Description
本発明は、ヒトTL1Aに特異的に結合し、かつDR3への結合をブロックし、それによってTL1A発現細胞においてそうでなければ生じるであろう免疫刺激シグナルを阻害する、単離抗体、特にモノクローナル抗体、例えばヒトモノクローナル抗体を開示する。単離抗体、かかる抗体の製造方法ならびに抗体または断片を含有するように製剤化された医薬組成物が、本明細書に提供される。免疫抑制のための抗体単独で、または他の免疫抑制剤と組み合わせて使用する方法もまた、本明細書に提供される。従って、本明細書中に記載される抗huTL1A抗体は、例えば、免疫系疾患を治療することを含む広範な治療適用に使用されてもよい。

本願は、免疫系疾患を治療することにおける治療剤としての使用に望ましい特性を有する完全ヒト抗huTL1A抗体を開示する。これらの特性は、高親和性でヒトTL1Aに結合する能力、カニクイザルTL1Aに結合する能力およびDR3結合(したがってシグナル伝達)をブロックする能力を含む。
10A4.F7.2E8などの、huTL1Aへの結合について本発明の抗体と競合する抗huTL1A抗体は、本明細書中に記載されるもの(実施例1)と類似の免疫処置プロトコールを使用して産生されてもよい。本明細書中に記載される抗huTL1A抗体と結合について競合する抗体はまた、ヒトTL1Aまたはその細胞外ドメイン(配列番号19の残基72-251)を含むコンストラクトでマウスを免疫することにより、または本明細書に開示された抗TL1A抗体(例えば10A4.F7.2E8)により結合されるエピトープを含有するヒトTL1Aの断片で免疫することにより、生成されてもよい。生じる抗体は、当技術分野で周知の方法、例えば、ELISAでTL1Aの細胞外ドメインおよび免疫グロブリンFcドメインの融合タンパク質への結合をブロックすること、または例えばFACSにより表面上でhuTL1Aを発現する細胞に結合する能力をブロックすることによりヒトTL1Aへの10A4.F7.2E8の結合をブロックする能力についてスクリーニングされ得る。様々な実施形態では、試験抗体を、10A4.F7.2E8の添加前、同時、または後にTL1A-Fc融合タンパク質(または表面上でhuTL1Aを発現する細胞)を接触させる。TL1A(Fc融合として、または細胞上のいずれか)への10A4.F7.2E8の結合を、特におよそ化学量論的な濃度で、減少させる抗体は、同じ、オーバーラップする、または隣接するエピトープに結合する可能性があり、したがって、10A4.F7.2E8の望ましい機能的な特性を共有してもよい。
本明細書に開示された抗体と同一の、または類似のエピトープに結合する抗huTL1A抗体は、本明細書中に記載されるもの(実施例1)と類似の免疫処置プロトコールを使用して産生されてもよい。生じる抗体は、ヒトTL1Aへの高親和性結合についてスクリーニングされ得る(実施例4)。選択された抗体は、抗体により結合される正確なエピトープを決定するために水素/重水素交換質量分析(HDX-MS)方法で研究され得る(実施例8)。ヒトTL1A上の、抗体10A4.F7.2E8と同一の、または類似のエピトープに結合する抗体は、10A4.F7.2E8の望ましい機能的な特性を共有する可能性が高い。

いくつかの実施形態では、本発明の抗huTL1A抗体は、本明細書に開示された抗huTL1A抗体のように、高親和性でhuTL1Aに結合し、有効な治療剤である可能性を高める。様々な実施形態では、本発明の抗huTL1A抗体は、10nM、5nM、2nM、1nM、300pMまたは100pM未満のKDでhuTL1Aに結合する。他の実施形態では、本発明の抗huTL1A抗体は、2nMおよび100pM間のKDでhuTL1Aに結合する。huTL1Aに対する抗体の結合能力を評価する標準アッセイは、ELISA、ウェスタンブロット、BIACORE(登録商標)SPR解析およびRIAを含む。
