発明の名称
循環器治療薬として有用な酸化窒素−第一級アミン安定化コンプレックス
背景技術
数多くの酸化窒素−求核コンプレックスが、例えばR,S、Drago、AC3
Adv、Chem、Ser、Vol、3B、p、143−149(1962)に
記載されている。これらのコンプレックスの成るものは、加熱あるいは加水分解
により酸化窒素を放出することが知られている。(例: T、J、Hansen
etal、、IARCSC1,PUBL、、Vol、41. p、2l−29
(1982)) 、また酸化窒素は、ある種の血管拡張神経の作用を促しくR。
M、J、Palmer、et al、、Nature Vol、327.p、5
24−526(1987))、脳の細胞間交流(intercellular
communication)のあるタイプの作用を促す(J、Garthwa
ite、et al、。
Nature Vol、336.p、385−388(1988))内皮細胞由
来平滑筋弛緩因子(EDRF)と同じものであると想定されている。酸化窒素−
第一級アミンコンプレックスを生理活性剤として用いることは、これまで開示さ
れていない。
発明の要約
本発明の一目的は、循環器病の治療に有用な有力な循環器治療薬を提供すること
である。本発明の循環器治療薬は、酸化窒素および第一級アミン、および、エス
テル、エーテル、あるいはその他の誘導体から生成したコンプレックスである。
これらの酸化窒素−第一級アミンコンプレックス、および、エステル、エーテル
、あるいはその他の誘導体は、酸化窒素を生体中(in vivo )に再生す
なわち放出し、このin vivo酸化窒素の放出により生理活性が得られる。
さらに、酸化窒素−第一級アミンコンプレックスおよびそのエステルからの酸化
窒素の放出速度は、pH,および、温度などのその他の要因により、殆ど瞬間か
ら非常に緩慢まで変わるので、本発明の第二の目的は、薬理学的試験により決め
られる許されつる態様で酸化窒素をin vivoに放出する安定化コンプレッ
クスを提供することである6本発明の第三の目的は、ここに開示する循環器治療
薬を哺乳動物に投薬する薬品組成物を提供することである。
従って、本発明は上記の目的に沿って、循環器病の治療法を提供するものである
。この治療法は、必要に応じて、次の式1の化合物を有効量哺乳動物に投薬する
ことより成る。
[R−N(H)N(NO)0−]yX 式1Rは、低級アルキル、アリール(a
ryl)、アリールアルキル、あるいは、シクロアルキルであり、これらのRは
いずれもハロ、ヒドロキシ、アルコキシ、アミノ、アミド、カルボニル、カルボ
キシ、あるいはニトロから選択された工ないし3傭の置換基(同じものでも異な
ったものでもよい)で置換されたものでもよく、Xは、生理的に受容できるカチ
オン、
生理的に受容できる金属中心、 或いは、生理的に受容できる低級アルキル、ア
シル、あるいはアミドがら選択された有機基であり、Yは、Xの結合価数に対応
して1または2である。
さらに、本発明は、次の(1)および(I I)より成る薬品組成物を提供する
。
[R−N()I)N(No)0−]、X 式1(ここで、Rは、低級アルキル、
アリール(aryl)、アリールアルキル、あるいは、シクロアルキルであり、
これらのR基はいずれもハロ、ヒドロキシ、アルコキシ、アミノ、アミド、カル
ボニル、カルボキシ、あるいはニトロから選択された工ないし3個の置換基(同
じものでも異なったものでもよい)で置換されたものでもよく、
Xは、生理的に受容できるカチオン、
生理的に受容できる金属中心、或いは、低級アルキル、アシル、あるいはアミド

