A.VHライブラリ構成

HCDR3シャッフリングされたVHドメインライブラリは次にmRNAライブラリに転写され、WO2010/011944に記載のようにdsDNAディスプレイ技術を用いた選択を受けた。選択は、4ラウンドにわたる交互のラウンドでヒトPDGFRβおよびマウスPDGFRβを用いて行なわれた。速度論的に制御されたオンおよびオフレート選択が連続ラウンドで適用されて、選択の厳しさを増大させ、およびしたがってPDGFRβについて高い親和性を有するVHドメインを選択した。具体的に、選択は以下のように行なわれた。ラウンド1(R1)は、10nMの固定されたヒトPDGFRβを用い、R2は、固定された100nMのマウスPDGFRβを用い、R3は、10nMの可溶性ヒトPDGFRβを用い、200nMの固定されたヒトPDGFRβと24時間および120時間競合され、R4は、10nMのマウスPDGFRβを用いた。R4結合プールは、DNA配列化のためにサブクローニングされた。R4結合プールの配列の分析は、XB1511のHCDR3がさまざまな異なるフレームワーク文脈の中に存在することを示した。分析された配列の組からは野生型親配列は得られなかった。選択されたVHドメインのアミノ酸配列を本明細書中では表3に記載する。

この実施例で述べる方法を用いて生成されたVκおよびVλ DNAライブラリは、T7Megascriptキット(Invitrogen、Cat#AM1334)を用いてmRNAライブラリに転写された。mRNAは、キットが提供する実験計画案に従って、RNeasy MinElute Cleanup Kit(Qiagen、Cat#74204)を用いて精製された。合計600pmolの
RNA(300pmolのVκおよびVλライブラリ)が、WO2010/011944に記載のようにdsDNAディスプレイリンカーおよび成分と結合され、アセンブルされた。アセンブルされたVLライブラリは、各々のVLドメイン(表現型)が安定してそのコーディング配列(遺伝子型)に融合される融合ライブラリを作り出すためにインビトロ翻訳された。35S Metを翻訳プロセスに組入れて、融合を放射性同位元素で識別した。次に、ライブラリはオリゴdTセルロースを用いて精製されて、後にフラグタグ精製を伴う逆転写、RNアーゼH消化、第2鎖DNA合成の標準的な分子生物学技術を用いてdsDNAディスプレイライブラリに変換された。
XB1511 VHドメインは、(翻訳反応への35S Metの組入れにより)遊離タンパク質として翻訳され、c末端フラグタグを通してアフィニティ精製された。XB1511 VHドメインおよび(以上のように調製された)精製されたVLドメイン融合ライブラリは次に等モル比で混合され、25Cで1晩インキュベーションされて、それらの疎水性パッチを通してVHおよびVL融合ドメインのインビトロ会合を可能にした。次に混合物は、エポキシ450ビーズ上または溶液の中で予め固定され、かつタンパク質Aビーズによって捕らえられたPDGFRβ標的と接するようにされた。固定されたPDGFRβ標的に結合した複合体は0.1NのKOHで洗浄され、溶出された。PCRは、VL特異的プライマーセットを用いて行なわれて、両者ともVH−VL対としておよび不対VLドメインとしてPDGFRβ標的に結合したVLを回収した。VLペアリングは、初めの2ラウンドについては厳しさを低くして(100nMのPDGFRβ)、3ラウンド目については厳しさを高くして(10nMのPDGFRβ)、3ラウンド行なった。XB2202 VHドメインも、2ラウンド同様にVLライブラリとペアリングされた。XB2202/VLペアリングおよび選択の各ラウンド毎に、厳しさは、速度論的に制御されたオンおよびオフレート戦略によって増大されて、安定してXB2202 VHドメインとペアリングされたVLドメインを同定してVH結合を向上させた。
