Abstract

1,236,801. Thyrocalcitonins. CIBA-GEIGY A.G. 20 May, 1968 [24 May, 1967; 12 July, 1967; 25 Jan., 1968], No. 23897/68. Heading C2C. [Also in Division A5] α-Thyrocalcitonin and/or #-thyrocalcitonin (see below) are prepared by extracting at least once, defatted thyroid gland tissue optionally pre-purified by extraction with a C 1-8 alkanol or aqueous trichloracetic acid, with a solvent system having a pH of from 1 to 6 and comprising water and a C 1-8 alkanol, the resulting solution being worked up by one or more of the following techniques used in any order; precipitation with acetone or trichloracetic acid, countercurrent distribution, gel - chromatography, countercurrent dialysis, lyophilization, partial evaporation, thin layer chromatography, ion exchange, electrophoresis, and, if desired, splitting the mixture so obtained into α-thyrocalcitonin and #-thyrocalcitonin (of specified formulµ) preferably by countercurrent distribution. The solvent system may contain methanol, ethanol, propanol, n or sec-butanol and an acid such as sulphuric, hydrochloric, formic or glacial acetic acid and/or an organic solvent such as acetone, pyridine or morpholine and/or inorganic salts such as sodium chloride. Physiologically tolerable salts of α-thyrocalcitonin and/or #-thyrocalcitonin are also mentioned. Thyrocalcitonins have the formula H - Cys - Ser - Asn - Leu - Ser - Thr - Cys - Val- Leu - Ser - Ala - Tyr - Trp - Arg - Asn - Leu- Asn - Asn - Phe - His - Arg - Phe - Ser - Gly - X - Gly - Phe - Gly - Pro - Glu - Thr - PrONH 2 ; α - compound X = Met; (3 - compound X = Met(O).
