【発明の詳細な説明】
組換え抗体−毒素融合タンパク質
本願は、参考のため本願に組み込まれる1986年9月24日付で出願された係
属出願第06/911,227号の一部継続である。親出願(第06/911,
227号)明細書は、特異的レセプターが細胞上に存在している認識タンパク質
をコードするDNA配列と融合された、修飾シュードモナス(Pseudomo
nas)外毒素(P E)遺伝子からの組換えタンパク質の産生について教示す
る。
PE遺伝子は、PE分子の様々なドメインの特定セグメント又は配列を変えるた
めに修正きれた。これは親出願においてIL−2−PE融合遺伝子の組立て及び
発現で例示された。本出願はPEを利用した改良毒素の産生、更に詳しくは適切
な抗原又はレセプターを有した細胞を有効かつ選択的に死滅させることができる
組換え抗体−毒素融合タンパク質の合成に関する。ここで記載された研究は米国
政府により一部援助された。
図面の簡単な説明

的な構築を示す。プラスミドpVc70108は、単一ポリペプチド鎖として、
抗Tacの様々なドメイン:H鎖のVu可変ドメイン(成熟H鎖の最初の116

t)3]、L鎖のVL可変ドメイン(成熟り鎖の最初の106アミノ酸)と転位
及びADPリボシル化活性を有するPHのアミノ酸253−613とをコードす
る融合遺伝子を含んでいる。コードされたタンパク質は抗体−毒素融合タンパク
質、又は更に具体的には抗Tac (Fv)−PE40と呼ばれる。融合遺伝子
はシャイン・ダルガノ領域及び開始コドン(PT7)に結合されたT7プロモー
ターのコントロール下にある。pVc7010gを有する大腸菌株BL21 (
λDE3)はI PTG誘導でハイブリッドタンパク質を発現させるために用い
られた。

A、Ava 1.PCRに用いられるオリゴヌクレオチドは水平矢印として示さ
れた。プラスミドpVc701において、4つの余分な塩基は斜線枠内に示され

第1図(b)は、ポリメラーゼ鎖反応(PCR)に用いられるオリゴヌクレオチ
ドを示し、VKO74、VICO75及びVKO76は各々2列で示されている
。
VK O73及びVKO75(7)一部はH及びL @ c D N Aの非コ
ード鎖に相補的であり、一方VKO74及びVKO76の一部はH及びL鎖cD
NAのコード鎖に各々相補的である。VKO73及びVKO74は抗Ta(H鎖
cDNAを用いてVu上セグメント増幅するために用いられ、一方VK675及
びvK076は第1図aで示されるように抗TacL鎖cDNAを用いてVL上
セグメント生成するために用いられた。すべてのオリゴヌクレオチドは5’−3
’方向に記載された。VL又はV u c D N Aのコード鎖又は非コード
鎖に相補的な配列は枠内に示されている。Vo及びVL間のペプチドリンカ−を
コードする配列は破線で下線が引かれている。
第2図(a)は、モノQカラムクロマトグラフィーによる復元可溶性抗Ta c
(Fv)−PE40の精製結果を示す。復元物質はモノQカラムに適用され、
タンパク質がNaC1勾配(0〜0.5M)で溶出され、4m1分画が集められ
た。活性モノマー抗Tac (Fv)−PE40の位置は垂直矢印で示されてい
る。
第2図(b)は様々な精製段階におけるサンプルのSDS −PAGEの結果を
示す。ゲルはクマシーブルーで染色された。1列目、全細胞ベレット:2列目、
スフェロプラスト;3列目、超音波処理スフェロプラストの100、OOOXg
ペレット;4列目、モノQカラムによる分画(32〜33)のブール;5列目、
TSK−250カラムによるピーク分画のブール;6列目、天然PE、Mr66
kDa、分子量マーカーはkDaとしてM「に関し付記されている。
