- 前記第1ポリペプチド、前記第2ポリペプチド及び前記第3ポリペプチドの間が可操作性に連結され、及び/又は前記応答要素、前記プロモーター及び前記転写されるべき核酸配列の間が可操作性に連結される、請求項1に記載の光制御可能な遺伝子発現システム。
- 前記第1ポリペプチドがヘリックス・ターン・ヘリックスDNA結合ドメイン、亜鉛フィンガーモチーフ又は亜鉛クラスターDNA結合ドメイン、ロイシンジッパーDNA結合ドメイン、翼状ヘリックスDNA結合ドメイン、翼状ヘリックス・ターン・ヘリックスDNA結合ドメイン、ヘリックス・ループ・ヘリックスDNA結合ドメイン、高速移動群DNA結合ドメイン、B3DNA結合ドメインから選ばれる、請求項1に記載の光制御可能な遺伝子発現システム。

- 前記第3ポリペプチドが酸性アミノ酸に富む転写活性化ドメイン、プロリンに富む転写活性化ドメイン、セリン/トレオニンに富む転写活性化ドメインとグルタミンに富む転写活性化ドメイン、及びKruppel相関ボックス転写抑制ドメインから選ばれる、請求項1に記載の光制御可能な遺伝子発現システム。
- 前記第3ポリペプチドが単純疱疹ウイルスグラニュリンVP16転写活性化ドメイン、酵母Ga14転写活性化ドメイン、NF−κB p65イリデン転写活性化ドメイン、酵母通用抑制蛋白4の転写活性化ドメイン及び亜鉛フィンガー蛋白354AのKruppel相関ボックス転写抑制ドメインから選ばれる、請求項8に記載の光制御可能な遺伝子発現システム。
- さらに前記組み換え光感受性転写因子の細胞核へ輸送することを促進する核定位シグナル第4ポリペプチドを含み、前記第4ポリペプチドが前記第1ポリペプチド、前記第2ポリペプチド、前記第3ポリペプチドと直接、又はリンカーペプチド(linker peptide)によって連結される、請求項1に記載の光制御可能な遺伝子発現システム。
- さらにホルモンによって制御される第5ポリペプチドを含み、前記第5ポリペプチドが前記第1ポリペプチド、前記第2ポリペプチド、前記第3ポリペプチドと直接、又はリンカーペプチドによって連結される、請求項1に記載の光制御可能な遺伝子発現システム。
- 前記応答要素が、前記第1ポリペプチドによって特異的識別及び結合可能なDNAモチーフである、請求項1に記載の光制御可能な遺伝子発現システム。
- 前記応答要素がGa14結合要素、LexA結合要素、TetR結合要素、LacI結合要素及びcI結合要素から選ばれる、請求項12に記載の光制御可能な遺伝子発現システム。
- 前記プロモーターがアデノウイルス後期プロモーター、サイトメガロウイルスCMV最小プロモーター、酵母Gall遺伝子のプロモーター及びSv40プロモーターから選ばれる、請求項1に記載の光制御可能な遺伝子発現システム。
- 前記第1ポリペプチドと前記第2ポリペプチドとが1つの光制御可能なDNA結合蛋白を構成する、請求項1に記載の光制御可能な遺伝子発現システム。
- 請求項1〜15のいずれか1項に記載の光制御可能な遺伝子発現システムを含有する真核発現ベクター。
- 前記真核発現ベクターが前記組み換え光感受性転写因子のコード遺伝子、及び/又は前記ターゲット転写ユニットを含み、前記ターゲット転写ユニットは応答要素−プロモーターを含有するが、転写されるべき核酸配列を欠く、請求項16に記載の真核発現ベクター。
- 前記組み換え光感受性転写因子のヌクレオチド配列が配列番号3、7、11、15、19、31、33、35、37、39、43、47、57、59、61、63、97、99、101、103、105、107、109、111、113、115、117、119から選ばれ、アミノ酸配列が配列番号4、8、12、16、20、32、34、36、38、40、44、48、58、60、62、64、98、100、102、104、106、108、110、112、114、116、118、120から選ばれる、請求項16に記載の真核発現ベクター。
- 前記光制御可能な遺伝子発現システムを真核プラスミド発現ベクターに構築するステップa)と、
制御される遺伝子を含有する宿主細胞を導入するステップb)と、
光照射で前記宿主細胞を誘導して、前記宿主細胞における制御されるヌクレオチドを発現させるステップc)と、を含む、請求項1〜15のいずれか1項に記載の光制御可能な遺伝子発現システムを用いて宿主細胞において遺伝子発現を制御する方法。 - 前記宿主細胞は細菌細胞、真菌細胞、酵母細胞、植物細胞、線虫細胞、ミバエ細胞、昆虫細胞、ゼブラフィッシュ細胞、動物細胞及び哺乳動物細胞を含む、請求項19に記載の遺伝子発現を制御する方法。
- さらに光源の選択及び照射方法の選択を含み、前記光源がLED灯、蛍光灯、レーザ及び白熱灯から選ばれ、前記照射方法が持続的な光照射又は非持続的な光照射である、請求項19に記載の遺伝子発現を制御する方法。
- 前記光源の選択及び照射方法の選択は光による走査、投影、又は光金型によって空間において異なる位置における細胞の遺伝子発現レベルを制御することを含む、請求項21に記載の遺伝子発現を制御する方法。
- 前記照射方法はさらに印刷の投影フィルム、又は中性灰色フィルターを用いて空間において異なる位置における細胞の遺伝子発現レベルを制御することを含む、請求項21に記載の遺伝子発現を制御する方法。
- 請求項16に記載の前記真核発現ベクター及び/又は前記組み換え光感受性転写因子を含有する真核発現ベクターにより形質転換された哺乳動物細胞、及び関係カタログが装入されている試薬キット。
- さらに、応答要素−プロモーターを含有するが、転写されるべき核酸配列が欠如しているターゲット転写ユニットの真核発現ベクターが装入されている、請求項24に記載の試薬キット。
