本発明は、クロロフィルの減少、ならびにバイオマスおよび脂質生産性の増大を有する光合成生物の突然変異体に関する。本発明はまた、こうした突然変異体を作製し、選択し、かつスクリーニングする方法に関する。
より高い生産性となるように、光合成生物を操作して、光合成効率を高めることは、植物および藻類生物学者の長年の目標である。米国特許出願公開第2014/0220638号および米国特許出願公開第2016/030489号(両方とも参照により本明細書に組み入れられる)は、弱光順化能力が損なわれている、クロロフィルの減少を有する「LIHLA」突然変異体と称される藻類突然変異体を得るための突然変異体スクリーニングについて記載している。すなわち、弱光であっても、強光適合細胞の低クロロフィル状態を保持する。米国特許出願公開第2014/0220638号には、光順化調節因子LAR1、LAR2、およびLAR3遺伝子に突然変異を有する藻類突然変異体について記載されており、米国特許第2016/0304896号では、葉緑体SRP54遺伝子に突然変異を有する藻類変異体について開示している。
本明細書に提供されるのは、Significant Growth Improvement 1(SGI1)ポリペプチドをコードしている遺伝子の減衰を有する突然変異光合成生物である。この突然変異光合成生物は、SGI1遺伝子の減衰を有さないこと以外は実質的に同一である対照光合成生物よりも、高い生産性を有する。より高い生産性は、バイオマス蓄積速度、またはバイオ産物、例えば脂質、タンパク質、炭水化物(例えば、1つ以上の糖またはアルコール)、色素、抗酸化物質、テルペノイド、ビタミン、またはポリマーなどの産生速度として測定することができる。本明細書に開示されているSGI1突然変異体は、光独立栄養条件下で生産性の増大を呈し得る。

本明細書に記載されるように、SGI1または「有意な成長改善1」ポリペプチドは、レスポンスレギュレーターレシーバーまたは「RR」ドメイン(pfam PF00072)およびMyb様結合ドメイン(本明細書において単に「myb」ドメイン(pfam PF00249)と称される)を含むポリペプチドであり、RRドメインは、mybドメインに対してN末端側に位置付けられている。RRドメインおよびmybドメインを包含するSGI1ポリペプチドのアミノ酸配列は、SRG1ポリペプチドの間ではあまり保存されていないかまたは保存されていないことが判明しているRRドメインとmybドメインとの間に生じるアミノ酸の伸長を含む。RRドメインとmybドメインとの間に生じるアミノ酸配列は、本明細書において2つのドメイン間のリンカーと称され得る。リンカーは任意の長さのものであってもよく、様々な実施例において1~約300個のアミノ酸、10~約200個のアミノ酸、または20~約150個のアミノ酸の長さの範囲であり得る。リンカー領域は、核局在化配列(NLS)を任意に含み得る。
SGI1遺伝子の減衰を有する、本明細書において提供される突然変異体としては、植物および藻類を含む、単細胞真核生物藻類(微細藻類)などの真核生物光合成生物などの光合成生物が挙げられる。
SGI1遺伝子の減衰を有する本明細書に提供される突然変異微生物は、ヒトの介入によって、例えば古典的突然変異誘発または遺伝子操作によって生成される突然変異体である。例えば、本明細書において提供される突然変異微生物は、実行可能な任意の突然変異誘発方法、これらに限定されないが、UV照射、ガンマ線照射、または化学的突然変異誘発、ならびにクロロフィルの減少および生産性の増加を有する突然変異体のスクリーニングなど、例えば、本明細書に開示された方法を用いて生成された突然変異体であり得る。例えば、クロロフィルの減少した突然変異体のスクリーニングでは、蛍光活性化細胞選別(FACS)、目視検査、および/またはクロロフィル吸収の抽出および測定を使用することができる。生産性の向上は、突然変異体の成長、および乾燥重量、湿潤重量、無灰乾燥重量、または全有機炭素(TOC)の測定によって評価され得る。古典的突然変異誘発および遺伝子操作を用いて光合成生物の突然変異体を生成する方法は周知である。