にクローニングされ、結果的に得られる細菌コロニーから、VLエンコーディングDNA配列が、M13フォワードおよびリバースプライマーを用いてPCRによって増幅された。個々の増幅されたVLエンコーディングDNA配列を次に配列化した。VLプールから得られた配列データは、VLの多様なレパートリがプロセスを通して濃縮されることを示した。複数のファミリーおよびフレームワークがプールの中に存在した。いくつかのVLは複製またはファミリーとして存在した。区別されるVLファミリーを同定することがで
き、いくつかのVLは何度も存在した。PDGFRβ結合VHドメインXB1511およびXB2202と対になる、本発明の方法を用いて同定された例示的なVL配列を本明細書中の表4に記載する。
同定されたVH−VL対の特性を評価するため、各々のプールからの10−12個のscFvが、インビトロ翻訳によってまたは大腸菌発現によって構築され、産生され、その後アフィニティ精製された。

定hPDGFRβに結合した(35S Metで標識付けされた)scFvの量は、シンチレーションカウンタを用いて数えられ、Graph Pad Prism 5を用いてKdが算出された。
試験されたXB1511由来scFvについては、XB1511 VH単独(図8)と比較して、2つのscFvが、8−10倍、より高いKdを示し、1つが、2.5倍、より高いKdを示し、4つが同様のKdを示した。1つのscFvのみが、XB1511 VH単独よりも低いKDを示した。図9に示されるように、XB2202 VH単独と比較
して、試験されたXB2202由来scFvの両者ともが、約8−10倍、より良好なKdを示した。
実施例2で選択されたR4フレームワークシャッフリングされたヒトPDGFRβおよびマウスPDGFRβ濃縮VHドメインプールは、大腸菌発現ベクターにクローニングされ、産生され、かつ精製された。ヒトPDGFRおよびマウスPDGFRへのVHドメインの結合キネティクスは、BiacoreT100での表面プラズモン共鳴を用いて測定された
。簡単に述べると、ヒトおよびマウスPDGFR−hIgG1−Fcキメラ融合タンパク質は、抗hIgG1 Fcモノクローナル抗体に結合されたSeries CM5センサチップ(C
M5)を用いて別個に固定された。各サイクル毎に、PDGFR融合タンパク質がまず捕らえられ、その後に、流量100uL/分で115秒間、VHを注射した(会合)。会合段階のすぐ後に、600秒の解離段階がある。表面は、3MのMgCl2(10uL/分、60秒)の単回注射で各サイクルに再生された。VHドメインの複数の濃縮物が注射され(0.55nM−40nM)、結果的に得られたセンサグラムがT100評価ソフトウェアで分析された。結合キネティクスは、1:1結合曲線当てはめを用いて測定された。ヒトPDGFRβおよびマウスPDGFRβに対するVHドメインクローンXB2202およびXB2708の結合キネティクスを、それぞれ図10、図11、および図12に示す。これらの結果は、XB2202およびXB2708が、親XB1511と比較して、50−150倍の親和性向上を有することを示す。具体的に、XB2202およびXB2708は、それぞれ249pMおよび93pMのKd、ならびにそれぞれ1.86×10-3および9.267×10-4のオフレート(Koff)を有する。XB2202およびXB2708の両者ともがヒトPDGFRβおよびマウスPDGFRβに結合した。それらは同じHCDR3を共有したが、XB2202は、VH1ファミリー生殖細胞系配列に由来し、XB2708は、VH3ファミリー生殖細胞系配列に由来したことに特に注目すべきである。
本明細書中に開示されるXB2202 VHドメインの、ヒトPDFGRbへのPDGFBBリガンドの結合に拮抗する能力は、BiacoreT100での表面プラズモン共鳴を用
いて評定された。簡単に述べると、ヒトPDGFR−hIgG1−Fcキメラ融合タンパク質は、抗hIgG1 Fcモノクローナル抗体と結合されたSeries CM5センサチップを用いて固定された。10nMのヒトPDGFBBは、予め捕らえられたヒトPDGFRβに注射されて、VHの不在下でPDGFRβに対する100%の結合応答ユニットを得た。各連続サイクル毎に、PDGFR融合タンパク質がまず捕らえられ、次に、VHドメインが次に120秒間注射された。未結合のVHドメインを洗浄した後、次に10nMのPDGFBBを120秒間注射した。表面は、3MのMgCl2(10uL/分、60秒)の単回注射によって各サイクルに再生された。VHの複数の濃縮物が注射され(0.46nM−60nM)、結果的に得られるセンサグラムがT100評価ソフトウェアで分析され、PDGFBB結合抑制が算出された。図13に示されるように、XB2202は、5nM未満のIC50でのヒトPDFGRbへのPDGFBB結合を抑制する。
