技術分野
本開示は、バイオ燃料生産のための生物工学アプローチに関し、特に、C1基質(メタンまたはメタノールなど)をバイオマスに変換し、次いで、バイオ燃料またはバイオプラスチックなどに変換するためのC1代謝微生物反応装置系の使用に関する。
化石燃料の枯渇、温室効果ガス生成量の増加、および気候変動に関する懸念の高まりに伴い、化石燃料の代替物としてのバイオ燃料(例えば、エタノール、バイオディーゼル)が産業的に注目されるようになった。しかし、今日までに生成されたバイオ燃料には、困難と懸念がある。第1世代のバイオ燃料は植物(例えば、デンプン;ショ糖;ならびにトウモロコシ油、ナタネ油、ダイズ油、ヤシ油、および他の植物油)に由来するが、これらの燃料用作物はヒトおよび動物が消費するために育てられる作物と競合する。利用可能な耕作地は、世界的規模の食物および燃料のニーズを満たすのに十分でない。したがって、第2の世代のバイオ燃料は、例えば、セルロースまたは藻類から生産されている。しかし、生産コスト高と共に技術的に生産が困難であることにより、第2世代のバイオ燃料を、これ以上費用効果を高くしたり利用しやすくしたりできていない。
1つの態様では、本開示は、(例えば、精製ユニット中で)C1代謝非光合成微生物由来の油組成物を精製して燃料を生産することによって燃料を作製する方法を提供する。さらに、本開示は、C1代謝非光合成微生物を主に含む培養物由来のバイオマスを油組成物に変換し、油組成物を燃料に精製することによる燃料を作製するための方法を開示する。さらに別の態様では、本開示は、油組成物がC1代謝非光合成微生物に由来する処理ユニットおよび油組成物を精製して燃料を生産するための精製ユニットを含むバイオリファイナリーを提供する。さらに別の態様では、本開示は、水素原子および炭素原子を含む分子を有し、水素原子および炭素原子が組成物の重量の少なくとも約50%〜約99%であり、組成物のδ13Cが約−35‰〜約−50‰、−45‰〜約−35‰、または約−50‰〜約−40‰、または約−45‰〜約−65‰、または約−60‰〜約−70‰、または約−30‰〜約−70‰の範囲である、油組成物またはバイオ燃料組成物を提供する。

本開示は、C1代謝微生物をバイオオイルの蓄積を最大にしたバイオマスを生成するように培養する、バイオ燃料およびバイオプラスチックを生成するための組成物、方法、およびシステムを提供する。例えば、規模を変更可能な商業的プロセスであるメタンからバイオ燃料への発酵プロセスを提供する。この新規のアプローチは、例えば、メチロトローフ細菌またはメタン資化細菌を、例えば、水素処理のためのエステル化したバイオディーゼルまたはアルカン燃料の形態またはバイオプラスチックのためのポリヒドロアルカノアート(PHA)の形態でバイオ燃料のためのバイオマスを生成するための新規の宿主系として使用することができる。さらに、目的の油組成物を、メチロトローフ細菌またはメタン資化細菌から得ることができる。何故なら、これらの生物は本明細書中に記載の条件下でかなりの量の膜脂質を蓄積することができ、さらに、これらの微生物は膜成分を大量に産生するからである。

本開示のC1代謝微生物は、天然であるか、適応させた株であるか(例えば、親株と比較して成長速度が改善され、総バイオマス収量が増加した株を選択するための発酵を実施する)、目的の脂質を産生するように(例えば、脂肪酸産生酵素、ホルムアルデヒド同化酵素、またはその組み合わせを発現するように遺伝子操作する)、成長速度を増加させるように、またはその両方のために組み換えによって改変することができる。一定の実施形態では、C1代謝微生物は、C1代謝非光合成微生物(藻類または植物など)ではない。