第3図は、IL2レセプターを発現するHUT −102細胞での抗Tac (
Fv)−PE40の細胞毒性の検討結果を示す。HUT−102細胞における抗
Tac (Fv)−PE40の細胞毒性は(・)抗T、ac(Fv)−PE40
; (0)抗Tac (Fv)−PE40+10Jzg抗Tac:(Δ)抗T
ac (Fv)−PE40+10μgovB3でタンパク質合成G II 定す
ることにより決定された。
第4図は、抗Tac (Fv)−PE40の競合結合分析について示す。HUT
−102細胞においてTac抗原と結合するl−125標識トレ一サー抗Tac
抗体による抗Tac (Fv)−PE40 (ロ)及び天然マウス抗Tac抗体
(・)の競合データが水声れている。実線は結合競合をモデル化したコンピュー
ター作成の理想曲線である。Roは競合物質の不存在下におけるトレーサーの結
合/遊離比であり、RO/2はトレーサー結合に関する50%阻害点であって、
それから抗Tac (Fv)−PE40に関して3.5X109M−17)結合
アフィニティが、天然抗Tacに関する9、7X109M″″lと比較して、計
算された。
発明の詳細な説明
本発明による組換え免疫毒素は抗体−PE40組換え融合タンパク質からなる。
組換え融合タンパク質は、この融合タンパク質の合成を指示するDNAセグメン
トを含んだ発現ベクター又はプラスミドにより産生される。
組成物は、抗体が結合するレセプター又は抗原を有した細胞を殺す上で有効な量
の抗体−PE40組換え融合タンパク質及び薬学上許容されるキャリアを含む。
標的細胞毒性を発揮する方法は、死滅される標的細胞を細胞毒性量の抗体−PE
40組換え融合タンパク質の組成物と接触させ、上記標的細胞は上記抗体が結合
するレセプター又は抗原を有した細胞であるが、但しその組成物は上記抗体の結
合のためのレセプター又は抗原を欠いた細胞に有意の細胞毒性を与えない(表2
)。
他にことわらないかぎり、ここで用いられるすべての技術及び科学用語は本発明
が属する当業者にとって普通に理解されるとおりの意味を有する。ここで記載さ
れたものと同様の又はそれに相当するいかなる方法及び物質も本発明の実施又は
試験で用いることができるが、ここでは好ましい方法及び物質が記載されている
。以下で記載されたすべての文献が参考のため本明細書に組み込まれる。他にこ
とわらないかぎり、ここで用いられる技術は当業者に周知の標準的方法である。
その物質、方法及び例は説明のみのためであって、限定のためではない。

19118のような文献で記載されている。抗体鎖をコードする遺伝子は当業者
に公知のいかなるクローニング操作によりcDNA又はゲノム形でクローン化し
てもよい。例えば、Maniatls et al、。

照。ここで用いられる用語“有意の細胞毒性なしに°とは、本発明の融合タンパ
ク質がいかなるはっきりとした程度又はいかなる異常なレベルにも非標的細胞の
機能に影響を与えないことを意味する。
組換え抗体−PE40融合タンパク質は1本又は2本のポリペプチド鎖から構成
される。一本領融合タンパク質の産生がここで説明されている。二本鎖融合タン
パク質を産生ずるためには、アミノ酸リンカ−はvH及びV 配列間に挿入され
ない。代わりに終結コドンがV。
配列後に挿入され、開始コドン及びリポソーム結合配列がvL配列前に挿入され
る。本発明のもう1つの態様において、■ 及びVu配列は、H鎖ヒンジドメイ
ンと共り
に又はそれなしに、一部又は全部のし及びH鎖不変領域、例えば全にL鎖不変領
域及びH鎖不変領域のCH1ドメインによって各々追随されている。v SvH
及びPE40遺伝子はプラスミドにおいていかなる順序であってもよく、このた