実施形態1は、SGI1ポリペプチドをコードする遺伝子の減衰を有する突然変異光合成生物であって、光合成生物は、バイオマスまたは少なくとも1つのバイオ産物を実質的に同じ条件下で成長または培養させた対照光合成生物よりも、より多く産生し、任意で、以下のうちの1つ、またはそれ以上を満たすものとする:
a) 突然変異光合成生物は、対照光合成微生物と比べてクロロフィルの減少を有する。
b) 突然変異光合成生物は、対照光合成生物と比べてクロロフィルa:b比の上昇を有する。
c) 突然変異光合成生物は、対照光合成生物と比べて、減少した光化学系II(PSII)アンテナサイズを有する。
d) 突然変異光合成生物は、対照光合成生物と比べて、より高い14C Pmaxを有する請求項1記載の突然変異光合成生物。
e) 突然変異光合成生物が、対照生物よりも高いFv/Fmを有する、請求項1記載の突然変異光合成生物。
f) 突然変異光合成生物が、対照生物よりも高いタンパク質含有量を有する。
g) 突然変異光合成生物が、対照生物よりも高いRubisco活性化酵素含有量を有する。

実施形態4は、SGI1遺伝子が突然変異している、実施形態1~3のいずれかの突然変異光合成生物である。任意で:

PM119は、以下を含む栄養豊富培地である:35pptのInstant Ocean Salt(Aquatic Eco Systems;Apopka、FL)、5xギラードF/2海洋水濃縮溶液(50Xストック、Sigma-Aldrich、St.Louis、MO、カタログ番号G0154;培地中の成分の最終濃度:4.413mM硝酸ナトリウム;一塩基性0.16mMリン酸ナトリウム;0.103μMビオチン;0.240μM塩化コバルト・6H2O;0.200μM硫酸銅・5H2O;0.0585mM EDTA二ナトリウム・2H2O;4.54μM塩化マンガン・4H2O;0.124μMモリブデン酸ナトリウム・2H2O;1.48μMチアミン・HCl;0.0185μMのビタミンB12;0.382μM硫酸亜鉛・7H2O)。
葉緑素、または緑藻種から突然変異体を単離するために、パラクロレラ株WT-1185の細胞を、UVにより突然変異誘発し、弱光順化後に、低クロロフィル蛍光に基づいて選択した。突然変異誘発に使用するためのパラクロレラ株WT-1185は、海洋環境から単離させた。パラクロレラWT-1185細胞を対数増殖期中期まで成長させ、次いで成長培地PM119で1x106細胞/mLに希釈した。細胞懸濁液をピペットで100mmペトリ皿に移し、STRATALINKER(登録商標)2400 UV架橋剤(Agilent Technologies、Santa Clara、CA)内に入れ、プレートの蓋を外した。UV照射は、10,000、25,000、および50,000μJ/cm2で行った。照射後、回収の間24時間にわたって光曝露を妨げるために、細胞懸濁液を箔で包み込んだ振とうフラスコにピペットにより入れた。
実施例1に記載したように突然変異誘発させ、回収した後、淡色コロニーからの細胞を選択し、弱光(光量子束密度100μmol m-2sec-1)において、1~5日の間成長させ、次いで、BD FACSAria II flow cytometer(BD Biosciences、San Jose、CA)を用いてフローサイトメトリーにより選別し、低クロロフィル蛍光を有する細胞を選択した。一般に、細胞の全集団と比較して、クロロフィル蛍光が最も少ない細胞の部分(約0.5~2%)を選択した。選別した細胞をプレーティングした後、視覚的に淡緑色または黄色のコロニーを選択することによって、フローサイトメトリーによって単離されたアンテナが減少している系統のさらなる一次スクリーニングを行った。他の色素減少突然変異体および偽陽性から推定上のアンテナ減少系統をスクリーニングするために、選択されたコロニーを中程度の処理量の二次培養スクリーニングに供し、光生理学的測定の前に分離株を弱光条件に順化させた。クロロフィル蛍光を弱光順化中にモニターして、強光順化状態の特徴である減少したクロロフィル蛍光特性を保持しているコロニーを選択した。選択されたクローンは、強光から弱光に移されたときに、クロロフィルのほんのわずかな増加(野生型細胞と比較して)を示した。