用語の意味を明確にするため、本明細書で用いる用語の幾つかを、次に説明する
。
本明細書で用いる「低級アルキル」という用語は、炭素原子数3乃至8の分岐鎖
および直鎖の基を意味し、例えば、プロピル、イソプロピル、ブチル、2−ブチ
ル、tert−ブチル、アミル、イソアミル、ヘキシル、ヘプチル、オクチル基
などである。
本明細書で用いる「アリール」という用語は、フェニル、ナフチル、ピロリル、
ピリジニル、キノリニル(quinolinyl)、イソキノリニルなどを意味
する。
本明細書で用いる「アリールアルキル」という用語は、炭素原子数1乃至3の直
鎖または分岐鎖の基で置換した上に定義したアリール基を意味する。
本明細書で用いる「シクロアルキル」という用語は、シクロプロピル、シクロペ
ンチル、シクロヘキシル、シクロヘプチル、シクロオクチル、ピロリジニル(p
yrrolidinyl)、ピペリジニル(piperidinyl)、 およ
および、ピペラジニル(piperazinyl)などを意味する。
本明細書で用いる「ハロJ或いは「ハロゲン」という用語は、弗素、塩素、臭素
、および沃素から選択されたハロゲン原子を意味する。
本明細書で用いるrヒドロキシ」あるいは「ヒドロキシル」という用語は、−O
Hを意味する。
本明細書で用いる「アミノ」という用語は、−Nl2を意味する。
本明細書で用いる「アミド」という用語は、−C(0)Nl(、を意味する。
本明細書で用いる[カルボニルJという用語は、−CH(0)を意味する。
本明細書で用いる「カルボキシ基という用語は、−c(o)oHを意味する。

本明細書で用いる「生理的に受容できるカチオン」という用語は、哺乳動物中で
生理的に相客れる任意のカチオンを意味し、例えばイソプロピルアンモニウムカ
チオン等のようなアルキルアンモニウムカチオン、例えばナトリウム、カリウム
、リチウム等のようなアルカリ金属、および、カルシウム、バリウム、マグネシ
ウム等のようなアルカリ土類金属が包含される0選択するカチオンの唯一の木質
的な特徴は、哺乳動物中で生理的に相客れないものではないことである。
本明細書で用いる「生理的に受容できる金属中心」という用語は、1価から3価
で、式1のY個の有機基(即ち[R−N(H)N(NO)0−コ)の各々の非金
属イオンの一つ以上と共有結合で結合した中心の金属イオンを意味し、二のよう
な金属中心の例は、式1のY個の有機基の各々にある二つの酸素原子、あるいは
、少なくとも一木明細書で用いる「中心金属イオン」という用語はナトリウム、
カリウム、リチウム等のようなアルカリ金属、カルシウム、マグネシウム、バリ
ウム等のようなアルカリ土類金属、鉄、銅、ニッケル、亜鉛等を含む遷移金属、
アルミニウム等を含む第1II族金属、および、ランタノイド系金属から選択し
た生理的に受容できる金属を包含する0選択した中心金属イオンについては、哺
乳動物中で生理的に相客れることが唯一の本質的な要求である。
本明細書で用いる「生理的に受容できる有機基」という用語は、式 工の有機基
(即ち[R−N()I)N(NO)O−])と共有結合して、エーテル、エステ
ルおよびその他の誘導体(例: R−1t(H)N(NO)0−X : 、:、
: テXGf、生理的に受容できる有機基である)を生成する生理的に受容でき
る有機基を意味する。このような、生理的に受容できる有機基は、例えば、低級
アルキル、アシル、アミド等である。
本明細書で用いる「アシル」という用語は、カルボキシ基の−OHが、他の置換
基で置換された有機基を意味する。このようなアシル基は、例えば−C(0)C
)Is−−C(0)CeHs等である。
本明細書で用いる「アルコキシ」という用語は、−0−CH3、−0−C2H,
、および−〇−低級アルキルを意味する。
本明細書で用いる「調剤上許容できる担体」という用語は、静脈注射あるいは日
経で薬品組成物を投薬したい時に、薬品業界で許容されるものとして知られてい
る賦形剤および担体を意味する。
発明の詳細な説明
式1の化合物は、安定な酸化窒素−第一級アミンコンプレックスおよびそのエス
テルであり、いずれも一般に、制御された形で、酸化窒素を生体中において(i
n vivo ) 放出する。このように酸化窒素を制御さされた形で放出でき
るので、薬剤として貴重であり、高血圧、動脈硬化症、脳動脈撃縮(cereb
ralvasospasm)、および冠状動脈血管痙@ (coronaryv
asospasm)のような循環器病の治療に有用なものとして、充分本発明に
おいて考慮されている。
式■に包含されるすべての化合物が循環器病の治療に価値があり、本発明の方法
において有効と考えられるが、一般的に、R基が炭素原子数3−6の(置換また
は未置換の)分岐低級アルキル基である式1の化合物が本発明に於いて最も好ま
しいと考えられる。しかしR基が、(置換または未置換の)シクロアルキルある
いはシクロ低級アルキルであるものも好ましい、さらに、これら好ましいR基が
置換されているならば、最も好ましい置換基は、ハロ、ヒドロキシ、アミハカル
ボニルあるいはカルボキシル基の一つ乃至三つ(同じものあるいは異なったもの
)であると考えられる。
また、Xとして、低級アルキルアンモニウムカチオン、アルカリ金属カチオン、
あるいはアルカリ土類金属カチオンを有する化合物が、本発明の方法に用いるの
に最も好ましいと考えられる。
本発明の方法に用いられる式■の化合物は、当業者に知られている種々の化学合
成法により製造でき、以下に示す方法は、式1の化合物を製造する方法の例に過
ぎず、これらは本発明の範囲を、如何なる意味に於いても限定するものではない
。
本発明の方法に用いられる酸化窒素−第一級アミンコンプレックスは、所望であ
れば、R,S、Drago et al。
J、Am、Chen+、Soc、、Vol、83. p、1819−1822
(1961)の方法で製造できる。 Dragoの教示する反応スキームは、次
のとうりである。この反応で、(BNO)は、転移状態若しくは、反応中に生成
する中間体である。

