Description
なお、吸光度の違いは血球成分含量の違いを反映しており、血球成分含量の違いにより、センサー基板への血球成分や夾雑物の付着量、あるいは測定対象物質とセンサー基板との接触の効率性が異なるため、吸光度の違いに応じて、これ以降の工程の処理条件を調整し、SPR又はSPFSによる測定が適切に行われるようにすることが適切である。
前記の設定したモードに応じて異なる処理条件としての好ましい例は、一次反応工程後に標識反応工程が行われる際には、該標識反応工程前に行われる洗浄工程における洗浄回数である(第1実施形態)。
また、前記モード設定工程が、全血試料用の全血モードと、血清試料又は血漿試料用の通常モードとを設定する工程であり、全血モードは通常モードよりも前記洗浄回数が多いことが好ましい。
また、前記モード設定工程が、全血試料用の全血モードと、血清試料又は血漿試料用の通常モードとを設定する工程であり、全血モードは通常モードよりも前記希釈率が高いことが好ましい。
さらに、前記モード設定工程が、全血試料用の全血モードと、血清試料又は血漿試料用の通常モードとを設定する工程であり、全血モードは通常モードよりも前記一次反応工程の時間が長いことが好ましい。
[第1実施形態:設定したモードに応じて異なる処理条件が洗浄回数の場合]
本実施の形態に係る表面プラズモンを利用した免疫測定方法は、図1に示すように、まず、試料の吸光度測定工程、処理モード(全血モードと血清・血漿モード)を設定するモード設定工程を行う。このモード設定工程では処理モードの切り分けを行う。ここで、吸光度測定結果に応じて処理モードが「全血モード」に設定された場合、通常の希釈率で試料を希釈する希釈工程、一次反応工程、通常の第1洗浄工程よりも洗浄回数が追加された第1洗浄工程、標識反応工程、第2洗浄工程、測定工程を順次行う。尚、SPR免疫測定方法に実施する際には、一般に上述のフローから標識反応工程と第2洗浄工程とを省略したフローとして考えればよい。
一方、処理モードが「血清・血漿モード」に設定された場合については後述する。
吸光度測定工程は、試料の吸光度を測定する工程である。吸光度の測定方法は特に限定されるものではなく、公知の各種の手段を用いることができるが、たとえば、一般的な分光光度計と同様のユニット(試料を収納する容器、白色光源ランプ、プリズム、スリット、光検出器など)を用いて測定することができる。吸光度測定工程は、通常、試料によって条件が変更される可能性のある各種の処理(たとえば希釈処理)の前に行われるが、試料希釈工程の直前でなくてもよく、また、各試料に共通の処理を行った後に行ってもよい。
モード設定工程とは、全血を対象とする全血モードと、血清または血漿を対象とする血清及び血漿モードとを選択して試料を自動的に判別する工程である。全血とは、ヒト又はその他の動物から採取した血液であり、血清や血漿を分離する処理を行っていないものを意味する。血漿とは、血液から血球成分を除いたものをいい、血清とは、血漿から線維素原と凝固因子を(凝固によって)除いたものをいう。
希釈工程とは、SPFSの測定に関する各工程を適切に行えるよう、試料を希釈する工程である。試料の希釈率は特に限定されるものではなく、測定システムの実施形態、採取される試料の量、センサー基板に送液する際の流量、試料の粘度(全血中の血球成分の量)などに応じて適宜設定することができるが、通常2〜10倍、好ましくは2〜6倍である。希釈方法も特に限定されるものではないが、常法に従って、適量の溶媒(PBS(リン酸緩衝生理食塩水)、TBS(トリス緩衝生理食塩水)など)を試料に添加すればよい。
一次反応工程とは、試料をセンサー基板に接触させる工程である。この工程により、試料中の測定対象物質であるアナライト(抗原)とセンサー基板表面に固相化されたリガンド(抗体)との複合体が形成される。
一次反応工程の時間、すなわち試料をセンサー基板に接触させる時間は、試料中に含まれる測定対象物質の量などによって調整される場合もあるが、通常3〜30分間、好ましくは通常5〜20分間である。
