Abstract

NEW MATERIAL:Protein RBF-PM, its cystine content: less than 0.1%, nitrogen content: ranging from 12% to 15%; isoelectric point: ranging in pH from 3 to 6; solubility: insoluble in dilute sodium chloride solution and dilute acid, soluble in dilute alkali.
USE: Antitumor agent: it is a protein of less cystine content and obtained as a fraction insoluble in a polar organic solvent of RBF-P. It remarkably supresses mouce tumors.
PREPARATION: The hot water-soluble fraction is removed from rice bran and the bran is extracted with a basic aqueous solution such as 1W10wt% sodium hydroxide aqueous solution. The extract is mixed with an organic solvent such as ethanol or acetone and the insoluble fraction is removed. Then, an acid such as hydrochloric or phosphoric acid is added to precipitate RBF-P. The resultant RBF-P is treated with a polar organic solvent such as methanol or acetone and the insoluble fraction is collected by filtration as RBF-PM.