→ [RNHzNzOzコ[RNHzNzOzコ + RNI(2→ RNI(
3” R)lHN20□−上記のDrago反応は、適当なアミンの冷(−78
℃)エーテル溶液中に酸化窒素を発泡させて行なうことができる。 Drago
が教示するところによれば、純固体生成物が沈澱するはずであり、この沈澱物は
所望によりクロロホルム溶液からエーテルで再沈澱することができる。さらに、
この反応でR,S、 DragoらがJ、Am、Chem、Soc、、Vol、
83. p、1819−1822 (1961)において教示する高圧技術によ
り、製品を理論値の約70−80%の収率で得ることができる。
と記の反応では、最初に生成する X基は必然的にアンモニウムカチオンすなわ
ちBNO3”である。このコンプレックスのカチオンをアルカリ金属あるいはア
ルカリ土類金属カチオンに変えるには、次の反応スキームを、用いることができ
る。ここで、「は、アルカリ金属カチオンか、あるいは、アルカリ土類金属カチ
オンかのいずれかである。

M” + RNI(LO□このようにして得られた所望の塩を、当業者に知ら
れた適当な有機溶媒から再結晶させ、あるいは単に適当な溶媒で洗浄して、精製
製品を得ることができる。
一旦得られれば、適当な金属中心、アルキル化剤、アシル化剤、あるいはカルバ
モイル化剤と反応させ、上記の塩を式■により包括される共有結合金属コンプレ
ックス、エーテル、あるいはエステルに転化することができる。これらを製造す
るいろいろな方法あるいは処置は、当業者に一般的に知られており、それらを式
1の化合物でX基が先に定義した生理的に受容できる金属中心あるいは生理的に
受容できる有機基である化合物を製造することは充分考慮されている。
上述のDrago反応メカニズムを用いる場合には、酸化窒素と反応すべきある
種の第一級アミンは、酸化窒素と反応させたくない余分な窒素、酸素、あるいは
その他のへテロ原子で水素で置換されたものを含むことがある。そのような場合
には、酸化窒素との反応の前に、これらの水素置換へテロ原子を適当なブロッキ
ング基でブロックせねばならない、ブロックされたヘテロ原子は、第一級アミン
と酸化窒素との Drago反応の後で、ブロックを解かれる。適当なブロッキ
ング剤および解ブロッキング剤およびその用法は当業者に一般的に知られており
、それらを式1の化合物でR基が、少なくとも一つの水素原子で置換されたヘテ
ロ原子を少なくとも一つ含有する、あるいは/および、そのような少なくとも一
つのへテロ原子で置換されたものである式1の化合物の製造に用いることは充分
考慮されている。
以下の製法例は、式1の化合物を製造するのに用いることができる本明細書記載
の方法をさらに説明するものであり、本発明の範囲を限定するものと解してはな
らない。

