Description
結果的に、クラフト温度未満の温度において、界面活性剤の溶解性は非常に低く、かつ界面活性剤は規則的な有機分子として機能する。溶解性が劇的に増加するクラフト温度において、ミセルが形成し、そして界面活性剤の表面活性特性が有用な様式で明示される。他方では、クラフト温度未満の温度において、界面活性剤の溶解性が低く、界面活性剤は多くの用途において実質的に有用ではない。以下に詳述されるように、界面活性剤を含有する製品は日常的な条件下で使用されるように一般に意図されるため、したがって、ほとんどの用途では、室温未満のクラフト点を有する界面活性剤が必要とされる。
これらの界面活性剤は、皮膜および粘膜に適合性があり、急性的および繰り返された用量毒性研究において非毒性であることが発見された[20]。グリコシドは、酸素原子を通して、アルデヒドまたはケトン炭素に置換基が結合された置換糖類である。したがって、グリコシドはアセタールであると考えられる。「糖類」という用語と同様に、「グリコシド」という用語は、分子中の数も糖類単位の同一性も定義しない。アルキルグリコシドに適用される一般的な短縮命名法はCnGmであり、式中、nは、アルキル鎖中の炭素原子の数として定義され、そしてmは、頭部基を構成する糖類単位(通常グルコース単位)の数を定義する。
(A)CnGm1およびCnGm2の2成分系混合物(式中、m1およびm2は、7および13または8および14のいずれかである)。以下、この種類の2成分系界面活性剤組成物を、短縮形表記CnGm1/m2と記載する。したがって、CnG7/13は、CnG7およびCnG13の2成分系混合物を指すが、CnG8/14は、CnG8およびCnG14の2成分系混合物を指す。例えば、この種類の2成分系混合物は、出発材料として商業的に入手可能なC16G2を使用して製造することができ、これによって、C16G8/14が得られる。
(B)分子CnG4、CnG5・・・、CnG20の混合物。以下、この種類の多分散系界面活性剤組成物を短縮形表記CnG4-20と記載する。したがって、C16G4-20は、4~20の範囲のnを有するC16Gn分子の混合物を参照する。例えば、この種類の多分散系混合物は、C16G1およびC18G1の商業的に入手可能な混合物(Evonikからの商標名TEGO Care CG16)から製造することができ、これによって、C16-18G4-20が得られる。
CnGm (I)

もちろん、他の種類の2成分系組合せが、本発明の範囲内で製造されてもよい。
CnGm (I)

mは、4~20の数である)を有する少なくとも1つのアルキルグリコシドを含んでなる界面活性剤組成物が開示される。
(1)7~14から選択されるmを有し、好ましくはCnG7またはCnG8を含んでなるアルキルグリコシドの少なくとも1つまたは混合物を含有するもの。混合物は、例えば、(CnG7)および(CnG13)、または(CnG8)および(CnG14)を含有してもよい。
(2)好ましくは存在するアルキルグリコシドの総量の少なくとも67%を表す、m=4-20を有する成分を有する、成分CnGmの多分散系混合物を含有するもの。
より長い頭部は、アルキルグリコシドの毒性も減少させて、粘膜および他の感応性組織において刺激物として作用するその傾向を低下させる。
例えば、ペプチドまたはタンパク質自己会合または自己凝集は低下する。また、被術者に投与される時、他のペプチドまたはタンパク質による会合または凝集も低下する。
広範な研究コースにおいて、C16G8/14混合物を含んでなる溶液は、何年間もの冷蔵条件(2℃)で貯蔵される場合でも、いずれの沈殿物も生成しないことが発見された。これは、C16G8およびC16G14のクラフト点が両方とも2℃未満であることを意味する。この値は、表1中で他のアルキルグリコシドと比較される[11,12,13]。同様に、C16-18G4-20の多分散系混合物は、室温で可溶性であることが発見され、そして長期間、冷蔵条件で貯蔵される場合に、沈殿物を生成しないことが発見された(表1)。