組換えタンパク質の発現は、その元の宿主の外部で困難であり得る。例えば、異なる細菌種間でコドン使用頻度の偏りの変動が認められている(Sharpら,Nucl.Acids.Res.33:1141,2005)。組換えタンパク質の過剰発現は、その未変性の宿主内でさえも困難であり得る。一定の実施形態では、本明細書中に記載の宿主に導入すべき核酸分子(例えば、チオエステラーゼ、fabD、accABCDをコードする核酸)を、宿主への導入前に確実にタンパク質発現を効率的にするか増強するためにコドン最適化に供することができる。コドン最適化は、非天然DNA分子によってコードされるポリペプチドを変更することなく宿主の典型的なコドン使用頻度を反映するための形質転換前の遺伝子または核酸のコード領域中のコドンの変更をいう。異種宿主中の最適な遺伝子発現のためのコドン最適化法は、以前に記載されている(例えば、Welchら,PLoS One 4:e7002,2009;Gustafssonら,Trends Biotechnol.22:346,2004;Wuら,Nucl.Acids Res.35:D76,2007;Villalobosら,BMC Bioinformatics 7:285,2006;米国特許出願公開第2011/0111413号および同第2008/0292918号(その方法の開示全体が本明細書中で参考として援用される)を参照のこと)。
本明細書中に記載の任意の組換えC1代謝微生物またはメタン資化性細菌を、新規または増強された活性(例えば、酵素活性)を有する宿主を得るために少なくとも1つの外因性核酸を含むように形質転換することができるか、当該分野で公知の任意の種々の方法を使用して内因性遺伝子機能を除去または実質的に低下させるように遺伝子改変することができる。
本明細書中に記載のように、本明細書中に記載の任意の組換えC1代謝微生物(例えば、メタン資化性細菌)を、組換えC1代謝微生物を作製するための親または基準の宿主細胞として使用することができる。一定の実施形態では、本開示は組換えC1代謝非光合成微生物を提供し、ここで、この微生物は脂肪酸生合成に関連する異種核酸配列を含み、発現異種核酸配列により、親または基準のC1代謝非光合成微生物と比較して組換えC1代謝微生物中で高レベルの脂肪酸を蓄積し、脂肪酸を過剰発現する。
種々の培養方法を、本明細書中に記載の微生物、細菌、および酵母のために使用することができる。例えば、C1代謝微生物(メタン資化細菌またはメチロトローフ細菌など)を、バッチ培養または連続培養法によって成長させることができる。一定の実施形態では、培養物を、制御培養ユニット(発酵槽、バイオリアクター、または中空繊維セルなど)で成長させる。一般に、対数期の細胞は、しばしば、いくつかのシステムにおいて目的の生成物または中間体の大量生産を担うのに対して、他のシステムでは静止期または指数期後に生産することができる。
背景として、安定同位体測定および物質収支アプローチは、メタンの総供給源および吸収源を評価するために広く使用されている(Whiticar and Faber,Org.Geochem.10:759,1986;Whiticar,Org.Geochem.16:531,1990を参照のこと)。酸化量を決定するために残留メタンのδ13C値を使用するために、メタンの微生物酸化に原因する同位体分留度を知ることが必要である。例えば、好気性メタン資化性菌は、特異的酵素であるメタンモノオキシゲナーゼ(methane monooxygenase(MMO))によってメタンを代謝することができる。メタン資化性菌は、メタンをメタノールに変換し、その後にホルムアルデヒドに変換する。ホルムアルデヒドを、CO2にさらに酸化して還元等価物(NADH)の形態で細胞にエネルギーを供給することができるか、光合成生物における炭素同化経路と直接的に類似するRuMPサイクルまたはセリンサイクル(Hanson and Hanson,Microbiol.Rev.60:439,1996)のいずれかによってバイオマスに組み込むことができる。より詳細には、タイプIメタン資化性菌は、バイオマス合成のためにRuMP経路を使用して完全にCH4に由来するバイオマスを生成するのに対して、タイプIIメタン資化性菌は、50〜70%のCH4由来の細胞炭素および30〜50%のCO2由来の細胞炭素を同化するセリン経路を使用する(Hanson and Hanson,1996)。炭素同位体組成の測定方法は、例えば、Templetonら(Geochim.Cosmochim.Acta 70:1739,2006)(その方法全体が本明細書中で参考として援用される)に示されている。バイオマス由来の油組成物の13C/12C安定炭素同位体比(「δ」値‰(δ13C)として示す)は、使用されるC1基質の供給源および純度に応じて変化し得る(例えば、図7を参照のこと)。
