Claims (1)
- 【特許請求の範囲】 1.1個またはそれ以上の細胞をポリマービーズ内に封入し; ビーズ内の生細胞の増殖に十分な時間、増殖を支持する培地中でビーズ内の細 胞をインキュベーションし; 次いで、ビーズ内の細胞の異なる属性に関連した物理的特性に関して、ビーズ またはビーズに近接する部分を非侵入的かつ物理的に試験する工程により特徴づ けられる、異なる属性を有する微生物を識別する方法。 2.選択された属性を有する細胞を含むビーズをかかる細胞を含まないビーズ から分離するさらなる1工程または複数の工程により特徴づけられる請求項1記 載の方法。 3.増殖した生細胞を含むビーズを生細胞を含まないビーズから分離するか、 1の形態の細胞を含むビーズを別の形態の細胞を含むビーズから分離するか、あ るいは生存率および/または増殖レベルの異なる細胞を含むビーズを分離するこ とにより特徴づけられる請求項1または2記載の方法。 4.単一細胞が各ビーズ内に封入される請求項1、2または3記載の方法。 5.ビーズがアルギン酸ポリマービーズ、特に、アルギン酸カルシウムビーズ であることにより特徴づけられる請求項1ないし4のいずれか1項に記載の方法 。 6.個々の粒子(102A)、(102B)を非侵入的かつ物理的に検出し、 粒子または粒子に近接した部分の物理的特徴に基づいて識別してクラス分けする のに用いられる検出手段(111)、(112)、(406)、(502)に粒 子を提供するための手段(104)、(403)、(501)、(601); 該クラスのうちの1のクラスの粒子(102A)、(102B)を別のクラス の粒子から物理的に分離するのに用いられる粒子配向手段(119)、(303 )、(304)、(305)、(306)、(310)、(311)、(403 )、(504)、(602)、および; 該検出手段の出力に応答して粒子配向手段をコントロールするのに用いられる 調節手段(122) からなる、物理的に識別可能な粒子(102A)、(102B)を分離するため の装置。 7.粒子(102A)、(102B)が、流体(103)の流れ中において、 検出手段に輸送および/または提供されることを特徴とする請求項6記載の装置 。 8.流体中の粒子(102A)、(102B)を懸濁するための手段からなる ことを特徴とする請求項7記載の装置。 9.粒子を検出手段に提供する手段が、懸濁液を1つまたはそれ以上の流体の 流れに分流するための手段(104)であって、流れの中で粒子(102A)、 (102B)がそれぞれ連続して処理され、かつ、流体の流れが検出手段(11 1)、(112)の近傍へと配向されている手段であることを特徴とする請求項 7または8記載の装置。 10.粒子がそれぞれ連続して処理される流れに懸濁液を分流する手段が、粒 子直径の2倍未満の内径を有する1つまたはそれ以上の流れ管(104)からな ることを特徴とする請求項9記載の装置。 11.検出および識別手段(111)、(112)の下流において、流れ管( 104)が開口端(104C)となって終了し、この開口端(104C)が粒子 受け(119)の導入口に対面しており、その結果、粒子(102B)が、流れ 管の開口端(104C)から開口端と導入口との間の間隙を越えて粒子受け(1 19)の導入口の中に入るものであり、流れ管の開口端(104C)および/ま たは導入口に流れ調節手段(122)が設置されて流れ管の開口端(104C) から該導入口への粒子の通過を許可または阻止することを特徴とする請求項10 記載の装置。 12.粒子受け(119)の導入口、および/または流れ管の開口端(104 C)が、互いに相対的に直面した位置関係および直面しない位置関係に移動可能 であり、その結果、それらが対面した位置関係にある場合には粒子(102B) が流れ管(104)から粒子受け(119)中へと通過できるが、対面した位置 関係にない場合には通過できないことを特徴とする請求項11記載の装置。 13.流れ管の開口端(104C)および粒子受け(119)の導入口がとも に正の気圧の領域(113)にあり、粒子受け(119)がより低い気圧の領域 (126)に連絡していることを特徴とする請求項11または12のいずれか1 項に記載の装置。 14.流れ管(104B)が「Y字」に分枝しているか、さもなくばフォーク 状の管の配置であり、懸濁液の流れが柄の中をフォークに向かって流れ、この柄 の一部をモニターする検出手段(111)、(112)を備え、さらに柄の中を 移動する選択された粒子をフォークの選択された枝(301)、(302)、( 308)、(309)、(313)、(314)に向かって流すための迂回手段 (303)、(304)、(305)、(306)、(310)、(311)が 装備されており、そのため上記枝のうちの1本またはそれ以上が粒子受けからな っていることを特徴とする請求項10記載の装置。 15.捕獲した粒子を収集装置へと配向させるために、粒子受け(119)が 粒子配向器(120)に連絡していることを特徴とする請求項11ないし14の いずれか1項に記載の装置。 16.粒子配向器(120)が、粒子受けに至る下流への伸長部分からなり、 伸長部分(120)に沿った正の気圧の変化によって粒子が配向されうることを 特徴とする請求項15記載の装置。 17.粒子受け(119)の開口端の下流への伸長部分(120A)および該 伸長部分の終端の上流の領域が、低真空の多岐管(126)により囲まれている ことを特徴とする請求項16記載の装置。 18.粒子を検出手段(406)に提供するための手段が、粒子を把握する形 態または粒子を吸引する形態の1つまたはそれ以上の粒子配向プローブ(403 )からなることを特徴とする請求項6記載の装置。 19.粒子(102A)、(102B)がプレート(501)、(601)を 渡るように流されて、その結果、粒子(102A)、(102B)の2次元配列 が形成され、選択された個々の粒子(102A)、(102B)に向けられるこ とのできる粒子配向プローブ(504)、(602)を用いて粒子を配列から取 り、それらを収集(130)、(131)へと配向させることを特徴とする請求 項6記載の装置。 20.配向手段が光学的検出手段(111)、(112)、(502)である ことを特徴とする請求項6ないし19のいずれか1項に記載の装置。 21.添付図面を参照して本明細書に実質的に記載された請求項6ないし20 のいずれか1項に記載の装置。 22.培養した1個またはそれ以上の細胞をポリマービーズ内に封入し; ビーズ内での増殖のための十分な時間、増殖を支持する培地中でビーズ内の細 胞をインキュベーションし; 次いで、請求項6ないし21のいずれか1項に記載の装置を用いて、選択され た属性を有する増殖した生細胞を含むビーズを、空であるかまたは選択された属 性を有しない細胞を有するビーズから分離することを特徴とする、異なる属性を 有する細胞を識別するための方法。 23.添付図面を参照して本明細書に実質的に記載された請求項1ないし5、 または請求項22のいずれか1項に記載の、異なる属性を有する細胞を識別する ための方法。
