Description
本発明は、少なくとも1種のBacillus coagulans 高温菌、或いは、該少なくとも1種のBacillus coagulans 高温菌と、少なくとも1種のBacillus subtilis 中温菌との組み合わせを、家畜敷料に添加し、敷料を発酵させる工程を含む、家畜敷料の殺菌方法を提供する。
本発明はまた、少なくとも1種のBacillus coagulans 高温菌、或いは、該少なくとも1種のBacillus coagulans高温菌と、少なくとも1種のBacillus subtilis中温菌との組み合わせを、有機廃棄物又は家畜敷料に添加し、有機廃棄物又は敷料を発酵させる工程を含む、有機廃棄物又は敷料の堆肥化促進方法を提供する。
1.微生物の分離及び同定
日本で採取した敷料を、滅菌水で懸濁して希釈し、標準寒天培地あるいはCYC寒天培地(ATCC medium 2591)に塗抹して、50℃あるいは60℃にて培養し、コロニーを形成させた。単離したコロニーを純化し、16S rRNA遺伝子配列の相同性に基づき同定した。
分離・同定した菌株の培養菌体を滅菌水で懸濁して希釈し、アンモニウム塩を添加したTS培地 および高アンモニウム塩培地に滴下した。50℃、約12時間培養し、アンモニア塩を含むTS培地および高アンモニウム塩培地でのコロニーの大きさを基準にして微生物を選択した。このアンモニア耐性を有する微生物を選択する手法により、Bacillus licheniformis 群およびBacillus coagulans 群を選択した。
Bacillus subtilis C-3102株(FERM BP-1096)を1 x 105 cfu/g含む鶏の飼育に使用した敷料600 kgに分離したBacillus licheniformis群およびBacillus coagulans群を添加し、混合した。4-5日後に切返し、さらに4日間発酵させ、Bacillus licheniformis 群、もしくはBacillus coagulans 群の添加により発酵時の熱量が向上するかどうか観察した。熱量は敷料堆積物(図1)の3箇所(図2)の温度を測定し、式1にて算出した(表1)。

次に、Bacillus coagulans群を構成する各菌株が実際に敷料の温度上昇に寄与するかどうか検討した。Bacillus subtilis C-3102株(FERM BP-1096)を1x 105 cfu/g含む鶏の飼育に使用した敷料500 kgに、Bacillus coagulans 群の構成菌株であるBacillus coagulans CP3424株およびCP3425株を添加し、混合した。4日後に切返し、さらに4日間発酵させ、熱量が向上するかどうか観察した。熱量は敷料堆積物(図1)の3箇所(図2)の温度を測定し、式1にて算出した(表2)。

