 개선하는데 초점을 두고 있다. 병의 생물학적인 지식에 대해서는 보다 많이 알고 있으나, 임상에로의 적용 결과는 아직 성공적이지는 않다.
하지만, 현재까지 다양한 뇌신경계 질환을 상대로 뛰어난 신경보호 효능과 뇌기능 향상 효과를 갖는 예방 및 치료용 물질에 대해 제품 개발이 필요한 실정이다.
본 발명의 목적은 실크 단백질의 분해에 의해 생성된 실크 펩타이드를 유효 성분으로 포함하여, 치매 및 뇌졸증의 예방 또는 개선 효과 및 인지 기능 개선 효과가 우수한 조성물을 제공하는 것이다.
본 발명의 다른 목적은 주기적 또는 장기적 복용에 의해서 부작용의 발생이 없는 천연 물질을 구성 성분으로 포함하는 인지 기능의 개선용 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 실크 펩타이드 및 천연 추출물을 유효 성분으로 이용하여 인지 기능의 개선을 위한 약제학적 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 실크 펩타이드 및 천연 추출물을 유효 성분으로 이용하여 인지 기능의 개선을 위한 기능성 식품 조성물을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명의 일 실시예에 따른 실크 펩타이드 및 천연 추출물을 포함하는 인지 기능 개선용 조성물은 실크 단백질의 분해에 의해 생성된 실크 펩타이드 및 천연 추출물을 포함하며, 상기 천연 추출물은 일년봉(Erigeron annuus (L.) Pers) 추출물, 보리지(Borage officinalis) 추출물, 엉겅퀴(Cirsium japonicum var. maackii (Maxim)) 추출물, 짚신나물(Agrimonia Pilosa) 추출물, 다릅나무(Maackia amurensis) 추출물 및 이들의 혼합물로 이루어진 군으로부터 선택될 수 있다.
상기 실크 단백질은 실크 세리신 또는 실크 피브로인이다.
상기 조성물은 실크 펩타이드 100 중량부에 대하여, 천연 추출물 0.1 내지 100 중량부로 포함할 수 있다.
상기 천연 추출물은 물, 탄소수 1 내지 10의 알코올 및 이들의 혼합 용매로 이루어진 군으로부터 선택된 용매로 추출되는 것이다.
상기 인지 기능은 학습 능력, 기억 능력 또는 집중력이다.
상기 조성물은 뇌신경질환에 수반되는 두뇌 또는 인지 기능의 악화를 개선하는 것이다.

이하, 본 발명을 더욱 상세하게 설명한다.
본 발명에서 실크 펩타이드는 실크에서 유래된 것으로, "실크"는 견사 곤충이 토사하여 만든 섬유를 의미하며, 바람직하게는 누에(Bombyx Mori)가 토사하는 가잠사를 의미한다.
본 발명에서 "천연 추출물"은 천연물로부터 분리된 활성 성분을 의미한다.
본 명세서에서 "신경 세포"는 중추 신경계, 뇌, 뇌간, 척수, 중추 신경계와 말초 신경계의 접합부분 등의 구조를 이루는 뉴런, 신경지지 세포, 글리아, 슈만 세포 등을 포함한다.

본 명세서에서 신경성 질환은 상술한 신경 세포 또는 신경 조직의 손상에 의해 유발되는 모든 질환을 의미하며, 특히 뇌 신경질환은 뇌의 신경 세포 또는 신경 조직의 손상에 의해 유발되는 모든 질환을 의미한다.
본 명세서에서 "예방"은 질환 또는 질병을 보유하고 있다고 진단된 적은 없으나, 이러한 질환 또는 질병에 걸리기 쉬운 경향이 있는 동물에서 질환 또는 질병의 발생을 억제하는 것을 의미한다.
본 명세서에서 "치료"는 (a) 질환 또는 질병의 발전의 억제; (b) 질환 또는 질병의 경감; 및 (c) 질환 또는 질환의 제거를 의미한다.
본 발명의 일 실시예에 따른 실크 펩타이드 및 천연 추출물을 포함하는 인지 기능 개선용 조성물은 실크 단백질의 분해에 의해 생성된 실크 펩타이드 및 천연 추출물을 포함하며, 상기 천연 추출물은 일년봉(Erigeron annuus (L.) Pers) 추출물, 보리지(Borage officinalis) 추출물, 엉겅퀴(Cirsium japonicum var. maackii (Maxim)) 추출물, 짚신나물(Agrimonia Pilosa) 추출물, 다릅나무(Maackia amurensis) 추출물 및 이들의 혼합물로 이루어진 군으로부터 선택될 수 있다.
구체적으로, 실크 단백질은 실크 세리신 또는 실크 피브로인이다. 실크 단백질은 피브로인 및 세리신으로 이루어져 있고, 피브로인과 세리신은 각각 평균 75%와 25%의 비율로 존재한다. 피브로인 단백질은 특이하게 글리신 및 알라닌 함량이 매우 높은 반면, 세리신은 피브로인과 전혀 다른 아미노산 조성을 나타내고 있으며, 세린이 전체 구성 아미노산 중의 1/3 이상을 차지하고 있다. 최근의 연구 결과에 의하면 실크 피브로인은 H-사슬 (350 kDa) 및 L-사슬 (26 kDa)이 S-S 결합을 하고, 당단백질인 P25 (30 kDa)가 상기 두 사슬과 비공유 결합으로 연결된 구조를 하고 있으며, 각각 6:6:1의 몰 구성을 하고 있는 2.3 BG101a의 거대 단백질로 규명되었으며, 이러한 구조로 인해 결정영역과 비결정영역이 연속 교환 배열된 블록 형태의 고분자 성질을 가지게 된다. 보다 구체적으로, 상기 실크 단백질은 실크 피브로인이지만, 상기 예시에 국한되지 않는다.
