Abstract

PURPOSE: To measure the activity of an alkali phosphatase contained in a living body sample, by using the change in amount of a coenzyme NADH2 formed.
CONSTITUTION: An alkali phosphatase in a living body sample is reacted with β- glycerophosphoric acid as a substrate, and the glycerol thus formed is reacted with glycerol dehydrogenase in the presence of NAD to measure the rate of formation of NADH2 and the activity of the alkali phosphatase. After fractionating the alkali phosphatase by electrophoresis with various supports, NADH2 is reacted with a diaphorase in the presence of nitroblue tetrazolium in the support, and the diformazan formed in the reaction is detected to detect the alkali phosphatase isoenzyme.
