Abstract

PURPOSE: In the titled measuring method utilizing oxidation and reduction reactions, the determination is made after the removal of inhibiting substances by making ascorbic acid oxidase act on ascorbic acid and glutathion-dehydroascorbic acid oxidoreductase on SH compounds.
CONSTITUTION: On the determination, before or when the oxidation and reduction reaction is effected, ascorbic acid oxidase is made to act on ascorbic acid and the glutathion dehydroascorbic acid oxidoreductase is made to act on both the dehydroascorbic acid formed and SH compounds, which is converted into the oxidation type SH compounds. The amounts of enzymes added can be decided regardless of the amount of the reductive substances, but a large amount of the enzymes removes the inhibiting substance in a shorter time. Thus, the inhibiting substances, that is, ascorbic acid and SH compounds, are removed and the concentration of the substrate or enzyme activity can be determined.