弱光条件下でクロロフィルの減少を有することが判明したパラクロレラ株において、生産性の増大について、2つの分離株(突然変異体NE-7843およびNE-13380)を分析した。生産性アッセイでは、突然変異体の光独立栄養培養物を、一定の光の半連続式(CL-SCPA)で数日間にわたって成長させた。培養サンプルは、バイオマス測定のために毎日取り出した。光は、1日24時間、一定の光量子束密度1900~2000μmol m-2sec-1に維持した。このアッセイでは、225cm2フラスコ中のPM119培地に、所定の突然変異株のシード培養物を接種した。1株につき3つの培養を開始した。フラスコは、培養物からバブリングされたCO2富化空気(1%CO2)を送達するためのシリンジフィルターに接続された管を有する栓を有していた。フラスコは、LEDライトバンクに対して幅(最も狭い寸法)により整列させた。光源から背部にわたるフラスコの「深さ」寸法は、13.7cmであった。フラスコの配置を考慮に入れると、光源の表面からのフラスコ内の細胞の最も遠い距離は、約15.5cmであった。培養物は、培養体積の65%を除去し、その分を新鮮なPM119培地に交換して、培養物において発生する蒸発による塩分の増加を調整するよう希釈することで、毎日希釈した。TOC分析用のサンプルは、希釈のために取り出した培養物から採取した。培養物が定常状態に達した後、半連続生産性アッセイを12日間実施した。
NE-13380およびNE-7843のゲノムを配列決定し、一塩基多型(SNP)または挿入/欠失突然変異(インデル)などの突然変異を同定する。これらのうちのいずれもが、この2つの株において観察される表現型の遺伝的基礎を形成し得る。SNPは、4つの異なる遺伝子のコード領域において突然変異が引き起こされたNE-13380のゲノムにおいて同定された(表1の最初の4行)。
T-2261遺伝子の突然変異が結果的に高生産性表現型をもたらしたことを明確に実証するために、T-2261遺伝子が、2番目のエクソンの3’末端を標的とするガイドRNAを用いて、Cas9媒介挿入突然変異誘発によってノックアウトされた3つの株を作製した(図2)。単一ガイドRNA(sg RNA)(SEQ ID NO:5)は、SEQ ID NO:4の標的配列(すなわち、CRISPR配列またはT-2261遺伝子の標的配列に対応する配列)を含んでいた。また、in vitro転写のために、T7プロモーターの配列を組み込んで二重鎖鋳型を生成する2つの相補的DNAオリゴマーをアニーリングすることによって生成した。製造業者のプロトコールに従って、MEGAshortscript(商標)T7キット(Life Technologiesカタログ番号AM1354M;Carlsbad、CA)を使用して、in vitro転写反応を実施した。得られたRNAは、製造業者のプロトコールに従って、Zymo-Spin(商標)V-Eカラム(Zymo Research;Irvine、CA;カタログ番号C1024-25)を使用して精製した。
1)パラクロレラのために最適化され、パラクロレラ由来のイントロンを含む、操作されたCas9遺伝子コドンを含有し、N末端FLAGタグ、核局在化シグナル、およびパラクロレラRPS17プロモーター(SEQ ID NO:77)およびパラクロレラRPS17ターミネーター(SEQ ID NO:78)に作動可能に連結したペプチドリンカー(SEQ ID NO:76)を含む、Cas9発現カセット;
