Claims (1)
- 【特許請求の範囲】 1.ヒトDNアーゼIアクチン−耐性変異体 2.天然ヒトDNアーゼIの結合親和性よりも少なくとも5倍小さいアクチン に対する結合親和性を有する請求項1記載の変異体。 3.天然ヒトDNアーゼIの結合親和性よりも少なくとも100倍小さいアク チンに対する結合親和性を有する請求項1記載の変異体。 4.図1に示した天然ヒトDNアーゼIのアミノ酸配列と少なくとも90%の 相同性を有するアミノ酸配列よりなる請求項1記載の変異体。 5.図1に示した天然ヒトDNアーゼIのアミノ酸配列と少なくとも95%の 相同性を有するアミノ酸配列よりなる請求項1記載の変異体。 6.図1の配列内の単一位置のみにおいて一のアミノ酸が他のアミノ酸に置換 されることより図1に示したアミノ酸配列とは異なるアミノ酸配列を有するヒト DNアーゼIアクチン−耐性変異体。 7.アミノ酸置換が天然ヒトDNアーゼIに存在しないグリコシル化部位を変 異体内に生成する請求項6記載の変異体。 8.アミノ酸置換が図1に示したアミノ酸配列内で以下の位置:His44、 Leu45、Val48、Gly49、Leu52、Asp53、Asn56、 His64、Tyr65、Val66、Val67、Glu69またはAla1 14のうちの1つにおけるものである請求項6記載の変異体。 9.図1の配列内の2以上の位置において一のアミノ酸が他のアミノ酸に置換 されることにより図1に示したアミノ酸配列とは異なるアミノ酸配列を有するヒ トDNアーゼIアクチン−耐性変異体。 10.アミノ酸置換の少なくとも1つが図1に示したアミノ酸配列内で以下の 位置:His44、Leu45、Val48、Gly49、Leu52、Asp 53、Asn56、His64、Tyr65、Val66、Val67、Glu 69、またはAla114のうちの1つにおいてなされた請求項9記載の変異体 。 11.アミノ酸置換の少なくとも1つが天然ヒトDNアーゼIに存在しないグ リコシル化部位を変異体内に生成する請求項9記載の変異体。 12.ヒトDNアーゼIアクチン−耐性変異体をコードする単離された核酸。 13.図1に示した天然ヒトDNアーゼのアミノ酸配列内に少なくとも90% の相同性を有するアミノ酸配列をコードするヌクレオチド配列を含む請求項12 記載の核酸。 14.図1に示した天然ヒトDNアーゼのアミノ酸配列内に少なくとも95% の相同性を有するアミノ酸配列をコードするヌクレオチド配列を含む請求項12 記載の核酸。 15.図1の配列内の単一位置のみにおいて一のアミノ酸が他のアミノ酸に置 換されることにより図1に示したアミノ酸配列とは異なるアミノ酸配列をコード するヌクレオチド配列を含む請求項12記載の核酸。 16.図1の配列内の少なくとも2つの位置において一のアミノ酸が他のアミ ノ酸に置換されることにより図1に示したアミノ酸配列とは異なるアミノ酸配列 をコードするヌクレオチド配列を含む請求項12記載の核酸。 17.治療上有効量のヒトDNアーゼIのアクチン−耐性変異体を患者に投与 することからなる肺の疾患または障害を有する患者を治療する方法。 18.該疾患または障害が嚢胞性線維症である請求項17記載の方法。 19.該疾患または障害が慢性気管支炎である請求項17記載の方法。 20.ヒトDNアーゼIのアクチン−耐性変異体および任意に医薬上許容され る賦形剤を含んでなる医薬組成物。 21.該組成物が液状形態である請求項20記載の組成物。 22.該組成物が粉末形態である請求項21記載の組成物。
