생후 7~8주령인 위스타 랫 수컷을 에틸에테르로 흡입마취한 후 경추도살하고 전립선을 떼어내서 액체질소로 급랭하였다. 전립선의 무게를 측정하여 0.32mM 수크로오스(sucrose;Amresco Inc, USA)와 1mM 디티오트레이톨(Dithiothreitol; Sigma, USA)을 함유한 pH 6.5의 20mM photassium phosphate buffer를 1g당 5ml씩 가하였다. 전립선을 호모게나이저(homogenizer)로 분쇄한 후 1000rpm으로 10분간 원심분리하여 여과하였다. 상층액을 회수하여 초원심분리기(Ultra centrifuge; Beckman, USA)로 원심분리한 후, 침전물을 회수하여 20mM photassium phosphate buffer를 가하여 녹였으며, 이것을 5알파-환원효소 분획으로 사용하였다. 단백질농도는 BSA(Bovine serum albumin)를 표준화하여 Bio-Rad protein assay kit(Bio-Rad, USA)를 사용하여 정량하였다. 에펜도르프튜브(Eppendorf tube)에 상기의 5알파-환원효소 분획 250㎍과 120nCi[1,2,6,7-3H]-testosterone, 200μM NADPH(Calbiochem, US), 제조예 1~5의 추출물을 pH 6.6의 40mM potassium phosphate buffer로 첨가해서 500㎕로 만들었다. 37℃에서 30분 동안 반응시킨 후 1ml의 초산에틸을 넣고 1000rpm에서 10분간 원심분리 하였다. 상층액만을 회수하여 감압건조한 후, 50㎕의 초산에틸을 가하여 녹이고, TLC(Thin layer chromatography, Merck, Germany)판에 점적하였다. 전개용매로는 50:50(초산에틸:사이클로헥산)을 사용하였다. 테스토스테론(Testosterone)은 UV램프(UVGL-58, Upland, USA)를 사용하여 254nm에서 확인하였고, 디하이드로테스토스테론(dihydrotestosterone ; DHT)은 10% 황산을 뿌린 후 가열하여 확인하였다. TLC판에서 테스토스테론 및 디하이드로테스토스테론 부위를 오려내어 신틸레이션 바이알(scintilation vial)에 넣고 5ml의 cocktail을 넣은 후, LSC(Liquid scintilation counter, Packard Instruments, USA)를 사용하여 cpm(counter per minute)을 측정하였다. 5알파-환원효소 활성에 의한 테스토스테론의 전환율을 DHT/(testosterone+DHT)의 비로 계산하여 대조군에 대한 %로 나타내었다(도 2). 도 2에서 확인되는 바와 같이 상기 제조예의 추출물에 의하여 5알파-환원효소의 활성이 감소하였으며, 특히 제조예 5의 추출물에 의하여 대조군과 비교하여 5알파-환원효소 활성이 크게 감소하였다.