C1代謝微生物による脂質産生のための培養条件およびバイオリアクター条件
例示的な本開示のC1代謝微生物(メタン資化性菌、メチロトローフ菌、Clostridia)を、試験管、バイアル、ボトル、プレート、またはバイオリアクター(発酵)内で培養した。種々の微生物についての成長条件、培地、および炭素源を、本実施例中に記載する。
血清瓶のために、細菌を、ヒギンス最少硝酸塩培地(NSM;Cornishら,J.Gen.Microbiol.130:2565,1984;Parkら,Biotechnol.Bioeng.38:423,1991)またはMM−W1培地中にて30℃で培養した。上部空間の組成を、メタン:空気の体積比を1:1に調整した。ボトルを200〜250rpmの速度で震盪した。あるいは、培養物を、1:1(v/v)メタン:空気ガス混合物を含む気密チャンバー中またはメタノール蒸気の存在下(パラフィルム密封プレートの蓋内の0.5mLメタノールによる)で成長させた1.5%w/v寒天を含むNSM−培地プレート上または0.5%メタノールを補足したNSM−培地プレート上に維持した。プレートを逆さにして30℃の加湿チャンバー中でインキュベートした。

Clostridium細菌を、ブチルゴム栓および200kPaの鉄鋼所排ガスを有するプラスチックコーティングした500ml−Schott Duran(登録商標)GL45ボトル内の100mLの改変PETC培地(ATCC培地1754)中にて37℃で嫌気的に培養する。600nmでの光学密度(OD600)の測定によって成長をモニタリングする。
Clostridium autoethanogenumの発酵を、例えば、米国特許出願公開第2011/0300593号に記載の方法に類似の方法を使用して行う。簡潔に述べれば、1.3Lの溶液A(3.083gのNH4Ac;0.61gのMgCl2・6H2O;0.294gのCaCl2・2H2O;0.15gのKCl;0.12gのNaCl(任意);これらを蒸留水で1Lにする)を含む2リットルのバイオリアクターに、N2ガスを噴霧する。H3PO4の85%溶液(2.025mL、30mM)を添加し、濃NH4OH水溶液を使用してpH5.3に調整する。次いで、13.5mLの溶液B(20.0mgのビオチン;20.0mgの葉酸;10.0mgのピリドキシンHCl;50.0mgのチアミンHCl;50.0mgのリボフラビン;50.0mgのニコチン酸;50.0mgのD−(*)−パントテン酸カルシウム;50.0mgのビタミンB12;50.0mgのp−アミノ安息香酸;50.0mgのチオクト酸;これらを蒸留水で1Lにする)を添加し、溶液にN2ガスを噴霧する。溶液の酸化還元電位(ORP)がおよそ−200mVに低下するまで塩化クロム(II)を添加し、ここで、レサズリン(1.35mLの2g/L溶液)を添加する。多硫化ナトリウム(5.4mLの3M溶液、以下を参照のこと)を添加し、溶液にN2を噴霧し、次いでCO含有ガス(1%H2;13%N2;71%CO;15%CO2)を噴霧する。金属硫化物溶液(150mL、以下を参照のこと)を添加し、およそ5%(v/v)のレベルの活発に成長しているC.autoethanogenum培養物の播種前にこの溶液をさらに30分間噴霧する。
