Description
a)強度が変化し、その変化の振幅は5〜1,000マイクロモル/m2秒、好ましくは5〜400マイクロモル/m2秒であり、その変化は1時間に2〜3,600回、好ましくは5〜400回である、不連続な照明および/または時間変化する照明という条件下でラビリンツロマイケテス綱、特にシゾキトリウム属の原生生物の1種類以上の株を混合栄養モードで培養するステップと、
b)培地の中で有機炭素含有基質の存在下にてこの培養物を数世代にわたって維持するステップと、必要に応じて
c)このようにして培養した原生生物を回収するステップ
を含むことを特徴とする。
d)原生生物から疎水性物質を回収する工程;及び任意で
e)回収された疎水性物質からDHA(ドコサヘキサエン酸)及びアスタキサンチンを抽出する工程
を含む。
本発明態様の原生生物のアスタキサンチンの生産性(1時間あたりの1リットルの培養物あたりの所望の産物の量)は、0.015 mg/L/h超、又は0.020 mg/L/h超、又は0.025 mg/L/h超であってもよい。
Claims (10)
- シゾキトリウム(Schizochytrium)属の原生生物中でDHA(ドコサヘキサエン酸)及びアスタキサンチンを生産する方法であって、以下の工程;
a)不連続な照明条件下にて混合栄養モードでシゾキトリウム属の1つ以上の株を培養するステップ、ここで当該不連続な照明は、フラッシュの形態で供給され、強度が変化することにより明相と暗相が切り替わり、その振幅は5〜1,000μmol.m−2.s−1であり、変化が1時間につき2〜3,600回起こり、当該暗相は、当該シゾキトリウムの株が培養される期間の半分以上を占める;
b)当該培養を、有機炭素含有基質の存在下、培養培地中で、数世代に渡り維持するステップ;及び
c)そのように培養したシゾキトリウム株を回収するステップ;及び
d)疎水性物質を回収するステップ;及び
e)回収された疎水性物質からDHA及びアスタキサンチンを抽出するステップ;
を含む方法。 - 前記培養を100mM〜1.5Mの濃度の有機炭素含有基質の存在下で実施し、当該有機炭素含有基質が、蔗糖、グリセロール及びグルコース、並びにこれら分子の混合物の中から選択されることを特徴とする、請求項1に記載の方法。
- 培地の中に存在する前記有機炭素含有基質が少なくとも300mMのグルコースおよび/またはグリセロールを含むことを特徴とする、請求項2に記載の方法。
- 前記強度変化の振幅が70〜300μmol.m−2.s−1であることを特徴とする、請求項1〜3のいずれか1項に記載の方法。
- フラッシュの持続時間が1/10秒間〜10分間であることを特徴とする、請求項1〜4のいずれか1項に記載の方法。
- フラッシュの持続時間が20秒間〜1分間であることを特徴とする、請求項5に記載の方法。
- フラッシュの数が、1時間につき5〜3,600回であることを特徴とする、請求項1〜6のいずれか1項に記載の方法。
- フラッシュの数が、1時間につき20〜50回であることを特徴とする、請求項7に記載の方法。
- 1時間あたりの全光照射量が、フォトンのマイクロモル数を単位として2,000〜600,000マイクロモル/m2であることを特徴とする、請求項1〜6のいずれか1項に記載の方法。
- 前記原生生物が、受入番号CCAP 4087/1でCCAP(Culture Collection of Algae and Protozoa)に供託されたFCC1104株である、請求項1〜9のいずれか1項に記載の方法。