第1洗浄工程とは、一次反応工程の後、標識反応工程の前に、洗浄液を送液等により供給してセンサー基板を洗浄する工程である。第1洗浄工程を行うことにより、一次反応工程においてセンサー基板表面に非特異的に吸着した、血中夾雑物(測定対象物質以外のタンパク質、糖脂質など)や血球成分を除去することができる。これにより、標識反応工程の際に用いる標識抗体が前記非特異的吸着物質にさらに非特異的に結合して、測定工程の際にノイズとなって表れることを抑制することができる。
第1洗浄工程に含まれる洗浄処理の回数は、洗浄の効果を考慮しながら適宜調節することができるが、血清・血漿モード(通常モード)では、通常2〜6回、好ましくは3〜4回、より好ましくは3回含まれる。
これに対して、全血モードの第1実施形態では、洗浄回数を血清・血漿モード(通常モード)にさらに1〜3回、より好ましくは2回追加洗浄を行なう。
標識反応工程とは、蛍光体で標識化された標識抗体をセンサー基板に接触させる工程である。一次反応工程において、センサー基板表面の固相化抗体に抗原が結合して複合体を形成している場合、その捕捉された抗原にさらに標識抗体が結合し、固相化抗体−抗原−標識抗体からなる複合体が形成される。この工程は、一般的なSPFS測定系にしたがって行うことができる。
第2洗浄工程とは、標識反応工程の後、測定工程の前に、洗浄液を送液する工程である。第2洗浄工程を行うことにより、標識反応工程においてセンサー基板表面に非特異的に吸着した標識抗体を除去することができ、測定の際にノイズとなって表れることを抑制することができる。
第2洗浄工程において洗浄液を往復送液等により供給する際の界面活性剤の種類および濃度、液量、時間などの諸条件は、第1洗浄工程と同様とすることもできるが、必要に応じて異なる条件を設定してもよい。
SPFS測定系において、測定工程とは、センサー基板の裏面から特定の励起波長を有する光を照射し、それにより発生する特定の蛍光波長を有する光の強度を、センサー基板の上面に設けられたPMT(光電子倍増管)などの光量検知センサーにより測定する工程である。試料中に所定の測定対象物質(抗原)が存在し、標識反応工程において固相化抗体−抗原−標識抗体が形成されていれば、その複合体中の蛍光体がSPFSによる増強されたエバネッセント波で励起されて発する蛍光を、PMTなどで測定することができる。この工程は、一般的なSPFS測定系にしたがって行うことができる。蛍光量の測定結果は適切な情報処理手段により出力、解析され、測定結果に基づいて試料中の測定対象物質の定量的または定性的な分析を行うことができる。
各工程についての説明は、上記の「全血モード」の場合と同様であるので、ここでは説明を省略する。
本実施の形態に係る表面プラズモンを利用した免疫測定方法は、図2に示すように、まず、試料の吸光度測定工程、処理モード(全血モードと血清・血漿モード)を設定するモード設定工程を行う。このモード設定工程では処理モードの切り分けを行う。
本実施の形態に係る表面プラズモンを利用した免疫測定方法は、図3に示すように、まず、試料の吸光度測定工程、処理モード(全血モードと血清・血漿モード)を設定するモード設定工程を行う。このモード設定工程では処理モードの切り分けを行う。
本実施の形態に係る表面プラズモンを利用した免疫測定方法は、図4に示すように、まず、試料の吸光度測定工程、処理モード(全血モードと血清及び血漿モード)を設定するモード設定工程を行う。このモード設定工程では処理モードの切り分けを行う。
異なる処理条件を「洗浄回数の変更(追加)」と「一次反応時間の変更(延長)」との組み合わせ、「希釈率の変更(増加)」と「一次反応時間の変更(延長)」との組み合わせ、「洗浄回数の変更(追加)」と「希釈率の変更(増加)」と「一次反応時間の変更(延長)」との組み合わせ等いずれの実施形態でも試料の免疫測定を行うことができる。
表面プラズモンセンサーは、少なくとも誘電体部材(プリズムまたは透明平面基板)と、当該誘電体部材の上層に形成された金属薄膜と、当該金属薄膜の上層に形成された固定化リガンドを含む層(反応層)とにより構成される、SPFS等によるシグナルを測定するための構造をいう。
センサーチップに用いられる誘電体部材は、ガラス製や、ポリカーボネート(PC)、シクロオレフィンポリマー(COP)などのプラスチック製のもの、好ましくはd線(588nm)における屈折率〔nd〕が1.40〜2.20の範囲にある材質のものを用いることができる。誘電体部材に透明平面基板を用いる場合の厚さは、例えば0.01〜10mmの範囲で調整することができる。また、金属薄膜を形成する前に、誘電体部材の表面は酸またはプラズマによる洗浄処理がなされていることが好ましい。
