Abstract
PURPOSE: To prepare the titled compound useful as an intermediate for the synthesis of pharmaceuticals such as β-adrenergic nerve blocking agent, in high yield, by using an epoxy compound and an isocyanate compound as starting raw materials, and reacting the compounds.
CONSTITUTION: The objective compound of formula I can be prepared by (1) reacting 6-(2,3-epoxypropoxy)-2-nitro-toluene of formula II with the compound of formula III (R is lower alkyl), (2) reacting the resultant compound of formula VII with the compound of formula IV[Z is group of formula IX or formula X (R1 and R2 are alkyl, aryl, etc.; X is halogen)]or the compound of formula V (Y is halogen) to obtain the compound of formula VIII, and (3) reducing the compound thus obtained. The reactions (1) and (2) are carried out pref. in the presence of a solvent (e.g. DMF) at 25W175°C for 0.5W20hr, and the reduction of the compound VIII is carried out pref. by conventional process such as the catalytic reduction with hydrogen in the presence of a catalyst (e.g. platinum).
