Description
Claims (1)
- 【特許請求の範囲】 1.非天然コンホメーションの目的とする外因性ポリペプチドを細胞から単離 する方法であって、細胞を、ポリペプチドを細胞から抽出しその溶解度を維持す るに十分な量のカオトロピック剤、および複数の水相(このうち一方は当該ポリ ペプチドに富み、且つ細胞に由来するバイオマス固体および核酸が涸渇している )を形成する相形成物質の有効量と接触させることからなる方法。 2.カオトロピック剤および相形成物質と接触する細胞が細胞培養中にある、 請求項1に記載の方法。 3.目的とするポリペプチドが封入体の形である、請求項2に記載の方法。 4.封入体が原核生物培養基由来である、請求項3に記載の方法。 5.ポリペプチドが細菌において産生される、請求項4に記載の方法。 6.添加が全て発酵容器において起こる、請求項1に記載の方法。 7.ポリペプチドを、ポリペプチドに存在する少なくとも1個のジスルフィド 結合を還元するに十分な量の還元剤とも接触させる、請求項1に記載の方法。 8.ポリペプチドが約0.1ないし15g/Lの濃度で存在する、請求項1に 記載の方法。 9.カオトロピック剤の濃度が約0.1ないし9Mである、請求項1に記載の 方法。 10.カオトロピック剤の濃度が約0.5ないし6Mである、請求項1に記載 の方法。 11.相形成試薬が添加される前にカオトロピック剤を培養基に添加する、請 求項1に記載の方法。 12.カオトロピック剤が尿素である、請求項1に記載の方法。 13.相を分離することにより該ポリペプチドを回収することをさらに含む、 請求項1に記載の方法。 14.相のpHを変化させることにより、または相に溶媒、ポリマー、または 塩を添加することにより、該ポリペプチドをそれに富む相から単離することをさ らに含む、請求項13に記載の方法。 15.ポリペプチドがIGF−Iであり、相のpHを低下させる、請求項14 に記載の方法。 16.ポリペプチドが哺乳動物のポリペプチドである、請求項1に記載の方法 。 17.ポリペプチドがIGF−Iまたは成長ホルモンである、請求項16に記 載の方法。 18.相形成物質が、ポリマー−ポリマー、ポリマー−塩、溶媒−塩、または ポリマー−溶媒の組み合わせである、請求項1に記載の方法。 19.相形成物質がポリマー−塩の組み合わせである、請求項18に記載の方 法。 20.塩が硫酸塩、燐酸塩、またはクエン酸塩である、請求項19に記載の方 法。 21.塩が硫酸塩である、請求項20に記載の方法。 22.ポリマーがポリエチレングリコール、ポリプロピレングリコール、ポリ ビニルピロリドン、または多糖類である、請求項19に記載の方法。 23.使用されるポリマーの濃度が約5%(w/w)から該ポリマーの溶解度 の限界までであり、そして使用される塩の濃度が約3%(w/w)から該塩の溶 解度の限界までである、請求項19に記載の方法。 24.使用されるポリマーの濃度が約5および18%(w/w)の間であり、 使用される塩の濃度が約4および15%(w/w)の間である、請求項23に記 載の方法。 25.形成される水相の数が2である、請求項1に記載の方法。 26.上相が該ポリペプチドに富み、且つバイオマス固体および核酸を涸渇し ている、請求項25に記載の方法。 27.約4ないし7%(w/w)の相形成塩および約12ないし18%(w/ w)の相形成ポリマーが添加される、請求項26に記載の方法。 28.濃厚になっているポリペプチドを活性コンホメーションに再折り畳みす ることをさらに含む、請求項1に記載の方法。 29.目的とする生物活性な外因性ポリペプチドを細胞から回収する方法であ って、細胞を、非天然コンホメーションをとっているポリペプチドを細胞から抽 出しその溶解度を維持するに十分な量のカオトロピック剤、および複数の水相( このうち一方は、当該ポリペプチドに富み且つ細胞に由来するバイオマス固体お よび核酸が涸渇している)を形成する相形成物質の有効量と接触させ、相を分離 することにより該ポリペプチドを回収し、そして、この回収されたポリペプチド を、約5−40%(v/v)のアルコール性または極性非プロトン性溶媒、約0 .2ないし3Mのアルカリ土類、アルカリ金属、またはアンモニウム塩、約0. 1ないし9Mのカオトロピック剤、および約0.01ないし15μMの銅または マンガン塩からなり、酸素供給源が導入されているpH7−12の緩衝液中でイ ンキュベートして、該ポリペプチドの再折り畳みがインキュベーション中に起こ るようにさせることからなる方法。 30.非天然コンホメーションをとっている目的とする外因性ポリペプチドに 富み且つこのポリペプチドが産生された細胞由来のバイオマス固体および核酸が 涸渇している相を含み、その水溶液がさらに、相形成物質および該ポリペプチド の溶解度を維持するに十分な量のカオトロピック剤を含む、多相水溶液。 31.二つの相を含み、ここで上相が該ポリペプチドに富み且つバイオマス固 体および核酸を涸渇している、請求項30に記載の溶液。 32.ポリペプチドが成長ホルモンまたはIGF−Iである、請求項30に記 載の溶液。