表面プラズモンセンサーの金属薄膜は、酸化に対して安定であり、かつ表面プラズモンによる電場増強効果が大きい、金、銀、アルミニウム、銅、および白金からなる群から選ばれる少なくとも1種の金属からなる(合金の形態であってもよい)ことが好ましく、特に金からなることが好ましい。透明平面基板としてガラス製平面基板を用いる場合には、ガラスと上記金属薄膜とをより強固に接着するため、あらかじめクロム、ニッケルクロム合金またはチタンの薄膜を形成することが好ましい。
表面プラズモンセンサーには、必要に応じて、金属薄膜による蛍光色素の金属消光を防止するため、金属薄膜と反応層(またはSAM)の間に誘電体からなるスペーサ層を形成してもよい。
表面プラズモンセンサーには、必要に応じて、金属薄膜(またはスペーサー層)と反応層の間にSAM(Self-Assembled Monolayer:自己組織化単分子膜)を形成してもよい。
表面プラズモンセンサー(特にセンサーチップの態様をとるもの)は、公知のSPFS等用の測定装置に装着して使用することができる。この測定装置は、基本的に光源、プリズム、光検出器などを備え、通常はさらに集光レンズ、カットフィルターなどを備える。各種の流体を所定の流速、タイミング等で所定の領域に送液するための手段(送液ポンプなど)や、各種の操作や情報処理を制御するコンピュータなどが上記測定装置に一体化されていてもよい。
(試料)
試料とは、SPFS等に供される物質であり、代表的な試料としては、ヒト、ヒト以外のほ乳類(モデル動物、ペット等)、その他の動物から採取される血液検体(全血、血清、血漿など)に由来する試料が挙げられる。分析の際、試料は必要に応じて、純水、生理食塩水、緩衝液、試薬溶液等の各種の溶媒、あるいは血液検体については抗凝固剤(ヘパリン等)などと混合して用いてもよい。このような混合液または試料そのもの、あるいは所定の目的のために調製した測定対象物質を含有する溶液など、SPFS等によるシグナルを測定するために表面プラズモンセンサーの所定の領域に送液される流体(溶液、懸濁液、ゾル、その他流動性を有する物質を含む。)のいずれもが、試料に含まれる。
SPFS等により定量ないし検出すべき対象となる物質を「測定対象物質」という。センサー表面に捕捉することのできる物体であれば特に限定されることなく測定対象物質となり得るが、代表的な測定対象物質としては、例えば腫瘍マーカーとなるような、タンパク質(ポリペプチド、オリゴペプチド等を含む)またはその複合体が挙げられる。また、リガンドに認識される部位(エピトープ等)を表面に有する細胞やウイルス等も測定対象物質となり得る。
また、抗原抗体反応を用いる免疫測定法に限らず、測定対象物質およびリガンドの間におきる同様の反応を利用して、核酸(一本鎖または二本鎖の、DNA、RNA、ポリヌクレオチド、オリゴヌクレオチド、PNA(ペプチド核酸)等を含む)、糖質(オリゴ糖、多糖類、糖鎖等を含む)、脂質などのその他の分子を、必要に応じてビオチン化等の処理をした上で、測定対象物質とする測定方法に拡張することも可能である。
測定対象物質と特異的に結合し得る分子を「リガンド」という。特に、センサー表面に固相化された、測定対象物質をセンサー表面に捕捉するためのリガンドを「固相化リガンド」(リガンドが抗体であれば「固相化抗体」)、測定対象物質を標識するために液中に存在する、蛍光体と結合したリガンドを「標識リガンド」(リガンドが抗体であれば「標識抗体」)という。なお、固相化リガンドと標識リガンドのリガンド部分は、同じでもよいし、異なっていてもよい。ただし、固相化リガンドがポリクローナル抗体である場合、標識リガンドはモノクローナル抗体であってもポリクローナル抗体であってもよいが、固相化リガンドがモノクローナル抗体である場合、標識リガンドはその固相化リガンドが認識しないエピトープを認識するモノクローナル抗体であるか、またはポリクローナル抗体であることが望ましい。
処理モードの選択により、一次反応後の第1洗浄工程における洗浄回数を変更して免疫測定反応を行った。図1に示すフローチャートに沿って測定を行った。
