Description
溶解性マイクロニードルは、溶液流延(solution casting)法で製造され、溶液をモールドにキャスティングして真空あるいは遠心分離で満たした後、乾燥して製造した。
架橋されたヒアルロン酸の乾燥後の再膨潤性の評価
本発明者らは、架橋されたヒアルロン酸(Hylasome EG10、Vantage)の再膨潤性を評価するため、Hyalsome EG10、Hyalsome EG10+トレハロース10重量%を混合した溶液を60℃のオーブンで乾燥した後、精製水を混合して再膨潤性を評価した。
架橋されたヒアルロン酸が含有された組成物による肌ボリューム増強効果の評価
本発明者らは、架橋されたヒアルロン酸(Hyalsome EG10、Vantage)が含有された組成物の肌ボリューム増強効果を評価するため、架橋されたヒアルロン酸が含有された組成物を60℃のオーブンで乾燥した後、豚肌を切開して乾燥物を入れて縫合した。
マイクロニードルパッチ内の架橋されたヒアルロン酸の含有形態の評価
架橋されたヒアルロン
酸(Hyalsome EG10、Vantage)がマイクロニードルパッチ内で如何なる形態で存在するかを評価した。まず、架橋されたヒアルロン酸がマイクロニードルに均一に含浸されるように、架橋されたヒアルロン酸をUltra Turrax(T-18、IKA)を用いて5分間細かく粉砕した。その後、架橋されたヒアルロン酸は透明であって確認し難いため、目視で確認するためにアルシアンブルー8GX(sigma-aldrich)0.1%を混ぜて架橋されたヒアルロン酸を染色した後、マイクロニードルパッチを製作し、顕微鏡(Leica DM 1000)を用いてマイクロニードルパッチに存在する架橋されたヒアルロン酸を確認した。
Claims (8)
- (i)マイクロニードルを形成する物質と、
(ii)架橋されたヒアルロン酸からなるフィラー粒子と、
(iii)糖と、
を含むフィラー粒子投与用溶解性マイクロニードルであって、
前記糖と架橋されたヒアルロン酸とは、1:0.00001~0.5(糖:架橋されたヒアルロン酸)の重量比で含まれ、
前記糖が、キシロース、スクロース、マルトース、ラクトース、グルコース、グルコサミン、グルクロン酸、ラムノース、マンノース、フコース、アラビノース、アセチルグルコサミン、イノシトール、トレハロース、イソマルト、キシリトール、エリトリトールまたはこれらのうち2つ以上の混合物であり、
前記フィラー粒子が皮内または皮下に投与された後、架橋されたヒアルロン酸が膨潤する、マイクロニードル。 - 前記マイクロニードルを形成する物質が肌内で溶解される、請求項1に記載のマイクロニードル。
- 前記糖が、マイクロニードルの総重量に対して1~70重量%で含まれる、請求項1に記載のマイクロニードル。
- 前記マイクロニードルを形成する物質が、ヒアルロン酸、カルボキシメチルセルロースナトリウム (Na-CMC)、ビニルピロリドン-ビニルアセテート共重合体、ポリビニルアルコール、ポリビニルピロリドンまたはこれらの2つ以上の混合物を含む、請求項2に記載のマイクロニードル。
- 前記マイクロニードルが、マイクロニードルを形成する物質の外に、可塑剤をさらに含む、請求項4に記載のマイクロニードル。
- 前記糖が、前記フィラー粒子内の架橋されたヒアルロン酸の膨潤性を向上させる、請求項1に記載のマイクロニードル。
- 前記マイクロニードルがしわ改善用である、請求項1から6のいずれかに記載のマイクロニードル。
- 請求項1から6のいずれかに記載のマイクロニードルを含むマイクロニードルパッチ。
