Description
Claims (14)
- 細胞体を操作及び/又は研究する方法であって、
流体媒体を保持するための保持空間を含む試料ホルダーを用意するステップ、
上記保持空間内の流体媒体中に1以上の細胞体を含む試料を用意するステップ
を含んでおり、
当該方法が、上記保持空間に、上記試料の少なくとも一部と接着させられる官能化された壁面部分を用意することを含んでおり、
上記官能化された壁面部分は試料の少なくとも一部が接着する傾向があるところの細胞体を含み、
当該方法が、上記保持空間内に共鳴バルク音波を発生させることを含んでおり、
前記共鳴バルク音波を加えるステップの少なくとも一部の間に、上記試料の少なくとも一部が上記官能化された壁面部分と接着し、
当該方法が、上記保持空間内で上記共鳴バルク音波を発生させて、該保持空間内の上記試料の1以上の細胞体に対して、上記官能化された壁面部分から離れる方向に、力を加え、及び、前記力によって官能化された壁面部分から離れる方向に保持空間内の上記試料の1以上の細胞体を促し、および
ここで、該方法は、官能化された壁面部分に対する1以上の細胞体の、上記力と接着強度との間の関係を決定することをさらに含む、上記方法。 - 上記保持空間への試料流体の導入及び/又は上記保持空間からの試料流体の除去の少なくとも1つ、例えば音波を加えるステップの少なくとも一部の間に上記保持空間を通して試料流体を流すこと、を含んでおり、特に導入された試料流体が1以上の細胞体を含んでいてもよい、請求項1に記載の方法。
- 上記保持空間内の音波の周波数及び振幅の少なくとも一方を、好ましくは時間依存的に、変化させることを含む、請求項1又は請求項2に記載の方法。
- 上記1以上の細胞体の1以上の特性を検出及び/又はモニタリングするステップをさらに含んでおり、
該1以上の特性が、細胞の完全性、上記官能化された壁面部分の少なくとも一部への細胞体の接着、1以上の細胞体の運動、蛍光発光、1以上の細胞体の生存能の兆候の少なくとも1つであるか或いはそれらを含み、
検出及び/又はモニタリングステップが、
写真撮影、動画撮影、顕微鏡検査の1以上による光学的検出、例えば光強度検出及び/又は光学イメージング、並びに
音響検出、例えば表面音響検出
の少なくとも1つを含む、請求項1乃至請求項3のいずれか1項に記載の方法。 - 細胞選別;
音響力、試料流体の流れ及び試料流体の組成の1以上の関数として、1以上の細胞体の運動の追跡;
1以上の細胞体の光学活性のモニタリング;
試料ホルダーの温度及び/又は温度プロファイルの変更;
試料ホルダーの照明及び/又は照明プロファイルの変更;
試料流体の組成の変更
の少なくとも1つを含む、請求項1乃至請求項4のいずれか1項に記載の方法。 - 1以上の細胞体と官能化された表面部分との間の接着強度を定量化するステップを含む、請求項1乃至請求項5のいずれか1項に記載の方法。
- 細胞体を研究するための操作システムであって、当該システムが、
流体媒体中に1以上の細胞体を含む試料を保持するための保持空間を含む試料ホルダー、
上記保持空間内で共鳴バルク音波を発生させて試料に力を加えるために試料ホルダーに接続された又は接続可能な音波発生器、
及び、コントローラー
を備えており、
上記試料ホルダーが、使用時に試料の少なくとも一部と接着させられる官能化された壁面部分を保持空間にもたらす壁を備えており、
上記官能化された壁面部分は試料の少なくとも一部が接着する傾向があるところの細胞体を含み、
上記音波発生器が、上記保持空間内に上記試料が含まれているときに、上記保持空間内の上記試料の1以上の細胞体に対して、上記官能化された壁面部分から離れる方向に、力を加えるように構成されており
前記コントローラーは、
官能化された壁面部分に付着した細胞体の量、
官能化された壁面部分への1以上の細胞体の接着強度、および
1以上の細胞体の位置および/または動き
のうちの少なくとも1を、官能化された壁面部分から離れる方向の力の関数として決定するように構成されている、
操作システム。 - 上記官能化された壁面部分が、試料の少なくとも一部と接着されられる複数の互いに異なって官能化された壁面部分を含む、請求項7に記載の操作システム。
- 上記試料ホルダーが、上記保持空間に流体及び/又はガスを導入するため及び/又は上記保持空間から流体及び/又はガスを除去するため、例えば保持空間を通して流体を流すため、フローシステムに接続されているか或いは接続可能であり、特に上記流体及び/又はガスが、試料物質、例えば試料流体及び/又は1以上の細胞体、を含む、請求項7又は8に記載の操作システム。
- 上記音波発生器が、上記保持空間内で調節可能な音波、好ましくは時間依存性のもの、を発生させるために、周波数及び振幅の少なくとも一方を調節するように制御可能なものであり、特に音波が定在波である、請求項7乃至請求項9のいずれか1項に記載の操作システム。
- 当該システムが、音波に対する1以上の細胞体の応答を検出するための検出器を備えており、該検出器が、音響検出器、例えば圧電素子、及び光学検出器、例えばフォトダイオード、フォトダイオードのアレイ、カメラ及び/又は顕微鏡、の1以上を含むことができる、請求項7乃至請求項10のいずれか1項に記載の操作システム。
- 当該システムが、
光源;
システムの操作の指標となるデータ及び/又は検出器からの信号を記憶するためのメモリ;
1以上の細胞体を追跡するためのトラッキングシステム、及び顕微鏡検査計算及び/又は顕微鏡法に関連する解析を実行するために、検出器に接続された又は接続可能なコントローラー;
センサー、及び該センサーからの信号に応答して音波発生器の動作を制御するために、音波発生器に接続された又は接続可能なコントローラー;
上記試料ホルダーの温度及び/又は温度プロファイルを調節するための熱素子、例えばペルチェ素子
の1以上を備える、請求項7乃至請求項11のいずれか1項に記載の操作システム。 - 焦点面のデジタル画像を生成するための検出器をさらに備えており、当該システムが、焦点が合っていない1以上の細胞体による干渉パターンの演算処理によって焦点面に垂直な方向における1以上の細胞体の位置を計算するための計算装置を備える、請求項7乃至請求項12のいずれか1項に記載の操作システム。
- 請求項1乃至請求項6のいずれか1項に記載の方法及び/又は請求項7乃至請求項13のいずれか1項に記載の操作システムで使用するための試料ホルダーであって、
流体媒体中に1以上の細胞体を含む試料を保持するための保持空間、及び
音波を上記保持空間内で生成して試料に力を加えるために当該試料ホルダーに接続された音波発生器を
備えており、当該試料ホルダーが、使用時に試料の少なくとも一部と接着させられる官能化された壁面部分を保持空間にもたらす壁を備えている、試料ホルダー。