본 발명의 일 실시예에 따르면, 실크 단백질의 분해는 (a) 칼슘염 용액에서의 분해, (b) 산 가수분해, (c) 효소에 의한 가수분해 또는 (d) 상기한 방법들의 조합을 통해 실시될 수 있으나, 이에 한정되는 것은 아니다. 상기한 방법에 의해 분해되어 생성된 실크 펩타이드의 중량 평균 분자량은 약 200-100,000의 값을 나타낸다.
상기 (a) 칼슘염 용액에서의 분해는 통상적으로 염화칼슘을 이용하는 데, CaCl2·에탄올을 이용한다. 이와 같은 칼슘염 용액에서의 분해에 의해 실크 피브로인은 분자량 25,000 내지 50,000 정도의 비교적 고분자 펩타이드로 분해된다. 이러한 고분자 펩타이드는 그 큰 분자량 때문에 화장품 성분으로는 적합하나, 생체 내 흡수율이 낮기 때문에 식품 및 약제로서는 적합하지 않다.
상기 (b) 산 가수분해는 통상적으로 무기산 (예: 염산)을 이용하여 실시되며, 강산을 통상적으로 이용하며, 얻어지는 펩타이드의 중량 평균 분자량은 200 내지 10,000정도이다. 이와 같은 분해 방법은 분자량이 작은 펩타이드를 형성시키는 장점은 있으나, 적당한 분자량의 펩타이드를 수득하기가 어렵다는 문제점이 있다. 본 발명의 일 실시예에 따르면, 산 가수분해에 의해 수득한 실크 펩타이드의 중량 평균 분자량은 200 내지 3,000이고, 보다 구체적으로 300 내지 1,500이다. 산 가수분해에 의해 수득된 본 발명의 펩타이드는 물에 대하여 실질적으로 100%의 용해도를 나타내며, 유기 용매 (예: 에탄올, 메탄올, 아세톤 및 디메틸포름 아마이드)에 대해서는 매우 낮은 용해도를 나타낸다.
상기 (c) 효소에 의한 가수분해는 단백질 분해 효소를 이용하여 실시된다. 최종적으로 수득되는 실크 펩타이드의 중량 평균 분자량이 200 내지 15,000, 보다 구체적으로 200 내지 4,000이며, 보다 더 구체적으로는 300 내지 2,000이고, 바람직하게는 300 내지 1,500의 범위에 있도록 실크 단백질을 분해할 수 있는 효소 또는 효소들의 조합을 이용하여 실시된다.
상기 분해 효소는 아미노산 서열에 특이적 또는 비특이적으로 작용하여 가수분해를 촉매한다. 상기 분해 효소는 트립신(trypsin), 펩신(pepsin), 알카라제 (alcalase), 써모아제(thermoase), 플라보자임(flavourzyme), 수미자임 (sumizyme), 플로타멕스(protamex) 및 프로틴(protin)으로 이루어진 군으로부터 선택되며, 상기 예시에 한정되는 것은 아니다.
상기 (d) 조합에 의한 분해의 경우에는 1차적으로 칼슘염에 의한 분해 또는 약산 또는 알칼리에 의한 가수분해를 한 후 2차적으로 효소로 가수분해하는 것이다. 보다 바람직하게는, 1차적으로 칼슘염에 의한 분해를 하고 2차적으로 써모아제, 플라보자임, 수미자임 또는 이들의 조합으로 이루어진 군으로부터 선택된 분해 효소로 분해를 하여 실크 펩타이드를 얻는 것이다. 이와 같은 조합적인 방법에 의해 얻어지는 실크 펩타이드의 중량 평균 분자량은 200 내지 15,000이고, 구체적으로는 200 내지 4,000이며, 보다 구체적으로는 300 내지 2,000이고, 가장 구체적으로는 400 내지 1,200이다. 이와 같은 방법에 의해 본 발명의 실크 펩타이드는 물에 대하여 실질적으로 100%의 용해도를 나타내며, 유기 용매 (예: 에탄올, 메탄올, 아세톤 및 디메틸포름 아마이드)에 대해서는 매우 낮은 용해도를 나타낸다.
본 발명에서의 실크 펩타이드는 (a) 실크 피브로인을 기질로 하여 산 가수분해를 통해 수득한 실크 펩타이드; 및 (b) 실크 피브로인을 기질로 하여 1차적으로 칼슘염 분해를 하고, 트립신 (trypsin), 펩신(pepsin), 알카라제 (alcalase), 써모아제, 플라보자임, 수미자임 또는 이들의 조합으로 분해를 하여 수득한 펩타이드이다. 보다 바람직한 펩타이드는 (a) 실크 피브로인을 기질로 하여, 20 내지 30%의 염산 수용액을 첨가하여 가수분해를 통해 수득한 실크 펩타이드 또는 (b) 실크 피브로인을 기질로 하여 1차적으로 칼슘염 분해를 하고, 플라보자임 및 수미자임으로 분해를 하여 수득한 실크 펩타이드이다. 이때, 플라보자임 및 수미자임은 1:1의 중량비율로 혼합하여 사용하였다.