めPE40遺伝子はL又はH鎖遺伝子いずれかの5′又は3′末端のいずれかに
結合される。当業者であれば、更に修飾、欠失、挿入等が抗体及びPE40遺伝
子に行われてもよいことを理解するであろう。
特に、欠失又は変更は、標的細胞に対する融合タンパク質の細胞毒性を増加させ
るか又は抗体に関する抗原のない細胞に対する非特異的細胞毒性を減少させるた
めに、PE40で又は抗体遺伝子をPE40に結合させるリンカ−で行ってもよ
い。すべてのこのような組立ては当業者に周知の遺伝子工学方法で行われ(一般
的にManlatiset al、、同上参照)、様々な臨床的又は生物学的応
用に特に適したものとする異なる性質のアフイニテイ、特異性、安定性及び毒性
を有したタンパク質を産生じてもよい。
0遺伝子は単一プラスミドに含まれている。更に、合成後の組換え抗体−PE4
0タンパク質は精製さ杵るまで大腸菌宿主細胞に内在したままである。本発明の
もう1つの態様において、Vt遺伝子及びいずれかのL鎖不変領域は1つのプラ
スミド上に存在するが、vH遺伝子、いずれかのH鎖不変領域及びPE40遺伝
子は第二のプラスミド上に存在する。いずれのケースでも、vL及び/又はVu
遺伝子は細胞から組換え融合タンパク質の分泌を指示するシグナル配列で前置さ
れていてよい(Better et al、、1988.5cience、24

本発明の融合タンパク質は大腸菌、他の細菌宿主、酵母並びにCO8,CHO1
HeLa細胞系及びミエローマ細胞系のような様々なより高等の真核細胞を含め
た様。
々な宿主細胞で発現される。組換えタンパク質遺伝子は各宿主に関して適切な発
現コントロール配列に作動可能に結合される。大腸菌の場合、これにはT7、t
rp又はλプロモーターのようなプロモーター、リポソーム結合部位、好ましく
は転写終結シグナルを含む。真核細胞の場合、コントロール配列にはプロモータ
ー、好ましくは免疫グロブリン遺伝子、SV40、サイトメガロウィルス等に由
来するエンハンサ−及びポリアデニル化配列を含み、更にスプライスドナー及び
アクセプター配列も含む。本発明のプラスミドは、大腸菌の場合塩化カルシウム
形質転換で、哺乳動物細胞の場合リン酸カルシウム処理又は電気ボレーションの
ような周知方法で、選択宿主細胞中に移入できる。プラスミドで形質転換された
細胞はamp、gpt、neo及びhyg遺伝子のようなプラスミドに含まれる
遺伝子で与えられる抗生物質耐性によって選択できる。
発現されると、組換え融合タンパク質は硫酸アンモニウム沈降、アフィニティ力
ラム、カラムクロマトグラフィー、ゲル電気泳動等を含めた当業界の標準操作に
従い精製できる〔一般的にR,5copes、”Protein Puriri
ca−tion’ 、Sprlnger−Verlag、N、Y、(1982)
参照〕。少なくとも約90〜95%均一率の実質上純粋な組成物が医薬用に好ま
しく、98〜99%以上の均−率が最も好ましい。
部分的に又は所望の均−率まで精製されると、ポリペプチドは治療に(体外を含
む)又はアッセイ操作を展開及び実施するような他の場合に用いられる〔一般的
にImmunologlcal Methods、Vols、I and II
ルef’kovits andPernls、eds、、Academlc P
ress、Nev York、N、Y、(19γ9 and1981)参照〕。
本発明の組換え融合タンパク質及び医薬組成物は、特に非経口投与、即ち皮下、
筋肉内又は静脈内投与に有用好ましくは水性キャリア、に溶解された抗体又はそ
のカクテルの溶液である。様々な水性キャリア、例えば水、緩衝液、0.4%塩
水、0.3%グリシン等が使用可能である。これらの溶液は無菌であって、通常