ピルアミンおよびエーテルの溶液を酸化窒素と一78℃の温度で約24時間反応
させ標記の酸化窒素コンプレックスを得た0反応は、高圧反応容器中で高圧下で
行い、酸化窒素は高圧貯槽を経て反応混合物に加えた6標記化合物は満足な収率
で得られた。

れた。

本発明に包含される成る化合物を、循環器治療薬としての活性を調べるために、
薬理学的モデルで試験した0本明細書で開示する化合物の薬理学的活性は、主と
して酸化窒素を生体中(in vivo )で発生することにあるのであり、式
1の化合物はすべて酸化窒素をin vivoで発生することができるのである
から、以下の例は、本発明で開示する化合物の数を限定するものと解してはなら
ない。本明細書で開示するすべての式1の化合物が、高血圧、動脈硬化症、脳動
脈筆線、および冠状動脈血管痙牽のような成る種の循環器病を、酸化窒素を生体
中(in vivo )で制御しつつ発生することにより有効に治療する一般的
能力を説明するものと解すべきである。
倒−上
製法例工の化合物すなわち
(C)1s)zcHNH,’″−0−NOJO)N(H)(−C)I(CH3)
2)を、正塩水で0.05 M溶液に調整した。麻酔したラット(350g)に
、この溶液0.04m1を、カテーテルを経て大腿静脈に、30秒間で注射した
ところ、ただちに血圧が80mm)Igから30mmHgに下がり、代償として
心拍数が320/分から340/分に上昇した。低下した血圧は、測定可能な増
加が見られるまで、 10分間そのままであった。注射後30分後までに完全に
もとの値に戻った。のちに、製法例工の化合物のより少ない投与量のポーラスを
同じ麻麻酔したラットに投与したところ、より大きな投与量の場合と同様なこと
になった。さらに後に、製法例■の化合物の0.0014 Mを連続的に1.4
m17分の速度で同じ麻酔したラットに注入した。もとの血圧が、10分間に
わたって、105/’13mmHgから83/85 +++n+Hgに低下した
。
また、連続投与(上記の速度で)中、製法例式1の化合物は、ラットに与えたア
ンギオテンシンII(angiotensin H)の0.5nmolの作用に
は影響を及ばさないが、製法例工の化合物を連続注入(上記の速度で)すると、
ラットに与えた0、 5nmolのエンドセリン(endothelin)のポ
ーラスの血管収縮作用を完全に廃絶したことが分かった。これらの結果から、製
法例工の化合物は有効な循環器治療薬であり、その作用は予測でき、且つ可逆的
であることが結論ずけられる。

の化合物と同様にして、麻酔したラットにカテーテルを経て投与すると、同様に
予測でき且つ可逆的な血圧低下が期待できる。

本明細書の式Iに包含される任意の化合物を、例1と同様な方法で麻酔したラッ
トに投薬すると、用いた式1の化合物による酸化窒素のin vivo発生によ
り、血圧が同様に低下することを予測できる。
薬品組成物
本発明の化合物で、Xが生理的に受容できるカチオンであるときは、その化学構
造のため、静脈注射で投薬するのが最も好ましく、Xが生理的に受容できる金属
中心あるいは有機基であるときは、静脈注射か経口かで投薬するのが好ましい0
本発明の化合物は、調剤上許容できる適当な担体あるいは希釈体とともに、注射
あるいは経口薬品組成物とするのが望ましい、さらに、本発明の式Iの化合物は
これらの投薬ルートに普通である方法で注射薬および経口薬に処方できる。以下
に述べる方法および賦形剤は、普通の許容できるものの例であるが、薬品組成物
に関して本発明の範囲を限定するものではない。
本発明の化合物は、水溶液、あるいは、植物油、合成脂肪酸グリセライド、高級
脂肪酸エステルあるいはプロとレンゲリコールエステルのような非水溶媒に、溶
解、懸濁、あるいは乳化させて、注射用に処方することができ、所望により、可
溶化剤、等張化剤、懸濁剤、乳化剤、安定剤、および保存剤のような一般的な添
加剤を一緒に用いることができる0本発明の化合物の非経口投薬は、デキストロ
ーゼ、注射用殺菌水、USPlあるいは正塩水のような調剤上許容できる担体を
有することができる。
経口処方の場合は、本発明の化合物は、単独で、あるいは、適当な、例えば、ラ
クトース、マンニトールコーンスターチあるいはポテトスターチ等の一般的添加
物:あるいは、結晶性セルロース、セルロース誘導体、アラビアゴム、コーンス
ターチ、あるいは、ゼラチン等のバインダー;コーンスターチ、ポテトスターチ
、あるいは、カルボキシメチルセルロースのような砕解剤;タルク、あるいは、
ステアリン酸マグネシウムのような潤滑剤;および、所望により、希釈剤、バッ
ファー剤、湿潤剤、保存剤、および芳香剤といったものと一緒に用いて、錠剤、
粉末、顆粒あるいはカプセルとすることができる6
循環器治療薬として用いるべき本発明の化合物の量は、もちろん、その循環器病
のタイプおよび投薬法により異なる。静訊注射による高血圧、動脈硬化症、脳動
脈牽縮、および冠状動脈血管連撃の治療に適当と考えられる量は、約0.01乃
至10.0 mg/kg(体重)/dayである。好ましい量は、その循環器病
を治療するのにちょうど充分な量であることは勿論であり、約0.05乃至5.
0 mg/kg/ dayであろう。
本発明の範囲は、請求の範囲によってのみ限定される。

