Description
RNAi(RNA干渉)は1998年にアンドリュー・ファイア(Andrew Z.Fire)らがカエノラブディティス・エレガンスでアンチセンスRNA抑制の実験を行った時に発見され、この過程をRNAiと呼んだ。この発見は『Science』誌により2001年の10大科学業績の一つに選定され、そして2002年の10大科学業績のトップとなった。それ以降、RNAiを作用機序とするsiRNAは潜在的な遺伝子治療薬として広く注目されてきて、2006年、アンドリュー・ファイアとクレイグ・メロー(Craig C.Mello)はRNAi機序研究における貢献によりノーベル生理・医学賞を受賞した。RNAiは動物、植物や真菌を含め多くの生物において二本鎖RNA(dsRNA)によってトリガーすることができ、RNAi過程において、「Dicer」と呼ばれるエンドヌクレアーゼは長鎖dsRNAを21~25ヌクレオチド長の小断片に切断又は「細断」する。これらの小断片は、低分子干渉RNA(siRNA)と呼ばれ、そのアンチセンス鎖(Guide strand)がArgonauteタンパク質(AGO2)にローディングされている。AGO2ローディングはRISC-loading複合体で発生し、このRISC-loading複合体は3元複合体であり、Argonauteタンパク質、Dicer、及びdsRNA結合タンパク質(TRBPと略称する)からなっている。ローディング中にAGO2によってセンス鎖(Passenger strand)が切断されて排出される。その後、AGO2はアンチセンス鎖を用いて、完全に相補的な配列を含むmRNAと結合し、その後、これらのmRNAの切断を触媒したところ、mRNAが分裂して翻訳テンプレートの作用を失い、さらに関連タンパク質の合成が阻止される。切断後、切断されたmRNAは放出され、アンチセンス鎖をローディングしたRISC-loading複合体は、別のラウンドの切断にリサイクルされる。
siRNAは血液や組織で不安定であり、ヌクレアーゼによって分解されやすく、siRNAの安定性を高めるためには、siRNA骨格を修飾してもよいが、これらの化学修飾はヌクレアーゼ分解から限定的な保護作用を提供するだけであり、最終的にsiRNAの活性に影響を与える恐れがある。そのため、siRNAが安全かつ効率的に細胞膜を通過することを確保するために、対応する伝達系が必要とされる。siRNAは分子質量が大きく、大量の負電荷を持ち、さらに水溶解性が高いため、そのもの自体が順調に細胞膜を通過して細胞内に到達することができない。
肝臓中のアシアロ糖タンパク質受容体(ASGPR)は肝細胞により特異的に発現される受容体であり、高効率なエンドサイトーシス型受容体である。生体内の生理状況下で各種糖タンパク質は酵素や酸でシアル酸を加水分解された後、露出したペナルティメートがガラクトース残基であるため、ASGPRが特異的に結合する糖はガラクトシル基であり、それでガラクトース特異的受容体とも呼ばれる。ガラクトース、ガラクトサミン、N-アセチルガラクトサミンなどの単糖や多糖分子はASGPRに対して高い親和性を有する。ASGPRの主な生理学的機能は血液中のアシアロ糖タンパク質、リポタンパク質などの物質の除去を媒介することであり、ウイルス性肝炎、肝硬変、肝臓癌などの肝疾患の発症や進行と密接な関係がある。このようなASGPRの特性の発見によって、肝疾患の診断や治療に重要な役割が果たされている(Ashwell G,Harford J,Carbohydrate specific Receptors of the Liver,Ann Rev Biochem 1982 51:531-554)。ガラクトース又はガラクトサミン及びそれらの誘導体を構造中に含む肝疾患治療薬は、ASGPRに特異的に親和性を有し、それにより肝を能動的に標的とする特性を有し、その送達に他のベクター系を必要としない。

mG、mA、mC及びmUは2’-メトキシ(2’-OMe)で修飾されたヌクレオチドであり、fG、fA、fC及びfUは2’-フッ素で修飾されたヌクレオチドであり、sはホスホロチオ酸ヌクレオシド間結合であり、残りのヌクレオチドモノマーはホスホジエステル結合を介して連結されている。詳細は以下のとおりである。