本明細書に開示された抗体配列におけるいくつかの可変性(variability)は、許容され、依然として抗体の望ましい特性を維持してもよい。CDR領域はカバットシステムを使用して記載されている(Kabat,E.A.,et al.(1991)Sequences of Proteins of Immunological Interest,Fifth Edition,U.S.Department of Health and Human Services,NIH Publication No.91-3242)。従って、本発明は、本明細書に開示された抗体(例えば10A4.F7.2E8)のCDR配列と少なくとも70%、75%、80%、85%、90%、95%、96%、97%、98%、または99%同一のCDR配列を含む抗huTL1A抗体をさらに提供する。本発明はまた、本明細書に開示された抗体(例えば10A4.F7.2E8)の重および/または軽鎖可変ドメイン配列と少なくとも70%、75%、80%、85%、90%、95%、96%、97%、98%、または99%同一の重および/または軽鎖可変ドメイン配列を含む抗huTL1A抗体を提供する。
抗原結合特異性が主にCDRによって決定されることを考慮すると、本明細書に開示された抗体(例えば10A4.F7.2E8)とCDR配列を共有する抗体は、それらの望ましい特性を共有する可能性が高い。
VHおよびVL領域
改変された抗体を遺伝子操作するために出発物質として本明細書に開示された1以上のVHおよび/またはVL配列を有する抗体を使用して調製され得る、遺伝子操作された抗体および改変された抗体もまた提供され、改変された抗体は出発抗体から変化した特性を有してもよい。抗体は、可変領域の1つまたは両方(すなわち、VHおよび/またはVL)内、例えば、1以上のCDR領域内および/または1以上のフレームワーク領域内、の1以上の残基を改変することにより遺伝子操作され得る。その上または代わりに、例えば、抗体のエフェクター機能を変えるために、抗体は、定常領域内の残基を改変することにより遺伝子操作され得る。
抗原への抗原結合ドメインの結合から生じる治療用抗体の活性(例えばアンタゴニスト抗体の場合、コグネイトリガンドまたは受容体タンパク質、またはアゴニスト抗体の場合、誘導されたシグナル伝達をブロックする)に加えて、抗体のFc部分は、任意の数の生物学的効果を引き出すために複雑な方法で一般に免疫系と相互作用する。免疫グロブリンのFc領域などのエフェクター機能は、抗原-依存性細胞傷害性(ADCC)、補体依存性細胞傷害(CDC)、および抗体-依存性細胞介在貪食(ADCP)などの多くの重要な抗体機能に関与し、異なる機構によるものではあるが標的細胞の死滅をもたらす。重鎖定常領域(IgA、IgG、IgD、IgE、IgM)の5つの主要なクラスまたはアイソタイプが存在し、特徴的なエフェクター機能をそれぞれ有する。これらのアイソタイプは、サブクラスにさらに細分することができ、例えば、IgGは、IgG1、IgG2、IgG3、およびIgG4として知られている4つのサブクラスに分かれている。IgG分子は、抗体のIgGクラスに特異的な3つのクラスのFcγ受容体(FcγR)、すなわちFcγRI、FcγRII、およびFcγRIIIと相互作用する。FcγR受容体へのIgGの結合についての重要な配列は、CH2およびCH3ドメインに位置することが報告されている。抗体の血清半減期は、新生児のFc受容体(FcRn)に結合する抗体の能力により影響される。
ADCC活性は、Fc領域を改変することによって減少されてもよい。或る実施形態では、Fc受容体への結合に影響する部位、好ましくはサルベージ受容体結合部位以外の部位は、除去されてもよい。他の実施形態では、Fc領域は、ADCC部位を除去するように改変されてもよい。ADCC部位は、当技術分野で知られている;例えば、IgG1におけるADCC部位に関してSarmay et al.(1992)Molec.Immunol.29(5):633-9参照。一実施形態では、ヒトIgG1のG236RおよびL328Rバリアントは、FcγR結合を効率的に除去する。Horton et al.(2011)J.Immunol.186:4223およびChu et al.(2008)Mol.Immunol.45:3926。他の実施形態では、FcγRへの減少した結合を有するFcは、アミノ酸置換L234A、L235EおよびG237Aを含んでいた。Gross et al.(2001)Immunity 15:289。