Clostridium ljungdahliiの発酵を、例えば、米国特許第5,173,429号および同第5,593,886号に記載の方法に類似の方法を使用して行う。簡潔に述べれば、バッチ発酵を、C.ljungdahliiの生物学的に純粋な培養物を使用して行う。培地((1)80.0mLのKH2PO4 3.00g/L、K2HPO4 3.00g/L、(NH4)2SO4 6.00g/L、NaCl 6.00g/L、MgSO4・2H2O 1.25g/Lを含む塩;(2)1.0gの酵母エキス;(3)1.0gのトリプチケース;(4)3.0mlのFeCl2・4H2O 1500mg、ZnSO4・7H2O 100mg、MnCl2・4H2O 30mg、H3BO3 300mg、CoCl2・6H2O 200mg、CuCl2・H2O 10mg、NiCl2・6H2O 20mg、NaMoO4・2H2O 30mg、Na2SeO3 10mgを含み、蒸留水で1LにしたPFN(Pfenning)微量金属溶液;(5)10.0mlの塩酸ピリドキサール10mg、リボフラビン50mg、塩酸チアミン50mg、ニコチン酸(Nictotinic acid)50mg、Ca−D−パントテイナート50mg、リポ酸60mg、p−アミノ安息香酸50mg、葉酸20mg、ビオチン20mg、シアノコバラミン50mgを含み、蒸留水で1LにしたビタミンB群;(6)0.5gのシステインHCl;(7)0.06gのCaCl2・2H2O;(8)2.0gのNaHCO3;(9)1.0mLのレサズリン(0.01%);および(10)920.0mLの蒸留水)の調製を、80%窒素および20%CO2の雰囲気下で嫌気的に行う。発酵中の培地のpHを制御し、HClで5.0に維持する。必要な場合、滅菌10%NaOH溶液または1.0%酢酸溶液を使用してpHを調整する。培地を157.5mL血清瓶に移し、ブチルゴム栓およびアルミニウムシールで密封する。次いで、ボトルを121℃で20分間オートクレーブする。
脂質産生の増強のために操作されたC1代謝微生物
宿主細胞を、内因性fadD遺伝子によってコードされる長鎖脂肪酸−CoAリガーゼ活性のノックアウトによって脂肪酸分解を最小にするか減少させるための遺伝子改変を有するように操作した。さらに、遊離脂肪酸(FFA)の生合成を、チオエステラーゼ(TE)遺伝子の本開示のメタン資化性菌(Methylococcus capsulatus)への導入によって増強した。かかる組換え変化を、本実施例にさらに記載する。
野生型FadDタンパク質をコードする核酸配列は、変異fadD遺伝子のデザインのための基準標準出発点であった。例えば、M.trichosporium OB3b、M.capsulatus Bath、M.methanica、M.extorquens、およびC.ljungdahliiによってコードされる野生型FadDタンパク質配列は、それぞれ、GenBank受入番号EFH00931.1、YP_114021.1、YP_004512148.1、YP_002964871.1、およびYP_003782065.1中に提供されている。それ故、上述のタンパク質をコードするfadD遺伝子の核酸分子を、M.trichosporium OB3b(配列番号1)、M.methanica(配列番号35)、M.extorquens(配列番号52)、およびC.ljungdahlii(配列番号85)由来の遺伝子の5’領域にいくつかの停止変異およびフレームシフトを組み込むように個別に合成した。M.capsulatus fadD遺伝子について、遺伝子の残存する5’末端および3’末端を元の読み枠(配列番号18)を維持するように連結することができるように内部欠失を含む核酸分子を合成した。
大腸菌由来のプラスミドのM.trichosporium OB3bまたはM.methanicaへの接合手順は、Martin and Murrellによって開発された方法(FEMS Microbiol.Lett.127:243,1995)に基づく一方で、大腸菌由来のプラスミドのM.capsulatusへの接合手順は、Ali and Murrellによって報告された方法(Micologiology 155:761,2009)に基づいた。