前記立体障害安定構造を含有する分岐鎖LはC3~C18の直鎖であり、該直鎖には1つ又は複数のカルボニル、アミド、ホスホリル、酸素原子又はこれらの基の組み合わせが含まれ、以下のような構造から選ばれる1種又は複数種としてもよい。
(1)リポソーム媒介siRNA送達と比較して:リポソーム媒介siRNA送達は、主にリポソームがsiRNAをリポソーム内に封じ込み、siRNAをヌクレアーゼによる分解から保護し、siRNAの細胞膜障害への通過効率を高め、細胞の吸収を促進する。、例えばアニオン性リポソーム、pH感受性リポソーム、免疫リポソーム、膜融合リポソーム(fusogenic liposome)及びカチオン性リポソーム等のリポソームは、ある程度の進展を遂げているにも係わらず、リポソーム自体は、炎症反応を起こしやすく、投与前に、発生可能な急性炎症反応を減少するようにセチリジンやデキサメタゾンなどの抗ヒスタミンやホルモン類を多く使用しなければならない。このため、実際の臨床応用において治療領域のすべてに適するわけではなく、特に慢性B型肝炎のような治療周期の長い疾患では、長期の使用による蓄積毒性は潜在的な安全上の潜在的な危険である恐れがある。
(2)革新的なN-アセチルガラクトサミン導入方法:
3つのN-ガラクトサミン構造を有するsiRNAのHBsAg阻害効果と較べて、現在I/II期にある慢性B型肝炎治療用siRNA薬としては、ARO-HBVとALN-HBV02がある。ARO-HBVはsiARNAのセンス鎖の5’末端に連結鎖を介して3個のN-アセチルガラクトサミンを導入し、ALN-HBV02はsiRNAセンス鎖の3’末端に連結鎖を介して3個のN-アセチルガラクトサミンを導入したが、これらの薬剤では、ガラクトサミンが導入された部位がいずれもセンス鎖にあり、そして3個のN-アセチルガラクトサミンが導入された。本発明から提供される化合物では、siRNAのセンス鎖の5’末端とアンチセンス鎖の3’末端に異なる数又は同じ数のN-アセチルガラクトサミンが同時に導入された。現在、センス鎖の5’末端とアンチセンス鎖の3’末端に同時に導入されたという報道が見られず、特にアンチセンス鎖の3’に3つのN-アセチルガラクトサミンを導入するのは革新的な導入態様である。実施例によれば、この導入態様によってsiRNAがHBV遺伝子を効率良く抑制するという効果を奏することが確認された。

mGを出発モノマー、C6NHホスホルアミダイトモノマーを末端モノマーとして、固相ホスホルアミダイト法によるカップリングによって異なるホスホルアミダイトモノマーを導入した。固相ホスホルアミダイト法の基本的なステップは以下を含む。1)脱保護:固相担体における酸素原子上の保護基(DMT)を除去する。2)カップリング:1番目のヌクレオチドモノマーを追加し、3’から5’への方向でカップリング反応を起こす。3)酸化:得られたヌクレオシドホスファイトを酸化して、より安定的なヌクレオシドホスフェート(即ち3価リンが酸化されて5価リンとなる)とする。4)ブロック:前のステップの未反応ヌクレオチドモノマー5’-OHにキャップをつけてブロックし、さらに反応に関与しないようにする。所望の配列が得られるまで上記ステップを繰り返す。合成後、メチルアミンエタノール溶液及びアンモニア水を用いて固相担体において化合物と最初のヌクレオシドとを連結するエステル結合を開裂させ、オリゴヌクレオチド上の各塩基とリン酸上の保護基であるシアノエチル(P)、ベンゾイル(mA、fA)、アセチル(mC、fC)、イソブチリル(mG、fG)及び4-メトキシトリチル(C6NH)とを除去した。HPLCにより分離して精製した後、生成物をろ過して殺菌し、凍結乾燥させた。

3、GBL-0401合成
Kys-01及びKyas-01溶液の濃度を精確に測定し、等モル濃度で混合した後、該体積に対して1/20体積の1 M PBS溶液を加えて再び均一に混合し、混合系を95℃に加熱して5min継続させた後、3hかけて40℃又は室温に自然降温すると、HPLC検出を行った。残留一本鎖<5%であれば、反応が完了したとしてもよい。

3、GBL-0410の合成
Kys-10及びKyas-10溶液の濃度を正確に測定し、合成ステップは実施例1の3、GBL-0401の合成と同様であった。
1.実験の群分け
空白対照群:2%FBSを含むDMEM培地を加えて72hインキュベートした。
試験品群:それぞれ濃度5nM、0.5nM又は0.05nMの試験品希釈液を加え、濃度ごとに重複ウエルを3つとし、37℃、5%CO2インキュベータで72h培養した。
2、実験材料
HepG2.2.15細胞
3、実験試薬
HepG2.2.15細胞は96ウェル細胞培養プレートで培養し、3日ごとに新鮮な培地に交換し、6日目に上述で調製した各濃度の薬物含有培養液を加え、9日目まで培養を続けた。上清液を収集し、測定キットで細胞上清中のHBsAg、HBeAg及びHBV DNAの含有量を測定した。また、対照非投与群とOD値を比較し、比の値に基づいて有効性を確定した。
7.実験結果
7.1 HepG2.2.15細胞中のHBsAgに対する抑制効果:図6参照