이와 같은 실크 펩타이드를 포함하는 본 발명의 조성물은 신경성 질환의 예방 또는 치료에 효능을 나타낸다. 이와 같은 본 발명의 작용은 대체적으로 신경 세포의 보호 작용을 통하여 나타난다.
본 발명의 일 실시예에 따르면, 본 발명의 실크 펩타이드를 얻기 위하여 이용되는 효소는 비특이적으로 아미노산 서열을 분해하는 효소이며, 보다 구체적으로, 써모아제, 플라보자임, 수미자임 및 이들의 조합으로 이루어진 군으로부터 선택되며, 보다 더 구체적으로 플라보자임, 수미자임 및 이들의 혼합이지만, 상기 예시에 국한되지 않는다.
한편, 상기 실크 펩타이드의 함량을 유지하면서 상호작용으로 인지기능 개선에 대한 상승효과를 나타내도록 하기 위해 상기 실크 펩타이드에 천연 추출물을 혼합사용하는 경우 일정한 종류의 천연 추출물이 일정한 범위에서 상호 작용에 의한 상승효과를 낼 수 있다는 것을 확인하였다. 따라서 하기의 천연 추출물을 혼합사용하는 경우 실크 펩타이드를 단독으로 사용한 경우에 비하여 동일한 함량으로 인지기능 개선에 있어 보다 우수한 효과를 낼 수 있다는 것을 알 수 있다.
상기 천연 추출물은 일년봉(Erigeron annuus (L.) Pers) 추출물, 보리지(Borage officinalis) 추출물, 엉겅퀴(Cirsium japonicum var. maackii (Maxim)) 추출물, 짚신나물(Agrimonia Pilosa) 추출물, 다릅나무(Maackia amurensis) 추출물 및 이들의 혼합물로 이루어진 군으로부터 선택될 수 있다.
상기 일년봉(Erigeron annuus (L.) Pers)은 개망초라고도 불리우며, 국화과 식물이며, 높이는 20 내지 100cm정도이고, 줄기는 직립하고 상부에서 분지한다. 북아메리카 원산으로 우리 나라 각처의 밭, 들, 길가에 나는 월년초로, 귀화식물이다. 전체에 털이 있고, 가지가 많이 갈라져 있다. 근생엽은 꽃이 필 때 시들고, 잎자루가 길며 난형으로 가장자리에 톱니가 있다.
상기 엉겅퀴(Cirsium japonicum var. maackii (Maxim))는 국화과에 속하는 여러해살이풀로, 한국, 일본, 중국 북동부에 분포하며, 가시나물 혹은 항가새라고도 한다. 높이 50 내지 100 ㎝이고 전체에 흰털과 더불어 거미줄과 같 은 털이 있다. 뿌리 잎은 꽃필 때까지 남아있고 줄기 잎보다 크다. 줄기 잎은 피침상 타원형으로 깃처럼 갈라지 고 밑은 원대를 감싸며 갈라진 가장자기가 다시 갈라지고 결각상의 톱니와 더불어 가시가 있다. 꽃은 6~8월에 피고 자주색에서 적색이다. 총포의 포편은 7 내지 8열로 배열하고 안쪽일수록 길어진다. 열매는 수과이며 관모는 길이 16 내지 19 ㎜이다. 잎이 좁고 녹색이며 가시가 다소 많은 것을 좁은 잎 엉겅퀴, 가시가 많은 것을 가시엉겅퀴라 한다.
상기 짚신나물(Agrimonia pilosa)은 다년생 숙근초로 장미과에 속하며, 북반구 온대와 브라질 및 남아메리카 등에 10 여종이 분포하는 것으로 알려져 있다. 우리나라의 경우에는 야산의 길가, 들판 등에 흔히 자라며, 선학초, 용아, 용아초, 황아초, 황용초, 지선초 등으로 부르기도 한다. 키는 15 내지 60 cm쯤 자라고 전체 흰 털이 있으 며 버들잎 모양 또는 긴 타원 꼴의 쪽잎이 어긋나게 붙는다. 6 내지 8월에 꽃대 위에 노란색의 작은 꽃이 모여서 핀다. 전통적으로 민간요법, 녹즙 등으로 널리 이용되어 왔고, 폐암, 간암, 식도암, 종양, 통증제거, 지형, 지사, 토혈, 혈토, 자궁출혈, 열기 등의 약용으로 널리 이용되어 왔다. 최근에는 짚신나물 추출물로부터 많은 물질들이 제약원료, 건강식품, 산채, 식물성 영양제 등으로 쓰이고 있으며 Vitatmin K, 탄닌, 정유, luteolin-7-glucosidase, apigeni-7-glucosidase, 페놀성배당체, agrimoniin, agrimonolide 등이 알려져있으며 특히 agrimoniin의 항암효과가 잘알려져 있다.