プラスミドのM.extorquensへの導入手順は、Uedaら、Appl.Environ.Microbiol.57:924、1991に記載の手順に基づく。簡潔に述べれば、野生型(wt)M.extorquensを、0.5%メタノールを補足したNSM培地中にて30℃で培養する。対数中期(1.4×109/ml)まで成長させたM.extorquens NR−2の細胞を6,000×gで10分間の遠心分離によって収集し、エレクトロポレーション緩衝液(10mMのTris−HCl、2mMのMgCl2・6H2O(H20)、10%[wt/vol]スクロース[pH7.5])で洗浄した。細胞を、7.0×1010/mlの細胞濃度で同一緩衝液中に再懸濁する。細胞懸濁液およびベクター(70μg/mL)をチューブ内で9:1(vol/vol)の比で混合し、次いで、10μLを、3分間平衡化したチャンバーの電極間の空間に移す。10パルスの10kV/cm電界に300μsec/パルス供した後、5μLのアリコートの混合物を清潔なチューブに移し、0.2mLのNSM培地を添加する。次いで、細胞懸濁液を30℃で2時間インキュベートして抗生物質耐性遺伝子を発現後、0.5μg/mLメタノールおよび20μg/mLカナマイシンを含むNSMプレート上にプレートする。
C.autoethanogenumまたはC.ljungdahliiの形質転換法を米国特許出願公開第2011/0236941号に記載のように行うかC.tyrobutyricumについては修正プロトコール(Zhuら、Biotechnol.Bioeng.90:154、2005)を使用して行う。簡潔に述べれば、コンピテント細胞を作製するために、本明細書中に記載の培養条件にしたがって50mLのC.autoethanogenum培養物を新鮮な培地に3日間連続して継代培養する。これらの細胞を使用して、40mMのDL−トレオニンを含む50mLのPETC培地にOD600値0.05で播種する。培養物がOD600値0.4に到達した時に細胞を嫌気性チャンバーに移し、4,700×gおよび4℃で収集する。培養物を氷冷エレクトロポレーション緩衝液(270mMのスクロース、1mMのMgCl2、7mMのリン酸ナトリウム、pH7.4)で2回洗浄し、最終的に600μlの新鮮なエレクトロポレーション緩衝液に懸濁する。この混合物を、1μgのベクター(Clostridium複製起点を欠き、目的の核酸分子を含み、クラリスロマイシン耐性をコードする)を含む0.4cm電極ギャップを備えた予冷エレクトロポレーションキュベットに移し、Gene pulser Xcellエレクトロポレーションシステム(Bio−Rad)を以下の設定で使用して直ちにパルスを発生させる:2.5kV、600μl、および25μF。時定数3.7〜4.0msが達成される。培養物を5mlの新鮮な培地に移す。細胞の再生を、チューブホルダを備えたSpectronic Helios Epsilon分光光度計(Thermo)を使用して波長600nmでモニタリングする。バイオマスの初期の減少後、細胞は再度成長し始める。一旦バイオマスがその時点から倍加すると、細胞を収集し、200μlの新鮮な培地に懸濁し、4μg/μlクラリスロマイシンを有する選択PETCプレート(1.2%Bacto(商標)寒天(BD)を含有)上にプレートする。30psi鉄鋼所ガスとの37℃で4〜5日間のインキュベーション後、コロニーが明らかに認められる。
抗生物質選択に対する細胞の耐性によって形質転換を確認し、PCR法、ノーザンブロット法、ウェスタンブロット法、またはELISA法によって遺伝子発現を確認する。例えば、移入を検証するために、プラスミドDNAを単離し、標準的条件(95℃で5分間;95℃で30秒間、50℃で30秒間、および72℃で1分間を32サイクル;72℃で10分間)を使用したillustra PuReTaq Ready−To−Go(商標)PCR Beads(GE Healthcare)を使用したPCRに供することができる。さらなるコントロールとして、1μlの各単離プラスミドを大腸菌XL1−Blue MRF’Kan(Stratagene,La Jolla,CA)に再形質転換し、これからプラスミドを清潔に単離し、制限消化によって検証することができる。