2)パラクロレラのために最適化されたアスペルギルス・テレウスコドン由来のブラストサイジン耐性遺伝子を含み、およびパラクロレラRPS4プロモーター(SEQ ID NO:80)に作動可能に連結されたパラクロレライントロン(SEQ ID NO:79)、およびパラクロレラRPS4ターミネーター(SEQ ID NO:81)を含む、選択マーカー発現カセット;ならびに
3)TurboGFP遺伝子(Evrogen、Moscow、Russia)(SEQ ID NO:82)を含み、パラクロレラACP1プロモーター(SEQ ID NO:83)によって駆動され、パラクロレラACP1ターミネーター(SEQ ID NO:84)によって終結される、GFPレポーター発現カセット
(例えば、同時係属中の米国特許出願公開第2016/0304896号、米国特許出願公開第2017/0073695号および米国特許出願公開第2017/0152520号(これらはすべて、その全体が参照により本明細書に組み込まれる)を参照されたい)。
パラクロレラSGI1遺伝子(SEQ ID NO:1、SEQ ID NO:2として提供されるコード配列)は、2つの主要な機能的ドメイン(両方とも、619個のアミノ酸タンパク質のN末端側半分において生じる)を含むポリペプチド(SEQ ID NO:3)をコードすることが見出された。レスポンスレギュレーターレシーバーまたは「RR」ドメイン(Pfam PF00072)の存在(パラクロレラSGI1ポリペプチド(SEQ ID NO:3)の約アミノ酸36からアミノ酸148まで及ぶ)は、CheY様ポリペプチドとして、SGI1のバイオインフォマティックアノテーションを担っている(表1および2参照)。このRRドメインはまた、保存ドメインデータベース(CDD)において「シグナルレシーバードメイン」cd00156としても特徴付けられ、約アミノ酸37からアミノ酸154まで及んでいる。このドメインは、タンパク質のCOG(Clusters of Orthologous Groups)データベースにおける「CheY様レシーバー(REC)ドメイン」COG0784として、およびInterproの「CheY様スーパーファミリー」ドメインIPR011006としても特徴付けられ、これらの特徴付けられたドメインは両方とも、SEQ ID NO:3のパラクロレラSGI1ポリペプチドの約アミノ酸33から約アミノ酸161まで及んでいる。RRドメイン(時に「レシーバー」または多くの場合「REC」ドメインと称される)は、細菌性二成分調節系(CheYとして公知であるポリペプチドを含む細菌性走化性二成分系など)に見られ、センサーパートナーからシグナルを受け取る。こうした系のRRドメインは、多くの場合DNA結合ドメインのN末端側に見出され、典型的にはリン酸化され得るホスホアクセプター部位を含む。リン酸化は、その活性化または不活性化の原因であり得る。
高生産性突然変異体のクロロフィル含有量は、細胞をメタノールで抽出し、上清を分光光度法で分析することによって決定した。簡単に説明すると、500μlアリコートの培養物を2.0mlツイストトップチューブにピペットで移し、卓上型微量遠心機を用いて15,000rpmで10分間ペレット化した。上清をペレットから吸引し、各ペレットを1.5mlの99.8%メタノール(予め炭酸マグネシウムで中和している)中に再懸濁した。0.2mlのガラスビーズ(直径0.1mm)を各バイアルに添加し、3分間ビーズビートした。1.0mlの上清を新しい1.7mlのフリップトップチューブに移し、卓上型微量遠心機で15,000rpmで10分間遠心分離した。得られたペレットは白色であり、完全な抽出が行われたことを示していた。各上清0.8mlを光学ガラスキュベットにピペットで移し、吸収波長を720nm、665nmおよび652nmの波長で直ちに読み取った。分光光度測定は、99.8%メタノールブランクを用いて二重ビームモードで行った。