Claims (20)
- 1.化学式[R−N(H)N(NO)O−]YX(ここで、 Rは、低級アルキル、アリール、アリールアルキル、あるいは、シクロアルキル であり、これらのR基はいずれも、ハロ、ヒドロキシ、アルコキシ、アミノ、ア ミド、カルボニル、カルボキシ、あるいはニトロから選択された同じあるいは異 なった1または3個の置換基で置換されたものでもよく、 Xは、生理的に受容できるカチオン、 生理的に受容できる金属中心、或いは、生理的に受容できる低級アルキル、アシ ル、あるいはアミドから選択された有機基であり、Yは、Xの結合価数に対応し て1または3である。)の化合物を、必要に応じて、有効量、哺乳動物に投薬す ることより成る循環器病の治療方法。
- 2.Rが、3〜6の炭素長の分岐鎖低級アルキル基であり、所望により、ハロ、 アミノ、ヒドロキシ、カルボニル、あるいはカルボキシルから成る群から選択さ れた同じあるいは異なった1乃至3個の置換基で置換されたものでもよいことを 特徴とする請求項1の方法。
- 3.Rが、シクロアルキルであり、所望により、ハロ、アミノ、ヒドロキシ、カ ルボニル、あるいはカルボキシルから成る群から選択された同じあるいは異なっ た1乃至3個の置換基で置換されたものでもよいことを特徴とする請求項1の方 法。
- 4.Rがイソプロピルである請求項1の方法。
- 5.上記化合物が、静脈注射により投薬されることを特徴とする請求項1の方法 。
- 6.Rがイソプロピルであり、上記化合物が、静脈注射により投薬されることを 特徴とする請求項1の方法。
- 7.Xが、生理的に受容できるカチオンである請求項1の方法。
- 8.Rが、3〜6の炭素長の分岐鎖低級アルキル基であり、所望により、ハロ、 アミノ、ヒドロキシ、カルボニル、あるいはカルボキシルから成る群から選択さ れた同じあるいは異なった1乃至3個の置換基で置換されたものでもよいことを 特徴とする請求項7の方法。
- 9.Rがイソプロピルである請求項7の方法。
- 10.Rがイソプロピルであり、上記化合物が、静脈注射により投薬されること を特徴とする請求項7の方法。
- 11.(I)化学式[R−N(H)N(NO)O−]YX(ここで、 Rは、低級アルキル、アリール、アリールアルキル、あるいは、シクロアルキル であり、これらのR基はいずれも、ハロ、ヒドロキシ、アルコキシ、アミノ、ア ミド、カルボニル、カルボキシ、あるいはニトロから選択された同じあるいは異 なった1乃至3個の置換基で置換されたものでもよく、 Xは、生理的に受容できるカチオン、生理的に受容できる金属中心、或いは、生 理的に受容できる低級アルキル、アシル、あるいはアミドから選択された有機基 であり、 Yは、Xの結合価数に対応して1または3である。)の化合物の循環器病の治療 に有効な量、および、(II)それに対し薬学的に許容できる担体、よりなる薬 品組成物。

- 15.組成物が、静脈注射で投薬されることを特徴とする請求項11の組成物。