メタン資化性菌について、1つ以上のチオエステラーゼ遺伝子をコードする遺伝子を含むかアセチル−CoAカルボキシラーゼ遺伝子を過剰発現するベクターで形質転換した野生型またはfadD−ノックアウトのM.trichosporium OB3b、M.methanica、M.extorquens、またはM.capsulatus Bathを使用して、20〜50mLのNSM培地および10μg/mLカナマイシンを含む100mLの血清瓶または培養管に播種する。M.extorquensについて、培地に炭素源として0.5%メタノールを補足するのに対して、M.trichosporium OB3b、M.methanica、およびM.capsulatus Bathについてはボトルの上部空間に炭素源として酸素およびメタンの1:1混合物をフラッシングする。ボトルをブチルゴム隔膜で密封し、圧着する。次いで、ボトルまたはチューブを、30℃(M.trichosporium OB3b、M.methanica、M.extorquens)または42〜45℃(M.capsulatus Bath)でのインキュベーション中に200〜250rpmの速度で連続して振盪する。
M.capsulatus Bathの脂肪酸プロフィールは、fadDのノックアウトならびに大腸菌チオエステラーゼ遺伝子の導入および発現によって変化した。第1に、周辺質ターゲティング配列が除去された大腸菌チオエステラーゼ遺伝子(TesA’)を、異なる発現レベルを有するバリアントを生成するためにデザインされた3つの異なるコドン組成(TesA’−3、配列番号102;TesA’−37、配列番号103;およびTesA’−20、配列番号104)を使用して合成した。TesA’バリアントを、IncPベースのプラスミド(Inc−P oriVおよびoriTを含む)にクローン化し、メタン資化性菌で機能するプロモーターに作動可能に連結した。TesA’を含む組換え発現ベクターを、本明細書中に記載のようにM.capsulatusに形質転換した。150mLの密封血清瓶中にて5mLのM.capsulatus培養物を、40mLメタンおよび80mL酸素と共に5日間成長させた。成長段階後、1mLの各培養物を、本明細書中に記載のように、GC/MSを使用して脂肪酸の濃度および組成についてアッセイした。遊離脂肪酸の測定値を、培養によるOD600に正規化した。C16:1という表記は少なくとも3つの異なる異性体から構成され、Δ9−シスパルミトレイン酸が最も豊富である(データ示さず)ことに留意のこと。
C1代謝微生物からの脂質抽出
収集した細菌バイオマス内に含まれる油組成物を、20℃(すなわち、室温)で1体積のメタノールを含む2体積のクロロホルムから作製された抽出溶液(CM溶液)中にて実施したFolch抽出プロトコール(Folchら,J.Biol.Chem.226:497,1957)の修正バージョンを使用して抽出した。約5gの湿細胞重量(WCW)のいずれかの新鮮な細菌バイオマス(または−80℃で保存し、その後に解凍した細菌バイオマス)を抽出のために使用した。100mLのCM溶液を細胞材料に添加し、混合物を分液漏斗中で強く抽出した。少なくとも10分後、3相に分離された。抽出脂質を含む有機相が分液漏斗の底部に沈殿し、これを清潔なガラスボトルに排出させた。中間層は主に溶解した細胞材料を含み、塩および他の可溶性細胞成分を含む軽質の水相から分離することができた。
C1代謝微生物由来の脂質の脂肪酸メチルエステル変換