Bacillus subtilis C-3102株(FERM BP-1096)とBacillus coagulans CP3425株について、それぞれの芽胞化した菌体を水に懸濁した液体を脱脂大豆500gと混合した。この混合物を発泡スチロール容器に充填して培養し、その中心部の温度変化を3日間測定した。熱量は発泡スチロール容器の中心部の温度を測定して、式1にて算出した(表3)。
Claims (23)
- 少なくとも1種のBacillus coagulans高温菌、或いは、該少なくとも1種のBacillus coagulans高温菌と、少なくとも1種のBacillus subtilis中温菌との組み合わせを、家畜敷料に添加し、敷料を発酵させる工程を含む、但し前記少なくとも1種のBacillus coagulans高温菌がBacillus coagulans CP3425株(国際寄託番号NITE BP-01693)又は同等の殺菌作用を有するその変異株を含むことを特徴とする、家畜敷料の殺菌方法。
- 少なくとも1種のBacillus coagulans高温菌、或いは、該少なくとも1種のBacillus coagulans高温菌と、少なくとも1種のBacillus subtilis中温菌との組み合わせを、有機廃棄物又は家畜敷料に添加し、有機廃棄物又は敷料を発酵させる工程を含む、但し前記少なくとも1種のBacillus coagulans高温菌がBacillus coagulans CP3425株(国際寄託番号NITE BP-01693)又は同等の殺菌作用を有するその変異株を含むことを特徴とする、有機廃棄物又は家畜敷料の堆肥化促進方法。
- Bacillus coagulans高温菌が、少なくとも40〜60℃の温度で生育可能である請求項1又は2に記載の方法。
- Bacillus subtilis中温菌が、少なくとも30〜50℃の温度で生育可能である請求項1又は2に記載の方法。
- 微生物の発酵熱のみを利用することを特徴とする請求項1〜4いずれか1項に記載の方法。
- Bacillus coagulans高温菌がさらに、Bacillus coagulans CP3424株(国際寄託番号NITE BP-01692)、又は同等の殺菌作用を有するその変異株を含む、請求項1〜3及び5のいずれか1項に記載の方法。
- Bacillus subtilis中温菌が、Bacillus subtilis C-3102株(国際寄託番号FERM BP-1096)又はその変異株である、請求項1、2、4及び5のいずれか1項に記載の方法。
- Bacillus coagulans高温菌がBacillus coagulans CP3425株(国際寄託番号NITE BP-01693)であり、及び、Bacillus subtilis中温菌がBacillus subtilis C-3102株(国際寄託番号FERM BP-1096)である、請求項1〜5及び7のいずれか1項に記載の方法。
- Bacillus coagulans CP3424株(国際寄託番号NITE BP-01692)。
- Bacillus coagulans CP3425株(国際寄託番号NITE BP-01693)。
- 少なくとも1種のBacillus coagulans高温菌、或いは、該少なくとも1種のBacillus coagulans高温菌と、少なくとも1種のBacillus subtilis中温菌との組み合わせを、有効成分として含む、但し前記少なくとも1種のBacillus coagulans高温菌がBacillus coagulans CP3425株(国際寄託番号NITE BP-01693)又は同等の殺菌作用を有するその変異株を含むことを特徴とする、家畜敷料の殺菌剤。
- Bacillus coagulans高温菌が、少なくとも40〜60℃の温度で生育可能である、請求項11に記載の殺菌剤。
- Bacillus subtilis中温菌が、少なくとも30〜50℃の温度で生育可能である、請求項11に記載の殺菌剤。
- Bacillus coagulans高温菌がさらに、Bacillus coagulans CP3424株(国際寄託番号NITE BP-01692)、又は同等の殺菌作用を有するその変異株を含む、請求項11又は12に記載の殺菌剤。
- Bacillus subtilis中温菌が、Bacillus subtilis C-3102株(国際寄託番号FERM BP-1096)又はその変異株である、請求項11又は13に記載の殺菌剤。
- Bacillus coagulans高温菌がBacillus coagulans CP3425株(国際寄託番号NITE BP-01693)であり、及び、Bacillus subtilis中温菌がBacillus subtilis C-3102株(国際寄託番号FERM BP-1096)である、請求項11〜13及び15のいずれか1項に記載の殺菌剤。
- 少なくとも1種のBacillus coagulans高温菌、或いは、該少なくとも1種のBacillus coagulans高温菌と、少なくとも1種のBacillus subtilis中温菌との組み合わせを、有効成分として含む、但し前記少なくとも1種のBacillus coagulans高温菌がBacillus coagulans CP3425株(国際寄託番号NITE BP-01693)又は同等の殺菌作用を有するその変異株を含むことを特徴とする、有機廃棄物又は家畜敷料の堆肥化促進剤。
- Bacillus coagulans高温菌が、少なくとも40〜60℃の温度で生育可能である、請求項17に記載の有機廃棄物又は家畜敷料の堆肥化促進剤。

- Bacillus coagulans高温菌がさらに、Bacillus coagulans CP3424株(国際寄託番号NITE BP-01692)、又は同等の殺菌作用を有するその変異株を含む、請求項17又は18に記載の堆肥化促進剤。
- Bacillus subtilis中温菌が、Bacillus subtilis C-3102株(国際寄託番号FERM BP-1096)又はその変異株である、請求項17又は19に記載の堆肥化促進剤。
- Bacillus coagulans高温菌がBacillus coagulans CP3425株(国際寄託番号NITE BP-01693)であり、及び、Bacillus subtilis中温菌がBacillus subtilis C-3102株(国際寄託番号FERM BP-1096)である、請求項17〜19及び21のいずれか1項に記載の堆肥化促進剤。
- Bacillus coagulans高温菌とBacillus subtilis中温菌からなる、但し前記Bacillus coagulans高温菌がBacillus coagulans CP3425株(国際寄託番号NITE BP-01693)又は同等の殺菌作用を有するその変異株であることを特徴とする、敷料殺菌または堆肥化促進用組成物。
