Claims (1)
- 【特許請求の範囲】 1.(a)混合物を非誘導シリカ粒子に通してポリペプチドをシリカ粒子に吸 着させ、 (b)シリカ粒子を洗浄して夾雑物を除去し、 (c)アルコール溶液もしくは極性非プロトン性溶媒、およびアルカリ土類塩、 アルカリ金属塩もしくは無機アンモニウム塩を含む緩衝液でシリカ粒子からポリ ペプチドを溶離する、工程からなる混合物中の疎水性の異なる成分から目的とす るポリペプチドを選択的に分離する方法。 2.ポリペプチドがIGF−I、成長ホルモン、ニューロトロピン、IGF結 合蛋白、血管内皮成長細胞因子、RANTES、またはヒトMIP−1−αであ る請求項1に記載の方法。 3.ポリペプチドがIGF−Iである請求項1に記載の方法。 4.IGF−Iが完全長IGF−Iである請求項3に記載の方法。 5.IGF−Iが脳IGF−Iである請求項3に記載の方法。 6.シリカ粒子のポアサイズが約200〜1000Åである請求項1に記載の 方法。 7.シリカ粒子のポアサイズが約225Åである請求項4に記載の方法。 8.アルカリ土類塩、アルカリ金属塩、または無機アンモニウム塩が塩化また は硫酸ナトリウム、カリウム、およびアンモニウムである、請求項1に記載の方 法。 9.塩が塩化ナトリウムである請求項8に記載の方法。 10.塩濃度が約0.2〜3Mである請求項8に記載の方法。 11.溶媒がメタノール、エタノール、イソ−プロパノール、n−プロパノー ル、またはアセトニトリルである請求項1に記載の方法。 12.溶媒濃度が約5〜40%(v/v)である請求項1に記載の方法。 13.粒子が工程(b)においてリン酸緩衝液中のアルコール溶媒を用いて洗 浄される請求項1に記載の方法。 14.アルコール溶媒がpH7のリン酸緩衝液中の約20%(v/v)エタノ ールである請求項13に記載の方法。 15.溶離用緩衝液のpHが約5〜8である請求項1に記載の方法。 16.工程(c)で用いる緩衝液がリン酸緩衝液である請求項1に記載の方法 。 17.(a)混合物を非誘導シリカ粒子にロードし、 (b)カラムを緩衝液で洗浄して夾雑物を除去し、 (c)約5〜40%(v/v)のアルコール溶液もしくは極性非プロトン性溶媒 、および約0.2〜3Mのアルカリ土類塩、アルカリ金属塩もしくは無機アンモ ニウム塩を含むpH約5〜8の緩衝液でカラムからIGF−Iを溶離する、工程 からなるIGF−Iを含む混合物からIGF−Iを選択的に分離する方法。 18.カラムをリン酸緩衝液中のアルコール溶媒で洗浄する請求項17に記載 の方法。 19.アルコール溶媒がpH7のリン酸緩衝液中の約20%(v/v)エタノ ールである請求項18に記載の方法。 20.溶媒濃度が約10〜30%(v/v)である請求項17に記載の方法。 21.溶媒がメタノール、エタノール、イソ−プロパノール、n−プロパノー ル、またはアセトニトリルである請求項17に記載の方法。 22.アルカリ土類塩、アルカリ金属塩、または無機アンモニウム塩の濃度が 約0.5〜2Mである請求項17に記載の方法。 23.アルカリ土類塩、アルカリ金属塩、または無機アンモニウム塩が塩化ま たは硫酸ナトリウム、カリウム、またはアンモニウムである請求項22に記載の 方法。 24.リン酸緩衝液(pH7)中の、溶媒が濃度約20%(v/v)のエタノ ールであり、塩が濃度約1Mの塩化ナトリウムである請求項23に記載の方法。 25.IGF−Iが完全長IGF−Iである請求項17に記載の方法。 26.IGF−Iが脳IGF−Iである請求項17に記載の方法。