乾燥固体の形態のM.capsulatus Bath、M.trichosporium OB3b、およびMethylomonas sp.16a培養バイオマスから抽出した脂質画分を個別に水酸化カリウム(KOH)を使用して加水分解し、単一工程でのメタノールとの反応によって脂肪酸メチルエステル(FAME)に変換した。10mLガラスボトル中の約5gの抽出固体脂質を、5mLのトルエン:メタノール(1:1 v/v)の0.2M KOH溶液を使用して溶解した。ボトルを強く振盪し、次いで、250rpmにて42℃で60分間混合後、溶液を周囲温度に冷却し、分液漏斗に移した。およそ5mLの蒸留水および5mLのCM溶液を分液漏斗に添加し、混合し、次いで、相を重力または5分間の遠心分離(3,000rpm、25℃)によって分離させた。溶解したグリセロールリン酸エステルを含む上部の水層を除去し、重い油相(底部)を回収し、回転蒸発または一定の窒素流によって濃縮乾固した。
C1代謝微生物由来の油組成物を使用したバイオ燃料産生
C1代謝微生物由来の抽出油組成物を、共同設置された精製所で処理するか、離れた精製所に輸送することができる。精製所を使用して、生体再生可能飼料由来のトリグリセリド(脂肪、グリース、およびメタン資化性菌油など)を脂質炭化水素燃料の混合物(主にバイオディーゼルおよびバイオジェット燃料、高品質の合成パラフィン系ケロシン(SPK))に変換する。この過程には、メタン改質を使用して施設内で生産することができるか、既存の精製所での発酵施設の共同設置によって供給される水素を必要とする。
C1代謝微生物由来の脂質中の安定炭素同位体分布
M.trichosporiumのバイオマスおよび脂質画分の乾燥サンプルを、IsoPrime100 IRMS(Isoprime,Cheadle,UK)を取り付けたCHNOS元素分析器(vario ISOTOPE cube,Elementar,Hanau,Germany)を使用した元素分析器/連続フロー型同位体比質量分析によって、炭素および窒素含有量(%乾燥重量)、ならびに炭素(13C)および窒素(15N)安定同位体比について分析した。発酵槽または血清瓶で培養したメタン資化性バイオマスのサンプルを遠心分離し、脱イオン水に再懸濁し、0.2〜2mgの炭素(約0.5〜5mg乾燥細胞重量)に対応する体積を5×9mmスズカプセル(Costech Analytical Technologies,Inc.,Valencia,CA)に移し、80℃で24時間乾燥させた。同様に、事前に抽出した脂質画分をクロロホルムに懸濁し、0.1〜1.5mgの炭素を含む体積をスズカプセルに移し、80℃で24時間蒸発乾固させた。0.1mgの炭素を含む標準によって信頼性の有るδ13C値を得た。
脂質における安定炭素同位体分布に及ぼすメタン供給源および純度の影響
天然ガス成分を含むメタンに対して成長させたメタン資化性菌を試験するために、100mL特定培地MMS1.0を含む一連の0.5リットル血清瓶に、メタンおよび空気の1:1(v/v)混合物を供給した同一培地中で成長させた血清瓶バッチ培養(5%v/v)由来のMethylosinus trichosporium OB3bまたはMethylococcus capsulatus Bathを播種した。MMS1.0培地の組成は以下であった:0.8mMのMgSO4・7H2O、30mMのNaNO3、0.14mMのCaCl2、1.2mMのNaHCO3、2.35mMのKH2PO4、3.4mMのK2HPO4、20.7μMのNa2MoO4・2H2O、6μMのCuSO4・5H2O、10μMのFeIII−Na−EDTA、および1mL/リットルの微量元素溶液(Lあたり以下を含む:500mgのFeSO4・7H2O、400mgのZnSO4・7H2O、20mgのMnCl2・7H2O、50mgのCoCl2・6H2O、10mgのNiCl2・6H2O、15mgのH3BO3、250mgのEDTA)。培地をオートクレーブし、冷却した後に、リン酸塩、重炭酸塩、およびFeIII−Na−EDTAを添加した。培地の最終pHは7.0±0.1であった。
Claims (16)
