Description
Claims (1)
- 【特許請求の範囲】 1.不死化哺乳動物細胞株であって、不死化遺伝子をフランキングする、少なく とも第一及び第二レコンビナーゼ標的部位を有する外来核酸を前記細胞のゲノム 中に含有し、前記標的部位が、レコンビナーゼと接触した時に、前記不死化遺伝 子の切出しを媒介できることを特徴とする細胞株。 2.前記外来核酸が更に選択マーカー遺伝子を有する請求項1に記載の細胞株。 3.前記選択マーカー遺伝子が、負の選択マーカー遺伝子である請求項2に記載 の細胞株。 4.前記核酸が、更に、停止部位を有する請求項2に記載の細胞株。 5.前記外来核酸が、更に、第一選択マーカー遺伝子と、第二選択マーカー遺伝 子とを含有する請求項1に記載の細胞株。 6.前記第二選択マーカー遺伝子が、第一及び第二レコンビナーゼ標的部位の間 にない請求項5に記載の細胞株。 7.不死化細胞のゲノムにおいて不死化遺伝子をフランキングするレコンビナー ゼ標的部位を含有する不死化細胞を形成する方法であって、 前記方法が、細胞を、 (a)第一レコンビナーゼ部位、 (b)不死化遺伝子、及び (c)第二レコンビナーゼ部位、 を含有する外来核酸で形質転換して、レコンビナーゼの不存在下において、前 記外来核酸を前記細胞のゲノム中に挿入することを特徴とする方法。 8.前記核酸が、更に、少なくとも一種の選択マーカー遺伝子を含有する請求項 7に記載の方法。 9.前記選択マーカー遺伝子が、負のの選択マーカー遺伝子である請求項8に記 載の方法。 10.不死化細胞のゲノム中に不死化遺伝子をフランキングするレコンビナーゼ標 的部位を含有する外来核酸を含有する不死化細胞を非不死化する方法であって、 前記方法が、前記レコンビナーゼ標的部位を、該レコンビナーゼ標的部位を 認識できるレコンビナーゼと接触することを特徴とする方法。 11.前記不死化細胞を前記レコンビナーゼをコードするレコンビナーゼ核酸で形 質転換し、該形質転換細胞を、前記レコンビナーゼが発現し、前記不死化遺伝子 が切出される条件下において維持することによって、前記接触を行う請求項10に 記載の方法。 12.不死化細胞の非不死化方法であって、 (a)(i)第一レコンビナーゼ標的部位、 (ii)不死化遺伝子、 (iii)負の選択マーカー遺伝子、及び (iv)第二レコンビナーゼ標的部位、 を有する外来核酸を、細胞のゲノム中に挿入して、前記レコンビナーゼ標的 部位間の配列の切出しによって、不死化遺伝子及び負の選択マーカーを切出すよ うな配向で切出し得る不死化遺伝子を含有する不死化細胞を製造し、そして (b)前記レコンビナーゼ標的部位を前記不死化標的部位を認識するレコンビナ ーゼと接触して、前記不死化遺伝子及び負の選択マーカー遺伝子を切出す、 ことを特徴とする方法。 13.前記不死化細胞を、前記レコンビナーゼが発現される条件下において前記レ コンビナーゼ標的部位を認識するレコンビナーゼをコードするベクターで形質転 換することによって前記接触を行う請求項12に記載の方法。 14.前記外来核酸が、更に、前記レコンビナーゼ標的部位を認識するレコンビナ ーゼの遺伝子を含有し、前記レコンビナーゼ遺伝子が、誘導性プロモーターと作 動可能に連結され、前記不死化細胞を前記レコンビナーゼが発現される条件下で 成長させることによって前記接触を行う請求項12に記載の方法。 15.前記方法が、更に、負の選択マーカー剤の存在下に非不死化細胞を選択する 工程を含有する請求項12に記載の方法。 16.前記外来核酸が、更に正の選択マーカー遺伝子を含有する請求項12に記載の 方法。 17.前記外来核酸が、更に、付加的な外来遺伝子を含有する請求項12に記載の方 法。 18.不死化細胞の非不死化方法であって、 (a)(i)第一レコンビナーゼ標的部位、 (ii)不死化遺伝子、 (iii)選択マーカー遺伝子、及び (iv)第二レコンビナーゼ標的部位、 を有する外来核酸を、細胞のゲノム中に挿入して、前記レコンビナーゼ標的 部位間の配列の切出しによって、不死化遺伝子を切出して、選択マーカーを発現 させ、そして (b)前記レコンビナーゼ標的部位を前記レコンビナーゼ標的部位を認識するレ コンビナーゼと接触して、前記不死化遺伝子を切出す、 ことを特徴とする方法。 19.前記核酸が、更に、停止部位を含有する請求項18に記載の方法。 20.前記不死化細胞を、前記レコンビナーゼが発現される条件下において前記レ コンビナーゼ標的部位を認識するレコンビナーゼをコードするベクターで形質転 換することによって前記接触を行う請求項18に記載の方法。 21.前記外来核酸が、更に、誘導性プロモーターと作動可能に連結された前記レ コンビナーゼ標的部位を認識するレコンビナーゼの遺伝子を含有し、前記不死化 細胞を前記レコンビナーゼが発現される条件下で成長させることによって前記接 触を行う請求項18に記載の方法。 22.前記方法が、更に、前記選択マーカー遺伝子を発現する非不死化細胞を選択 する工程を含有する請求項18に記載の方法。 23.不死化細胞の非不死化方法であって、 (a)(i)第一レコンビナーゼ標的部位、 (ii)不死化遺伝子、 (iii)第一選択マーカー遺伝子、 (iv)第二レコンビナーゼ標的部位、及び (iv)第二選択マーカー遺伝子、 を有する外来核酸を、細胞のゲノム中に挿入して、前記不死化遺伝子が発 現される場合に、第一の選択マーカー遺伝子も発現され、前記第二選択マー カー遺伝子は発現されず、前記不死化遺伝子が切出される場合には、前記第二選 択マーカー遺伝子が発現されるような配向で、切出し得る不死化遺伝子を含有す る不死化細胞を製造し、そして (b)前記レコンビナーゼ標的部位を前記レコンビナーゼ標的部位を認識するレ コンビナーゼと接触して、前記不死化遺伝子を切出す、 ことを特徴とする方法。 24.前記不死化細胞を、前記レコンビナーゼが発現される条件下において前記レ コンビナーゼ標的部位を認識するレコンビナーゼをコードするベクターで形質転 換することによって前記接触を行う請求項23に記載の方法。 25.前記外来核酸が、更に、前記レコンビナーゼ標的部位を認識するレコンビナ ーゼの遺伝子を含有し、前記レコンビナーゼ遺伝子が、誘導性プロモーターと作 動可能に連結され、前記不死化細胞を前記レコンビナーゼが発現される条件の下 で成長させることにより前記接触を行う請求項23に記載の方法。 26.前記核酸が、更に、付加的な外来遺伝子を含有する請求項24に記載の方法。 27.前記外来核酸が、更に、停止部位を含有する請求項24に記載の方法。
