Abstract

PURPOSE: To obtain an enzyme electrode by which the time required for measurement is shortened and which is excellent in sensitivity in an electrochemical measurement of the substrate concentration of an enzyme, by a method wherein oxidoreductase and coenzyme are fixed as film directly on a collecting body by a cross- linking reagent.
CONSTITUTION: A phosphate buffer solution of dehydrogenase (e.g. lactic dehydrogenase, LDH) and coenzyme thereof (e.g. nicotinamide adenine dinucleotide, NAD) is applied on to a collecting body (e.g. a graphite disc) and dried. Then, a cross- linking reagent is applied thereto, and the dehydrogenase and the coenzyme are stuck fast and fixed directly on to the collecting body as an insoluble film as a whole, to obtain an enzyme electrode. As the above-mentioned cross-linking reagent, glutaraldehyde, dimethyl adipoimidate or the like is employed.