以下の式を用いてクロロフィル濃度を算出した:クロロフィルa[g m-3]=16.72(A665-A720)+9.16(A652-A720)およびクロロフィルb[g m-3]=34.09(A652-A720)-15.28(A665-A720)。クロロフィルaおよびbの量は、細胞ごとおよびTOCごとに標準化した。表5は、細胞当たりの総クロロフィル量は、SGI1突然変異体間で幾分変動したが、野生型細胞と比べて約30%~約65%の範囲の量だけ普遍的に減少したことを示す。これは、観察されたアンテナサイズの減少と一致している。TOCごとに、野生型細胞に対するSGI1突然変異体中の総クロロフィルの減少は、約30%~約50%の範囲であった。
を用いてフィッテイングさせた。式中、ECSMは、最大ECSシグナルであり;Itは、光子密度(単位photon/m2)であり;σPSIは、PSIの機能的断面積である。野生型のパラクロレラ(WT-1185)のPSI機能的断面積の得られた値は、(4.0+/-0.5)×10-18m2であった。これらの値は、同じ条件下で成長させたPSIIの機能的断面積について得られたものに近接している(σPSII=(4.3+/-0.1)×10-18m2)。これらのパラメータの誤差は、20%を超えないと推定した。
式中、[Chl/TOC]は、サンプルのクロロフィル/TOCであり;OD(λ)は、波長λで測定されたサンプルの光学密度であり;ΔIは、キュベット内の測定光路長(1cm)であり;I(λ)は、波長λで藻類を成長させるために使用される光源の強度である。
アンテナ減少株のスクリーニングおよび生産性アッセイを使用した高生産性変異体NE-7843およびNE-13380の発見、ならびにその後のこれらの高生産性変異体が、より高い最大光合成収率(FV/FM)、PSIIアンテナサイズσPSII)の減少(σPSII)、およびPSII代謝回転速度(1/τ’Qa)の上昇など、表5および6に記載の特徴的な光生理学的特性を有するという発見に基づき、本明細書では「FACS-FIRE」スクリーニングと呼ばれる、高生産性藻類突然変異体のスクリーニングを考案した。このスクリーニングは、FACSを使用して、祖先藻類集団から低クロロフィル変異体を選択することを含み、実施例6に記載のFIRe装置を用いて低クロロフィル蛍光についてFACSにより選択された集団から、個々の系統のスクリーニングを行い、1つ以上の波長でのPSIIアンテナサイズ(σPSII)、FV/FM、およびPSIIのアクセプター側での電子輸送速度(1/τ’Qa)を決定した。PSIIアンテナのサイズの減少、FV/FMの上昇、および1/τ’Qaの上昇を有する系統を拡大し、再試験し、炭素吸収速度(Pm(14C))および培養中の生産性についてアッセイした。
SGI1減衰突然変異体の全バイオマス組成を決定するために、毎日40%希釈を行い、半連続モードで成長させた培養物からのサンプルの定量分析を行い、半連続培養における細胞の脂質、タンパク質および炭水化物の含有量を決定した。培養物が定常状態に達した後、毎日希釈のために取り出した培養物のアリコートを脂質、タンパク質、および炭水化物を分析するために使用した。藻類培養サンプルの全有機炭素(TOC)は、2mLの細胞培養物をDI水で総量20mLに希釈することによって決定した。全炭素(TC)および全無機炭素(TIC)を決定するために、測定1回当たり3回の注入物をShimadzu TOC-Vcsj Analyzerに注入した。燃焼炉は720℃に設定し、TOCはTCからTICを差し引くことによって決定した。4点較正範囲は、2ppm~200ppmまでであり、これは、非希釈培養物の20~2000ppmに対応し、相関係数はr2>0.999であった。