望ましくない物質を含まない。これらの組成物は慣用的な周知の滅菌技術で滅菌
される。組成物はpH調整及び緩衝剤、毒性調整剤等のような大体の生理条件に
必要な薬学上許容される補助物質、例えば酢酸ナトリウム、塩化ナトリウム、塩
化カリウム、塩化カルシウム、乳酸ナトリウム等を含んでいてもよい。これらの
処方剤における融合タンパク質の濃度は様々であって、即ち重量で約0.5%以
下、通常的1%又は少なくとも約1%から多くて15又は20%までであり、選
択される具体的投与方式に従い液体容量、粘度、体重等に基づき主に選択される
。
このように、筋肉内注射用の典型的医薬組成物は無菌緩衝水11及び融合タンパ
ク質10Bからなる。静脈内注入用の典型的組成物は無菌リンゲル液250m1
及びタンパク質10+egからなる。非経口投与用組成物の実際の製造方法は当
業者に公知又は明白であって、RelingtOn’S PharlaCeut
ieal 5cience、15th ed、、Mackpublishlng
Co5pany、Easton、Penn5ylvania(1980)のよ
うな文献で更に詳細に記載されているが、これは参考のためここに組み込まれる
。
本融合タンパク質又はそのカクテル(即ち他のタンパク質と一緒)を含有した組
成物は、予防及び/又は治療処置のために投与できる。治療適用の場合、組成物
は疾患及びその合併症を治癒又は少なくとも部分的に抑制するために充分な量で
疾患のある患者に投与される。これを果たす上で充分な量は“治療有効量”とし
て規定される。この用途に有効な量は疾患の程度及び患者健康の一般的状態に依
存している。
予防適用の場合、本抗体又はそのカクテルを含有した組成物は発病を予防するた
め、まだ発病していない患者に投与される。このような量は“予防有効量1とし
て規定される。このケースにおける正確な量も患者の健康状態及び一般的免疫レ
ベルに依存している。
本組成物の単回又は複数回投薬は患者によって必要とされかつ耐えられる用量及
び頻度に応じて行われる。いかなる場合でも、本組成物は患者を有効に治療する
上で充分な本発明のタンパク質の量を与えるべきである。
本発明の組換え融合タンパク質の様々な用途の中には、タンパク質の毒性作用に
より排除されうる特定ヒト細胞起因性の様々な病状が含まれる。1つの好ましい
適用例は、T及びB細胞に起因する移植片対宿主疾患、臓器移植拒絶、I型糖尿
病、多発性硬化症、リウマチ様関節炎、全身性紅斑性狼癒、重症筋無力症等のよ
うな自己免疫症状の治療である。ここで開示された例は例示にすぎない。
他の好ましい適用例は様々なタイプの悪性細胞に起因する癌の治療用である。融
合タンパク質は、インビトロで、いられる。融合タンパク質の抗体部分は所定の
用途に応じて選択される。抗体の標的として機能するT細胞の膜上のタンパク質
としてはCD2 (Tl 1) 、CD3、CD4及びCD8がある。標的とし
て機能するB細胞上で主にみられるタンパク質としてはCD 10 (CALL
A抗原)、CD19及びCD20がある。CD45はリンパ球細胞上に広く存在
しうる標的である。抗体に関するこれら及び他の可能な標的リンパ球抗原はLe
ucocyteTyping Ill、A、J、McMlcheal、ed、、
0xford UniversityPress、 1987 で記載されてい
る。抗体の標的として機能する癌細胞上でみられる抗原としては癌胎児性抗原(
CE−A)、トランスフェリンレセプター、P−糖タンパク質、c−erbB2
及びAbstracts of the Th1rdInternat1ona
l Conf’erence on Monoclonal Antibody
