本明細書に記載される二重特異性抗体の調製のために、当技術分野において公知である任意の好適な方法(例えば、標準的な組換え技術)を用いることができる。以下に例を提示する。
本明細書に記載される二重特異性抗体またはコード核酸もしくは核酸のセットは、診断および治療を目的として使用できる。それらはまた、基礎研究および治療研究において研究ツールとしても使用できる。
本明細書に記載されるような二重特異性抗体(またはコード核酸もしくは核酸のセット)を医薬的に許容される担体(賦形剤)(緩衝剤を含む)と混合して、標的疾患の治療に使用するための医薬組成物を作製できる。「許容される」は、担体が組成物の有効成分に適合する(および好ましくは有効成分を安定化することが可能である)必要があり且つ治療される対象に有害でない必要があるということを意味する。医薬的に許容される賦形剤(担体)(緩衝剤を含む)は、当技術分野において周知である。例えば、Remington:The Science and Practice of Pharmacy 20th Ed.(2000) Lippincott Williams and Wilkins, Ed. K.E.Hooverを参照のこと。
本明細書に記載される二重特異性抗体(またはコード核酸もしくは核酸のセット)は、二重特異性抗体が結合する抗原の一方または両方に関連する疾患または障害の治療に有用である。例えば、二重特異性抗体がpKalおよびFXIIaに結合してpKalおよびFXIIaの活性を遮断することが可能である場合、その二重特異性抗体は、接触活性化系の調節不全に関連する疾患(例えば、HAEおよび血栓症)の治療に使用できる。
本開示はまた、標的の疾患または障害の軽減に使用するためのキットも提供する。このようなキットは、本明細書に記載される二重特異性抗体のうちの1種以上および/または1種以上の単離された核酸または核酸のセットを含む、1つ以上の容器を含んでよい。

親抗体としてDX-2930と抗FXIIaクローン559C-M0071-F06、559C-M0184-B04、559C-M0179-D04、559C-M0181-C02および559C-M0180-G03のうちの1つとを用いて、クローンX0120-A01、X0120-C01、X0120-E01、X0120-G01、X0121-A03、X0121-C01、X0121-E01、X0121-G01、X0122-A01およびX0122-C01を含む、いくつかの例示的な抗pKal/抗FXIIa二重特異性抗体を構築した。以下の表2を参照のこと。
抗pKal/抗FXIIa二重特異性抗体の別の例示的なセットを以下のように構築した。分子のIgG部分は、実施例1で使用されたものと同じ(すなわち、DX-2930)であった。抗FXIIa成分については、36種の単離物を選択し、軽鎖/重鎖の方向および重鎖/軽鎖の方向の両方でscFvに変換した。SGGGS(配列番号22)リンカーを用いてscFvをDX-2930 IgGに融合した。scFvを構築する際、(G4S)4リンカーを用いて抗FXIIaの可変重鎖ドメインおよび可変軽鎖ドメインを互いに融合した。二重特異性抗体の配列を以下に提示する。
これらの凝集体に対抗するために、VHの残基44(C44)とVLの残基100(C100)との間のジスルフィド結合を4種のクローン620I-X0177-A01(620I-X0173-A11)、620I-X0177-C01(620I-X0173-C07)、620I-X0177-E01(620I-X0173-E07)および620I-X0177-G01(620I-X0173-G11)のscFv領域内に設計した。ジスルフィドを有するscFvを含む二重特異性体のSEC分析は、高分子量のピークの劇的な減少を示し、その範囲は1~2%に低下した(図4)。この凝集の減少は、試験された全ての濃度にわたって当てはまった。これらの二重特異性クローンのビアコアは、pKalおよびFXIIaに対する強い特異的結合を示した(図9)。これらの単離物の血漿阻害は0.5~8nMの範囲にわたっていた(図10)。
例示的な二重特異性抗体(620I-X0177-A01、620I-X0177-C01、620I-X0177-E01、620I-X0177-G01)を、生産および製造可能性について評価した。分析の前に様々なpHにて48時間、室温で二重特異性抗体のサンプルをインキュベートした。次に、サンプルを、例えば図15で二重特異性抗体620I-X0177-A01について示されるように、SDS-PAGEタンパク質ゲル上で分離するか、またはサイズ排除クロマトグラフィーによって分析した(図16A~図16C)。還元条件下では、30kDaおよび50kDaのバンドのpH依存的増加ならびに80kDaのバンドの減少が観察された(図15、レーン2~6)。これらの予想外のバンドの出現は、二重特異性抗体が、予期せぬタンパク質分解的切断を受けているということを示した。非還元条件下における同じ30kDaの種の出現は、切断された種が単量体であるということを示した(図15、レーン6~8)。SEC分析によって、正しく形成された二重特異性抗体を表すピークが15.7~16.1分に、DX-2930を表すピークが17分に、切断された単鎖抗体を表すピークが22分に観察された(図16)。
本明細書で開示される特徴は全て、任意の組み合わせで組み合わされてよい。本明細書で開示される特徴はそれぞれ、同じ目的、同等の目的または類似の目的を果たす代替的特徴で置き換えられてよい。従って、特に明記しない限り、開示される特徴はそれぞれ、同等または類似の特徴の一般的な連なりの一例に過ぎない。