C16G8/14混合物を含んでなる界面活性剤組成物のCMCは、テンシオメトリーによって決定された(図1)。測定によると、CMCは37mg/Lであり、これは、この特定の混合物に関して、24μMに等しい。この値は、それぞれ、2.1、2.3および3.9μmのCMC値を示す、PEGをベースとする界面活性剤C16E9、C16E12およびC16E21の相当する値と比較されてもよい[14]。この値は、13-15mg/LにおけるPolysorbate 80のCMCと比較されてもよい(10~11μM)[15]。これらの結果によって、同様のPEGをベースとする界面活性剤と比較して、C16G8/14混合物を含んでなる界面活性剤組成物はより親水性であることが確認された。驚くべきことに、CMC(49mN/m)より高い濃度でC16G8/14混合物を含んでなる溶液の表面張力は、PEGをベースとする界面活性剤のもの(30~35mN/m)、および短い頭部による従来のアルキルグリコシドのもの(32~37mN/m)より有意に高いことが発見された[16]。高い表面張力は、C16G8/14混合物の生体適合性の鍵となり得る。
疎水性モデル表面へのC16G8/14混合物の吸着のエリプソメーター研究によって、3mg/m2(3μモル/m2)に相当する非常に効率的な表面被覆率が明らかになる(図2)。比較のために、疎水性シリカ基板へのPolysorbate 80の吸着は、0.028mg/mlにおいて1.4mg/m2(1.1μモル/m2)である[17]。したがって、吸着された質量に関して、C16G8/14は、そのより低い表面活性(テンシオメトリーによって決定される;上記参照)にもかかわらず、Polysorbate 80の約2倍効率的である。
臨界ミセル濃度(CMC)より高い、およびそれより低い濃度で、C16G8/14混合物を含んでなる界面活性剤組成物の細胞毒性を評価し、そして他の界面活性剤の数値と比較した。細胞毒性のパラメーターとして、細胞代謝はXTT変換によって評価した。XTTアッセイは細胞のミトコンドリア活性に基づき、そして細胞がどのように活性であり、それによってどのように存続可能であるかを反映する[24,25]。XTT化合物(ナトリウム3’[1-フェニルアミノカルボニル]-3,4-テトラゾリウムビス[4-メトキシ-6-ニトロ]ベンゼンスルホン酸水和物)は、ミトコンドリアによって還元されて、オレンジ色のホルマザン色素を形成する。黄色からオレンジ色への色変化は、450nmにおいて分光光度計で測定される。
結果を、対照培養物(未処置の細胞)の%として観察された吸光度として示す。図15aで分かるように、細胞生存能力は、polysorbate 80およびC16G2の両方におけるよりも、C16G8/14(図中C16G8として示される)の組成物において高い。細胞生存能力が50%減少する濃度(IC50)は、polysorbate 80に関する0.13mMおよびC16G2に関する0.017mMと比較して、C16G8/14組成物に関して0.43mMであった。
酸化に対して感応性の水性調製物におけるC16G8/14混合物の化学的安定性を、Polysorbate 80(高度精製および医薬品グレード)の2つの異なるグレードのものと比較した。酸化傾向があるモデル化合物として、ブチルヒドロキシトルエン(BHT)が使用された。BHTは容易に酸化し、明るい黄色の3,3’,5,5’-テトラ-ビス-(tert-ブチル)-スチルベンキノンとなる。分散剤として0.2mg/mlの界面活性剤を使用して、0.5mg/mlの微粒状化BHTを150mM NaCl溶液中で分散した。調合物を室温および40℃で安定して配置した。調製物をHPLCによって分析し、規則的な時間点において視覚的に検査した。研究の間、瓶の視覚による検査によって、段階的な黄変色は、ポリソルベートを含有する溶液に関してより顕著であることが明らかとなり、このことは、明らかに、ポリソルベートの化学安定性がC16G8/14混合物のものより低いことを示した。図4に示されるHPLCの結果は、この結論の定量的形態を提供する。明白であるように、C16G8/14混合物は、調製物の化学的安定性に関して、ポリソルベートの2つのグレードより非常に優れている。実際には、28週間の研究において、加速条件(40℃)下でさえ、C16G8/14混合物系のBHT含有量は減少しなかった。他方、ポリソルベート系において、BHT含有量が経時的に劇的に減少することが発見された。