상기 다릅나무(Maackia amurensis Rupr.)는 콩과에 속하는 낙엽교목으로 높이가 15 m에 달한다. 잎은 호생하며 홀수 1회 우상복엽이고 소엽은 타원형 또는 장란형이며 짧은 점첨두이고 원저이며 길이 5 내지 8 cm로 양면에 털이 없다. 꽃차례는 가지 끝에 달리고 총상화서 또는 원추화서로서 길이 10 내지 20 cm이며 위로 향하고 꽃은 7월에 피며 지름 8 mm로서 밀착한다. 꼬투리는 넓은 선형이고 털이 없으며 길이 3.5 내지 5.0 cm, 너비 7 내지 9 mm이고 열매 꼭지는 길이 5 내지 10 mm이다. 종자는 길이 6 mm로서 신장형에 가까우며 열매는 9월에 익는다. 잎 뒷면에 털이 있고 꽃차례에 황갈색 또는 회갈색의 짧은 털이 밀생한 것을 개물푸레나무(Maackia amurensis Rurp. var. buergeri (Maxim.) C.K.Schneid.)라고 한다. 각지의 깊은 산골짜기에서 자라며, 줄기를 자른 단면 은 심재가 흑갈색을 띄는 것이 특징이다. 다릅나무는 다름나무라고도 불리며, 쇠코둘개나무, 개박달나무, 소터 래나무, 다즙나무로도 불리고 있으며, 민간에서는 선화삼이라 하여 임파선염, 임파선암, 폐암, 자궁근종, 신경 쇠약, 생리불순, 편두통 등에 사용되고 있다.
상기 인지 기능 개선용 조성물은 상기 실크 펩타이드 100 중량부에 대하여 천연 추출물 0.1 내지 100 중량부를 포함하며, 바람직하게는 실크 펩타이드 100 중량부에 대하여 천연 추출물 3 내지 5 중량부를 포함할 수 있지만, 상기 예시에 국한되지 않는다.
실크 펩타이드에 천연 추출물을 혼합한 혼합물은 상호 작용으로 단순 실크 펩타이드만 이용하는 경우와 비교하여, 인지 기능 개선에 효과를 크게 증대시킬 수 있다. 실크 펩타이드 100 중량부에 대하여 천연 추출물 3 중량부 미만으로 포함할 경우, 실크 펩타이드에 천연 혼합물을 혼합함에 따른 인지 기능 개선의 증대 효과가 미비하며, 천연 추출물을 5 중량부 초과하여 포함할 경우, 천연 추출물을 과다 포함함에 따라, 실크 펩타이드 및 천연 추출물의 인지 기능 개선 효과를 저해하는 문제가 발생한다.
바람직하게 상기 천연 추출물은 보리지(Borage officinalis) 추출물, 엉겅퀴(Cirsium japonicum var. maackii (Maxim)) 추출물, 짚신나물(Agrimonia Pilosa) 추출물 및 다릅나무(Maackia amurensis) 추출물을 혼합한 복합 추출물로 포함될 수 있다.
상기 실크 펩타이드에 천연 추출물을 각각 혼합하는 경우에도, 인지 기능 개선 효과가 실크 펩타이드만을 단독으로 사용하는 경우에 비해 상승됨을 확인하였으나, 복합 추출물을 실크 펩타이드와 혼합하는 경우, 그 효과가 더욱 상승함을 확인하였다.
이때 복합 추출물은 보리지 추출물, 엉겅퀴 추출물, 짚신나물 추출물 및 다릅나무 추출물을 1:1:1:1의 중량 비율로 혼합하여 포함하는 것을 특징으로 한다. 동일 중량비로 혼합하여 사용하는 경우, 최상의 효과 상승 효과가 나타남을 확인하였다.
구체적으로, 일년봉, 보리지, 엉겅퀴, 짚신나물, 다릅나무 및 이들의 혼합물로 이루어진 군으로부터 선택된 천연물를 분쇄하는 단계; 유기 용매를 사용하여 상기 분쇄물을 침출시키는 단계; 시료를 침출 후 건조시키는 단계; 건조된 시료를 유기 용매를 사용하여 재 침출시키는 단계; 시료를 침출 후 건조시키는 단계; 물을 이용하여 침출시키는 단계; 및 침출하는 단계를 포함하여, 천연 추출물을 획득할 수 있다.
상기 유기 용매를 사용하여 추출한 천연 추출물을 유기 용매를 사용하여 분획을 실시하는 단계를 더 포함할 수 있다.
상기 추출물을 제조하는 방법은 초음파 추출법, 침출법 및 환류 추출법 등 당업계의 통상적인 추출 방법일 수 있다. 구체적으로 세척 및 건조로 이물질이 제거된 천연물을 물, 탄소수 1 내지 6의 알코올 또는 이들의 혼합 용매로 추출한 추출물일 수 있으며, 상기 용매들을 순차적으로 시료에 적용하여 추출한 추출물일 수도 있다.
상기 환류 추출법은 탄소수 1 내지 6의 알코올 100mL기준으로, 천연물의 분쇄물 10 내지 30g, 환류 시간 1 내지 3시간 및 50 내지 100%의 탄소수 1 내지 6의 알코올에 의한다. 보다 구체적으로, 탄소수 1 내지 6의 알코올 100mL기준으로, 천연물의 분쇄물 10 내지 20g, 환류 시간 1 내지 2시간 및 70 내지 90%의 탄소수 1 내지 4의 알코올에 의한 것이다.