前記第2の抗体は、活性型第XII因子(FXIIa)に結合し、前記第2の抗体のV H は、配列番号132に記載される相補性決定領域(CDR)1、配列番号133に記載されるCDR2および配列番号134に記載されるCDR3を含み、前記第2の抗体のV L は、配列番号34に記載されるCDR1、配列番号36に記載されるCDR2および配列番号37に記載されるCDR3を含む、

- 前記第2の抗体においてV H がV L のN末端に融合されている、請求項1~8のいずれかに記載の二重特異性抗体。
- 前記第2の抗体が、V H 領域とV L 領域との間にペプチドリンカーを含み、前記ペプチドリンカーが(G 4 S) 4 である、請求項9に記載の二重特異性抗体。
- 前記第2の抗体のV H 鎖のC 44 とV L 鎖のC 100 との間にジスルフィド結合が形成されている、請求項1~10のいずれかに記載の二重特異性抗体。
- 前記第1の抗体のV H が、配列番号1のアミノ酸配列を含み、
前記第1の抗体のV L が、配列番号2のアミノ酸配列を含み、
前記第2の抗体のV H が、配列番号123のアミノ酸配列を含み、
前記第2の抗体のV L が、配列番号127のアミノ酸配列を含む、

- 前記第2の抗体のV H が、配列番号123のアミノ酸配列を含む、および/または
前記第2の抗体のV L が、配列番号127のアミノ酸配列の残基1~111を含む、
請求項12~14のいずれか1項に記載の二重特異性抗体。 - 前記第1のポリペプチドが、配列番号10のアミノ酸配列を含み、
前記第2のポリペプチドが、配列番号141、142、151および152からなる群より選択されるアミノ酸配列を含む、
請求項1に記載の二重特異性抗体。 - 血漿カリクレイン(pKal)に結合する第1の抗体と第XIIa因子に結合する第2の抗体とを含む、二重特異性抗体であって、
前記第1の抗体は、配列番号1および配列番号2と同じ重鎖と軽鎖の相補性決定領域を含み、
前記第2の抗体は、配列番号132に記載される相補性決定領域(CDR)1、配列番号133に記載されるCDR2および配列番号134に記載されるCDR3を含む重鎖可変領域(V H )と、配列番号34に記載されるCDR1、配列番号36に記載されるCDR2および配列番号37に記載されるCDR3を含む軽鎖可変領域(V L )と、を含む、
二重特異性抗体。 - 前記第2の抗体のV H が、配列番号123のアミノ酸配列を含む、および/または
前記第2の抗体のV L が、配列番号127のアミノ酸配列を含む、
請求項17に記載の二重特異性抗体。 - 請求項1~18のいずれかに記載される第1のポリペプチドをコードする第1のヌクレオチド配列と請求項1~18のいずれかに記載される第2のポリペプチドをコードする第2のヌクレオチド配列とを含む、単離された核酸または核酸のセットであって、
前記第1のヌクレオチド配列および前記第2のヌクレオチド配列は、2つの別個の核酸分子上に位置するか、または前記第1のヌクレオチド配列および前記第2のヌクレオチド配列は、1つの核酸分子上に位置する、
核酸または核酸のセット。 - 請求項19の核酸または核酸のセットを含む、ベクターまたはベクターのセット。

(ii)医薬的に許容される担体、
を含む医薬組成物。 - 接触活性化系に関連する疾患の治療における使用のための医薬組成物であって、
(i)請求項1~18のいずれか1項に記載の二重特異性抗体、請求項19の核酸または核酸のセット、あるいは請求項20のベクターまたはベクターのセット、および
(ii)医薬的に許容される担体、
を含む医薬組成物。 - 前記疾患が遺伝性血管性浮腫または血栓症である、請求項24に記載の医薬組成物。