本明細書中に記載される抗体は、軽または重鎖可変領域のいずれかにおける1以上のグリコシル化部位を含有できる。かかるグリコシル化部位は、抗体の増加した免疫原性または変化した抗原結合に起因する抗体のpKの変化をもたらしてもよい(Marshall et al(1972)Annu Rev Biochem 41:673-702;Gala and Morrison(2004)J.Immunol 172:5489-94;Wallick et al(1988)J Exp Med 168:1099-109;Spiro(2002)Glycobiology 12:43R-56R;Parekh et al(1985)Nature 316:452-7;Mimura et al.(2000)Mol Immunol 37:697-706)。グリコシル化は、N-X-S/T配列を含有するモチーフで起こることが知られている。いくつかの例では、可変領域グリコシル化を含有しない抗TL1A抗体を有することが好ましい。これは、可変領域におけるグリコシル化モチーフを含有しない抗体を選択することによる、またはグリコシル化領域内の残基を突然変異させることによる、いずれかで達成され得る。
本明細書中に記載される別の態様は、本明細書中に記載される抗体をコードする核酸分子に関する。核酸は、全細胞、細胞溶解物、または部分的に精製された、もしくは実質的に純粋な形態で存在してもよい。核酸は、アルカリ性/SDS処理、CsClバンド形成、カラムクロマトグラフィー、制限酵素、アガロースゲル電気泳動および他の当技術分野で周知のものを含む標準技術により、他の細胞成分または他の混入物、例えば、他の細胞核酸(例えば、他の染色体DNA、例えば、天然の単離されたDNAに結合している染色体DNA)またはタンパク質から精製される場合、「単離された」または「実質的に純粋である」。F.Ausubel,et al.,ed.(1987)Current Protocols in Molecular Biology,Greene Publishing and Wiley Interscience,New York参照。本明細書中に記載される核酸は、例えば、DNAまたはRNAであり得、かつイントロン配列を含有しても、しなくてもよい。或る実施形態では、核酸はcDNA分子である。
本発明の様々な抗体、例えば本明細書に開示された抗ヒトTL1A抗体と同一のエピトープと、競合または結合する抗体は、Kohler and Milstein,Nature 256:495(1975)に記載された標準体細胞ハイブリダイゼーション技術などの様々な既知の技術を使用して産生され得る。体細胞ハイブリダイゼーション法が好ましいが、原理上、モノクローナル抗体を産生する他の技術、例えば、Bリンパ球のウイルス性または発癌性形質転換、ヒト抗体遺伝子のライブラリーを使用するファージディスプレイ技術もまた、利用され得る。
ヒトTL1Aに対する完全ヒト抗体を作製するために、例えば、Lonberg et al.(1994)Nature 368(6474):856-859;Fishwild et al.(1996)Nature Biotechnology 14:845-851およびWO98/24884によって、その他の抗原について記載されるように、ヒト免疫グロブリン遺伝子を含有するトランスジェニックまたはトランスクロモソーマルマウス(例えば、HCo12、HCo7またはKMマウス)を、TL1A抗原および/またはTL1Aを発現する細胞の精製または濃縮された調製物を用いて免疫処置できる。あるいは、マウスをヒトTL1AをコードするDNAを用いて免疫処置できる。好ましくは、マウスは、第1の注入の際に6~16週齢とする。例えば、HuMAbマウスを腹膜内に免疫処置するために、組換えTL1A抗原の精製または濃縮された調製物(5~50μg)を使用できる。TL1A抗原の精製または濃縮された調製物を使用する免疫処置が抗体をもたらさない事象では、TL1Aを発現する細胞、例えば、細胞系統を用いてマウスを免疫処置して、免疫応答を促進することもできる。例示的細胞系統として、TL1A過剰発現性安定CHOおよびRaji細胞系統が挙げられる。