Ismunoconjugates I’or Cancer(San Dle
go、CA、198g)で記載された抗原がある。当業者であれば、更に他のタ
イプの細胞で発現される抗原、例えば膜糖タンパク質又は表皮増殖因子レセプタ
ー等のような増殖因子もしくはホルモンレセプター、と結合する抗体も選択して
よいと理解している。
物質及び方法
下記例は限定ではなく説明のために示されている。
抗Tac (Fv)−PE40に関する発現プラスミドの構築
ここでpVc70108と命名されたプラスミドは、開始メチオニンをコードす
るATG%VLをコードする318bpDNAセグメントに45bpリンカ−で
結合された抗TacVHをコードする3 48 bpD N Aセグメントを含
むように組立てられ、次いでVLはPEのアミノ酸253−613をフードする
DNAセグメントに結合さレタ(第1図)、V −V −IJシンカ−PE40
)IL
DNAセグメント及びコードされた抗Tac’(Fv)−PE40融合タンパク
質の全配列が表1に示されている。
■ 及びVLの配列は米国特許出願節182.682号明細書で示されているが
、これは特に参考のため本明細書に組み込まれる。
第1図a及びbで示されるように、2組のオリゴヌクレオチドが以下のものを作
成するため、ポリメラーゼ鎖反応(PCR)に基づ< (Perkfn Els
er Cetus、 GeneAmp kitを用いた)増幅に用いられた:(
a ) V n遺伝子の5′末端におけるNde1部位、Vn ドメイン及びリ
ンカ−の最初のアミノ末端9コドンしかる後Kpn1部位(プラスミドpHTa
c2において抗TacH鎖cDNAと共にVKO73及びVKO74を用いる)

(b)Kpn 1部位を有するペプチドリンカ−の7カルボキシ末端コドン、■
ドメイン及びVLコード遭伝り
子の3′末端におけるHindlN部位(プラスミドpLTac2においてL
@ c D N Aと共にVKO75及びVKO76を用いる)。
pUc18における):coRIインサートとして抗TacのH及びL鎖をコー
ドする全鎖長cDNAを有したプラスミドが鋳型として用いられた。PCR増幅
後、2つの断片、即ち(1)H鎖の可変ドメイン及びペプチドリンカ−のアミノ
末端部分を含む413bp断片; (2)ペプチドリンカ−のカルボキシ末端部
分及びL鎖の可変ドメインを含む396bp断片が、単離された。413J)断
片はNdeI及びKpnlで切断され、377bp断片を得た。396bp断片
はKpnl及びHindlllで制限され、340bp断片を生じた。それとは
別に、PE由来PE43をコードする遺伝子を有したプラスミドp V C20
(Chaudhary et al、、Proc、Natl、Aead、Sci
。
USA、85:29H−2943,1988)が、Ndel及びHi ndll
lで制限され、脱リン酸化されて、3.75Kp断片を単離した。次いでこれは
、先に単離された377bp及び340bp断片と結合された。得られたプラス
ミドpvC701はT7プロモーター下で融合遺伝子を含んでいる。
この遺伝子はペプチドリンカ−中に4つの余分な塩基を含む。プラスミドpVc
701はKpnlで切断され、3′突出部分を除去するためT4DNAポリメラ
ーゼで処理されて、単離されたプラスミドpVc701020に結合された。p
Vc701020 (4,4Xb) は抗Tacの可変ドメイン及びPE43か
ら構成される融合タンパク質をコードしている。
pVc701020はHindlllで制限され、5′突出部分を除去するため
S1ヌクレアーゼで処理され、BamHIで切断され、脱リン酸化されて、3.
