Description
a)添付の配列リストの配列番号1、3または5;
b)配列番号1、3または5に相補的な核酸配列;
c)a)およびb)で定められる核酸配列またはそれらの相補鎖とストリンジェントな条件下でハイブリッド形成する、核酸配列。

c)配列番号1、3または5で示される核酸分子の逆鎖にストリンジェントな条件下でハイブリッド形成する核酸分子によりコードされる、配列番号2、4または6で示されるアミノ酸配列のオルソログの、アミノ酸配列;および
(d)少なくとも10連続アミノ酸を含み、α−1,3−フコシルトランスフェラーゼ活性を有する、配列番号2、4または6で示されるアミノ酸配列の断片。
a.本発明によるα−1,3−フコシルトランスフェラーゼ活性があるポリペプチドを提供し、
b.このポリペプチドがドナー基質からアクセプター基質へのフコース残基の転移を触媒し、それによりフコシル化オリゴ糖、(糖)タンパク質または(糖)脂質が生成する条件下で、フコース残基を含むドナー基質と、単糖またはオリゴ糖、(糖)タンパク質または(糖)脂質を含むアクセプター基質と、を含む混合物と、段階aのα−1,3−フコシルトランスフェラーゼ活性のあるポリペプチドを接触させる、段階。
a.フコシルオリゴ糖、(糖)タンパク質および/または(糖)脂質の生成が可能な、およびα−1,3−フコシルトランスフェラーゼ活性を有するポリペプチドの発現が可能な適切な栄養条件下で、上述のような宿主細胞を培養し;
b.段階aと同時にまたはこれと連続して、フコース残基を含むドナー基質と、オリゴ糖、(糖)タンパク質または(糖)脂質を含むアクセプター基質と、を提供して、前記宿主細胞において発現されるα−1,3−フコシルトランスフェラーゼがドナー基質からアクセプター基質へのフコース残基の転移を触媒し、それにより、フコシル化オリゴ糖、(糖)タンパク質または(糖)脂質を生成させるようにし;
c.宿主細胞またはその増殖培地から、前記フコシル化オリゴ糖、(糖)タンパク質および/または(糖)脂質を単離する、段階。
a)i)同封の配列表からの配列番号1、3または5、ii)配列番号1、3または5に相補的な核酸配列およびiii)適切な栄養条件下で、i)およびii)またはそれらの相補鎖で定められる核酸配列にストリンジェントな条件下でハイブリッド形成する核酸配列から選択される核酸配列を含むように形質転換または形質移入された宿主細胞を培養して、α−1,3−フコシルトランスフェラーゼ活性を有するペプチドとして、i)、ii)およびiii)から選択される核酸配列が発現されているようにし;
b)段階aと同時にまたはこれと連続して、フコース残基を含むドナー基質と、オリゴ糖、(糖)タンパク質または(糖)脂質を含むアクセプター基質と、を提供して、前記宿主細胞において発現されるα−1,3−フコシルトランスフェラーゼがドナー基質からアクセプター基質へのフコース残基の転移を触媒し、それにより、フコシル化オリゴ糖、(糖)タンパク質または(糖)脂質を生成させるようにし;
c)宿主細胞またはその増殖培地から、前記フコシル化オリゴ糖、(糖)タンパク質および/または(糖)脂質を単離する、段階。
Claims (12)
- フコシル化オリゴ糖の生成に使用する宿主細胞であって、前記宿主細胞は外来のポリヌクレオチド配列により形質転換または形質移入されており、
前記ポリヌクレオチド配列はベクターに含まれ、
前記ポリヌクレオチド配列が、a)添付の配列リストの配列番号1;b)a)で定められる核酸配列またはその相補鎖とストリンジェントな条件下でハイブリッド形成する核酸配列、からなる群より選択され、