実施例3に記載した一定の光の半連続培養アッセイ(CL-SCPAまたは「CL2000」アッセイ)で培養した野生型およびSGI1突然変異株NE-7843およびNE-13380のmRNA抽出物についてトランスクリプトーム解析を行った。SGI1突然変異体のトランスクリプトーム解析では、野生型と比べて異なるように調節された多数の遺伝子が示された。SGI1突然変異体がクロロフィルの減少および光合成アンテナの減少を示したように、集光クロロフィル結合タンパク質(LHC)遺伝子が、野生型祖先細胞株と比べて、突然変異体においてより少ない転写産物の存在量を有していたかを確認することは興味深いものであった。パラクロレラゲノムにおいて15のLHC遺伝子が同定され、PSII反応中心に会合するLHCポリペプチドであるCP26をコードする遺伝子を除いて、野生型の発現レベルと比べて、同定されたパラクロレラLHC遺伝子のすべてが、SGI1突然変異体において著しく下方調節されることが見出された(図9)。LHC mRNAは、平均して、WT-1185と比較してSGI1の存在量において約1.7倍の減少を示した。これらのトランスクリプトームデータからの別の驚くべき観察は、Rubisco活性化酵素がSGI1突然変異体において、約6倍高い転写産物存在量を有することがわかったことである。
南カリフォルニアの春における自然の昼光条件を模倣した光レジメン下で半連続系におけるSGI1突然変異体の生産性を評価するために、スケールアップ培養を用いて、225cm2の長方形組織培養フラスコに接種した。そのそれぞれは、接種後に、培養液550mlの最終総量を含んでいた。典型的な接種量は、約350mlのPM119培地に添加された約200mlのスケールアップ培養物であった。培養物は、照度がピークになった日中、体積の65%を取り除き、その分を同体積のアッセイ媒体、および追加の10mlの脱イオン水と交換して、蒸発を補うことにより、毎日希釈した(補充培地に含まれる)。半連続アッセイは、典型的には10~14日間実行した。毎日の脂質およびバイオマス生産性は、定常状態に達した培養物についてのみ算出した(このアッセイでは、成長の増加は、希釈率に等しかった)。
SGI1突然変異体NE-13380の生産性も、窒素豊富バッチモードで培養しながら1週間にわたって評価した。NE-13380および野生型WT-1185の培養物を、窒素豊富培養培地中で成長させたシード(スケールアップ)培養物からの初期OD730 0.5まで接種した。
SGI1突然変異体も、窒素制限下においてバイオマスおよび脂質蓄積について評価した。バッチ生産性アッセイにおいて、SGI1ノックアウト株NE-7843および野生型祖先株WT-1185を、窒素欠乏培地で培養した。すなわち、培地は細胞成長のための窒素源を有していなかった。生産培養物を、8.8mMの硝酸塩を含むPM074培地で成長させたシード(スケールアップ)培養物から、初期OD730 0.5まで接種した。
本明細書に開示されているSGI1突然変異体の成長速度の改善は、複数の培養条件において観察されている(表8)。栄養素を豊富に含む新鮮な培地を導入し、等量の培養物を排出する連続希釈システムによって、培養物を一定の強光下で高希釈度において維持した。パラクロレラSGI1突然変異体は、中程度から強光条件(500~1,400μE)において、18~20%上昇した成長速度を示し、非常に強い光(5,000μE)においては、SGI1突然変異体は、野生型祖先株の約5倍の速度で成長することが観察された(表8)。高光条件下での成長速度の劇的な差は、SGI1突然変異体を選択するために使用され得る。

