Claims (1)
- 【特許請求の範囲】 1.1つ以上の治療用遺伝子を、その発現を可能とする調節要素の制御下に含ん で成る組換え核酸を含む単球−マクロファージ株の細胞を含んで成る細胞組成物 。 2.1つの治療用遺伝子または複数の治療用遺伝子が、新規な、または増強され た抗感染性、抗癌性または免疫刺激特性を細胞に与えることができるタンパク質 の全部または一部をコードすることを特徴とする、請求の範囲第1項に記載の細 胞組成物。 3.遺伝子がインターフェロン、腫瘍壊死因子、インターロイキンおよびコロニ ー刺激因子(G-CSF、M-CSF、GM-CSF等)をコードする遺伝子、MDR遺伝子、ならび に感染性粒子の抗原または腫瘍に特異的な抗原をコードする遺伝子から選択され ることを特徴とする、請求の範囲第1または第2項に記載の細胞組成物。 4.組換え核酸がウイルスベクターに保持されていることを特徴とする、前記請 求の範囲のいずれか1項に記載の細胞組成物。 5.ウイルスベクターがアデノウイルス、アデノ伴生ウイルス、ヘルペスウイル ス、ワクシニアウイルスまたはサイトメガロウイルス(CMV)に由来するベクター であることを特徴とする、請求の範囲第4項に記載の細胞組成物。 6.ウイルスベクターがアデノウイルスに由来するベクターであることを特徴と する、請求の範囲第5項に記載の細胞組成物。 7.ウイルスベクターが欠陥組換えウイルスであることを特徴とする、請求の範 囲第4ないし第6項のいずれか1項に記載の細胞組成物。 8.1つ以上の治療用遺伝子を、その発現を可能とする調節要素の制御 下に含んで成る欠陥組換えアデノウイルスを含む、単球−マクロファージの前駆 細胞。 9.造血系の幹および祖先細胞から選択されることを特徴とする、請求の範囲第 8項に記載の前駆細胞。 10.請求の範囲第1ないし第9項のいずれか1項に記載の1つ以上の細胞組成 物を有効成分として含んで成る、医薬組成物。 11.体内から腫瘍細胞の全部または一部を排除する特性をマクロファージに与 えるタンパク質の全部または一部をコードする組換え核酸を含む単球−マクロフ ァージ系の細胞を含んで成る、請求の範囲第10項に記載の医薬組成物。 12.遺伝子がインターフェロン−γ遺伝子および腫瘍壊死因子アルファ遺伝子 から選択されることを特徴とする、請求の範囲第11項に記載の医薬組成物。 13.INF-γ遺伝子を持つ欠陥組換えアデノウイルスを含む単球−マクロファー ジ株の細胞を含んで成る、医薬組成物。 14.TNFα遺伝子を持つ欠陥組換えアデノウイルスを含む単球−マクロファー ジ株の細胞を含んで成る、医薬組成物。 15.欠陥組換えアデノウイルスが膜状態のTNFαをコードする遺伝子を持つこ とを特徴とする、請求の範囲第14項に記載の医薬組成物。 16.ウイルス、レトロウイルス、寄生虫または細菌のような感染原による感染 の予防または発病を治療する特性をマクロファージに与えるタンパク質の全部ま たは一部をコードする組換え核酸を含む、単球−マクロファージ株の細胞を含ん で成る、請求の範囲第10項に記載の医薬組成物。 17.組換え核酸がウイルスまたは細菌の表面抗原をコードすることを特徴とす る、請求の範囲第16項に記載の医薬組成物。 18.GP160タンパク質の全部または一部のようなHIVウイルスの表面抗原を発現 する欠陥組換えアデノウイルスを含む単球−マクロファージ株の細胞を含んで成 る医薬組成物。 19.注入可能な状態であることを特徴とする、請求の範囲第10ないし第18 項のいずれか1項に記載の医薬組成物。 20.静脈または腫瘍内注射を目的とする、請求の範囲第18項に記載の医薬組 成物。 21.潅流を目的として配合された、請求の範囲第18項に記載の医薬組成物。 22.投与量あたり105-109細胞を含んで成ることを特徴とする、請求の範囲第 10ないし第21項のいずれか1項に記載の医薬組成物。 23.請求の範囲第1ないし第9項のいずれか1項に記載の単球−マクロファー ジの調製法であって、 −血液または骨髄から単球−マクロファージまたはその前駆体を取り出し、そ して単離し、 −これらの細胞を培養し、 −これらの細胞を上記定義の組換え核酸で形質転換させ、そして適当な場合に は、 −得られた細胞をパッケージングおよび/または保存する、 工程を行うことを特徴とする上記方法。