상기 침출법은 15 내지 30℃, 24 내지 72시간 동안 및 50 내지 100%의 탄소수 1 내지 6의 알코올에 의한 것이다. 보다 구체적으로는 20 내지 25℃, 30 내지 54시간 동안 및 70 내지 80%의 탄소수 1 내지 6의 알코올에 의한 것이다.
상기 초음파 추출법은 30 내지 50℃, 0.5 내지 2.5시간 동안 및 50 내지 100%의 탄소수 1 내지 6의 알코올에 의한 것이다. 구체적으로는 40 내지 50℃, 1 내지 2.5시간 동안 및 70 내지 80%의 탄소수 1 내지 6의 알코올에 의한 것이다.
상기 추출 용매는 시료의 중량 기준으로 2 내지 50배를 사용할 수 있으며, 보다 구체적으로는 2 내지 20배이다. 추출을 위해 시료는 추출 용매에서 침출을 위해 1 내지 72 시간 동안 방치될 수 있으며, 보다 구체적으로 24 내지 48시간 동안 방치될 수 있다.
추출 후, 추출물은 새로운 분획 용매를 순차적으로 적용하여 분획할 수 있다. 분획시 사용하는 분획 용매는 상기 용매는 물, 헥산, 부탄올, 에틸아세트산, 에틸 아세테이트, 메틸렌클로라이드 및 이들의 혼합물로 이루어진 군으로부터 선택된 어느 하나 이상이며, 바람직하게는 에틸아세테이트 또는 메틸렌클로라이드이다.
추출물 또는 분획물을 얻은 후에는 농축 또는 동결건조 등의 방법을 추가적으로 사용할 수 있다.
본 발명의 조성물이 적용될 수 있는 질환의 구체적인 예는 신경 퇴행성 질환, 허혈 또는 재관류에 의한 질환 및 정신질환 등을 포함하나, 이에 한정되는 것은 아니다. 보다 구체적으로는 치매, 알츠하이머병, 헌팅톤 질병, 파킨슨씨 질병 및 근위 축성 측삭 경화증과 같은 신경 퇴행성 질환; 뇌졸중 (특히, 허혈성 뇌졸중)과 같은 허열 또는 재관류에 따른 신경세포의 손상에 의한 질환; 그리고 (c) 정신분열증 및 심적 외상후 스트레스 장애와 같은 정신질환 등의 예방 또는 치료에 이용될 수 있다.
본 발명의 일 실시예에 따른, 실크 펩타이드 및 천연 추출물을 포함하는 조성물은 허혈 또는 재관류에 따른 신경 세포의 손상에 의한 질환(예: 뇌졸중)의 예방 또는 치료에 특히 유용하다.
상기와 같은 효능은 대체적으로 실크 펩타이드 및 천연 추출물을 포함하는 조성물의 신경 세포 보호 작용을 통해 나타난다. 상기의 실크 펩타이드 및 천연 추출물을 포함하는 조성물에 의한 신경 세포의 보호 작용은 다양한 기전을 통해 발휘될 수 있으며, 예를 들어 신경 세포의 사멸을 억제함으로써 발휘되며, 이러한 신경 세포의 사멸은 신경 세포의 괴사 (necrosis) 및 아폽토시스 (apotosis)를 포함한다. 신경 세포의 아폽토시스를 억제하는 경우, 본 발명의 실크 펩타이드 및 천연 추출물을 포함하는 조성물의 타겟 중 하나는 카스파아제 (caspase, Guy et.al., Cell 91:443-446(1987))이며, 상기 효소의 활성을 억제하여 아폽토시스를 억제하게 된다.
상기의 실크 펩타이드 및 천연 추출물을 포함하는 조성물은 인지 기능 (cognitive function)의 증진에도 매우 우수한 효능을 나타낸다. 본 발명의 실크 펩타이드 및 천연 추출물을 포함하는 조성물은 인지 기능의 증진을 뒷받침하는 다양한 지표, 예컨대, 기억지수 (MQ), 학습 기울기, 기억유지도, 인출 효율성, 그리기/기억 일치도, 언어/시각 일치도, 지능/기억 일치도, 그리고 단기 기억과 주의 집중력에서 모두 우수한 효능을 발휘하여, 전체적으로 인간의 인지 기능을 크게 증진하는 작용을 한다.
또한, 본 발명의 일 실시예에 따른, 실크 펩타이드 및 천연 추출물을 포함하는 조성물은 상기한 신경성 질환에 수반하는 인지 기능의 악화를 개선하거나 예방하는 데 우수한 효능을 나타낸다.
특히, 실크 펩타이드 및 천연 추출물을 포함하는 조성물은 뇌에서의 아세틸콜린 양의 감소를 억제하여 인지 기능의 증진을 달성한다.
또한, 실크 펩타이드 및 천연 추출물을 포함하는 조성물은 뇌졸중에 의해 발생되는 신경 세포의 사멸을 억제하여 인지 기능의 손상을 방지한다.
