Abstract
비오틴 생산 능력을 갖는 스핀고모나스 속 미생물의 배양액을 배지에 제조하고, 배지에 생산되고 축적된 비오틴을 수집하는 것으로 구성된 비오틴의 제조방법이 개시되어 있다. 또한, 본 발명의 제조방법을 위해 유용한 비오틴 생산능력을 갖는 스핀고모나스 속 미생물이 개시되어 있다.

본 발명은 비오틴의 제조방법 및 이 방법에서 사용하기 위한 미생물에 관한 것이다.
비오틴은 인간, 동물, 식물 및 각종 미생물에서 요구되는 비타민의 하나이다. 미생물을 이용한 비오틴의 제조방법으로는, 종래, 스트렙토마이세스(Streptomyces) 또는 미크로모노스포어(Micromonospore)(일본국 특허 공고 제41-21756호); 스포로보로마이세스(Sporobolomyces)(일본국 특허 공고 제42-3074호); 바실러스(Bacillus)(일본국 특허 공개 제56-160998호); 에스케리키아(Escherichia)(일본국 특허 공개 제61-149091호); 세라티아(Serratia)(일본국 특허 공개 제2-27980호); 및 브레비박테리움(Brevibacterium)(일본국 특허 공개 제3-240489호) 속의 미생물과 같은 미생물을 사용한 각종 방법이 공지되어 있다.
그러나, 종래의 방법들은, 이러한 미생물에 의해 생산되는 비오틴의 수율이 낮기 때문에, 항상 만족스러운 것은 아니다. 따라서, 효능이 더욱 높은 미생물을 사용하여, 공업적 규모로 비오틴을 제조하는데 적절한 비오틴의 제조방법을 개발하는 것이 요망되어 왔다.
이러한 상황하에서, 본 발명자들은, 미생물의 침착 설비내에 유지된 천연의 각종 배양 수집물에 대하여, 증가된 비오틴 생산 능력을 갖는 미생물을 집중적으로 탐구하였다. 그 결과, 본 발명자들은 스핀고모나스(Sphingomonas) 속의 미생물이 매우 많은 양의 비오틴을 배지에 축적할 수 있음을 알아내었으며, 이에 의해 본 발명을 완성하였다.
즉, 본 발명은 비오틴 생산능력을 갖는 스핀고모나스 속 미생물을 적절한 배지에 배양하고, 배지에 축적된 비오틴을 수거함을 특징으로 하는 비오틴의 제조 방법을 제공한다. 또한, 이 방법에서 사용하기 위한 미생물이 제공된다.
본 발명의 방법에서 사용하기 위한 미생물은 비오틴 생산능력을 갖는 스핀고모나스 속 미생물이며, 그의 예로는 스핀고모나스 아드해시바(Sphingomonas adhaeshiva), 스핀고모나스 파라포시모빌리스(Sphingomonas parapaucimobilis), 및 스핀고모나스 포시모닐리스(Sphingomonas paucimonilis)가 있다. 이들 미생물중에서, 스핀고모나스 sp. SC-42405가 가장 바람직하다. 일반적으로, 스핀고모나스 속 미생물은 예를 들어 하기 특징을 갖는다:
(1) 그람-음성 간균이고; (2) 카탈라제 시험에서 양성을 나타내고; (3) 황색 콜로니가 주로 형성되고; (4) 인지질로서 스핀고리피드를 함유하고; (5) 이소프레노이드 퀴논계가 Q10이고; (6) 세균 지방산으로서 2-히드록시 산을 함유하며; (7) DNA의 G+C몰%가 약 60~70의 범위이다.
스핀고모나스 sp. SC-42405는 본 발명자들에 의해 일본국 오이따껭 토양에서 분리된 세균으로부터 단리된 미생물이며, 매우 많은 양의 비오틴을 배지에 축적하는 것으로 발견되었다. 이 미생물은 부다페스트 조약하에 일본국 통상 산업성 미생물 공업 기술 연구소(“생명 공학 공업 기술 연구소”로 명칭이 바뀜)에 1992년 9월 3일자로 수탁번호 FERM BP-3995로 기탁되었다.
