Abstract
PURPOSE: To determine hydrogen peroxide, by using an enzymatic reagent obtained by freeze-drying an aqueous solution containing enzymes and 4-aminoantipyrine, in combination with a color-developing reagent composed of a buffer solution containing N-ethyl-N-[hydroxy-substituted sulfo-(lower alkyl)]-m- toluidine.
CONSTITUTION: An enzymatic reagent is prepared by freeze-drying a aqueous solution of enzymes and 4-aminoantipyrine. The enzymes are the combination of uricase and peroxidase, cholesterol-ester-hydrase, etc. and peroxidase, or phospholipase D, etc. and peroxidase, etc. Separately, a buffer solution containing N-ethyl-N-[hydroxy-substituted sulfo-(lower alkyl)]-m-toluidine is prepared. In the determination, the former enzymatic reagent is dissolved in the latter color- developing reagent, the solution is mixed with the specimen, and the amount of the hydrogen peroxide is determined from the absorbance of the mixture.