본 발명의 일 실시예에 따르면, 본 발명의 실크 펩타이드 및 천연 추출물을 포함하는 조성물로부터 발휘되는 인지 기능의 개선은 학습 능력 및/또는 기억 능력의 증진이다.
아세틸콜린의 농도를 증가시켜 인지 능력의 향상과 치매 진행을 막아 치매 치료 및 예방에 탁월한 효과가 있는 것으로 알려지면서, 환자를 대상으로 콜린 효현제나 콜린 에스테라제 억제제를 사용하고 있다. 현재까지 개발된 약물들에는 아세틸콜린 전구체 (acetylcholine precursor)로 레시틴 (Lecithin), 수용체 활성제 (Receptor agonist)로 RS-86 및 니코틴 등이 있으며, 아세틸콜린 분해 억제제(acetylcholinesterase inhibitor)로 FDA의 승인을 받아 국내에서도 시판 사용 중인 타크린 (Tacrine)과 최근에 승인된 아리셉트 (Aricept) 등이 있으나, 효과가 일시적이고 미약하며 심각한 독성 때문에 아직 사용에 논란의 여지가 많은 상태이다. 그러나, 본 발명의 실크 펩타이드 및 천연 추출물을 포함하는 조성물은 인체에 대한 독성이 극히 미약하기 때문에 인지 기능을 증진하기 위한 의약 또는 기능성 식품으로서 매우 유용하다.
본 발명의 약제학적 조성물에 포함되는 약제학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로서, 탄수화물류 화합물 (예: 락토스, 아밀로스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 셀룰로스, 등), 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 염 용액, 알코올, 아라비아 고무, 폴리에틸렌 글리콜, 메틸 셀룰로스, 메틸히드록시 벤조에이트, 프로필히드록시 벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함하나, 이에 한정되는 것은 아니다.
본 발명의 약제학적 조성물은 경구 또는 비경구로 투여할 수 있고, 비경구 투여인 경우에는 정맥내 주입, 피하 주입, 근육 주입 등으로 투여할 수 있다.
본 발명의 약제학적 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하며, 보통으로 숙련된 의사는 소망하는 치료 또는 예방에 효과적인 투여량을 용이하게 결정 및 처방할 수 있다. 본 발명의 바람직한 구현예에 따르면, 적합한 투여량은 성인 기준으로 1일 1회 50 ㎎-10 g이다.
본 발명의 약제학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액 또는 유화액 형태이거나 엑스제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
한편, 본 발명의 기능성 식품 조성물은 식품 제조 시에 통상적으로 첨가되는 성분을 포함하며, 예를 들어, 단백질, 탄수화물, 지방, 영양소 및 조미제를 포함한다. 예컨대, 드링크제로 제조되는 경우에는 본 발명의 실크 펩타이드 및 천연 추출물 이외에 구연산, 액상과당, 설탕, 포도당, 초산, 사과산, 과즙, 두충 추출액, 대추 추출액, 감초 추출액 등을 추가로 포함시킬 수 있다. 식품에 대한 용이한 접근성을 고려한다면, 본 발명의 식품은 신경성 질환의 치료 또는 예방, 산화 스트레스에 의해 초래되는 질환의 치료 또는 예방 및 인지 기능의 개선에 매우 유용하다.
본 발명의 조성물은 상술한 바와 같이 다양한 효능을 나타낼 뿐만 아니라, 천연물인 실크 펩타이드 및 천연 추출물을 유효성분으로 포함하고 있기 때문에, 인체에 대한 부작용이 화학적 합성 의약보다 극히 적다.
본 발명의 실크 펩타이드를 포함하는 치매, 뇌졸증 및 인지 기능 개선용 조성물에 의하면 천연물 유래의 추출물을 활용하여 치매 및 뇌졸증의 예방 또는 치료 및 인지 기능의 개선에 효과가 있다.
본 발명의 실크 펩타이드 및 천연 추출물을 포함하는 인지 기능 개선용 조성물은 약제학적 조성물로 제공되어, 종래 실크 펩타이드만을 이용하는 경우에 비해, 인지 기능의 개선 효과가 높아진 조성물을 제공할 수 있다.
또한, 본 발명의 기능성 식품 조성물에 의하는 경우, 인지 기능의 개선 효과를 내면서도 기호도 높은 기능성 식품 조성물로 제공될 수 있다.
도 1은 허혈에 의한 뇌경색 부위에 대한 본 발명의 실크 펩타이드 및 천연 추출물의 혼합물에 의한 효과를 나타내는 그래프에 관한 것이다.

이하, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시예에 대하여 상세히 설명한다. 그러나 본 발명은 여러 가지 상이한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.
[제조예 1: 기능성 실크 펩타이드의 제조]
순수 실크 피브로인의 정제
견 단백질은 가잠 (Bombyx mori)을 전령 상엽으로 사육하여 얻은 누에고치를 정련하여 사용하였다. 우선, 세리신을 제거하기 위하여 고치 내에 남아있는 번데기를 제거하였고, 물 8 ℓ, 탄산나트륨 0.45 g 및 마르셀 비누 0.75 g을 혼합한 용액에 누에 고치 150 g을 넣어 40분씩 2번 비등 처리를 하였다. 그 후, 적당량의 물로 20분 정도 2번 다시 비등 처리를 한 후, 3회 정도 수세를 하여 건조를 하면 수용성의 세리신이 용해되어 나간다. 이때의 감소 무게는 37 g(약 25% 감소) 였다. 수세 후 자연 건조시켜 순수한 피브로인 단백질을 얻었다.
