Claims (11)
- 1.生物流体試料中のリガンド(L)の量を測定する方法において、 被測定リガンド(L)を含有している該生物流体が、液相に於て、不溶形に変換 することが出来る溶解されたリガンド誘導体(L−IgG)の存在下で、被測定 リガンド(L)及びリガンド誘導体(L−IgG)が標識化された特異的バイン ダー(Ak■)について競争するようにそしてリガンド誘導体(L−IgG)が 不溶形に変換されるように、被測定リガンド(L)及びリガンド誘導体(L−I gG)の両方に結合する標識化された特異的バインダー(Ak■)の化学量論量 未満とともに培養され、 そして、液相中に残っている検定系の成分から検定系不溶成分を分離した後に、 該生物流体中の被測定リガンド(L)の量が、リガンド誘導体(L−IgG)に 対し結合されている標識化された特異的バインダー(Ak■)の量から計算され る方法に於て、 リガンド誘導体(L−IgG)として基体タンパク質とリガンドとのコンジュゲ ートが使用され、 該反応が該コンジュゲートのリガンド部分及び特異的バインダー(Ak■)の間 の結合に影響を与えることなしにリカンド誘導体(L−IgG)の基体タンパク 質部分に特異的に結合するタンパク質物質(抗1gG)で壁が被覆されている試 験管内で実施されることを特徴とする方法。
- 2.リガンド(L)が一価の抗原又はハブチンであることを特徴とする請求項1 に記載の方法。
- 3.リガンド誘導体(L−IgG)の基体タンパク質部分が免疫グロブリンであ ることを特徴とする請求項1又は2に記載の方法。
- 4.タンパク質材科(抗IgG)が、リガンド誘導体(L−lgG)の基体タン パク質部分に対する抗イディオタイプ免疫グロブリン又は抗イディオタイプポリ 又はモノクローナル抗体によって形成され、そしてリガンド誘導体(L−IgG )の基体タンパク質部分とタンパク質材科(抗1gG)との間の結合が、リガン ド(L)又はリガンド誘導体(L−IgG)のリガンド部分と標識化特異的バイ ンダー(AK■)の間の結合よりもゆっくりと行われる請求項1、2、又は3の いずれか1に記載の方法。
- 5.次の培養段階の順序で実施されることを特徴としている請求項1〜4のいず れか1に記載の方法。 (a)リガンド誘導体(L−IgG)の基体タンパク質部分と特異的に結合する タンパク質材料(抗1gc)の過剰量で試験管壁が被覆されている試験管を準備 し、(b)未知の量の被測定リガンド(L)を含有している生物流体又はリカン ド(し)の標準溶液を添加し、(c)リガンド誘導体(L−IgG)の既知の量 を含有している溶液又は分散液を添加し、そして (d)既知の量の標識化された特異的バインダー(Ak■)を含有している溶液 又は分散液を添加することを特徴とし、 そして適当な培養期間の後、液相を試験管から除去し、試験管を洗浄し、そして 次に調べている試料中の被測定リガンド(L)の量の割合を、標準試料の結果を 参照して、試験管壁に結合された標識化された特異的バインダー(Ak■)の量 から決定することを特徴とする請求項1〜4のいずれか1に記載の方法。
- 6.生物流体中の被測定リガンド(L)が部分的に遊離型で、そしてまた部分的 に生理学的結合タンパク質と結合した形で存在し、この方法が遊離型の割合の排 他的な測定法であり、そしてりかンド誘導体が生理学的結合タンバク質と比較し てかなり減少した結合能力しか有さないものであることを特徴とする請求項1〜 5のいずれか1に記載の方法。
- 7.この方法がチロキシン(FT4)の遊離割合を測定する方法であって、リガ ンド誘導体(L−IgG)がチロキシン−IgGコンジュゲートであり、試験管 壁に結合したタンパク質材科(抗IgG)がリガンド誘導体(L−IgG)のI gG部分に対する抗イディオタイブ(anti−idiotypical)なI gGであることを特徴とする請求項6に記載の方法。
- 8.リガンド誘導体(L−IgG)がチロキシン−ウサギIgGコンジュゲート であり抗イディオタイブIgG(抗−IgG)がヤギ抗ウサギIgG又はヤギ抗 ウサギ抗体である請求項7に記載の方法。
- 9.使用されるリガンド誘導体(L−IgG)の合計量を完全に結合するのに少 なくとも十分なタンパク質材料(抗IgG)の量で試験管壁が被覆されており、 使用されるリガンド誘導体(L−IgG)の量が普通の患者で期待される被測定 リガンド(L)の量に基づくモル量の0.5〜20倍のオーダーの量であり、試 料中に存在する該バインダーの量と特定の親和性を表わしている、被測定リガン ド(L)及びリガンド誘導体(L−IgG)の合計量の特定のフラクションのみ が標識化された特異的バインダー(Ak■)と反応するような、化学量論量以下 の量で標識化された特異的バインダー(Ak■)が使用されることを特徴とする 請求項1〜8のいずれか1に記載の方法。
- 10.リガンド・免疫グロブリンコンジュゲートの形態のリガンド誘導体の既知 の量、標識化された抗リガンド抗体の既知の量、及びもし要求されるならば、リ かンドの標準溶液及び適当な緩衝液及び希釈溶液、並びに該リガンド誘導体の免 疫グロブリン部分に対する抗イディオタイブ免疫グロブリンの過剰で壁が被覆さ れている試験管を含有していることを特徴とする請求項1〜9のいずれか1に記 載の方法を実施する為のキット。
- 11.リガンド誘導体、標識化抗体、もし要求されるならばリガンド標準試料、 及び被覆された凍結乾燥形の試験管を含有することを特徴とする請求項10に記 載のキット。