本明細書に記載されるヒトモノクローナル抗体を産生するハイブリドーマを作製するために、免疫処置されたマウスから脾細胞および/またはリンパ節細胞を単離し、マウス骨髄腫細胞系統などの適当な不死化細胞系統と融合できる。得られたハイブリドーマを抗原特異的抗体の産生についてスクリーニングできる。例えば、50% PEGを用いて、免疫処置マウス由来の脾臓リンパ球の単細胞懸濁液を、Sp2/0非分泌性マウス骨髄腫細胞(ATCC、CRL 1581)と融合できる。細胞を、およそ2×105で平底マイクロタイタープレートにプレーティングし、続いて、10%胎児クローン血清、18%「653」コンディショニング培地、5%オリゲン(origen)(IGEN)、4mM L-グルタミン、1mM ピルビン酸ナトリウム、5mM HEPES、0.055mM 2-メルカプトエタノール、50ユニット/mlペニシリン、50mg/mlストレプトマイシン、50mg/mlゲンタマイシンおよび1X HAT(Sigma)を含有する選択培地で2週間インキュベートする。およそ2週間後、細胞をHATがHTと置換されている培地で培養できる。次いで、個々のウェルを、ヒトモノクローナルIgMおよびIgG抗体についてELISAによってスクリーニングできる。広範なハイブリドーマ成長が起こると、10~14日後に普通に培地を観察できる。抗体を分泌するハイブリドーマを再プレーティングし、再度スクリーニングでき、ヒトIgGについて依然として陽性である場合に、制限希釈によってモノクローナル抗体を少なくとも2回サブクローニングできる。次いで、安定なサブクローンをインビトロで培養して、特性決定のために組織培養培地において少量の抗体を作製できる。
TL1Aに対するモノクローナル抗体を産生するトランスフェクトーマの作製
配列が提供される特異的抗体およびその他の関連抗TL1A抗体の両方を含めた、本発明の抗体を、当技術分野で周知であるように、例えば、組換えDNA技術および遺伝子トランスフェクション法の組合せを使用して宿主細胞トランスフェクトーマにおいて産生できる(Morrison,S.(1985)Science 229:1202)。
本明細書に記載される抗体は、TL1Aとの結合について、例えば、標準ELISAによって試験できる。手短には、マイクロタイタープレートを精製されたTL1Aを、PBS中1~2μg/mLで用いてコーティングし、次いで、PBS中5%ウシ血清アルブミンを用いてブロッキングする。各ウェルに抗体の希釈物(例えば、TL1A免疫処置マウスから得た血漿の希釈物)を添加し、37℃で1~2時間インキュベートする。プレートをPBS/Tweenを用いて洗浄し、次いで、西洋ワサビペルオキシダーゼ(HRP)とコンジュゲートしている二次試薬(例えば、ヒト抗体、ヤギ抗ヒトIgG Fc特異的ポリクローナル試薬)とともに37℃で1時間インキュベートする。洗浄した後、プレートをABTS基質(Moss Inc、product:ABTS-1000)を用いて発色させ、OD415~495で分光光度計によって分析する。次いで、免疫処置されたマウスから得た血清を、TL1Aを発現しないコントロール細胞株とではなく、ヒトTL1Aを発現する細胞株との結合についてフローサイトメトリーによってさらにスクリーニングする。手短には、抗TL1A抗体の結合を、TL1A発現性CHO細胞を1:20希釈の抗TL1A抗体とともにインキュベートすることによって評価する。細胞を洗浄し、結合を、PE標識された抗ヒトIgG Abを用いて検出する。FACScanフローサイトメトリー(Becton Dickinson,San Jose,CA)を使用してフローサイトメトリー分析を実施する。最高の力価を発生するマウスが、融合に使用されることが好ましい。
容される担体と一緒に製剤化された、本明細書に記載される抗TL1A抗体またはその抗原結合断片(単数または複数)の1つまたは組み合わせを含有する組成物、例えば、医薬組成物がさらに提供される。
