Abstract

PURPOSE: To measure the substrate concentrations ob enzyme or activity of enzyme rapidly and readily by forming the enzyme electrode through directly and chemically bonding (particularly covalent bonding) coenzyme to current collectors and fixing the same and using this.
CONSTITUTION: Nicotineamide Adenine Dinucleotide (NAD, coenzyme) is reacted with, e.g., succinic acid anhydride to introduce carboxylic groups. On the other hand, SnO2 NESA glass (current collector) is subjected to hydrophilic treatment to form hydroxyl groups on the SnO2 surface, after which the above-mentioned denatured NAD is reacted to fix the NAD on the NESA glass through ester bond. After the NAD fixed electrode is formed by directly chemically bonding 4 the coenzyme 2 to the current collector 3 in the above-mentioned manner, it is dipped in a solution containing enzyme (e.g., lactic acid dehydrogenase) 1 to prepare the NAD- enzyme bond, which is then treated with crosslinking reagent (glutaraldehyde, etc.) to fix the enzyme.
