따라서 피부미용관점에서는 피하지방을 이식할 때, 이식되는 지방조직을 유지하거나 증가시킬 수 있는 방법이 필요하다. 또한 이식이 아닌 다른 방법으로 지방세포를 증가시켜 피하지방층을 증가시킴으로써 피하지방이 감소되는 것을 억제하는 것이 필요하다. 이러한 피하지방층 증가를 주름개선 및 피부의 처짐 방지 등의 미용성형 분야에 유용하게 사용될 수 있다 (Hu S et al., Ann Plast Surg. 2007 Jun;58(6):602-607; Yoshimura K et al., Aesthetic Plast Surg. 2008 ;32(1):48-55). 최근 피부 진피와 피하지방의 지방조직이 외부 세균이나 피부 상재균으로부터 피부를 보호하는 자연면역(natural immunity, innate immunity)에 중요한 역할을 한다는 사실이 알려져 있다 (Zhang et al., Science. 2015, 347:67-71). 상기 실시예 2에서와 같이 중간엽줄기세포로부터 지방세포의 분화를 유도한 후 마지막 배지 교체를 한 후 이틀 뒤 세포배양 상층액에서 아디포넥틴의 발현양을 측정하여, 지방분화 촉진 효과에 대한 정량적 비교를 실시하였다. 아디포넥틴(adiponectin)은 지방세포에서 분비되는 대표적인 아디포카인(adipokine) 중 하나로서, 항당뇨작용을 비롯하여 다양한 효능을 나타내는 것으로 보고되어 있다. 예를 들어 당뇨병에 있어서 아디포넥틴은 인슐린 감수성을 증가시켜 혈당을 낮춤으로써 당뇨병을 예방하는 효능을 나타낸다. 이러한 아디포넥틴은 지방세포가 분화되면서 발현이 증가하는 단백질로 알려져 있다. 아디포넥틴과 다른 사이토카인의 측정은 알엔디시스템사(R&D Systems, Minneapolis, MN, USA)의 DuoSet Kit를 제조자의 방법대로 사용하였다. 결과로 도 2에서 볼 수 있는 바와 같이, IDX 에 의해 지방세포로의 분화를 유도한 경우, 솜나물 추출물이 처리되지 않은 것에 비해 중간엽줄기세포의 지방세포 분화가 솜나물 추출물이 처리된 실험군에서 아디포넥틴의 발현양이 통계적으로 유의성 있게 증가하는 것으로 나타났다. 통계는 일원분산분석(ANOVA) 후, post hoc test를 수행하여 검증하였으며, p 값이 0.05 이하일 때 유의적인 차이가 있는 것으로 표시하였다.