健常人Aの全血サンプルを準備し、その一部を遠心分離(微量高速遠心機:himac CF15RXII,日立工機社製、1600g×15分)することにより血漿を得た。同健常人の全血および血漿サンプル100ulをミオグロビン測定用の試薬カートリッジ(免疫反応1回分の各種試薬が充填されているカートリッジ)のサンプルウエルに分注した後、該試薬カートリッジをSPFS免疫測定システムにセットし、測定を開始した。全血測定および血漿測定を各6回繰り返し測定し、CV値(変動係数:測定値の標準偏差/平均、単位%)を算出した。
なお、CV値<10%の場合、精度、確度の高い測定であり、臨床的価値を有する測定値である。
本実施例では、試料として、全血、血清、ヘパリンナトリウム血漿を用いた。
表1より、全血を処理する際に、一次反応後の洗浄回数を追加した本実施例においては、CVが4.2%と低値であることから十分に安定した測定が行なわれていることが判る。
比較例1では、全血を強制的に通常モード条件下で処理した。その結果を表1に示す。
全血を通常モードで処理した場合は、測定平均値が低値となり、またCVは13.0%と高値であることから、全血の洗浄が不十分であり安定した測定を行なえていないことが判った。
処理モードの選択により、試料の希釈率を通常の2倍に変更して免疫測定反応を行った。図2に示すフローチャートに沿って測定を行った。
健常人Bの全血サンプルを準備し、その一部を遠心分離(微量高速遠心機:himac CF15RXII,日立工機社製、1600g×15分)することにより血漿を得た。本SPFS免疫測定システムでは、通常モード処理の場合は装置内でサンプルと希釈液を1:1で混合し、2倍希釈された液を用いて免疫反応を行なっている。本実施例では装置内におけるサンプルと希釈液の混合比率を変化させることにより、サンプルの4倍希釈を実現した。実施例1と同様に、処理モード切替基準により、同健常人の全血及び血漿サンプルをSPFS免疫測定システムにおいてミオグロビン濃度を6回繰り返して測定した。その結果を表2に示す。
処理モードの選択により、試料の希釈率を通常の3倍に変更し、サンプルを6倍希釈にして免疫測定反応を行った他は、実施例2と同様にして測定を行った。その結果を表3に示す。
表3より、全血を処理する際に、サンプルの希釈率を通常モードより高くした本実施例においては、CVが5.0%と、10%以下の低値であることから十分に安定した測定が行われていることが判る。
比較例2では、全血を強制的に通常モード条件下で処理した。その結果を表2、3に示す。全血を通常モードで処理した場合は、測定平均値が低値となり、またCVは15.7%と高値であることから、全血の希釈が不十分なことから安定した測定を行なえていないことが判った。
Claims (11)
- 表面プラズモンを利用する免疫測定方法であって、
試料の吸光度を測定する吸光度測定工程と、該吸光度測定工程で測定した吸光度の結果に対応して異なる処理モードを設定するモード設定工程と、を含む表面プラズモンを利用した免疫測定方法。 - 前記モード設定工程の後に、該モード設定工程で設定した処理モードに応じて処理条件が異なる、一または複数の工程を含む、請求項1に記載の表面プラズモンを利用した免疫測定方法。
- 前記処理条件が、一次反応工程後であって、標識反応工程が行われる際には該標識反応工程前に行われる洗浄工程における洗浄回数である、請求項2に記載の表面プラズモンを利用した免疫測定方法。
- 前記処理条件が、希釈工程における試料の希釈率である、請求項2又は3に記載の表面プラズモンを利用した免疫測定方法。
- 前記処理条件が、一次反応工程の時間である、請求項2〜4のいずれか1項に記載の表面プラズモンを利用した免疫測定方法。
- 前記モード設定工程が、全血試料用のモードと、血清試料又は血漿試料用のモードとを設定する請求項1〜5のいずれか1項に記載の表面プラズモンを利用した免疫測定方法。
- 前記モード設定工程が、全血試料用の全血モードと、血清試料又は血漿試料用の通常モードとを設定する工程であり、全血モードは通常モードよりも前記洗浄回数が多い、請求項3に記載の表面プラズモンを利用した免疫測定方法。