本明細書でテトラセルミスWT-105と称される、テトラセルミス種の野生型藻類の環境単離物のゲノムは、施設内で完全にその配列が決定された(Synthetic Genomics、Inc.、La Jolla、CA、USA)。ゲノムの分析は、三倍体であるWT-105のテトラセルミス属と最も一致していた。テトラセルミスWT-105ゲノムの翻訳された配列についての照会として、パラクロレラSGIポリペプチド配列を用いた相互BLASTに基づいて、3つの遺伝子が同定された。T-5172(SEQ ID NO:85、SEQ ID NO:86のタンパク質をコードする配列(またはcDNA配列)を有する)、T-5185(SEQ ID NO:87、SEQ ID NO:88のタンパク質をコードする配列(またはcDNA配列)を有する)、およびT-5230(SEQ ID NO:89、SEQ ID NO:90のタンパク質をコードする配列(またはcDNA配列)を有する)。これらは、テトラセルミスWT-105 SGI1遺伝子の対立遺伝子とみなされる。対立遺伝子T-5185(SEQ ID NO:14)およびT-5230(SEQ ID NO:16)によってコードされるタンパク質は高度に相同性であり、BLASTにより98%のアミノ酸配列同一性が示されている。BLAST分析によれば、T-5172対立遺伝子によってコードされるタンパク質(SEQ ID NO:15)は、T-5230対立遺伝子によってコードされるタンパク質(SEQ ID NO:16)と90%同一であり、またT-5185対立遺伝子によってコードされるタンパク質(SEQ ID NO:14)と92%同一である。各対立遺伝子は、RRドメインおよびmybドメインを有すると決定された(表3参照)。表3にも提供されたとおり、テトラセルミスSGI1対立遺伝子ポリペプチドT-5172、T-5185およびT-5230は、少なくとも400、すなわち、SGI1ポリペプチドを同定するために開発されたHMMに対して、それぞれ、403.0(e値2e-118)、403.6(e値9e-119)、および402.9(e値3e-118)のスコアを有することが見出された(実施例6)。
これらのクローンの特徴を明らかにするために、表8に列挙された10個のクローンのうちの5個(クローン2、6、7、8、および10)を、24時間/日、弱光(光量子束密度140mol m-2s-1)下で、還元炭素源を含まず、窒素源として硝酸塩を含む豊富PM074培地において、光独立栄養により培養した。培養物は、毎日希釈して、固定細胞数を維持した。クロロフィルを抽出し、実施例7に記載したようにFIReおよび14C Pmax測定を行った。この結果を図16A、B、およびC;図17A、B、C、およびD、ならびに図18A、B、およびCに示す。これらの弱光条件下では、変異体のすべてについて、野生型株と比較してクロロフィルa:b比が増加し(図16C)、クローン2、7および10は、総クロロフィル、特に、TOC(図16A)およびセル(図16B)を基準にした両方においてクロロフィルbが減少したことを示した。クローンはすべて、450nmおよび520nmの両方の励起で測定されたPSII断面サイズの減少を示し、クローン2、7、および10は、野生型と比較してPSII断面サイズの最大の減少を示す(図17Aおよび17B)。クローン2、7および10もまたFv/Fmの最大の上昇を示し(図17C)、すべてのクローンが、光合成代謝回転時間PSIIの短縮を示した(図17D)。14C Pmaxは、細胞およびTOCの両方を基準として、すべてのクローンにおいて上昇し(図18Aおよび図18B)、すべてのクローンは、野生型よりも高い成長速度を有した(図18C)。
CL-SCUBA(一定の光の半連続尿素系アッセイ)において、テトラセルミスSGI1突然変異体の光独立栄養培養物を、一定の光半連続モードで数日間にわたって成長させた。培養サンプルは、バイオマスを決定するために、各日、同じ時に取り出した。光は、光量子束密度2000μmol m-2sec-1にプログラムされた。このアッセイでは、500mlの四角フラスコ中の420mlのPM153培地(窒素源として尿素を含む)に所定の株のシード培養物を接種した。1株につき3つの培養を開始した。フラスコは、培養物からバブリングされたCO2富化空気(1%CO2)を送達するためのシリンジフィルターに接続された管を有する栓を有していた。0.0875m2の開口部を有するフラスコを光に向けて整列させ、光源から背部にわたるフラスコの「深さ」寸法は8cmであった。培養物は、培養体積の40%を取り出し、その分を新鮮なPM153培地と交換して、培養物に生じる蒸発による塩分の増加を調整することにより、毎日希釈した。TOC分析用のサンプルは、培養物が定常状態に達した後に、希釈のために取り出した培養物から採取した。
