Abstract

PURPOSE: To determine the amount to hydrogen peroxide in a solution, in high accuracy, by oxidizing a detecting agent added to the solution with the hydrogen peroxide in the presence of an immobilized peroxidase as a catalyst, and determining the oxidized detecting agent.
CONSTITUTION: Hydrogen peroxide in a solution is determined by (1) oxidizing an intermediate electron transfer material such as metal ions having two or more ionic valences, with the hydrogen peroxide in the presence of an immobilized peroxidase as a catalyst, (2) separating the immobilized enzyme from the solution containing the oxidized intermediate electron transfer material, (3) adding a detecting agnet to the solution thereby oxidizing the agent, and (4) determining the amount of the oxidized detecting agent in the solution. A compound such as saccharides, purine derivatives, etc. can also be determined by (1) oxidizing the compound in the presence of an immobilized oxidase, and (2) carrying out the above procedures using the produced hydrogen peroxide as an oxidizing agent.
