Abstract

PURPOSE: To determine aminotransferase in high sensitivity, by the use of a reagent kit for the determinaton of hydrogen peroxide composed of a p-halophenol and 4- aminoantipyrine.
CONSTITUTION: The activity of aminotransferase in a living specimen is determined by measuring hydrogen peroxide with a reagent kit composed of a p-halophenol of formula (X is halogen) and 4-aminoantipyrine. For example, a substrate containing alanine or aspartic acid and α-ketoglutaric acid is treated with aminotransferase, the produced oxalacetic acid is treated with an enzyme which eliminates carbon dioxide from oxalacetic acid, produced pyruvic acid is oxidized to acetyl phosphoric acid and hydrogen peroxide with pyruvic acid oxidase, and the hydrogen peroxide thus produced is determined by the above method.