本明細書に記載される抗体、抗体組成物および方法は、例えば、TL1Aシグナル伝達をブロックすることによる免疫応答の抑制またはTL1Aの検出に絡む多数のインビトロおよびインビボ有用性を有する。好ましい実施形態では、本明細書に記載される抗体は、ヒト抗体である。例えば、本明細書に記載される抗TL1A抗体は、インビトロもしくはエキソビボで培養細胞に、または種々の疾患において免疫性を抑制するために、例えば、インビボでヒト対象に投与できる。したがって、対象において免疫応答が改変されるように、対象に、本明細書に記載される抗体またはその抗原結合断片を投与することを含む、対象において免疫応答を改変する方法が、本明細書において提供される。好ましくは、応答は、阻害され、抑制され、または減少する。
TL1A免疫処置および融合
KM免疫処置:
TL1Aへの完全ヒトモノクローナル抗体を生成するために、KMマウスTMを精製された組み換えTL1A抗原(R&D systems 1319-TL/CF)で免疫処置した(表1)。
KMマウスTMから単離されたリンパ球を電気融合によりマウス骨髄腫細胞株と融合した。電気融合は、CytopluseTM融合キュベット中の電極間にリンパ球および骨髄腫細胞を整列させるよう電流を使用し、かつ細胞膜を横切る電位を短時間上昇させることにより達成した。電流の短いパルスは、隣接する細胞間の孔を開く膜を不安定化させる。このプロセスの間に、隣接する細胞の膜は融合し、ハイブリッド骨髄腫:リンパ球(hydbridoma)細胞をもたらす。
本開示の抗体は、例えば、標準ELISAによりTL1Aへの結合について試験され得る。簡潔には、マイクロタイタープレートはPBS中で1.0μg/mlの精製されたTL1Aでコーティングされ、PBS/tween中で1%ウシ血清アルブミンでブロックされる。抗体の希釈液(例えば、TL1A-免疫処置マウスからの血漿、または細胞培養上清の希釈液)は、各ウェルに加えられ、周囲温度で1-2時間インキュベートした。プレートをPBS/Tweenで洗浄し、次いで西洋ワサビペルオキシダーゼとコンジュゲートした二次試薬(例えば、ヒト抗体、ヤギ抗ヒトIgG Fc特異的ポリクローナル試薬)と周囲温度で1時間インキュベートした。洗浄後、プレートをABTS(2,2’-アジノ-ビス-(3-エチルベンズチアゾリン(ethylbenzthiazoline)-6-スルホン酸)基質(Moss Substrates、1.46mmol/L)で発色し、OD405で解析した。
抗TL1A抗体精製
ハイブリドーマからの元の抗TL1A抗体、1490-2596-10A4.F7.2E8は、CHO細胞中で組み換えタンパク質として発現され、TL1A.2と呼ばれる。このタンパク質は、G4P Fcバージョン並びにG1.1 f Fcバージョンとして発現される。CHO細胞から得られる上清はMabSelectSureプロテインAカラムで精製した。簡潔には、CHO上清をPBS(リン酸緩衝液生理食塩水)、pH7.4で予め平衡化したプロテインAカラムにかけた。サンプルローディング後にPBSで広範にカラムを洗浄し、その後0.1Mクエン酸緩衝液、pH3.0を使用して結合タンパク質を溶出した。溶出したタンパク質を、適切な量の1M Tris緩衝液の添加により直ちに中性pHにした。次いで、試料を広範な透析によりPBSへ緩衝液を交換した。
ベクター構築および発現
10A4 VKを、鋳型としてVKクローンMP.1_06132012-A06を利用するPCRにより増幅し、かつオステオネクチンシグナル配列およびヒトカッパ定常領域を含有するベクターpICOFSCneoKにクローン化し、プラスミドpICOFSCneoK(TL1A.10A4)を生成した。
親和性測定についてのプロトコール