日周のSCUBA(半連続尿素系アッセイ)では、突然変異体の光独立栄養培養物を数日にわたって毎日、光半連続モードで成長させた。光は、暗所から正午の光量子束密度2000μmol m-2sec-1まで及ぶように、カリフォルニアのインペリアルバレーの平均的な春の日を模倣するようにプログラムされた。サンプルは毎日「夕暮れ」に採取した。このアッセイでは、500mlの四角フラスコに入った尿素ベースのPM153培地420mlに、所定の突然変異株のシード培養物を接種した。1株につき3つの培養を開始した。フラスコは、培養物からバブリングされたCO2富化空気(1%CO2)を送達するためのシリンジフィルターに接続された管を有する栓を有していた。フラスコを光に向かって0.0875m2の開口部と整列させ、光源から背部にわたるフラスコの「深さ」寸法は8cmであった。培養物は、培養体積の40%を除去し、その分を新鮮なPM153培地と交換して、培養物中で生じる蒸発による塩分の増加を調整することによって、毎日希釈した。培養物が定常状態に達した後に、培養サンプルをバイオマス測定のために毎日取り出した。TOC分析用のサンプルは、希釈のために取り出した培養物から採取した。
高等植物におけるSGI1突然変異の影響を決定するために、最初にシロイヌナズナ内のパラクロレラSGI1遺伝子の最も近い関連種を同定した。実施例6に開示されたバイオインフォマティック解析を用いて、パラクロレラSGI1に最も近いシロイヌナズナ相同体はARR2(SEQ ID NO:22)であると決定された。
Claims (34)
- SEQ ID NO:3、14、15、および16からなる群より選択されるポリペプチドに対して少なくとも90%のアミノ酸配列同一性を有するSGI1ポリペプチドをコードする遺伝子の減衰を有する突然変異緑藻類生物であって、
該突然変異緑藻類生物が、バイオマスを、同じ条件下で成長または培養させた対照緑藻類生物よりも多く産生し、
該対照緑藻類生物が、該SGI1遺伝子の減衰を有さない、
突然変異緑藻類生物。 - 同じ条件下で成長または培養させた対照緑藻類生物よりも、少なくとも15%多くの脂質を産生する、請求項1に記載の突然変異緑藻類生物。
- 対照緑藻類生物と比べて、減少したクロロフィルを有する、請求項1~2のいずれか一項に記載の突然変異緑藻類生物。
- 対照緑藻類生物と比べて、上昇したクロロフィルa:b比を有する、請求項1~3のいずれか一項に記載の突然変異緑藻類生物。
- 対照緑藻類生物と比べて、より高い14C Pmaxを有する、請求項1~4のいずれか一項に記載の突然変異緑藻類生物。
- 対照緑藻類生物よりも高いFv/Fmを有する、請求項1~5のいずれか一項に記載の突然変異緑藻類生物。
- 対照緑藻類生物よりも高いタンパク質含有量を有する、請求項1~6のいずれか一項に記載の突然変異緑藻類生物。
- 対照緑藻類生物よりも高いrubisco活性化酵素含有量を有する、請求項1~7のいずれか一項に記載の突然変異緑藻類生物。
- 対照緑藻類生物と比べて、減少した光化学系II(PSII)アンテナサイズを有する、請求項1~8のいずれか一項に記載の突然変異緑藻類生物。
- PSIIアンテナサイズの断面単位サイズが、対照緑藻類生物のPSIIアンテナの断面単位サイズと比べて、少なくとも25%減少している、請求項9に記載の突然変異緑藻類生物。
- 対照緑藻類生物と比べて、上昇したPmax(14C)を有する、請求項6~10のいずれか一項に記載の突然変異緑藻類生物。