ペプチドおよびタンパク質凝集の阻害は、物理的安定性および安全性のため、特に注射(インジェクタビリア)のための医薬品に関して重要である。したがって、界面活性剤は通常、そのような調製物におけるペプチドおよびタンパク質凝集の抑制剤として適用される。溶液中でのペプチドおよびタンパク質凝集を抑制するC16G8/14混合物の能力は、モデルペプチドとしてインシュリンを使用して調査された。研究では、C16G8/14混合物を、Polysorbate 20(インジェクタビリアのために現在承認される標準界面活性剤)およびC14G2(テトラデシルマルトシド;Aegis Therapeuticsによってこの用途に関して最近示唆されたAnatrace(Affymetrix)から購入した新規賦形剤)と比較した([21、22]を参照のこと)。この研究では、インスリン濃度は0.4mg/mlであり、界面活性剤濃度は1.4mg/mlであった。溶液のpHは、pH 6.8(加速条件)またはpH 7.4のいずれかにおいて、クエン酸塩によって緩衝された。溶液を25および40℃で3回安定に配置し、2、4、8および12週間後に視覚による検査によって分析した。この研究結果を表2に要約する。明らかであるように、C14G2は、非加速条件下でさえ沈殿を抑制することができないことが証明された。Polysorbate 20は、かなり良好な性能を示すが、pH 6.8またはpH 7.4のいずれかにおいて40℃で沈殿を停止しなかった。本研究において、3つの界面活性剤のうち、C16G8/14混合物が優れていることが証明された。この界面活性剤組成物に関しては、沈殿は最高加速条件(40℃、pH6.8)のみで観察された。
微粒状化された疎水性粒子のために効率的な分散剤として作用する選択された界面活性剤組成物の傾向を、モデル化合物として2種のステロイド薬、ブデソニドおよびベクロメタゾンジプロピオネート(BDP)を使用して、医薬品製剤において試験した。本研究において、以下の新規アルキルグリコシド組成物:C12G8/14、C16G8/14およびC16-18G4-20が含まれた。これらの組成物を、従来のアルキルグリコシド組成物TEGO Care CG90(主にC16-18G1からなる)、C16-18G1-3およびC16G2と比較した。
本発明による界面活性剤組成物によって調製された懸濁液の熱安定性は、微粒状化されたブデソニド(0.5mg/ml)の懸濁液を水浴上で30分間90℃まで、およびオートクレーブ中で8分間125℃まで加熱することによって調査した。懸濁液は、分散剤として0.2mg/mlのC16G8/14混合物を使用して、上記のとおり高剪断混合によって調製された。粒径分布は、レーザー回折測定によって調査した。また比較のために、分散剤としてPolysorbate 80を使用して、微粒状化されたブデソニドの懸濁液を調製した。
エマルジョン特性を比較するため、1.2%の乳化剤(C16G8/14混合物;Polysorbate 80;またはC12G8/14混合物)および20または50%のカノーラ油を含有するモデルエマルジョンを製造した。24mg/mlの濃度の乳化剤貯蔵液が調製された。次いで、1mlの貯蔵液を0.6mlの水と混合し、0.4mlのカノーラ油に添加して、20%の油エマルジョンを製造した。さらに、1mlの貯蔵液を1mlのカノーラ油と混合し、50%の油エマルジョンを製造した。均質化は、SONICS,Vibracellの超音波プローブを使用して、40%振幅で15秒間、続いて、15秒間の休止、合計1分の活性超音波処理で実行した。この手続きを、温度変動を最小化するために、繰り返しの間により長い休みを用いて、もう2回繰り返した。この手順によって、乳化剤Polysorbate 80およびC16G8/14混合物を含んでなる界面活性剤組成物の、なめらかな白色のエマルジョンが得られた。C16G8/14混合物の2つのバッチを試験したところ、最初の2ヵ月の間は全てのエマルジョンが安定であった。しかしながら、約3ヵ月後、Polysorbate 80を含有するエマルジョンおよびC16G8/14バッチの一方の相分離が生じた。したがって、C16G8/14混合物がPolysorbate 80より安定性のあるエマルジョンを製造することを結論づけることができる。C12G8/14混合物に関して、上記の体積の半分のみが使用され、なめらかな白色の安定性のある(少なくとも3週間)エマルジョンを製造するために、追加的な15秒間の70%振幅の超音波処理が必要とされた。明らかに、C12G8/14組成物は、C16G8/14組成物およびポリソルベート80の両方よりも有効ではない乳化剤である。

- 2つのアルキルグリコシドを含有する界面活性剤組成物の、ペプチドまたはタンパク質の会合または凝集を低下、防止または減少させるための使用であって、前記2つのアルキルグリコシドは、

をそれぞれ有し、
C16G8とC16G14の質量比が50:50~95:5であり、