65Kp断片を単離した。それとは別に、pVc701020はAvalで切断
され、S1ヌクレアーゼで処理され、BamHIで切断されて、750 bp断
片を単離した。
3.65Kp及び750bp断片は互いに結合されて、プラスミドpVc701
08を生シタ。p V C70108に!、15アミノ酸リンカ−でV に結合
されたV 、及び、L H
PEのアミノ酸253−613に結合されたvLをコードする融合遺伝子を有す
る。メチオニンコドンはVoの5′末端で作成された。
先に示されてように、構築された遺伝子はT7プロモーターのコントロール下に
ある。プラスミドのコード領域の正確性はDNA配列決定で確認された(データ
示さず)。
プラスミドpVc70108の寄託は1989年3月31日付でメリーランド州
ロックビルのATCCに受託番号67913として行われた。寄託物は寄託臼か
ら30年間又は寄託サンプルの最終請求日から5年間のいずれか長い期間にわた
り生存維持され、それが死滅したならば交換されるべきであり、法律の規定に従
い無制限に本出願による特許権の発行により公に利用される。特許商標局長は請
求により寄託物を入手することになる。
抗Ta c (Fv)−PE40の精製及び特徴IPTO誘導で、プラスミドp
vc70108を有する大腸菌株BL21 (スDE3)は、5DS−PAGE
(1列目、第2図b)で示されるように、多量の約65kDaのタンパク質を産
生じた。免疫プロットにおいて、65kDaキメラタンパク質はPHに対する抗
体と反応した(データ示さず)。融合タンパク質は超音波処理スフェロプラスト
(2列目、第2図b)の100.OOOXgペレット(3列目、第2図b)に大
部分食まれていた。
このベレットは抗Tac (Fv)−PE40の製造源として用いられた。塩酸
グアニジン変性しかる後迅速な希釈の技術がキメラタンパク質を溶解かつ復元す
るために用いられた(Chaudhary et al、、19g11.Nat
ure、835.889−s’y2: Siegall et al、、198
8.Proe、Natl、Aead、Sc1.DNA。
85.9738−9742)。DTTは変性緩衝液から省略され、復元は16時
間行われた。復元及び透析後、サンプルはモノQカラム(HRIO/10)に3
ml/sinで適用された。カラムは緩衝液A(20mMトリス塩酸、pH7
,6)40m+で洗浄され、2001直線勾配(0〜0.5M NaC1)で展
開された。溶出したタンパク質は280n−でモニタ−された。分画(41)が
集められ、HUT−102細胞の細胞毒性に関して試験された。5DS−PAG
Eのため、サンプルはレムリ(Laeanl 1)サンプル緩衝液と共に煮沸さ
れ、10%ゲル上で電気泳動された。モノマー形の融合タンパク質は0.2〜0
.22M NaC1で溶出した(第2図す、4列目及び第2図a)。高分子量凝
集物は高イオン強度側で溶出された(分画42−50、第2図a)。更にキメラ
タンパク質の精製がTSK−250ゲル濾過カラムで行われ、キメラタンパク質
は65kDaタンパク質に関して予想された位置で対称ピークとして溶出した(
データ示さず)。SDS −PAGEではそのタンパク質が純度95%以上であ
ることを示しく5列目、第2図b)、N末端アミノ酸配列分析ではそのタンパク
質が予想N末端配列met、gin、valを有することを示した。約200μ
gの高度精製モノマー抗Tac (Fv)−PE40が誘導前に0.6の0D8
50まで増殖された細胞11から得られた。
抗Tac (Fv)−PE40の細胞毒性試験HUT−102細胞は無血清培地
で2回洗浄され、24ウエルプレート内の5%牛脂児血清含有RPMII640
培地に3×105細胞/ウエルでいれられた。様々な希釈率の抗Tac (Fv
)−PE40が0.2%ヒト血清アルブミン含をPBSで調製され、適切なウェ
ル−ロイシンで標識され、細胞ペレットのTCA沈降物中における放射能が測定
された。結果は毒素無添加のコントロールの%として表示される。競合の場合に
は10Mgの抗Tac (Fv)−PE40゜
抗Ta c (Fv)−PE40の競合結合分析トレーサーとしてのl−125