스핀고모나스 sp. SC-42405의 세균학적 특징은 다음과 같다:

Bacteriol., 40, 320~321(1990)에 개시된 데이타와 비교하고, 본 발명자들에 의해 발견된 세균 균주를 스핀고모나스 속 미생물로 동정하였으며, 스핀고모나스 sp. SC-42405라 명명하였다. 아직까지, 비오틴 생산능력을 갖는 어떠한 스핀고모나스 속 미생물도 공지되지 않았음을 주지해야 한다. 이러한 점에서, 스핀고모나스 sp. SC-42405는 신규 균주인 것으로 간주된다. 또한, 이 균주의 변이주, 즉 스핀고모나스 sp. SC-42405에서 유래된 변이주, 세포 융합 균주 또는 재조합 균주의 어느것이라도 본 발명의 방법을 위해 사용될 수 있다.
본 발명의 방법에서 사용하기 위한 미생물의 배양액은 탄소 및 질소원, 유기 및/또는 무기염 등을 함유하는 각종 배지에서 제조될 수 있으며, 이들 배지는 모두 통상의 세균 배양액을 제조하기 위해 널리 사용된다.
탄소원의 예로는 글루코스, 말토스, 글리세롤, 전분, 덱스트린, 슈크로스, 동물성 및 식물성유, 및 당밀이 있다.
질소원의 예로는 브로쓰, 펩톤, 효모 추출물, 맥아 추출물, 대두분, 옥수수 침지액, 면실 분말, 건조 효모 및 카사미노 산과 같은 천연 유기 질소원; 및 암모니아, 염화 암모늄, 질산 나트륨, 질산 암모늄, 황산 암모늄, 아세트산 암모늄 및 우레아와 같은 기타 유기 또는 무기 질소원이 있다. 천연 유기질소원은 또한 탄소원으로 이동될 수도 있다.
유기 및 무기염의 예로는 칼륨, 나트륨, 마그네슘, 철, 망간, 코발트 및 아연과 같은 원소의 염화물, 황산염, 아세트산염, 탄산염 및 인산염이 있다. 이들의 특정예로는 염화 칼륨, 염화 나트륨, 황산 마그네슘, 황산 제1철, 황산 망간, 염화 코발트, 황산 아연, 황산구리, 아세트산 나트륨, 탄산 칼슘, 탄산 나트륨, 인산 일수소칼륨, 인산이수소칼륨, 인산일수소나트륨 및 인산이수소 나트륨이 있다.
본 발명의 방법에서 사용되는 미생물은, 비오틴을 공업적 규모로 제조하기 위해 유리한 각종 배지에서, 뛰어난 비오틴 생산 능력을 갖는다. 본 발명의 방법에서 사용되는 미생물의 비오틴 생산 능력을 증가시키기 위하여, 알라닌과 같은 아미노산 또는 피멜산과 같은 지방산을 배양 배지에 첨가할 수도 있다. 예를 들면, 이 화합물을 배양 배지 1000㎖당 약 1㎎ 내지 약 5㎎의 양으로 사용할 수도 있다.
본 발명의 방법에서 사용되는 미생물의 배양액은, 통상의 세균에 대해 사용되는 편리한 방법에 따라서, 시험관, 왕복 진탕기 또는 회전 진탕기를 이용한 진탕 배양액 및 통 발효기 또는 발효 탱크를 이용한 기타 배양액과 같은 고체 배양액 또는 액체 배양액의 형태로 제조된다.
미생물의 배양액을 통상 호기 조건하에서 배양한다. 특히, 통 발효기 또는 발효 탱크를 사용할때, 무균 공기를 통상 1분당 배양액 부피의 약 0.1 내지 약 2배의 비율로 도입하는 것이 필요하다.