前記ポリヌクレオチド配列は、ベクターで形質転換された宿主細胞により認識される配列を制御するために操作可能に連結されており、前記ポリヌクレオチド配列は、α−1,3−フコシルトランスフェラーゼ活性を有するポリペプチドをコードしており、
前記宿主細胞は、酵母を含む真菌、及び細菌からなる群より選択される、宿主細胞。 - 宿主細胞が、大腸菌(Escherichia coli)細胞であることを特徴とする、請求項1に記載の宿主細胞。
- α−1,3−フコシルトランスフェラーゼ活性を有するポリペプチドをコードする核酸が、個々の細胞のコドン使用に適応させられていることを特徴とする、請求項1又は2に記載の宿主細胞。
- α−1,3−フコシルトランスフェラーゼ活性を有するポリペプチドをコードし、且つa)添付の配列リストの配列番号1;b)a)で定められる核酸配列またはその相補鎖とストリンジェントな条件下でハイブリッド形成する核酸配列からなる群から選択される配列を含むポリヌクレオチドの、又は(a)配列番号2で示されるアミノ酸配列;(b)配列番号2で示されるアミノ酸配列に対して95%以上のアミノ酸配列同一性を有するアミノ酸配列、からなる群から選択されるアミノ酸配列からなるα−1,3−フコシルトランスフェラーゼ活性を有するポリペプチドの、フコシル化オリゴ糖の生成のための使用。
- 前記フコシル化オリゴ糖の生成が、α−1,3−フコシルトランスフェラーゼをコードするポリヌクレオチドの異種または同種発現により行われることを特徴とする、請求項4に記載の使用。
- 生成されるフコシル化オリゴ糖が3−フコシルラクトースであることを特徴とする、請求項4または5に記載の使用。
- a.(a)配列番号2で示されるアミノ酸配列;及び、(b)配列番号2で示されるアミノ酸配列に対して95%以上のアミノ酸配列同一性を有するアミノ酸配列、からなる群から選択されるアミノ酸配列からなる、α−1,3−フコシルトランスフェラーゼ活性があるポリペプチドを提供し、
b.該ポリペプチドが、ドナー基質からアクセプター基質へのフコース残基の転移を触媒し、それによりフコシル化オリゴ糖が生成する条件下で、フコース残基を含むドナー基質と、単糖またはオリゴ糖を含むアクセプター基質と、を含む混合物と、段階aのα−1,3−フコシルトランスフェラーゼ活性があるポリペプチドを接触させる段階を含む、フコシル化オリゴ糖を生成させるための方法。 - a.フコシル化オリゴ糖の生成可能なおよびα−1,3−フコシルトランスフェラーゼ活性を有するポリペプチドを発現可能な適切な栄養条件下で、請求項1から3のいずれかに記載の宿主細胞を培養し;
b.段階aと同時にまたは連続して、フコース残基を含むドナー基質と、単糖もしくはオリゴ糖を含むアクセプター基質と、を提供して、該α−1,3−フコシルトランスフェラーゼポリペプチドが該ドナー基質から該アクセプター基質へのフコース残基の転移を触媒し、それにより、フコシル化オリゴ糖を生成させるようにし;
c.該宿主細胞またはその増殖培地から、前記フコシル化オリゴ糖を単離する段階を含む、フコシル化オリゴ糖を生成させるための方法。 - ドナー基質がGDP−フコースであることを特徴とする、請求項7または8に記載の方法。
- 該GDP−フコースが、該宿主細胞で同時発現される酵素によるかまたは該宿主細胞の代謝により提供されることを特徴とする、請求項7から9のいずれかに記載の方法。
- 該アクセプター基質が、N−アセチルグルコサミン、N−アセチルラクトサミン、ガラクトース、フコース、シアル酸、グルコース、ラクトース又はこれらのあらゆる組み合わせであることを特徴とする、請求項7から10のいずれかに記載の方法。
- 生成されるフコシル化オリゴ糖がヒト乳オリゴ糖(HMO)である、請求項4または5に記載の使用、または請求項7から11のいずれかに記載の方法。