標識抗Tac(2μC1/μg)が、結合緩衝液(10%牛脂児血清、100μ
g/lヒトIgG、0.1%ナトリウムアジド含有RPM11640)0.2m
l中で、様々な濃度の競合物質とTac抗原源としての4X105HUT−10
2細胞と共に、1. 5ng/アッセイで用いられ、室温(22−24℃)で2
時間混ぜながらインキユベートされた。これらの条件下で、トレーサーの濃度は
50MM、Tacペプチドは5009Mである。遊離トレーサーは計算によると
109Mであり、遊離トレーサーが競合分析の仮定として(抗TacのKaに関
して101101Oを用いた場合)1/Ka−100pM以下であるという条件
を満足する。
アッセイはコントロールの冷抗T a、 c抗体と並行して行われ、結合/遊離
トレーサー結合50%阻害点(Ro/2)対log競合物質濃度の曲線シフトが
定量された。
濃度は横座標の真数から得られ、構造体Xに関するアフィニティ定数Kxは下記

72) −17KX −1/Ka上記において[x]112はトレーサー結合が
Ro / 2であるときの競合物質の濃度を示す。抗Tacとの結合データに関
する標準スカッチャード(Scatehard)プロッティングでは、直線のグ
ラフ及び9.7×109M−’のKaを示したが、これは他の研究者により得ら
れたものと同等である[Robb et al、、1984.J、Exp、Me
d、、1B0.1I2B−1146) 、抗Tac (Fv)−PE40に関す
る3、5×109M−1のKaは前記式から計算された。すべての濃度はヒトI
gGとの標準曲線に対するブラッドフォード(Bradrord)タンパク質マ
イクロアッセイで測定された。
競合分析の場合、これらの濃度は、横座標に関する結合性タンパク質の濃度を得
るため過剰のHUT−102細胞と共に、放射性同位元素標識抗Tac (Fv
)−PE40 (0,44)及び放射性同位元素標識抗Tac(0,8)との別
々の試験で得られた結合性分画に基づき標準化された。
抗Tac (Fv)−PE40の選択的細胞毒性抗Tac抗体はHUT−102
細胞に多量に存在するIL−2レセプターのp55サブユニット(Tac抗原)
と結合することから[Llchiyama et al、、lHl、J。
Ismunol−,126,1393−1397) 、キメラタンパク質は前記
のようにHUT−102細胞に対する細胞毒性に関して最初に試験された。抗T
ac CFv)−PE40は20時間アッセイにおいて、0. 15ng/5l
(2,3X 10−”’M)のID5oで用量依存的にタンパク質合成を阻害し
た(第3図及び表2)。4 ng/gg1以上の濃度で、タンパク質合成の完全
阻害があった。数回の特異性コントロールが行われた。過剰の抗Tac(10M
g/ml)の添加はHUT−102細胞において抗Tac (Fv)−PE40
の細胞毒性を妨げたが、一方卵巣癌細胞上でみられる抗原に対するコントロール
モノクローナル抗体0vB3はそうでなかツタ(第3図) 。HUT102より
少数のIL2レセプターを有するもう1つのヒトT細胞系Cr tl、2も、2
、 7 ng/slのID5oで抗Tac(Fv)−PE40に対して非常に
感受的であった(表2)。更に、T細胞系、CEM、A431、KB及び0VC
AR−3を含めたIL2レセプターのない数種のヒト細胞系は、1pg/mlで
あっても抗Tac (Fv)−PE40により影響されなかった(表2)。
PE(抗Tac−PE)又はPE40と化学的に複合化された抗TacはHUT
102細胞を死滅させたことが報告された(Kondo eL al、、19
88.J、Biol、ches、。

Invest、、74.9138−971)。千オニーチル複合体が製造された
場合、抗Tac−PEは約1 、 2 ng/mlのID50を有し、同様に製
造された抗Ta c −PE40は13 ng/mlのIDを有した。抗Tac
(Fv)−PE40 (65kDa)は抗Tac−PEの分子量(216kD
a)の約30%であるため、本発明のキメラ毒素はモルベースでみて抗Tac−
PEよりも数倍活性でかつ抗Tac−PE40よりも著しく活性である。化学的