배양 온도는 사용되는 미생물이 배양액에서 생육할 수 있는 온도 범위내에서 변할 수도 있다. 예를 들어, 배양액을 약 20℃ 내지 약 40℃, 바람직하게는 약 25℃ 내지 약 35℃, 더욱 바람직하게는 약 28℃ 내지 약 33℃의 온도에서 배양한다. 바람직하게는, 배양 pH를 약 6 내지 약 8로 조절한다.
배양 시간은 다양한 조건에 의존하여 변할 수도 있고, 배양액을 통상 약 1 내지 약 7일, 바람직하게는 약 3 내지 약 4일동안 배양한다.
배양의 완결후에, 천연원으로 부터 목적 화합물을 추출 및 정제하기 위한 종래의 방법에 따라서, 비오틴의 특성을 이용함으로써 배양액으로부터 비오틴을 수집한다. 예를 들면, 원심분리와 같은 기술에 의해 배양액으로부터 세균 세포를 제거하고, 배양액에 축적된 비오틴을 활성목탄 또는 거대 공극 비-이온성 교환 수지상에 흡착시킨 다음, 이온 교환 수지 또는 비오틴과 불순물간의 용해도 차를 이용한 재결정법에 의해 정제한다.
본 발명을 하기 실시예에 의해 더욱 예증할 것이며, 실시예가 본 발명의 범위를 제한하는 것으로는 결코 해석하지 말아야 한다.
[실시예 1]
[진탕 배양에 의한 비오틴의 생산]
1 백금이의 스핀고모나스 sp. SC-42405(FERM BP-3995)를 10㎖의 예비-배양 배지(1% 글리세롤, 2% 폴리펩톤, 0.15% K2HPO4, 0.15% MgSO4·7H2O, pH 7.2)를 함유한 큰 시험관(22㎜φ× 220㎜)에 접종하고, 200rpm으로 진탕하면서 3일동안 30℃에서 배양한다. 이어서, 이 예비-배양액의 2㎖를 50㎖의 배양배지(2% 글리세롤, 2% 효모추출물, 0.5% 비타민-유리 카사미노산, 0.15% K2HPO4, 0.05% KCl, 0.05% MgSO4·7H2O, 0.001% FeSO4·7H2O, 0.001% MnSO4·4-6H2O, 20㎍/ℓ 티아민 히드로클로라이드, pH 7.0)을 함유한 500㎖ 사까구찌 플라스크에 접종하고, 130rpm으로 진탕하면서 4일동안 30℃에서 배양한다. 배양이 끝난후 균체 농도(OD650)은 22.0이며, 이때, 배지에 생산되고 축적된 비오틴의 농도를 락토바실러스 플란타룸(Lactobacillus plantarum) IFO 3070을 이용한 정량적 미생물 생물검정(이즈미 및 야마다, Methods of Vitaminological Experiments II, Water-soluble Vitamins, pp. 481~499, Vitaminology Society of Japan, 도꾜 가가꾸 도오진 발생, 1985년)에 의해 결정한 결과, 3.3㎎/ℓ임을 알수 있었다.
[실시예 2]
[통기 교반 배양에 의한 비오틴의 생산]
큰 시험관내에서 스핀고모나스 sp. SC-42405의 배양액을 실시예 1에 기재된 것과 동일한 방식으로 예비-배양한다. 예비-배양액을 100㎖의 동일한 예비-배양 배지를 각각 함유하는 세개의 500㎖ 사까구찌 플라스크내에 2㎖ 분량씩 접종하고, 130rpm으로 진탕하면서 3일동안 30℃에서 배양한다. 이어서, 이 배양액의 300㎖를 15ℓ의 배양배지(2% 글리세롤, 2% 효모추출물, 0.5% 비타민-유리카사미노 산, 0.15% K2HPO4, 0.05% KCl, 0.05% MgSO4·7H2O, 0.001% FeSO4·7H2O, 0.001% MnSO4·4-6H2O, 20㎍/ℓ 티아민 히드로클로라이드, pH 7.0)를 함유하는 30ℓ 통발효기내에 접종하고, 통기 및 교반하면서 4일동안 30℃에서 배양한다. 통기 속도는 15ℓ/분이고, 교반 속도는 200~500rpm이다(배지중의 용존 산소의 농도가 2ppm 이상이 되도록 조절됨). 기포 억제제 Einol (Biotto Co.)를 첨가함으로써 자기-기포제거를 달성한다. 배양이 끝난 후 균체 농도(OD650)는 38.0이며, 이때 배지에 생산되고 축적된 비오틴의 농도를 실시예 1에 기재된 것과 동일한 정략적 미생물 생물 검정에 의해 결정한 결과 2.3㎎/ℓ임을 알수 있었다.
