Abstract
Description
도 2는 정상 인간 멜라노사이트(normal human melanocytes; NHMs) 및 생체 외(ex vivo) 인간 피부 상에서 내피세포의 효과를 나타낸다. (A) HUVECs 및 NHMs의 최적 공동-배양 배지를 선별하기 위해서, HUVECs 및 NHMs는 HGM, F12 또는 MCDB153에서 3일 동안 배양되었다. 세포 형태가 확인되었고(왼쪽 패널), 세포독성은 MTT 분석을 사용하여 분석하였다(오른쪽 패널). (B) 트랜스웰 플레이트를 사용하여 NHMs은 HUVECs와 함께 MCDB153 배지에서 5일 동안 공동 배양되었다. 멜라닌 함량 및 티로시나제 활성을 측정하였다. (C) mRNA 및 MITF와 티로시나제 단백질의 발현 수준을 나타낸다. (D) NHMs은 HUVEC-유래 CM으로 5일 동안 처리되었고, 멜라닌 함량 및 티로시나제 활성 수준을 측정하였다. (E) 생체 외(ex vivo) 인간 피부는 HUVEC-유래 CM으로 3일 동안 유지하였다. 표피 부위 당 색소 부위(pigmented area per epidermal area; PA/EA) 비율 수치를 측정하였다(Fontana-Masson stain). 모든 수치는 3번의 독립된 실험의 평균±SD로 나타냈다.
도 3은 여러 배지 조성을 변경하여 HUVEC 및 멜라노사이트의 증식 효과를 확인한 결과이다.
도 4는 색소 형성에 있어서 UV-조사된 내피세포의 효과를 나타낸다. (A) Bend3 세포는 UVB 50 mJ/cm2로 조사되었고, 세포 형태를 확인하였다. (B) 세포 펠렛 분석 결과를 나타낸다. B16 melanoma 세포는 UV-조사 또는 비-조사된 Bend3 세포 유래 CCM으로 3일 동안 처리하였다. (C) 세포 펠렛 상에서 멜라닌 함량 및 티로시나제 활성 수준(왼쪽 패널) 및 MITF와 티로시나제의 단백질 발현(오른쪽 패널)을 분석하였다. (D) NHMs는 UV-조사된 HUVECs 유래 CCM으로 5일 동안 처리하였고, 멜라닌 함량 및 티로시나제 활성 수준을 측정하였다.
도 5(A)는 UV-조사된 HUVECs 유래 CCM에서 3일 동안 유지된 생체 외(ex vivo) 인간 피부를 나타내고, 도 5(B)는 UV-조사된 HUVECs 존재하에서 3일 동안 유지된 생체 외(ex vivo) 인간 피부를 나타낸다. 표피 부위 당 색소 부위(pigmented area per epidermal area; PA/EA) 비율 수치를 측정하였다(Fontana-Masson stain). 모든 수치는 3번의 독립된 실험의 평균±SD로 나타냈다.
도 6은 인간 진피 미세 혈관내피세포(Human dermal microvascular endothelial cells; HDMEC)에 UVA 처리 후 배양액을 걷어 농축시킨 후, 멜라노사이트에 3일 처리한 결과를 나타낸다.
도 7은 UV-조사된 내피세포에 의해 분비되는 멜라닌 생성 인자로서 엔도텔린-1(endothelin-1)의 확인 결과를 나타낸다. (A) RNA 서열 분석을 통한 UV-조사 및 비-조사된 Bend3 세포의 차등적 유전자 발현 프로파일링을 나타낸다. (B) UV-조사 또는 비-조사된 Bend3 세포의 ET-1 mRNA의 발현 수준을 실시간 PCR 및 RT-PCR로 분석하였다. (C) B16 melanoma 세포는 UV-조사된 CCM과 함께 BQ-123(1μM)을 3일 동안 처리하거나 처리하지 않았다. 멜라닌 함량 및 티로시나제 활성 수준을 측정하였다. 모든 수치는 3번의 독립된 실험의 평균±SD로 나타냈다.
Claims (14)
- 혈관내피세포 또는 이의 배양물을 유효성분으로 포함하는 시험관 내(in vitro) 피부 색소 형성 억제용 시약 조성물.
