Claims (8)
- 효모균주를 숙주로 하여 재조합 인체 부갑상선호르몬(hPTH)을 생산하는 방법에 있어서, 앱신계통의 단백질 분해효소 YPS1, YPS2, YPS3에서 선택된 적어도 하나가 결손된 효모균주를 숙주로 하여 부갑상선 호르몬 유전자(hPTH)를 함유하는 발현벡터로 형질전환시켜 배양함으로써 hPTH를 생산함을 특징으로 하는 재조합 인체 부갑상선 호르몬의 생산방법.
- 제 1항에 있어서, 상기 단백질 분해효소의 결손은 YPS1, YPS2, YPS3에서 선택된 적어도 하나의 유전자를 효모선별표지를 사용하여 파쇄한 것임을 특징으로 하는 재조합 인체 부갑상선 호르몬의 생산방법.
- 제 1항 또는 제 2항에 있어서, YPS1과 YPS3 두 유전자가 동시에 파쇄된 효모변이주를 숙주로 한 것임을 특징으로 하는 재조합 인체 부갑상선 호르몬의 생산방법.
- 제 1항 또는 제 2항에 있어서, YPS1, YPS2, YPS3 세 유전자 모두가 동시에 파쇄된 효모변이주를 숙주로 한 것임을 특징으로 하는 재조합 인체 부갑상선 호르몬의 생산방법.
- 효모 yps1Δyps3Δ변이주를 숙주로 하고 상기 숙주에 인체 부갑상선 호르몬 유전자(hPTH)를 포함하는 발현벡터로 형질전환된 형질전환효모.
- 제 5항에 있어서, 상기 형질전환효모는 인체 부갑상선 호르몬 발현벡터 pG10-hPTH1으로 형질전환된 효모 사카로마이세스 세레비지애 SLH16/pG10-hPTH(KCTC 0815BP)임을 특징으로 하는 형질전환효모.
- 효모 yps1Δyps2Δyps3Δ변이주를 숙주로 하고 상기 숙주에 인체 부갑상선 호르몬 유전자(hPTH)를 포함하는 발현벡터로 형질전환된 형질전환효모.
- 제 7항에 있어서, 상기 형질전환효모는 인체 부갑상선 호르몬 발현벡터 pG10-hPTH1으로 형질전환된 효모 사카로마이세스 세레비지애 SLH18/pG10-hPTH(KCTC 0816BP)임을 특징으로 하는 형질전환효모.