プロテインGチップ(CM5)を、それを酢酸緩衝液(pH2.9)を使用して~400Rusでコーティングすることにより作製した。10A4.F7 mab(7.5ug/mL、12uL)をプロテインG表面上で10uL/分の流速で捕捉した。複数の濃度(250、200、150、100&50n)でのHu-TL1A-His抗原(Lot#50182AS151)を、捕捉されたmab上に25μl/分で5分間流し、かつ、7.5分間解離させた。プロテインG表面を100uL/分の流速で10uLの50mM NaOHおよび5uLの25mM HCLで再生した。データはBiaevaluation 3.1を使用することにより解析した。(図3)
エピトープ結合についてのプロトコール
10A4.A7(4.6KRUs)、17H11.C2(6.3KRUs)および10A6.B6(9.6KRUs)をCM5チップ上でコーティングした。Mabを30ug/mLから滴定し(1:3)、~2時間25n Hu-TL1A-His抗原でインキュベートした。mab上に注入された抗体-抗原複合体が2.5分間表面を覆った。表面を25mM NaOHで再生した。プロットをlog[Ab]対応答を使用して生成し、ここで、抗体-抗原複合体シグナルの減少は、同一のエピトープビンを示す。(図4)

可変領域シーケンシング
総RNAをハイブリドーマクローン1490.2596.10A4.F7.2E8から調製し、かつVHおよびVK cDNAを2通り調製した。抗体の可変領域を、5’RACEユニバーサルプライマーミックスと対の3’ヒト-特異的な定常領域プライマーを使用してcDNA末端の迅速増幅(rapid amplification of cDNA ends)(RACE)手法により増幅した。可変領域を含有する得られたPCR産物をpCR4-TOPO ベクターにクローニングした。Templiphi試料をTOPOクローンから調製し、かつDNAシーケンシングに供した。得られたDNA配列をインフレームの再編成および他の抗体特性について解析した。クローンMP.1_06132012-E02からのVH配列(図7および8)およびクローンMP.1_06132012-A06からのVK配列(図6および9)を代表的な配列として選択した。
HDX-MSによるTL1A/10A4エピトープマッピング
水素/重水素交換質量分析(HDX-MS)方法は、主鎖アミド水素原子の重水素交換の速度および程度をモニターすることによって溶液でのタンパク質立体配座および立体配座動態を調べる。HDXのレベルは、主鎖アミド水素原子の溶媒接触性およびタンパク質水素結合に依存する。HDXのタンパク質の質量増加は、MSによって正確に測定することができる。この技術を酵素消化と組み合わせると、ペプチドレベルでの構造特徴を解明することができ、内部に折りたたまれたペプチドと表面露出ペプチドを区別できる。典型的には、重水素標識およびその後のクエンチング実験が行われ、その後にオンラインペプシン消化、ペプチド分離およびMS解析が行われる。
コンピュータモデリングによるTL1A/10A4エピトープマッピング
TL1A三量体の構造を用いて、コンピュータ解析を行った。TNF様リガンド1A(TL1A)は、そのコグネイト受容体DR3およびデコイ受容体DcR3に結合する。TL1Aは、従来のTNFリガンドファミリーに属し、現在は、8つの他のメンバー:FasL、LIGHT、TNFα、LTα、LTβ、TRAIL、RANKLおよびCD40Lを含む。
T細胞増殖アッセイ
1μg/ml-0.219μg/mlのTL1A抗体10A4.F7.2E8 huIgG4の用量応答曲線は、StemCell Kit(19052)富化CD4+ヒトT細胞を加える前に37℃で30分間、5% CO2で抗ヒトCD3(Biolegend 300314)とインキュベートした。37℃、5% CO2でのインキュベーションの4日の後、3H-チミジン(0.5μCi/ウェル)を最後の18-20時間添加した。プレートをPackard Filtermate harvesterを使用してUnifilter GF/Cプレート(Packard 5007185)上に回収し、乾燥させた。50μl/ウェル PerkinElmer Microscint-20 scintillantを加え、プレートをPackard TopCountシンチレーションカウンターで計数した。EC50値を、GraphPad Prismソフトウェアを使用して最大マイナスバックグラウンドのパーセントをプロットすることにより計算した。