실크 펩타이드(BG101)의 제조
상기의 방법으로 얻은 피브로인 35 g을 기준으로 하여, 염화칼슘 (CaCl2, 1급, Mw=110.99)·H2O·에탄올(1:8:2 몰 함량)을 첨가하고 90℃에서 5시간 반응시켜 용해한 다음, 거즈 및 부직포로 이물질을 완전히 여과한 후 증류수로 2배 희석하였다. 직경 10 cm, 길이 1 m의 GradiFac 시스템 (Pharmacia Biotech, Sephadex G-25 Media, HiLoad P-50 Pump UV-1 Monitor, 스웨덴)을 사용한 겔 여과장치로 중성염을 제거하였다. 그런 다음, 얻은 피브로인 용액에 대하여 플라보자임 및 수미자임이 혼합(1:1)된 단백질 가수분해효소를 1% 가하고 55℃에서 5시간 가수분해를 행하였다. 그리고 나서, 100℃에서 5-10분간 열처리를 하여 효소의 활성을 없애고 동결 건조에 의하여 분말을 제조하였다.
[제조예 2: 천연 추출물의 제조]
(1) 보리지 추출물(BE)의 제조
1) 시료의 추출
건조된 보리지를 70% 에탄올을 사용하여 각각 상온에서 48 시간 동안 침출시킨 후 시료를 여과(filter)하여 농축하였다.
2) 분획물의 획득
70% 에탄올로 추출한 보리지 추출물 1g을 증류수 30ml에 녹인 후 헥산을 이용하여 3회 분획을 실시하고 분리된 물층을 다시 3회 분획하였다. 상기 3회 분획 단계에서 보리지 추출물은 메틸클로라이드를 이용하여 분획하였다. 3회 분획한 이후, 분리된 물층을 다시 에틸아세테이트을 이용하여 3회 분획하였으며, 분리된 물층을 다시 n-부탄올을 이용하여 3회 분획 하였으며, 얻어진 용액을 농축하여 각 분획물을 확보하였다.
(2) 기타 천연 추출물의 제조
상기 보리지 추출물과 동일한 방법을 이용하여 엉겅퀴(CE), 짚신나물(AE) 및 다릅나무(ME) 추출물을 제조하였다.
[제조예 3: 실크 펩타이드 및 천연 추출물을 포함하는 인지 기능 개선용 조성물의 제조]
상기 실크 펩타이드(BG101) 및 천연 추출물을 하기 표 1과 같은 조성 범위로 혼합하여 인지 기능 개선용 조성물을 제조하였다. MX는 보리지 추출물, 엉겅퀴 추출물, 짚신나물 추출물 및 다릅나무 추출물을 1:1:1:1의 중량비율로 혼합한 복합 혼합물이다.

[실험예 1: 허혈 동물 모델 실험]
상기 제조예에서 제조된 실크 펩타이드 및 천연 추출물을 포함하는 조성물이 허혈로 인한 뇌경색 면적에 미치는 영향을 확인하기 위해 하기의 실험을 진행하였다.
실험예 1-1: 국부적 허혈 유도 동물 모델
A. 약물 처리 및 국부적 허혈 유도 동물 모델의 제작
체중 200 내지 250 g인 웅성 백서 (Sprague-Dawley rat)를 이용하여 국부적 허혈 유도 동물 모델을 제작하였다. 약물은 실험번호 S1 내지 S11 1g/㎏을 허혈 유도 1시간 전에 1회 또는 허혈 유도 1시간 후에 1회 구강 투여하였고(구강 투여군, n=6), 허혈 대조군(n=6)은 약물처리군과 같은 용량의 BG101을 투여하였다. 실험동물에 케타민(ketamine) 30 내지 40 ㎎/㎏을 근육주사 하여 마취시킨 다음에 앙와위에서 목부분에 피부절개를 가하고 경동맥(common carotid artery), 외경동맥(external carotid artery) 및 내경동맥 (internal carotid artery)을 찾아 주위조직과 분리시켰다. 우선 외경동맥의 분지인 상부 상갑선 동맥과 후두와 동맥을 전기 소작시키고, 내경동맥의 분지인 익돌구개(pterygopalatine) 동맥을 전기소작하고 외경동맥을 잘라 4-0 나일론실(ETHICON, INC)을 외경동맥을 통하여 내경동맥으로 삽입한 다음에 경동맥 분지에서 나일론 실이 16 내지 18 ㎜가 들어가게 하여 위치시켜 중대뇌 동맥(Middle Cerebral Artery) 기시부를 폐색하였다.
B. 허혈에 의한 뇌경색 부위 감소 효과 실험
허혈 유도 6시간 후 동물을 단두하여 뇌를 적출하였다. 적출된 뇌는 전방극 (anterior pole)으로부터 2 ㎜ 간격으로 절편하고, 2% 트리페닐 테트라졸리움 클로라이드 (triphenyl tetrazolium chloride: TTC, Sigma) 용액으로 37℃에서 30분간 반응시킨 후 4% 파라포름알데히드(Sigma) 용액에 넣어 고정시켰다. 염색된 조직절편을 촬영한 후 사진을 MCID image processing system (Imaging Research Inc.)을 이용하여 적색으로 염색된 정상부위와 달리 뇌경색이 발생하여 백색으로 변한 부위의 면적을 측정하였으며, 전체 뇌 절편의 면적 대비 뇌경색 면적 비율을 구한 후 평균치를 계산하였다. 또한 각각의 뇌 절편에서 전체 부피에서 뇌경색 부위가 차지하는 부피의 비율을 측정하였다.