本明細書中に開示されるXB2708 VHドメインの、インビトロPDGFBB誘導周細胞移動に拮抗する能力が測定された。一次ヒト網膜周細胞がCell Systems Corporation(Kirkland、WA)から得られ、CSC完全成長媒質を用いて、製造者の提案に従って培養された。ほぼ125個の細胞(2−5継代)がヒト血漿フィブロネクチン(PBS中5μg/ml)でコーティングされ、0.1%のBSAを含有する無血清媒質中のBSA(PBS中の1%)で阻害された384ウェルBIND(著作権)バイオセンサプレートの各々のウェルに播種された。細胞は付着を許され、次に1晩血清飢餓とされた。血清飢餓の後に続いて、細胞は、組織培養インキュベータ中で、ヒトPDGFRβ受容体に対するVHのさまざまな濃縮で1時間インキュベーションされた。移動は、無血清培質中5ng/mlの最終濃度に対するPDGF−BBの添加によって、抗体の予めのインキュベーションの終わりに刺激された。ウェルの画像は、5%のCO2および>75%の湿度で3
7℃で組織培養インキュベータ中で20時間、18分毎にBIND(著作権)スキャナを用いて取得された。集められたデータを、Matlabに基づく重心識別および追跡アルゴリズムを用いて分析して、10時間と16時間との間の細胞の速度を算出した。図14に記載される結果は、XB2708が、0.54nMのIC50でPDGF−BB誘導周細胞移動に拮抗することができることを示す。
XB1511 VHおよびD8 VLは、293T細胞中の異種四量体IgG中にともに発現された。細胞培養上澄みは48時間および96時間後に集められ、発現されたIgGは、タンパク質Aアガロースビーズを用いて精製された。IgGは、最適化を何ら行なわずに、8mg/Lで産生された。XB1511/D8 IgGの生物学的活性を評価するため、HFF−1ヒト包皮線維芽が384ウェルBINDバイオセンサに播種され、無血清培質中で1晩付着を許された。次に、繊維芽細胞は、5ng/mLまたは10ng/mLのPDGFBBリガンドで刺激され、100nMのXB1511/D8 IgGの存在または不在下で、18時間移動を許された。BINDスキャナ画像は15分毎に捕捉され、ソフトウェア分析ツールを用いて個々の細胞移動応答の軌跡長さを測定した。軌跡長さは、青(移動なし)から赤(最大移動)への「ヒートマップ」によって表わされる。図15に示されるように、XB1511/D8 IgGは、ヒト線維芽のPDGF−BB誘導移動を完全に阻害することができた。
XB2202VHおよびXB2202/A4 scFvの熱安定性を定めた。具体的に、1mg/mLのXB2202およびXB2202−A4が、12時間、4℃、37℃、60℃、および70℃でインキュベーションされ、インキュベーション後にPDGFRβ結合ELISAを行なってタンパク質の結合活性を試験した。図16に示されるように、XB2202 VHドメインは、60℃でのインキュベーション後にかなりのPDGFRβ結合活性を失い、70℃でのインキュベーション後に結合活性を完全に失った。XB2202のTmは約62℃であると測定された。これに対し、XB2202/A4 scFvは、70℃での12時間のインキュベーション後に完全に活性であり、このことは、XB2202 scFvのTmが70℃よりも高いことを示した。
XB1511/D8およびXB2202/A4 VH/VL対は、293T細胞中の完全長異種四量体IgG1抗体として別個に発現され、精製された。XB1511/D8およびXB2202/A4 IgG1抗体の重鎖および軽鎖のアミノ酸配列を本明細書中では表7に記載する。