[실시예 3]
[생성물의 단리 및 동정]
실시예 2에서 수득된 15ℓ의 배양액을 연속 원심 분리하고, HCl의 첨가에 의해 pH 3으로 조정한 후, 상층액을 500g의 활성탄(Wako Pure Chemical Industries, Ltd)으로 충진된 컬럼을 통해 통과시킨다. 컬럼을 15ℓ의 물로 세척한 후, 5ℓ의 0.1N 암모니아 수로 용출시킨다. 용출액을 농축하고, 100㎖의 도웩스 1 × 8(The Dow Chemical Co.)로 충진된 컬럼을 통해 통과시킨 후, 2ℓ의 물로 세척하고 2ℓ의 1N 아세트산으로 용출시킨다. 용출액을 농축하고, 100㎖의 도웩스 50W-8(The Dow Chemical Co.)로 충진된 컬럼을 통과시킨다음, 2ℓ의 물로 용출시킨다. 용출액을 농축하고 50g의 활성탄(Wako Pure Chemical Industries, Ltd)으로 충진된 컬럼을 통해 통과시킨다. 컬럼을 500㎖의 물로 세척한 후, 500㎖의 0.1N 암모니아수로 용출시킨다. 용출액을 증발건조시키고, 잔류물을 5㎖의 0.1N 수산화나트륨 수용액에 용해시킨 후, 50㎖의 세파덱스 G-10(Pharmacia AB)이 충진된 컬럼을 사용하여 겔 여과 크로마토그래피를 통해 정제한다. 용출된 비오틴을 함유하는 분획을 합하고, 0.05N HCl의 첨가에 의해 pH 3으로 조절한 후, 합한 분획을 서늘한 장소에서 밤새 방치시킨다. 침전된 결정을 수집하고 감압 건조시키면 13㎎의 비오틴 결정이 얻어진다. 이렇게 수득된 비오틴은 비오틴의 표준 제조에서와 동일한 물리화학적 특성(예. 질량 스펙트럼, IR 흡수 스펙트럼)을 나타낸다.

스핀고모나스 속에 유사한 속의 각종 미생물을 각각 1백금이씩 5㎖의 예비-배양배지(1% 글리세롤, 2% 폴리펩톤, 0.15% K2HPO4, 0.15% MgSO4·7H2O, pH 7.2)를 함유하는 큰 시험관(22㎜φ × 220㎜)에 접종하고, 250rpm으로 진탕하면서 3일동안 30℃에서 배양한다. 배양이 끝난후, 배양배지에 생산되고 축적된 비오틴의 농도를 실시예 1에서와 동일한 정량적 미생물 생물검정에 의해 결정한다. 결과를 표 1에 나타낸다.

Claims (5)
- (a) 비오틴 생산능력을 갖는 스핀고모나스 속 미생물의 배양액을 배지에 제조하고; (b) 배지에 생산되고 축적된 비오틴을 수집하는 단계로 이루어진 비오틴의 제조방법.
- 제1항에 있어서, 비오틴 생산 능력을 갖는 스핀고모나스 속 미생물이 스핀고모나스 sp. SC-42405(FERM BP-3995)인 방법.
- 비오틴 생산능력을 갖는 스핀고모나스 속의 비오틴 생산용 미생물.
- 제3항에 있어서, 미생물이 스핀고모나스 sp. SC-42405(FERM BP-3995)인 비오틴 생산용 미생물.
- 스핀고모나스 sp. SC-42405(FERM BP-3995).