- 組換え偏性メタン資化細菌であって、前記組換え偏性メタン資化細菌が、チオエステラーゼをコードする異種ポリヌクレオチド、または、チオエステラーゼとマロニルCoA−アシルキャリアタンパク質トランスアシラーゼおよび/もしくはアセチル−CoAカルボキシラーゼとの組み合わせをコードする異種ポリヌクレオチドを含み、
前記異種ポリヌクレオチドの発現が、野生型偏性メタン資化細菌と比較した増大したレベルの脂肪酸の蓄積をもたらす、組換え偏性メタン資化細菌。 - 前記野生型偏性メタン資化細菌が、Methylomonas sp.16a、Methylosinus trichosporium OB3b、Methylosinus sporium、Methylocystis parvus、Methylomonas methanica、Methylomonas albus、Methylobacter capsulatus Y、Methylococcus capsulatus Bath、Methylomonas sp.AJ−3670、またはMethylomicrobium alcaliphilumである、請求項1に記載の組換え偏性メタン資化細菌。
- 前記チオエステラーゼが、前記組換え偏性メタン資化細菌にコドン最適化されている、請求項1または2に記載の組換え偏性メタン資化細菌。
- 前記チオエステラーゼをコードする前記異種ポリヌクレオチドが、周辺質ターゲティング配列を欠く大腸菌tesAである、請求項1〜3のいずれか1項に記載の組換え偏性メタン資化細菌。
- 前記マロニルCoA−アシルキャリアタンパク質トランスアシラーゼがコドン最適化した大腸菌fabDである、請求項1〜4のいずれか1項に記載の組換え偏性メタン資化細菌。
- 前記アセチル−CoAカルボキシラーゼがコドン最適化した大腸菌accA、accB、accC、accD、またはその任意の組み合わせである、請求項1〜5のいずれか1項に記載の組換え偏性メタン資化細菌。
- 前記組換え偏性メタン資化細菌が、脂肪酸−CoAリガーゼ活性を最小にするか排除する変異をさらに含む、請求項1〜6のいずれか1項に記載の組換え偏性メタン資化細菌。
- C1基質が気相で固相発酵中の微生物バイオフィルムに送達させる制御培養ユニットであって、前記微生物バイオフィルムが、請求項1〜7のいずれか1項に記載の組換え偏性メタン資化細菌を含む、制御培養ユニット。
- 前記C1基質が天然ガス、非従来型天然ガス、またはメタンから選択される、請求項8に記載の制御培養ユニット。
- 請求項1〜7のいずれか1項に記載の組換え偏性メタン資化細菌を含むバイオマスであって、前記組換え偏性メタン資化細菌が、炭素源としてC1基質を使用して培養される場合、δ13Cが約−30‰〜約−70‰の範囲にわたる、バイオマス。
- 前記バイオマスのδ13Cが約−40‰〜約−60‰の範囲である、請求項10に記載のバイオマス。
- 前記バイオマスが、(a)前記組換え偏性メタン資化細菌が成長した培養培地と共に前記組換え偏性メタン資化細菌の培養物を含むか、(b)成長し前記培養培地から回収された前記組換え偏性メタン資化細菌を含むか、または(c)前記組換え偏性メタン資化細菌の培養物由来の使用済み培地上清組成物を含む、請求項10または11に記載のバイオマス。
- 前記バイオマスが前記使用済み培地上清組成物を含み、油組成物を前記使用済み培地上清組成物から抽出するか濃縮する、請求項12に記載のバイオマス。
- 前記C1基質が天然ガス、メタン、またはメタノールを含む、請求項10〜13のいずれか1項に記載のバイオマス。
- 請求項10〜14のいずれか1項に記載のバイオマスを生成する方法であって、前記方法は、制御培養ユニット中で、C1基質を含む供給材料の存在下で、バイオマスを生成するのに十分な条件下および時間で、前記組換え偏性メタン資化細菌を培養する工程を含む、方法。
- 前記C1基質を含む前記供給材料が、天然ガス、メタン、またはメタノールを含む、請求項15に記載の方法。
