Claims (14)
- 1.以下の特性を有すること、即ち、 −分子量約80kDaを有する、 −糖タンパク質gp300に対するポリクローナル抗体によって認識される、 −ヒトのSLAまたはAIDSを発症させ得るHIV−2型ヒトレトロウイルス のゲノムによってコードされたトランスメンブランエンベロープ糖タンパク質g p36の2つの完全体を二量体形態に会合させて含む、または、 −前記gp80に対する抗体と共に抗原−抗体型の免疫複合体を形成し得る同程 度の分子量のすべての糖タンパク質を含む、 ことを特徴とする抗原性糖タンパク質gp80。
- 2.以下の特性を有すること、即ち、 −HIV−2型ヒトレトロウイルスに感染した細胞及び/またはHIV−2型の ビリオン及び/またはウイルスに会合し得る、−1%のTriton X−10 0のような非イオン性界面活性剤の存在下ではin vitroで解離せず、弱 酸性pHの1%のSDSのようなイオン性界面活性剤の存在下ではHIV−2型 レトロウイルスのトランスメンプラン糖タンパク質gp36を形成すべくinv itroで解離する、 ことを特徴とする請求項1に記載の糖タンパク質gp80。
- 3.−一方でHIV−2型レトロウイルスに感染した細胞中に存在するgp30 0、gp140、gp125及び請求項1または2に記載のgp80を特異的に 認識し、他方でHIV−2型レトロウイルスの抽出物中に存在するgp125、 請求項1または2に記載のgp80及びgp36を特異的に認識し、−健常細胞 またはHIV−1型レトロウイルスに感染した細胞中に存在するタンパク質また は糖タンパク質を認識しないことを特徴とするポリクローナル抗体。
- 4.請求項1または2に記載の糖タンパク質gp80と特異的に反応し、HIV −1型レトロウイルスに感染した細胞のタンパク質及び糖タンパク質またはHI V−1のウイルス集塊とは反応しないことを特徴とするモノクローナル抗体。
- 5.更に、HIV−2の糖タンパク質的gp300gp、gp140及びgp3 6を認識することを特徴とする請求項4に記載のモノクローナル抗体。
- 6.請求項4または5に記載のモノクローナル抗体を産生するハイブリドーマ。
- 7.HIV−2型ヒトレトロウイルス感染を、該感染後に形成された抗体を含む と予想される生物サンプルからin vitroで特異的に診断するための、請 求項1または2に記載の糖タンパク質gp80を少なくとも1つ含むことを特徴 とする抗原性組成物。
- 8.請求項1または2に記載の糖タンパク質gp80の少なくとも1つを、ワク チン成分として許容される医薬担体と共に含有することを特徴とする免疫原性組 成物。
- 9.−HIV−2型レトロウイルス感染後に産生する抗体を含むと予想される生 物サンプル請求項1まは2に記載の糖タンパク質gp80または請求項7に記載 の抗原性組成物とを抗原−抗体型の免疫複合体が形成され得る適当な条件下に接 触させ、 −場合によっては形成される抗原−抗体複合体を検出する段階を含むことを特徴 とするHIV−2型レトロウイルスに特異的に起因する感染のin vitro 診断方法。
- 10.−ヒトレトロウイルスHIV−2に感染した細胞を溶解し上清と感染細胞 とを分離するか、または遠心によって調製されたウイルス集塊を溶解させ、 −細胞抽出物及び/またはウイルス抽出物を、例えばヒトレトロウイルスHIV −2感染患者の血清即ちHIV−2のエンベロープタンパク質と強力に反応する 能力を有する血清から得られた精製抗体を好ましくは適当な担体に固定させて含 む免疫吸着剤を収容したイムノアファニティカラムで、バッファの存在下に抗原 −抗体免疫複合体が形成される十分な時間インキュベートし、 −カラムをバッファで洗浄して担体に保持されなかった分子を除去し、 −所望の抗原性タンパク質を回収する段階を含むことを特徴とする請求項1また は2に記載の抗原性糖タンパク質gp80の製造方法。
- 11.糖タンパク質の回収を電気泳動及び電気溶出によって行なうことを特徴と する請求項1または2に記載の糖タンパク質を製造するための請求項10に記載 の方法。
- 12.糖タンパク質を回収するために、−前記イムノアフィニティカラムに固定 されたタンパク質を溶出させ、 −前記の溶出産物を、分離担体に固定されたgp80認識モノクローナル抗体を 含むカラムクロマトグラフィーで精製することを特徴とする請求項1または2に 記載の糖タンパク質を製造するための請求項10に記載の方法。
- 13.−請求項1または2に記載のタンパク質及び/または抗原性糖タンパク質 少なくとも1つ、または、−請求項7に記載の組成物、または −これらの種々の成分の混合物と、 −被検生物サンプル中に場合によっては存在する抗体と前記抗原との間の免疫複 合体の形成反応を生じさせる手段、特に必要に応じて1種または複数のインキュ ベーションバッファと、 −陰性対照サンプルと、 −形成された抗原−抗体複合体の顕示手段とを含むことを特徴とするヒトレトロ ウイルスHIV−2に感染した疑いのある患者の血清またはその他の生物サンプ ル中の抗体検出用キット。
- 14.−細胞内タンパク質が露出するように生物サンプルを処理し、 −例えば電気泳動、または、gp80と免疫的に反応性の抗体を用いた免疫検定 によって、露出した細胞内タンパク質中のタンパクgp80存在を検出する段階 を含むことを特徴とする生物サンプル中のレトロウイルスHIV−2感染の検出 方法。