に製造された抗Tac−PE40又は組換え融合タンパクtIL−2−PE40
[Lorberbous−Galskl et al、、198g、Proc
、Nat、Acad 。
Sci、USA、85.1922−1926)よりも約10倍高い、標的HUT
102細胞に対する抗Tac (Fv)−PE40の非常に高い活性は予想外で
あり、先行技術から予想しえなかった。特に、抗Tac(Fv)抗体はIL−2
に関するK a −10”M−’よりも著しく低い、前記のようなKa−3,5
x109M−’の完全!L−2レセプターに対するアフィニティを有するため、
抗Tac (Fv)−PE40が他のいかなるIL−2−PE融合タンパク質よ
りも細胞毒性であることは予想外である(Tsudo etal、、19J17

)。
競合結合の検討では、抗Tac (Fv)−PE40に関して3゜5X109M
−’のアフィニティを示したが、これは9.7x109M”で測定された抗Ta
cの場合と比較して約1/3であった(第4図)。これは抗ウシ成長ホルモンの
Fv構遺体対Fab断片に関するアフィニティの1 / 4 [Blrd et
al、、198g、5cience、242.428−426)及び抗ジゴキ
シンのFv構遺体対抗ジゴキシンのFab断片に関して1 / 6 (Hous
ton et al、、198g、Proc、Nat。
Acad、Sei、USA、85.5879−5883 )に匹敵しうる。
Fv結合部位によるアフィニティの相対的保存度は抗BGH及び抗ジゴキシン抗
体で観察された場合と比較して抗Tacの方が明らかに優れている。
要約するに、ここで示されたデータは、抗Tac可変領域をコードするcDNA
と修飾形のシュードモナス外毒素をコードするDNAの断片との融合による大腸
菌内における活性組換え免疫毒素の合成を明らかに証明している。この業績は、
当業者に周知の大腸菌又は他の発現ベクターを用いて他の抗体との同様の活性組
換え免疫毒素の調製をここに可能とさせる。
ここで記載された例及び態様は説明目的のみのためであって、それからみて様々
な修正又は変更が当業者に示唆されかつ本出願の精神及び範囲内並びに請求の範
囲内に含まれることが理解される。

細胞系0VCAR3、KB及びA431は毒素添加前日に24ウエルプレートに
IX1057mlで接種した。
HUT−102及びCEMは2回洗浄して、24ウエルプレートに3X105/
mlで接種した(第3図参照)。
様々な希釈率の毒素調製物を加え、20時時間線胞を1.5時間かけて3H−ロ
イシンで標識した。細胞のTCA沈降物中における放射能を測定した。ID5o
とは毒素無添加コントロールと比較してタンパク質合成を50%阻害する毒素濃
度である。すべてのアッセイは重複して行われ、3回繰返された。

Claims (10)
- 1.適切な発現ベクター中で粗換え融合タンパク質の合成を指示するDNAセグ メントを含んでなる構築物であって、 上記融合タンパク質が抗体−PE40であることを特徴とする構築物。
- 2.抗体が抗Tac(Fv)である、請求項1に記載の構築物。
- 3.DNAセグメントがプラスミド中に含まれる、請求項2に記載の構築物。
- 4.ATCCNo.67913のプラスミドを含んでなる、請求項3に記載の構 築物。

- 6.PEの少なくとも転位及びADPリボシル化活性を有する、抗体−PE40 組換え融合タンパク質。
- 7.タンパク質が単一ポリペプチド鎖を有する、請求項6に記載の融合タンパク 質。
- 8.抗体が抗Tac(Fv)である、請求項7に記載の融合タンパク質。
- 9.抗体が結合するレセプター又は抗原を有した細胞を死滅させる上で有効な量 の請求項6に記載の融合タンパク質及び薬学上許容されるキャリアを含んでなる 、組成物。
- 10.標的細胞毒性を発揮するための方法であって、死滅させようとする標的細 胞を細胞毒性量の請求項9に記載の組成物と接触させ、上記標的細胞は抗体が結 合するレセプター又は抗原を有した細胞であるが、但し組成物は上記抗体の結合 のためのレセプター又は抗原を欠く細胞に有意の細胞毒性がないことを特徴とす る方法。