본 발명의 실크 펩타이드는 허혈에 의한 뇌경색 부위를 유의성 있는 수준으로 감소시키고 있다. 도 1에서 각각의 뇌 절편을 비교한 결과에서 뇌경색 감소 효과가 특정 부위에 치중되어 있지 않고 전반적으로 나타남을 관찰하였다(P<0.05). 또한, 도 2에서 전체 뇌 절편의 부피 대비 뇌경색 부피 비율을 측정해본 결과, BG101과 비교하여 S1 내지 S11의 전처리가 국부적 허혈에 의한 뇌경색 부위 감소에 유의한 효과를 보였다. 첨부한 도 1 및 도 2에서 각 측정값은 평균±표준오차를 나타내는 것이다. 따라서, 본 발명의 실크 펩타이드는 허혈성 뇌졸중의 예방과 치료에 관련된 기능성 식품 및 약물에 유용하게 사용될 수 있음을 알 수 있다.
[실험예 2: 인지기능 개선에 대한 동물 모델 실험]
실험예 2-1: 실험 동물 준비
실험 동물은 웅성 백서 (Sprague Dawley, 140 내지 180 g, 대한실험동물센터)로서, 이들은 일정한 환경 (실내온도 25±1℃, 상대습도 60±10%)에서 일주일 동안 하나의 사육 상자 당 4 마리씩 넣어 물은 제한 없이 제공하였으며, 이때, 몸무게의 80%를 유지하도록 제한 급식을 실시하였다. 본 실험에서는 총 50 마리를 사용하였다. 훈련 3일 전부터 5분씩 미로에 적응시켜 검사를 진행하였다.
실험예 2-2. 교대 지연 검사(delayed alternation test)
총 세 개의 통로(왼쪽, 오른쪽, 중앙부)가 있으며, 각 통로 입구는 실험자가 동물의 출입을 통제할 수 있도록 칸막이를 설치한 T자 모양의 미로를 준비하였다.
첫 시행에서는 출발 상자에 1분간 적응시켰다. 첫 시행은 '강제 선택 시행'으로, 칸막이로 한쪽 통로를 막아놓고 반대편 통로에 동물이 들어가면 칸막이로 후방을 막았다. 동물의 사지가 통로 입구를 통과하면 동물이 들어간 것으로 간주하였다. 60초간 동물이 해당 통로에서 먹이를 먹도록 하였다.
다음 시행부터는 '자유 선택 시행'으로 동물이 출발상자에서 출발하여 자유롭게 통로의 방향(왼쪽 혹은 오른쪽)을 선택하도록 하였다. 동물은 바로 이전의 시행에서 들어갔던 통로와 반대편 통로로 들어갔을 경우에만 먹이 보상을 하였다.
시행 간격은 5초 및 20초로 지연 시간(delay)를 두고 구성되며, 매 시행의 제한시간(cut-off time)은 60초로 하였다. 동물이 강화가 주어지는 통로에 정확하게 들어간 횟수와 통로까지 들어가는데 걸린 시간을 측정하였다. 하루에 10 시행식 9일 동안 동일한 절차를 수행하여 평가하였다.
투여 방법
본 발명의 실험번호 BG101, S1 내지 S11을 100 ㎎/㎖의 용액으로 제조하여 구강 투여하였다. 모든 시료들은 증류수에 용해하여 구강 투여하였다.
인지 기억 평가 결과
BG101, S1 내지 S11을 투여한 군에 대해 T-maze 실험을 진행한 결과는 도 3 및 도 4와 같다. 도 3은 시행 간격을 5초로 구성하여 실험을 진행한 것으로, 9일 간의 훈련 기간 동안 성능이 점차적으로 향상되었다. 특히, S6 및 S7 투여 군에서 우수한 성능 향상 효과를 확인하였다.
도 4는 시행 간격을 20초로 구성하여 실험을 진행한 것으로, 9일 간의 훈련 기간 동안 성능이 점차적으로 향상됨을 확인하였으나, 5초로 구성하였을 때와 비교해서 성능 향상 효과가 적게 나타나는 것을 확인하였다. S6 및 S7 투여 군에서는 시행 간격을 5초로 구성하였을때와 마찬가지로 우수한 성능 향상 효과를 확인하였으나, 다른 투여 군에서는 성능 향상 효과가 일부 나타났으나, S6 및 S7 투여 군에 못 미치는 것으로 확인되었다.
이상에서 본 발명의 바람직한 실시예에 대하여 상세하게 설명하였지만 본 발명의 권리범위는 이에 한정되는 것은 아니고 다음의 청구범위에서 정의하고 있는 본 발명의 기본 개념을 이용한 당업자의 여러 변형 및 개량 형태 또한 본 발명의 권리범위에 속하는 것이다.