Q XLカラム(GE Healthcare)を用いてイオン交換クロマトグラフィによってさらに精製された。精製された抗体は、pH7のPBS中に保存された。
とによって定められた。精製された抗体の純度および品質は、サイズ排除高性能液体クロマトグラフィ(SEC−HPLC)によって定められた。これらの実験の結果は、本明細書中では表8に記載されている。これらのデータは、XB1511/D8およびXB2202/A4 VH/VL対が完全長異種四量体IgG1抗体としてフォーマットされた場合に、結果的に得られる抗体が、それらが高レベルで発現され、容易に高純度に精製され、凝集をほとんど呈しないという点で製造性が高いことを示す。
XB1511/D8およびXB2202/A4 IgG1抗体の熱安定性は、蛍光に基づくアッセイを用いて測定された。具体的に、5mg/mlの精製されたXB1511/D8 IgG1、XB2202/A4 IgG1、またはヒトIgG1対照がサイプロ(登録商標)オレンジ染料(シグマ)と混合され、混合物の温度は、25℃から95℃へ1度の増分で上昇された。サイプロオレンジ染料は、温度が上昇してIgGがアンフォールドすると、IgGの中に混ざる。IgGとのサイプロオレンジ染料の会合によって発生する蛍光シグナルは、BioRad CFX96器具を用いてモニタされた。このアッセイでは、サイプロオレンジシグナルの負の回帰を用いて、各々のタンパク質毎にピーク融点(すなわちTm)を同定した。
ヒトPDFGRに対するXB2202 VHドメイン、XB2202/A4 scFv、およびXB2202/A4 IgG1の結合キネティクスは、BiacoreT100での表
面プラズモン共鳴を用いて測定された。簡単に述べると、組換えヒトPDGFR−hIgG1−Fcキメラ融合タンパク質(R&D、♯385−PR−100/CF)は、(VHおよびscFvアッセイについては)抗hIgG1 Fcモノクローナル抗体と、または(IgGアッセイについては)抗6His抗体と結合されたSeries CM5センサチップ上に固定された。XB2202 VHドメイン、XB2202/A4 scFv、およびXB2202/A4 IgG1が、異なる濃度(75、50、25、10、5、および1nM)で3分間、50または100ul/分で表面の上に流され、10分間解離するようにされた。データは、1:1モデルを用いてBiacoreT100分析ソフトウェアを用いて分析
された。正確な測定を可能にするように、大量輸送がチェックされ、回避された。すべてのデータは、Biacore標準実験計画案に従って、二重参照された。
本明細書中に開示される抗PDGFRβ抗体の、インビボでのPDGF誘導血管新生を抑制する能力は、(その全体が本明細書に引用により援用される)Nobuo et al. Am. J. Path, (2006) 168(6), 2036-2052に記載の成長中の網膜血管系モデル、角膜新血管新生モデル、および/または脈絡膜の新血管新生モデルを用いて評価される。これらのアッセイでは、抗体は、VHドメイン、scFv、および/または完全長IgGとしてマウスに投与される。
Claims (24)
- PDGFRβに特異的に結合する安定したVH/VL対を備え、
(A)前記VHドメインはSEQ ID NO:318のアミノ酸配列を備え、かつ、
前記VLドメインはSEQ ID NO:370、372、373、376、380、382、387、390、396、400、404、420、422、431および443からなる群より選択されるアミノ酸配列を備え、または
(B)前記VHドメインはSEQ ID NO:321のアミノ酸配列を備え、かつ、
前記VLドメインはSEQ ID NO:412、414、421、425、435、443および449からなる群より選択されるアミノ酸配列を備える、単離された結合ポリペプチド。 - PDGFRβの活性を抑制する、請求項1に記載の結合ポリペプチド。
- PDGFRβの活性を、PDGFRβへのPDGF結合に拮抗することによって抑制する、請求項1に記載の結合ポリペプチド。
