Claims (22)
- 1.a)DNAポリメラーゼおよび少なくとも1種のdNTPが標識されている 1種ないし3種のdNTPの存在下で、配列決定すべきDNAの領域に相補的な ポリヌクレオチドプライマーと配列決定すべきDNAとを接触させて、1種また はそれ以上の該dNTPを該プライマーに付加させることにより該プライマーを 伸長させて、伸長したプライマーを形成し、b)該プライマーを該DNAより解 離させ、c)段階a)およびb)を複数回繰り返し、ついでd)DNAポリメラ ーゼ、4種のdNTPおよび鎖伸長終結試薬の存在下で、該伸長したプライマー を該DNAと接触させる段階からなるDNA配列決定法。
- 2.該ポリメラーゼがTaqDNAポリメラーゼである請求項1記載の方法。
- 3.該ポリメラーゼがΔTaqDNAポリメラーゼである請求項2記載の方法。
- 4.該ポリメラーゼが、低エキソヌクレアーゼ活性を有するT7DNAポリメラ ーゼ、クレノウ(Klenow)、AMV逆転写酵素、BstDNAポリメラー ゼ、TthDNAポリメラーゼ、Ventポリメラーゼ、エキソヌクレアーゼ活 性のないVentポリメラーゼおよびエキソヌクレアーゼ活性のないクレノウか ら選択される請求項1記載の方法。
- 5.該標識が35Sである請求項1記載の方法。
- 6.該標識が蛍光標識である請求項1記載の方法。
- 7.該標識が化学発光標識である請求項1記載の方法。
- 8.該標識が間接的な酵素結合アッセイにより検出されるリガンドである請求項 1記載の方法。
- 9.該複数回が10ないし200回の間である請求項1記載の方法。
- 10.さらに該方法が、段階d)の後にさらに該DNAから該伸長したプライマ ーを分離する段階e)からなる請求項1記載の方法。
- 11.該段階d)およびe)をさらに複数回繰り返す請求項10記載の方法。
- 12.該複数回が10ないし200回の間である請求項11記載の方法。
- 13.該DNAがM13DNAである請求項1記載の方法。