- 제1항에 있어서, 상기 혈관내피세포는 Bend3 세포, 인간 제대정맥내피세포(Human umbilical vein endothelial cells; HUVEC), 인간 진피 미세 혈관내피세포(Human dermal microvascular endothelial cells; HMVEC-D 또는 HDMEC), 인간 심장 내피세포(Human cardiac endothelial cells), 인간 대동맥 내피세포(Human aortic endothelial cells; HAEC), 인간 관상 및 장골 동맥 내피세포(Human coronary and iliac artery endothelial cells; HIAEC 또는 HCAEC), 인간 심장 미세 혈관내피세포(Human cardiac microvascular endothelial cells), 인간 방광 및 자궁 미세 혈관내피세포(Human bladder and uterine microvascular endothelial cells), 인간 폐동맥 내피세포(Human pulmonary artery endothelial cells; HPAEC 또는 PASMC), 인간 폐 미세 혈관내피세포(Human lung microvascular endothelial cells; HMVEC-L), 인간 완두 동맥 내피세포(Human brachiocephalic artery endothelial cells; HBcAEC), 인간 목동맥 내피세포(Human carotid artery endothelial cells; HCtAEC), 인간 진피 림프 미세 혈관내피세포(Human dermal lymphatic microvascular endothelial cells; HDLMVEC), 인간 내흉 동맥 내피세포(Human internal thoracic artery endothelial cells; HITAEC), 인간 쇄골하 동맥 내피세포(Human subclavian artery endothelial cells; HScAEC) 및 인간 제대 동맥 내피세포(Human umbilical artery endothelial cells; HUAEC)로 이루어진 군에서 선택되는 어느 하나인 것을 특징으로 하는 시험관 내(in vitro) 피부 색소 형성 억제용 시약 조성물.
- 제1항에 있어서, 상기 피부 색소는 멜라닌인 것을 특징으로 하는 시험관 내(in vitro) 피부 색소 형성 억제용 시약 조성물.
- 혈관내피세포 또는 이의 배양물을 유효성분으로 포함하는 과색소침착증 예방 또는 치료용 조성물.
- 혈관내피세포 또는 이의 배양물을 유효성분으로 포함하는 피부 미백용 화장료 조성물.
- UV-조사된 혈관내피세포 또는 이의 배양물을 유효성분으로 포함하는 시험관 내(in vitro) 피부 색소 형성 유도용 시약 조성물.

- 제6항에 있어서, 상기 UV-조사된 혈관내피세포는 엔도텔린-1(Endothelin-1; ET-1), 조직 형태 플라스미노겐 활성화인자(Tissue type plasminogen activator; tPA), ET-1 수용체 a(ET-1 receptor a; ET-1Ra), 골 형성 단백질 2(Bone morphogenetic protein 2; BMP-2) 및 간세포 성장인자(Hepatocyte growth factor; HGF) 유전자가 상향 발현되고, ET-1 수용체 b(ET-1 receptor b; ET-1Rb), 혈관내피 성장인자(Vascular endothelial growth factor; VEGF) 및 기본 섬유아세포 성장인자(Basic fibroblast growth factor; bFGF) 유전자가 하향 발현되는 것을 특징으로 하는 시험관 내(in vitro) 피부 색소 형성 유도용 시약 조성물.
- 제6항에 있어서, 상기 피부 색소는 멜라닌인 것을 특징으로 하는 시험관 내(in vitro) 피부 색소 형성 유도용 시약 조성물.
- 제6항에 있어서, 상기 UV-조사는 20 내지 100 mJ/cm2의 UVA 또는 UVB로 조사하는 것을 특징으로 하는 시험관 내(in vitro) 피부 색소 형성 유도용 시약 조성물.
- UV-조사된 혈관내피세포 또는 이의 배양물을 유효성분으로 포함하는 백반증 또는 백모증 예방 또는 치료용 조성물.
- (1) 혈관내피세포에 UV를 조사하는 단계;
(2) 상기 UV-조사된 혈관내피세포를 멜라닌 세포와 공동배양하거나, 상기 UV-조사된 혈관내피세포의 배양물을 멜라닌 세포에 혼합하여 배양하는 단계;
(3) 상기 (2) 단계의 공동배양물 또는 혼합배양물에 시험물질을 접촉시키는 단계;
(4) 상기 시험물질을 접촉한 공동배양물 또는 혼합배양물의 멜라닌 함량 또는 티로시나제 활성을 측정하는 단계; 및