1.実験案
適齢のHBVトランスジェニック雄マウス(HBsAg発現が顕著であることを要求する)を実験評価に用い、体重が25g前後のマウスを90匹選び、18群に無作為に分け、各群を5匹とし、Day0でそれぞれ皮下注射により3mg/kg投与し、投与体積は100~200μLであった。投薬前にマウス血HBsAgを測定し、各群のHBsAgの平均レベルをできるだけ一致させた。
2.試験品及び試薬
抑制効果を図9に示す。
1.実験案
適齢のHBVトランスジェニック雄マウス(HBsAg発現が顕著であることを要求する)を実験評価に用い、体重が25g前後のマウスを10匹選び、2群に無作為に分け、各群を5匹とし、それぞれ対照群と投与群とし、Day0でそれぞれ皮下注射により3mg/kg投与し、投与体積は100~200μLであった。投薬前に採血してHBsAgを測定し、各群のHBsAgレベルをできるだけ一致させた。マウス眼窩静脈叢から全血を、投与前(day 0)、投与後-1週目、2週目、3週目、4週目、5週目、6週目の時点で採取し、HBsAgを検出し、HBV遺伝子発現に対するGBL-0401の抑制の持続性を考察した。
具体的な投与情報は下表のように示されている。
結果から、GBL-0401は1週目に最適な抑制効果である99.08%を達成し、2週目から3週目までは低下傾向にあったが、90%前後の高い抑制率を示し、4週目から6週目までは低下傾向にあったが、75%前後の抑制効果を維持した。GBL-0401はHBV HBsAg発現に対して持続的な抑制効果があり、6週間程度発現を安定的に抑制することができた。HBV HBsAgに対するGBL-0401のインビボ抑制効果を図10に示す。
Claims (9)
- HBV遺伝子発現を抑制する干渉核酸、トランジションポイント及びその末端修飾鎖を構造中に含む化合物であって、
前記末端修飾鎖は、連結鎖Dと、リンカーBと、分岐鎖Lと、肝臓を標的とする特異的配位子Xとから構成され、前記末端修飾鎖と前記干渉核酸はトランジションポイントR1/R2を介して連結され、その構造は一般式(I)に示され、
式中、
nが1である場合、mは3であり、又は、nが2である場合、mは2であり、
R1は-NH(CH2)xCH2-であり、ここでxは3~10の整数であり、
R2は-NHCH2CH(OH)CH2(OH)-、又は一級アルコールと二級アルコールを持つピロール環又はピペリジン環であり、
前記肝臓を標的とする特異的配位子XはN-アセチルガラクトサミンであり、
前記分岐鎖LはC3~C18の直鎖であり、該直鎖にはカルボニル、アミド、ホスホリル、酸素原子又はこれらの基の組み合わせが含まれ、
前記リンカーBは以下の構造式から選ばれ、
前記連結鎖DはC5~C20を含有し、アミノ、カルボニル、アミド、酸素原子、硫黄原子、ホスホリル、又はこれらの基の組み合わせを含有し、
前記干渉核酸は抗B型肝炎ウイルスsiRNAである、化合物。 - 前記siRNAの配列は以下から選ばれる、請求項1に記載の化合物。
1) センス鎖SEQ ID NO.1とアンチセンス鎖SEQ ID NO.2との組み合わせ配列
2) センス鎖SEQ ID NO.5とアンチセンス鎖SEQ ID NO.6との組み合わせ配列
3) センス鎖SEQ ID NO.7とアンチセンス鎖SEQ ID NO.8との組み合わせ配列
4) センス鎖SEQ ID NO.9とアンチセンス鎖SEQ ID NO.10との組み合わせ配列
5) センス鎖SEQ ID NO.13とアンチセンス鎖SEQ ID NO.14との組み合わせ配列
6) センス鎖SEQ ID NO.17とアンチセンス鎖SEQ ID NO.18との組み合わせ配列
7) センス鎖SEQ ID NO.27とアンチセンス鎖SEQ ID NO.28との組み合わせ配列
8) センス鎖SEQ ID NO.31とアンチセンス鎖SEQ ID NO.32との組み合わせ配列。
- 下記のGBL-0401~GBL-0418から選ばれる、請求項1~3のいずれか1項に記載の化合物。
前記末端修飾鎖の組み合わせのコードは以下の通りである。
- 肝関連疾患は、急性・慢性肝炎、肝臓癌、遺伝性肝疾患、肝硬変、脂肪肝、又は糖尿病である、肝関連疾患を治療する医薬品の製造における請求項1~4のいずれか1項に記載の化合物の適用。
- 前記HBV遺伝子発現の抑制によって治療されるHBV感染は、慢性B型肝炎ウイルス感染、急性B型肝炎ウイルス感染を含む、HBV感染関連疾患を治療する医薬品の製造における請求項1~4のいずれか1項に記載の化合物の適用。
- 前記肝臓を標的とする特異的配位子Xは、肝臓中のアシアロ糖タンパク質受容体(ASGPR)に対して特異的なものであり、前記HBV感染関連疾患は慢性B型肝炎であり、前記化合物は、HBVのHBsAg、HBeAg及びHBV DNAの発現を持続的に抑制することが可能である、ことを特徴とする請求項6に記載の適用。
- 請求項1~4のいずれか1項に記載の化合物と薬学的に許容される補助材料とを含む、医薬組成物。
- 前記医薬組成物の剤形は、皮下注射剤である、請求項8に記載の医薬組成物。