ヒトPBMC IFNg阻害アッセイ(可溶性のTL1A stim)
3ug/ml-0.3ng/mlのTL1A抗体10A4.F7.2E8 huIgG4の用量応答曲線を、ヒト全血から単離されたPBMCを有する96ウェル丸底プレート中で50ng/mlヒトTL1A(社内)プラス0.25ng/ml hIL-12(Peprotech)および1ng/ml hIL-18(R&D Systems)とインキュベートした。37℃、5% CO2で一晩のインキュベートした後、上清を回収し、IFNgレベルをマッチペアサンドイッチELISA抗体(Thermo Scientific)で試験した。EC50値を、GraphPad Prismソフトウェアを使用して最大マイナスバックグラウンドのパーセントをプロットすることにより計算した。
ヒトPBMC IFNg阻害アッセイ(TL1A発現CHO細胞 stim)

カニクイザルPBMC IFNg阻害アッセイ
ヒトpNFkB阻害アッセイ
TL1A抗体10A4.F7.2E8 huIgG4の用量応答曲線を0.5ug/mlヒトTL1A(社内の)と37℃で15分間;5% CO2、96ウェルディープウェルプレートにおいてヘパリン処置ヒト全血でインキュベートした。刺激後、細胞を溶解/固定し、透過処理し、抗体の適切なパネルで染色した。測定はフローサイトメーターで行い、かつ解析はTreeStarのFlowJo解析ソフトウェアで行った。EC50値はGraphPad Prismソフトウェアを使用して計算した。
Claims (15)
- ヒトTL1A(TNF様リガンド1A)に結合し、かつ、
a)以下を含む重鎖可変ドメイン:
配列番号7の配列を含むCDRH1;
配列番号8の配列を含むCDRH2;および
配列番号9の配列を含むCDRH3;
ならびに
b)以下を含む軽鎖可変ドメイン:
配列番号12の配列を含むCDRL1;
配列番号13の配列を含むCDRL2;および
配列番号14の配列を含むCDRL3
を含む、
単離抗体またはその抗原結合断片。 - 021 TVVRQTPTQHFKNQF116(配列番号16)の1以上の残基または166EIRQAGRPNKPDSIT180(配列番号17)の1以上の残基を含むエピトープでTL1Aに結合する、請求項1に記載の単離抗体または抗原結合断片。
- 691 QAGR172 の1以上の残基および 131 KNQF116 の1以上の残基を含むエピトープでTL1Aに結合する、請求項2に記載の単離抗体または抗原結合断片。
- 配列169QAGR172および/または113KNQF116を含むエピトープでTL1Aに結合する、請求項2に記載の単離抗体または抗原結合断片。
- DR3へのヒトTL1Aの結合を阻害する、請求項1-4のいずれか一項に記載の単離抗体または抗原結合断片。
- ヒトおよびカニクイザルTL1Aの両方に結合する、請求項1-4のいずれか一項に記載の単離抗体または抗原結合断片。
- a)重鎖が、配列番号6の配列と少なくとも90%配列同一性を有する重鎖可変領域を含み;および
b)軽鎖が、配列番号11の配列と少なくとも90%配列同一性を有する軽鎖可変領域を含む、
1以上の重鎖および1以上の軽鎖を含む、請求項1-6のいずれか一項に記載の単離抗体または抗原結合断片。 - 抗体が:
(a)ヒトIgG4 Fcバリアント;
(b)ヒトIgG1またはヒトIgG3 Fcバリアント;または

- 請求項11に記載の宿主細胞を、抗体またはその抗原結合断片の産生を可能にする条件下で培養すること、および細胞から抗体またはその抗原結合断片を精製することを含む、抗TL1A抗体またはその抗原結合断片の製造方法。
- 請求項1-8のいずれか一項に記載の単離抗体または抗原結合断片および医薬上許容される担体を含む、医薬組成物。
- 炎症性または免疫疾患の治療のための、請求項13に記載の医薬組成物。
- 炎症性または免疫疾患が、アレルギー/喘息、関節リウマチ、多発性硬化症、クローン病、炎症性腸疾患、全身性エリテマトーデス(SLE)、乾癬、1型糖尿病および移植片拒絶からなる群から選択される、請求項14に記載の医薬組成物。
